Eicosanoid regulation of oosporogenesis by Lagenidium giganteum.

Cell proliferation is inhibited in many biological systems by lipid peroxides and related products derived from polyunsaturated fatty acids. Using developmentally synchronized cultures of Lagenidium giganteum (Oomycetes: Lagenidiales), a facultative parasite of mosquito larvae, it has been documented that oxidative lipid metabolism is necessary for the induction and subsequent maturation of its sexual stage, the oospore. Addition of lipoxygenase inhibitors to liquid cultures of the fungus results in the stage-specific disruption of antheridial induction, gametangial fusion, induction of meiosi

Severe meningoencephalitis secondary to calvarial invasion of Lagenidium giganteum forma caninum in a dog.

BACKGROUND: The oomycete Lagenidiumgiganteum forma caninum is an uncommon cause of severe dermal and subcutaneous infections in dogs with possible vascular invasion and other fatal sequelae. Infection within the central nervous system of affected dogs has not been previously reported. CASE DESCRIPTION: A 6-year-old spayed female mixed-breed dog was evaluated at a referral institution with a 2-month history of suspected fungal infection in the region of the right mandibular lymph node that was refractory to surgical resection and empiric medical therapy. Physical examination identified a 6-cm

Cutaneous Lagenidium deciduum infection in a patient with relapsed acute myeloid leukemia.

BACKGROUND: Lagenidium deciduum is an oomycete that can cause infections in mammals that present similarly to pythiosis and mucormycosis. Most of the existing case reports have occurred in canines and have been fatal. In animals, medical therapy has not been successful, so surgical excision is the mainstay of treatment. Lagenidium sp. infections in humans are rare. There is only one case of a human Lagenidium sp. infection in the literature, and it presented as an ocular infection. The human ocular infection was resistant to medical therapy and required a penetrating keratoplasty for cure. Add

Sterol and steryl ester regulation of phospholipase A2 from the mosquito parasite Lagenidium giganteum.

Lagenidium giganteum, a facultative parasite of mosquito larvae, cannot synthesize sterols, and requires an exogenous source of these lipids in order to enter its reproductive cycle. This parasite grows vegetatively in the absence of sterols, but requires cholesterol or structurally related compounds to produce motile zoospores, which are the only stage capable of infecting mosquitoes. Sterols structurally related to cholesterol and some steryl esters inhibited the activity of L. giganteum phospholipase A2 (PLA2), an enzyme that hydrolyzes fatty acids from the sn-2 position of glycerophospholi

Clinicopathologic findings associated with Lagenidium sp. infection in 6 dogs: initial description of an emerging oomycosis.

An oomycotic pathogen in the genus Lagenidium was isolated from tissues obtained from 6 dogs with progressive cutaneous disease. Initial clinical findings in 5 dogs included multifocal cutaneous lesions, subcutaneous lesions, or both associated with regional lymphadenopathy: the 6th dog initially was presented for evaluation of mandibular lymphadenopathy. Cutaneous lesions were ulcerated, exudative regions (often with necrosis and draining tracts) or multiple firm dermal or subcutaneous nodules. Two dogs subsequently developed hemoabdomen from great vessel rupture and died acutely. Four dogs w

From plants to animals: Pythium, Lagenidium, Aphanomyces, and other oomycete pathogens of animals seen through a one-health lens.

Progress in treating oomycete infections of animals has been slow. Much relevant biological and therapeutic knowledge originates from plant pathology and agricultural science, while medical and veterinary literature develops independently. This review integrates these fields to highlight key knowledge gaps and translational opportunities in animal health. Oomycetes are eukaryotic Stramenopiles, distinct from true fungi; their hyphae are diploid and coenocytic, with cellulose and glucan cell walls. They reproduce via motile zoospores and durable oospores, enabling environmental persistence. Alt

Highly sensitive multiplex PCR for convenient quantification and differentiation of canine Oomycota pathogens: Pythium insidiosum, Lagenidium giganteum f. caninum, and Paralagenidium karlingii.

Oomycota mammalian pathogens, including Pythium (P.) insidiosum, Lagenidium (L.) giganteum f. caninum, and Paralagenidium (Para.) karlingii, are increasingly recognized as causes of life-threatening infections in dogs, with diagnosis complicated by their clinical and phenotypic resemblance to fungal pathogens. Traditional diagnostic methods, such as serology, histopathology, and culture, often lack sensitivity and specificity, delaying appropriate management. Here, we developed a multiplex PCR assay with remarkable sensitivity, detecting as few as one DNA copy per reaction for P. insidiosum an

Improving noninvasive diagnosis of cutaneous and gastrointestinal forms of Pythium insidiosum infections in dogs: complementary roles of serum-based ELISA and polymerase chain reaction.

OBJECTIVE: To evaluate the diagnostic performance of ELISA and PCR for detecting pathogenic oomycete infections and to explore associations with clinical presentation and treatment status. METHODS: Canine sera (n = 796) submitted to Pythium Serology Diagnostic Laboratory (from 2023 through 2025) were analyzed by ELISA to detect anti-Pythium antibodies and a PCR targeting Pythium insidiosum, Lagenidium spp, and Paralagenidium spp. Clinical records were reviewed for a subset of submissions. Analyses included χ2 tests, a linear mixed-effects model, and Cohen κ for ELISA-PCR agreement. RESULTS:

Occurrence of fungi and fungus-like organisms in the Horodnianka River in the vicinity of Białystok, Poland.

Studies of fungi and fungus- like organisms in the northeastern Poland have mainly concentrated on running waters in the vicinity of Białystok, including the Horodnianka River. The main objective was to investigate biodiversity of fungi and fungus-like organisms which take part in decomposition of organic matter commonly found in inland waters. To obtain a complete picture of species composition of fungi and fungus-like organisms in running waters we decided to explore representative sites of the Horodnianka River such as Olmonty, Hryniewicze and Horodniany with close localization of landfill.

Studies on Fungal Cultural Filtrates against Adult Culex quinquefasciatus (Diptera: Culicidae) a Vector of Filariasis.

Entomopathogenic fungi have significant potential to control mosquito population. The culture filtrates of Fusarium oxysporum, Lagenidium giganteum, Trichophyton ajelloi, and Culicinomyces clavisporus were evaluated against adults of Cx. quinquefasciatus. The culture filtrates were obtained by filtering the broth through Whatman-1 filter paper. These culture filtrates of C. clavisporus have been found significantly pathogenic with LC(50)-2.5, LC(90)-7.24, and LC(99)-8.7 ML, respectively, after exposure of 24 h. However, the culture filtrates when were combined, in ratios 1 : 1 : 1 of Fusarium

Pythiosis, lagenidiosis, and zygomycosis in small animals.

Pythiosis, lagenidiosis, and zygomycosis affect animals living in temperate, tropical, and subtropical climates, and these diseases are often fatal. Although Pythium insidiosum, Conidiobolus species, and Basidiobolus species have been recognized as pathogens for several years, members of the genus Lagenidium have been identified as a cause of oomycosis in dogs only recently. Pythiosis, lagenidiosis, and zygomycosis share similar clinical and histologic characteristics, making them difficult to distinguish from one another; however, distinguishing between these pathogens is important because of

Entomopathogenic fungi for mosquito control: a review.

Fungal diseases in insects are common and widespread and can decimate their populations in spectacular epizootics. Virtually all insect orders are susceptible to fungal diseases, including Dipterans. Fungal pathogens such as Lagenidium, Coelomomyces and Culicinomyces are known to affect mosquito populations, and have been studied extensively. There are, however, many other fungi that infect and kill mosquitoes at the larval and/or adult stage. The discovery, in 1977, of the selective mosquito-pathogenic bacterium Bacillus thuringiensis Berliner israelensis (Bti) curtailed widespread interest i

Hyphae, pseudohyphae, yeasts, spherules, spores, and more: A review on the morphology and pathology of fungal and oomycete infections in the skin of domestic animals.

Fungi are among the most common infectious agents affecting the skin of animals. The skin can serve as a port of entry for fungal infections, which can eventually become disseminated. In some regions of the world, oomycetes, such as Pythium and Lagenidium, are also responsible for a significant number of severe cutaneous infections. Histologic evaluation of fungal morphology, including size, shape, septation, branching, and budding characteristics, combined with the distribution of inflammatory infiltrates within different skin layers can potentially identify etiologic agents, guiding selectio

Parasitism of Xiphinema rivesi and X. americanum by Zoosporic Fungi.

Living Xiphinema americanum (Xa) and X. rivesi (Xr) extracted from soil samples and stored for 1-5 days at 4 or 20 C contained aseptate fungal hyphae. The fungi directly penetrated the nematode's cuticle from spores encysted near the head. Penetration through the stoma, vulva, or anus was rare. Catenaria anguillulae (Cat), Lagenidium caudatura (Lag), Aphanomyces sp. (Aph), and Leptolegnia sp. (Lep) were isolated into pure culture from infected nematodes. The pathogenicity of these zoosporic fungi was determined by incubating mixed freshly extracted Xa and Xr in 2% soil extract (pH = 6.7, condu

The medical importance of riceland mosquitoes and their control using alternatives to chemical insecticides.

The medical importance, ecology and control of riceland mosquitoes using alternative strategies is reviewed. Over 135 pest and vector anopheline and culicine mosquito species found in association with riceland habitats and their medical importance are presented. Malaria and Japanese encephalitis are the two most serious human diseases transmitted by riceland mosquitoes, but they have been incriminated as vectors of dozens of arboviruses and other parasites and pathogens including the causal agents of West Nile and Rift Valley Fevers and lymphatic filariasis. Control of vector and pest mosquito
