Selective Protein and Lipid Cargo Sorting into Extracellular Vesicles of Lacticaseibacillus paracasei subsp. paracasei.

Extracellular vesicles (EVs) are nanoscale lipid bilayer particles that transport bioactive cargo. EVs from lactic acid bacteria show health benefits and drug-delivery potential, yet cargo encapsulation remains understudied. Here, EVs were isolated, purified, and characterized from the bacteriocin-producing strain Lacticaseibacillus paracasei subsp. paracasei YCL29, which was isolated from fermented Yucha. Proteomic and lipidomic analyses were subsequently conducted to compare the EVs with their parental L. paracasei YCL29 cells. Nanoflow cytometry indicated EVs with mean diameter 86.0 ± 18.8

Food-grade Lacticaseibacillus paracasei postbiotics suppress oral Streptococcus mutans biofilm formation and cariogenicity.

Dental caries is a highly prevalent non-communicable disease driven by dysbiosis of the oral microbiota, in which Streptococcus (S.) mutans plays a keystone role. We discovered that cell-free supernatant (CFS) from food-associated Lacticaseibacillus (L.) paracasei disrupts S. mutans membranes, causing leakage, reduced viability, and decreased surface hydrophobicity. CFS also inhibits biofilms by decreasing biomass, metabolic activity, chain length, and exopolysaccharide (EPS) accumulation. Neutralization experiments revealed organic acids as the primary antibacterial factors: activity weakened

Probiotics of Lacticaseibacillus paracasei SD1 and Lacticaseibacillus rhamnosus SD11 attenuate inflammation and β-cell death in streptozotocin-induced type 1 diabetic mice.

Probiotics provide health benefits in various aspects and are believed to modulate the immune system by balancing gut microbiota homeostasis, termed the "microbiota-immune axis". Recent evidence supports that several Lactobacillus strains possess glucose-lowering and anti-inflammatory effects in an animal model of type 1 diabetes (T1D). Although probiotics of Lacticaseibacillus paracasei SD1 (SD1) and Lacticaseibacillus rhamnosus SD11 (SD11) exert human oral health benefits by reducing harmful bacterial populations, their clinical application regarding hypoglycemic-related traits as well as th
