The antimicrobial activity of liposomal lauric acids against Propionibacterium acnes.

This study evaluated the antimicrobial activity of lauric acid (LA) and its liposomal derivatives against Propionibacterium acnes (P. acnes), the bacterium that promotes inflammatory acne. First, the antimicrobial study of three free fatty acids (lauric acid, palmitic acid and oleic acid) demonstrated that LA gives the strongest bactericidal activity against P. acnes. However, a setback of using LA as a potential treatment for inflammatory acne is its poor water solubility. Then the LA was incorporated into a liposome formulation to aid its delivery to P. acnes. It was demonstrated that the an

In vivo treatment of Propionibacterium acnes infection with liposomal lauric acids.

Propionibacterium acnes (P. acnes) is a Gram-positive bacterium strongly associated with acne infection. While many antimicrobial agents have been used in clinic to treat acne infection by targeting P. acnes, these existing anti-acne agents usually produce considerable side effects. Herein, the development and evaluation of liposomal lauric acids (LipoLA) is reported as a new, effective and safe therapeutic agent for the treatment of acne infection. By incorporating lauric acids into the lipid bilayer of liposomes, it is observed that the resulting LipoLA readily fuse with bacterial membranes,

Ethanol promoted lauric acid interaction with rice starch during steaming.

Fatty acids could form inclusion complexes with starch with the aid of mechanical treatment to reduce starch digestibility. However, methods to enhance the interaction between starch and fatty acids without extensive mechanical treatment remained largely unexplored. This study investigated the effects of synchronous complexation of lauric acid (LA) and ethanol on the structure and digestibility of starch during steaming. The complexation of either ethanol or LA alone significantly enhanced the ordered structures of starch, which increased the content of resistant starch (RS) from 13.17 % to 19

Structural insights into the binding of lauric acid to CYP107L2 from Streptomyces avermitilis.

Streptomyces avermitilis is an actinobacterium known to produce clinically useful macrolides including avermectins. CYP107L2 from S. avermitilis shares a high sequence similarity with the PikC (CYP107L1) from S. venezuelae. To elucidate the structural features of CYP107L2, we conducted biochemical and structural characterization of CYP107L2 from S. avermitilis. The CYP107L2 gene was cloned, and its recombinant protein was expressed and purified. The CYP107L2 showed a low-spin state of heme, and the reduced form yielded the CO difference spectra with a maximal absorption at 449 nm. Binding of p

Lauric Acid, a Dietary Saturated Medium-Chain Fatty Acid, Elicits Calcium-Dependent Eryptosis.

Cardiovascular diseases (CVD) are a leading cause of mortality worldwide, and dietary habits represent a major risk factor for dyslipidemia; a hallmark of CVD. Saturated fatty acids contribute to CVD by aggravating dyslipidemia, and, in particular, lauric acid (LA) raises circulating cholesterol levels. The role of red blood cells (RBCs) in CVD is increasingly being appreciated, and eryptosis has recently been identified as a novel mechanism in CVD. However, the effect of LA on RBC physiology has not been thoroughly investigated. RBCs were isolated from heparin-anticoagulated whole blood (WB)

Solid Lipid Nanoparticles Loaded with Retinoic Acid and Lauric Acid as an Alternative for Topical Treatment of Acne Vulgaris.

Topical therapy is the first choice for the treatment of mild to moderate acne and all-trans retinoic acid is one of the most used drugs. The combination of retinoids and antimicrobials is an innovative approach for acne therapy. Recently, lauric acid, a saturated fatty acid, has shown strong antimicrobial activity against Propionibacterium acnes. However, topical application of retinoic acid is followed by high incidence of side-effects, including erythema and irritation. Solid lipid nanoparticles represent an alternative to overcome these side-effects. This work aims to develop solid lipid n

Synergistic Flotation Mechanism of Apatite Using Lauric Acid-Lauryl Alcohol Combined Collector.

To investigate the synergistic effect of fatty alcohols on the flotation performance of fatty acid collectors, a novel combined collector comprising lauric acid and lauryl alcohol was prepared. Flotation results of apatite indicated that the combined collector with 30% lauryl alcohol exhibited the best flotation performance. Under the conditions of pH 9 and a combined collector dosage of 50 mg/L, the flotation recovery of apatite exceeded 99% in the temperature range of 5-45 °C. In contrast, lauric acid alone produced a lower flotation recovery and exhibited strong temperature dependence. Ther

Lauric acid provides neuroprotection against oxidative stress in mouse model of hyperglycaemic stroke.

During ischemic stroke, higher glucose level linked worse outcomes were reported even in patients without pre-existing diabetes. Evidence suggest that such worse stroke outcomes were mainly due to production of reactive, toxic glucose metabolites that expands oxidative damage inside the brain. As a consequence of high oxidative stress, microvasculature structures and tight junctions compromised their functionally, infarct volume expands and brain edema exacerbates. In a mouse model of ischemic stroke with induced acute hyperglycaemia, Lauric acid (LA) as a natural saturated fatty acid demonstr

The Therapeutic Potential of Butyrate and Lauric Acid in Modulating Glial and Neuronal Activity in Alzheimer's Disease.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder marked by amyloid-β plaque accumulation, tau tangles, and extensive neuroinflammation. Neuroinflammation, driven by glial cells like microglia and astrocytes, plays a critical role in AD progression. Initially, these cells provide protective functions, such as debris clearance and neurotrophic support. However, as AD progresses, chronic activation of these cells exacerbates inflammation, contributing to synaptic dysfunction, neuronal loss, and cognitive decline. Microglia release pro-inflammatory cytokines and reactive oxygen

Integrated Silica-Encapsulated Lauric Acid Phase Change Material into Acid-Activated Geopolymer for Thermal Energy Storage Applications.

This paper investigates the development of a sustainable thermal energy storage material for buildings by combining silica-encapsulated lauric acid phase change materials (PCMs) with an acid-activated geopolymer matrix. The study addresses two significant environmental concerns: the high CO2 emissions associated with Portland cement production and the large energy consumption of buildings for heating and cooling. Lauric acid, a biobased fatty acid PCM, was selected as a by-stander because of its high latent heat capacity and suitable melting temperature facilitating encapsulation and leakage c

Lauric-acid-rich Hermetia illucens meal modulates lipid metabolism in the liver and the fatty acid profile in piglet tissues.

This study investigated whether a diet rich in lauric acid, based on Hermetia illucens (HI) larvae meal, would induce measurable changes in lipid-related metabolic pathways in the liver and in the fatty acid composition of tissues. Twenty-four weaned piglets were assigned to three dietary groups: control (0% HI), 2.5% HI, and 5% HI. Multilevel analyses, including histology, biochemical assays, fatty acid profiling, and gene expression, were performed to assess liver function and metabolic adaptations of animal organisms. Dietary HI markedly reshaped the tissue lipid composition, increasing the

Lauric Acid Is an Inhibitor of Clostridium difficile Growth in Vitro and Reduces Inflammation in a Mouse Infection Model.

Clostridium difficile is a Gram-positive, spore-forming anaerobic human gastrointestinal pathogen. C. difficile infection (CDI) is a major health concern worldwide, with symptoms ranging from diarrhea to pseudomembranous colitis, toxic megacolon, sepsis, and death. CDI onset and progression are mostly caused by intestinal dysbiosis and exposure to C. difficile spores. Current treatment strategies include antibiotics; however, antibiotic use is often associated with high recurrence rates and an increased risk of antibiotic resistance. Medium-chain fatty acids (MCFAs) have been revealed to inhib

Acute Treatment with Lauric Acid Reduces Blood Pressure and Oxidative Stress in Spontaneously Hypertensive Rats.

The effects of acute administration of lauric acid (LA), the most abundant medium-chain fatty acid of coconut oil, on blood pressure, heart rate and oxidative stress were investigated in spontaneously hypertensive rats (SHR). Intravenous doses of LA reduced blood pressure in a dose-dependent fashion (1, 3, 4, 8 and 10 mg/kg) in both SHR and Wistar Kyoto rats. LA (10-8 to 3 × 10-3 M) induced vasorelaxation in isolated superior mesenteric artery rings of SHR in the presence (n = 7) or absence (n = 8) of functional endothelium [maximum effect (ME) = 104 ± 3 versus 103 ± 4%]. Potent Inhibitory Effects of Lauric Acid and Glycerol Monolaurate Against CyHV-2 and the Viral Envelope.

Lauric Acid (LA) and Glycerol Monolaurate (GML) are naturally present in several plant-derived oils, particularly coconut and palm kernel oil. These compounds have been widely applied in swine production owing to their proven antiviral activity against Porcine Reproductive and Respiratory Syndrome Virus (PRRSV), Porcine Epidemic Diarrhoea Virus (PEDV), and African Swine Fever Virus (ASFV). In this study, we investigated the potential of LA and GML as antiviral agents against Cyprinid herpesvirus 2 (CyHV-2), the causative agent of Herpesviral haematopoietic necrosis (HVHN), which poses signific

Lauric acid reduces apoptosis by inhibiting FOXO3a-signaling in deoxynivalenol-treated IPEC-J2 cells.

Deoxynivalenol (DON) is the most common mycotoxin contaminant of food or feed worldwide and causes disease in animals. Lauric acid (LA) is a medium-chain fatty acid useful for barrier functions such as antimicrobial activity in the intestine of monogastric animals. However, the molecular mechanisms by which lauric acid exerts its effects on the deoxynivalenol-exposed small intestine have not been studied. We used an intestinal porcine epithelial cell line (IPEC-J2) as an in vitro model to explore the molecular mechanism of lauric acid in alleviating deoxynivalenol-induced damage. We found that
