Strain-resolved biology of Lactobacillus johnsonii across host systems.

Lactobacillus johnsonii is widely described as a probiotic species, but current evidence supports a strain-resolved view. Its biological effects depend on strain background, host niche, diet, challenge context and delivery format, including live cells, cell-derived structures and postbiotic fractions. This review synthesizes comparative genomic, mechanistic and intervention studies on L. johnsonii, using weaned piglets as the main translational reference because barrier injury, inflammation, microbial disruption, diarrhea and impaired growth often coincide after weaning. Piglet studies show th

Regulation effect of Lactobacillus johnsonii on chickens infected with Ascaridia galli.

Ascaridia galli, a highly prevalent intestinal parasite of poultry globally, poses significant challenges to prevention and control strategies in poultry industry due to its increasing resistance to anthelmintic. Therefore, this study aims to investigate the regulatory effects of Lactobacillus johnsonii on growth performance and intestinal health of broilers infected with A. galli. In this study, 80 one-day-old broilers were randomly assigned to a 2 × 2 factorial arrangement of treatments, which included two levels of L. johnsonii (0 or 1 × 10⁹ CFU/mL) and two infection statuses (A. galli chal

Lactobacillus johnsonii HL79 mitigate plateau environment-induced hippocampal dysfunction in mice.

Plateau environment represents a common terrestrial characterized by multistress conditions including hypobaric hypoxia, low temperature, and intense radiation, yet sustain over 100 million permanent or transient inhabitants. While this extreme environment exerts profound impacts on cerebral architecture and gut microbiota homeostasis, precipitating cognitive deficits and microbiome-derived intestinal pathologies, the mechanistic interplay between plateau environment adaptation and microbial dynamics remains contentious. Here, we employ a microbiota-gut-brain axis framework to investigate whet

Bile promotes Lactobacillus johnsonii N6.2 extracellular vesicle production with conserved immunomodulatory properties.

Recently, Lactobacillus johnsonii N6.2-derived extracellular vesicles (EVs) were shown to reduce apoptosis in human beta cell lines and stimulate insulin secretion in human islets. Our goal was to identify a physiologically relevant environmental condition that induces a hypervesiculation phenotype in L. johnsonii N6.2 and to evaluate if transcriptional changes are involved in this process. Culturing this strain in the presence of 0.2% bovine bile, which mimics a stressor encountered by the bacterium in the small intestine, resulted in approximately a 100-fold increase in EVs relative to cells

Development of a gene-editing strategy to overcome genetic intractability in Lactobacillus johnsonii.

Genetic editing in lactobacilli is often constrained by their robust defense mechanisms against foreign DNA and low transformation efficiency, resulting in pronounced species- and strain-specific variability in the performance of genetic tools. In this study, we developed a markerless gene editing system for Lactobacillus johnsonii MR1, a Jackson mouse isolate with a closed genome and well-documented host immunomodulatory properties. Initial testing revealed that existing transformation and gene-editing protocols produced low transformation efficiency, poor reproducibility, and exhibited poor

Mulberry water extract alleviates osteoarthritis via Lactobacillus johnsonii-dependent bile acid restoration.

BACKGROUND: Knee osteoarthritis (KOA) is a chronic degenerative joint disorder associated with dysregulated polarization of synovial macrophages. Natural plant-based medicines, characterized by their multi-component and multi-target properties, exhibit unique advantages in immune regulation and gut microbiota modulation. Despite this potential, the therapeutic roles of such interventions in the management of KOA remain inadequately explored. METHODS: This study investigated the therapeutic potential of mulberry water extract (MWE), a safe and sustainable preparation derived from mulberry frui

Synergistic protective effects of β-glucan and Lactobacillus johnsonii NSP009 in ulcerative colitis.

The combination of probiotics and prebiotics has emerged as a promising strategy for managing ulcerative colitis (UC). Among these, β-glucan (BG) and Lactobacillus johnsonii have individually shown potential in alleviating colitis, with BG notably enriching L. johnsonii populations, suggesting a synergistic therapeutic potential when combined. This study investigates the protective effects of a novel synbiotic formulation, BG and L. johnsonii NSP009 (LJ), termed BGLJ, in a dextran sulfate sodium (DSS)-induced colitis mouse model. Our results reveal that BGLJ significantly mitigated DSS-induced

Lactobacillus johnsonii and 3-indolepropionic acid improve the depression-like behaviors via inhibiting neuroinflammation.

Gut microbiota-derived metabolites play a crucial role in depression. This study aimed to elucidate the role of tryptophan metabolites herein. In a CSDS mouse model, we identified eight differential species, twelve altered neurotransmitters, and two up-regulated inflammatory factors (IL-6 and IL-1β). Notably, 3-indolepropionic acid (IPA) levels were consistently reduced in feces, colon, blood, and hippocampus of CSDS mice. The decreased abundance of Lactobacillus johnsonii (L. johnsonii) was correlated closely with depression-like behaviors (DLBs), reduced fecal IPA, and elevated IL-6 and IL-1

Lactobacillus johnsonii is a dominant Lactobacillus in the murine oral mucosa and has chitinase activity that compromises fungal cell wall integrity.

UNLABELLED: The oral microbiome is a critical determinant of health and disease, as interactions between oral microorganisms can influence their physiology and the development or severity of oral infections. Lactobacilli have a widely recognized antagonistic relationship with Candida albicans and may exhibit probiotic properties that limit oral fungal infection. We previously reported that Lactobacillus johnsonii strain MT4, an oral strain isolated from C57BL/6 mice, can induce global changes in the murine oral microbiome and has anti-Candida activity in vitro. To build on this information, we
