Accidental poisoning of 17 dogs with lasalocid.

Over a period of 10 days, 17 dogs became weak and developed neurological deficits of different degrees of severity. About 12 hours before these clinical signs appeared they had all eaten a particular brand of commercial dog food from a recently opened bag. They were all quadriparetic and hyporeflexic, and some of them also showed additional systemic or neurological signs, including dyspnoea, a high body temperature, tongue laxity, hyperaesthesia and anisochoria. Serum biochemical abnormalities included high activities of creatine kinase, lactate dehydrogenase and aspartate aminotransferase. An

Performance of beef heifers supplemented with sodium lasalocid.

This study was conducted on 78 13-month-old crossbred beef heifers that weighed 215 kg in Southern Rio Grande do Sul (RS) State, Brazil. We evaluated the performance of beef heifers that were reared in a pasture system that received a mineral supplement energy-type protein with added sodium lasalocid (LAS). The heifers were randomly and uniformly divided into 2 groups, with 39 animals in each group. One group of animals received a mineral supplement energy-type protein without sodium lasalocid (CON), and the other group received a mineral supplement energy-type protein with added LAS. The mean

Efficacy of lasalocid against murine Pneumocystis carinii pneumonitis.

The efficacy of the ionophore lasalocid against Pneumocystis carinii pneumonitis in corticosteroid-immunosuppressed Sprague-Dawley rats was investigated. Lasalocid was effective in the prevention of the pneumonitis in a dose-dependent manner. At dosages of 0, 5, 10, and 20 mg/kg/day, P. carinii infection rates were 92, 60, 20, and 0%, respectively, during dexamethasone immunosuppression. Also, lasalocid compared favorably with other drugs known to have anti-P. carinii activity, including trimethoprim-sulfamethoxazole, atovaquone, and dapsone-trimethoprim.

Acquired immunity to Eimeria tenella in lasalocid-treated chickens.

The effect of restricted medication with lasalocid sodium on the development of acquired immunity against Eimeria tenella was evaluated. The medication was allowed for all or part of 6-day test period (one day before until 4 days after infection). The parameters used for such evaluations were lesion score, caecal, bursal and splenic weights. The optimum treatment time for the drug was clearly indicated by lesion score which was very low when the medication was initiated 1 day before until 1 day after inoculation, but only partly effective if given on Day 2 post-inoculation. The challenge with

In vitro and in vivo efficacy of lasalocid for treatment of experimental cryptosporidiosis.

In vitro viability of purified Cryptosporidium parvum oocysts, exposed for 30, 60, 90 and 120min to 0.27mg/ml lasalocid suspension was evaluated by inclusion or exclusion of two fluorogenic vital dyes and an excystation technique. Continuously, preventive and curative efficacies at different doses (9, 6.75, 5.625 and 4.5mg/kg body weight) and regimens of lasalocid against cryptosporidial infection were evaluated on an experimental neonatal mice model. In vitro assays demonstrated a decrease in the oocyst viability related to an increase in exposure time for exposure to the lasalocid suspension

Chemotherapeutic effect of azithromycin and lasalocid on Cryptosporidium infection in mice.

Prednisolone-immunosuppressed mice (ICR, 7-wk-old female) were each inoculated with 1 x 10(5) oocysts of Cryptosporidium parvum. Medication with azithromycin (400 mg/kg/day) or lasalocid (64, or 128 mg/kg/day) was started 13 h after inoculation and continued for 3 days. The number of oocysts discharged by each mouse was calculated on days 4-12 post-inoculation. Compared with non-medicated controls, oocyst production by the medicated mice was markedly reduced; some mice did not discharge oocysts and the remaining mice discharged less than 1/100 the number of oocysts of the control mice. These r

Trypanosoma brucei: trypanocidal and cell swelling activities of lasalocid acid.

Chemotherapeutic treatment of human and animal trypanosomiasis is unsatisfactory because only a few drugs are available. As these drugs have poor efficacy and cause adverse reactions, more effective and tolerable medications are needed. As the polyether ionophore antibiotic lasalocid acid is used as medicated feed additive in cattle, the compound was tested for its trypanocidal and cytotoxic activity against bloodstream forms of Trypanosoma brucei and human myeloid HL-60 cells. The concentrations required of lasalocid acid to reduce the growth rate of trypanosomes by 50% and to kill the parasi

Pathologic changes associated with experimental lasalocid and monensin toxicosis in cattle.

The acute toxicity of lasalocid and monensin was studied in 36 Holstein steers. The cattle were given (orally) a single dosage of lasalocid (1, 10, 50, or 100 mg/kg of body weight) or monensin (25 mg/kg of body weight) or rice hulls. Animals were observed once a day until they died or were euthanatized at 32 days after the dose was given. All cattle were necropsied. Heart, kidney, adrenal gland, liver, spleen, pancreas, lungs, brain, sciatic nerve, skeletal muscle, small intestine, large intestine, and rumen tissue sections, stained with hematoxylin and eosin, were studied microscopically. Revisiting Old Ionophore Lasalocid as a Novel Inhibitor of Multiple Toxins.

The ionophore lasalocid is widely used as a veterinary drug against coccidiosis. We found recently that lasalocid protects cells from two unrelated bacterial toxins, the cytotoxic necrotizing factor-1 (CNF1) from Escherichia. coli and diphtheria toxin. We evaluated lasalocid's capacity to protect cells against other toxins of medical interest comprising toxin B from Clostridium difficile, Shiga-like toxin 1 from enterohemorrhagic E. coli and exotoxin A from Pseudomonas aeruginosa. We further characterized the impact of lasalocid on the endolysosomal and the retrograde pathways and organelle in

Spectroscopic, crystallographic and theoretical studies of lasalocid complex with ammonia and benzylamine.

A natural antibiotic--Lasalocid is able to form stable complexes with ammonia and organic amines. New complexes of lasalocid with benzylamine and ammonia were obtained in the crystal forms and studied using X-ray, FT-IR, (1)H NMR, (13)C NMR and DFT methods. These studies have shown that in both complexes the proton is transferred from the carboxylic group to the amine group with the formation of a pseudo-cyclic structure of lasalocid anion complexing the protonated amine or NH4(+) cation. The spectroscopic and DFT studies demonstrated that the structure of the complex formed between Lasalocid

Anticryptosporidial activity of lasalocid and other ionophorous antibiotics in immunosuppressed rats.

The anticryptosporidial activities of three ionophorous antibiotics were assessed in dexamethasone-immunosuppressed rats infected with Cryptosporidium parvum. When administered prophylactically, lasalocid prevented infection in a dose-dependent manner, whereas monensin and salinomycin were ineffective. Therapeutically, lasalocid eliminated established overt infections of the intestine, although infection persisted in the common bile duct and intestinal infection recurred after lasalocid treatment was stopped. These findings suggest that lasalocid is a potentially useful anticryptosporidial age

Chemoprophylaxis of cryptosporidiosis in chickens, using halofuginone, salinomycin, lasalocid, or monensin.

Chemoprophylaxis of Cryptosporidium baileyi infections was attempted by feeding 4 groups of chicks diets containing 3 mg of halofuginone/kg of feed, 60 mg of salinomycin/kg, 75 mg of lasalocid/kg, or 110 mg of monensin/kg. Rations were fed 5 days before oral or intratracheal inoculation with oocysts and were continued for 20 days. None of the drugs prevented C baileyi infections. Clinical signs of respiratory tract disease and gross lesions of airsacculitis were observed in intratracheally inoculated birds in all treatment groups and nonmedicated controls. Orally inoculated birds did not devel

Paralytic syndrome attributed to lasalocid residues in a commercial ration fed to dogs.

Lasalocid, accidently introduced into a commercial dog food, was found to be the cause of neuromuscular toxicosis in 10 dogs. Toxicosis was confirmed in 4 principal dogs. The history, clinical signs, and pattern of onset of lasalocid-induced toxicosis in the 14 dogs were similar to those reported for botulism. The signs, which were related to a generalized lower motor neuron deficit, were generally different from ionophore-induced toxicosis reported in other species. Supportive therapy and close monitoring were sufficient to bring about a gradual improvement in all of the dogs, despite the sev

The decrease of lasalocid residue in the edible tissues by silymarin supplementation of chicken diet.

Widespread use of coccidiostats, in spite of beneficial control of protozoan infections in poultry, implies a risk of residues in edible tissues, and there is increasing interest in the development of strategies for prevention of veterinary drugs residue in food-producing animals. The aim of this study is assigned to clarify the impact of silymarin addendum in the diet on lasalocid concentration in the liver and breast muscles from the broiler. Four groups of chickens received a feed with lasalocid at levels between 75 and 200 mg kg-1. Other four groups received a feed with lasalocid (75-200 m

Safety evaluation of lasalocid use in Chinese ring-necked pheasants (Phasianus colchicus).

Coccidiosis remains a significant threat to the welfare of game farm-reared pheasants in the United States. Although lasalocid has been demonstrated to be effective against pheasant specific coccidia, information regarding its safety in this species is lacking. The purpose of this study was to gather data on the safety of lasalocid when fed to Chinese ring-necked pheasants at one, two, and three times the recommended high dose of lasalocid used for prevention of coccidiosis in other poultry at three times the normal treatment period. Pheasant chicks (approximately 1 day-old; n = 160) were rand

Lasalocid Acid Antibiotic at a Membrane Surface Probed by Sum Frequency Generation Spectroscopy.

Carboxyl polyether ionophores (CPIs) are widely used as veterinary antibiotics and to increase food utilization in ruminating animals. Furthermore, CPIs can target drug-resistant bacteria, but detailed knowledge about their mode-of-action is needed to develop agents with a reasonable therapeutic index. It has been suggested that ionophores bind to membranes and incur large structural changes to shield a bound ion from the hydrophobic environment of the lipid bilayer for transport. One crucial piece of information is missing, however: Is it necessary for the free ionophore to adsorb on the memb

Lasalocid A selectively induces the degradation of MYD88 in lymphomas harboring the MYD88 L265P mutation.

Myeloid differentiation primary response protein 88 (MYD88) is a key adaptor molecule in the signaling pathways of toll-like receptor and interleukin-1 receptor. A somatic mutation resulting in a leucine-to-proline change at position 265 of the MYD88 protein (MYD88 L265P) is one of the most prevalent oncogenic mutations found in patients with hematological malignancies. In this study, we used high-throughput screening to identify lasalocid A as a potent small molecule that selectively inhibited the viability of lymphoma cells expressing MYD88 L265P and the associated activation of NF-κB. In vitro comparison of the dermal penetration of three different topical formulations containing lasalocid.

BACKGROUND: Topical antimicrobial preparations are of utmost importance in treating suspected and confirmed meticillin-resistant Staphylococcus pseudintermedius (MRSP) infections due to the increasing incidence of widespread resistance to systemic antimicrobials. Lasalocid is active against MRSP in vitro and this may become an important topical antimicrobial for the treatment of canine pyoderma. HYPOTHESIS/OBJECTIVES: To determine effects of various formulation types on penetration and retention of lasalocid applied to canine skin in vitro. ANIMALS: Normal canine skin was collected from the

Relaxing and metabolic inhibitory action of X537A (Lasalocid) on the taenia of the guinea-pig caecum.

1. In the isolated guinea-pig taenia, application of X537A to the muscle caused a relaxation, while A23187 caused a sustained contraction. 2. Treatment with adrenergic blocking agents and tetrodotoxin had no effect on the relaxing response to X537A. 3. In the presence of a low concentration (10(-6) M) of X537A or under hypoxic conditions (bubbled with 95% N2: 5% CO2), the taenia lost the ability to respond to a high concentration (40 mM) of potassium with a sustained tonic contraction, although the rapid phasic contraction was still present. 4. When a low concentration of X537A was applied the
