Magnetically encoded luminescent composite nanoparticles through layer-by-layer self-assembly.

Sensitive and rapid detection of multiple analytes and the collection of components from complex samples are important in fields ranging from bioassays/chemical assays, clinical diagnosis, to environmental monitoring. A convenient strategy for creating magnetically encoded luminescent CdTe@SiO2 @n Fe3 O4 composite nanoparticles, by using a layer-by-layer self-assembly approach based on electrostatic interactions, is described. Silica-coated CdTe quantum dots (CdTe@SiO2 ) serve as core templates for the deposition of alternating layers of Fe3 O4 magnetic nanoparticles and poly(dimethyldiallyl a

Layer-by-Layer Hollow Mesoporous Silica Nanoparticles with Tunable Degradation Profile.

Silica nanoparticles (SNPs) have shown promise in biomedical applications such as drug delivery and imaging due to their versatile synthetic methods, tunable physicochemical properties, and ability to load both hydrophilic and hydrophobic cargo with high efficiency. To improve the utility of these nanostructures, there is a need to control the degradation profile relative to specific microenvironments. The design of such nanostructures for controlled combination drug delivery would benefit from minimizing degradation and cargo release in circulation while increasing intracellular biodegradatio

Electric field directed layer-by-layer assembly of highly fluorescent CdTe nanoparticles.

An electric field directed layer-by-layer assembly technique has been developed by combining electrophoretic deposition and the layer-by-layer self-assembly method. This technique is employed to realize spatially selective deposition of CdTe nanoparticles on electrodes of indium-tin oxide (ITO). The fluorescence measurements prove that the selectivity of the film deposition increases against applied voltage and reaches 99% when the voltage is 1.4 V. Upon further increase of the voltage, the selectivity slightly decreases. This decrease occurs because high voltage leads to a degradation of CdTe

Convection-Enhanced Delivery of Auristatin-Conjugated Layer-by-Layer Nanoparticles for Glioblastoma Treatment.

Glioblastoma (GBM) has limited treatment options, as the restrictive blood-brain barrier (BBB) prevents most therapeutics from accumulating at sufficient levels in the brain. Convection-enhanced delivery (CED) offers a method for administering therapeutics directly into brain tumor tissue, but free drugs can be cleared rapidly and may be toxic to off-target cells. Drug-loaded nanoparticles (NPs) are a promising platform to prolong the residence time and improve cellular targeting of therapeutics. We designed drug-conjugated NPs comprising a liposomal core modified with a layer-by-layer (LbL) p

Modeling lung adenocarcinoma using layer-by-layer nanoparticles mitigates innate immune cell activation.

Lung adenocarcinoma, driven frequently by KRAS and p53 mutations, remains a leading cause of cancer mortality. Current state-of-the-art genetically engineered mouse models often rely on viral delivery of recombinases, such as Cre recombinase, to initiate transformation. However, viral particles can infect and activate innate immune cells, thus potentially impacting studies of tumor-immune dynamics. Here, we develop a layer-by-layer (LbL) polyplex platform using poly(β-aminoester) (PBAE) polymers layered with poly-L-aspartic acid (PLD) to deliver Cre mRNA to lungs while avoiding immune cell tra

Layer-by-layer polyelectrolyte coating of low molecular weight poly(lactic acid) nanoparticles.

Low molecular weight (M(w)) poly(L-lactic acid) (PLA) nanoparticles were coated with polyelectrolytes (PEs) by layer-by-layer (LbL) technique using a filtration approach. Poly(allylamine hydrochloride) and poly(sodium 4-styrenesulfonate) were applied as PEs in coating. LbL coating is aimed to use in producing (nano)particulate drug delivery systems with improved biocompatibility and sustained or targeted release of drug substances. Nanoparticles of rapidly biodegradable polymers, like the low M(w) PLA, open up a possibility to control the release of the encapsulated substance by the coating, b

Polymer Molecular Weight Influences Cancer Cell Surface Retention and Cytokine Presentation by Layer-by-Layer Nanoparticles.

Engineering the surface chemistry of nanoparticles (NPs) via the electrostatic layer-by-layer (LbL) assembly approach is a viable strategy for developing targeted drug delivery vehicles. However, how assembly solution conditions and polymer molecular weight (MW) govern LbL-NP properties remains poorly understood. We investigated the influence of layering buffer ionic strength and polymer MW on the functional performance of PLR/PLE bilayer-coated LbL-NPs designed to promote cancer cell targeting and NP retention on the extracellular membrane to deliver the cytokine interleukin-12 (IL-12). Layer-by-Layer Delivery of Multiple Antigens Using Trimethyl Chitosan Nanoparticles as a Malaria Vaccine Candidate.

Developing a safe and effective malaria vaccine is critical to reducing the spread and resurgence of this deadly disease, especially in children. In recent years, vaccine technology has seen expanded development of subunit protein, peptide, and nucleic acid vaccines. This is due to their inherent safety, the ability to tailor their immune response, simple storage requirements, easier production, and lower expense compared to using attenuated and inactivated organism-based approaches. However, these new vaccine technologies generally have low efficacy. Subunit vaccines, due to their weak immuno

Aptamer based photoelectrochemical cytosensor with layer-by-layer assembly of CdSe semiconductor nanoparticles as photoelectrochemically active species.

A label-free photoelectrochemical cytosensor for highly sensitive and specific detection of Ramos cell was developed based on photoactive films. The films were fabricated by a layer-by-layer (LBL) assembly of positively charged poly(dimethyldiallylammonium chloride) (PDDA) and negatively charged CdSe semiconductor nanoparticles (NPs) capped with mercaptoacetic acid. The resulting modified electrodes were tested as sensors for Ramos cell through the recognition of DNA aptamer which was covalently bound to the electrode using the classic coupling reactions between -COOH groups on the surfaces of

Design and in vitro release study of siRNA loaded Layer by Layer nanoparticles with sustained gene silencing effect.

OBJECTIVES: Clinical translation of siRNA therapeutics has been severely limited due to the lack of stable and sustained siRNA delivery systems. Furthermore, when nanocarrier systems with siRNA are administered systemically to treat diseases, insufficient doses reach the target tissue. Here we report the successful development of a new nanocarrier system for the management of fibrosis. METHODS: The new carrier has a hydroxyapatite core, with alternating layers of siRNA and a cationic peptide. The siRNA used here targets secreted protein acidic and rich in cysteine (SPARC), a key matricellular

Layer-by-layer nanoparticle outer polyelectrolyte impacts protein corona formation.

Nanoparticles (NPs) can be engineered to achieve targeted delivery with strategies based on surface modifications. These include layer-by-layer (LbL) NPs, modular electrostatically assembled carriers with tunable surface properties altered by changes to the outer polyelectrolyte layer. Variations in these polymers dictate intracellular trafficking and biodistribution patterns. As NPs are administered, a layer of protein adsorbs to their surfaces, forming a protein corona that affects NP properties, alters biodistribution, and ultimately impacts therapeutic efficacy. We hypothesized that some d

Layer-by-layer assembled multilayers of polyethylenimine-stabilized platinum nanoparticles and PEDOT:PSS as anodes for the methanol oxidation reaction.

Polyethylenimine-capped platinum nanoparticles (PEI-capped Pt NPs) are synthesized by photoreduction and qualified as a component for electrostatic layer-by-layer assembly and subsequent electrocatalysis. The PEI-capped Pt NPs are characterized for size and charge using scanning force microscopy, transmission electron microscopy, dynamic light scattering and zetapotential. Well-defined multilayers are produced via thin film electrostatic assembly of PEI-capped Pt NPs with the conducting polymer: poly(3,4-ethylenedioxythiophene):poly(p-styrenesulfonate) [(PEDOT:PSS)(-)Na(+)]. Modular Layer-by-Layer Nanoparticle Platform for Hematopoietic Progenitor and Stem Cell Targeting.

Effective delivery of drug and gene cargos to hematopoietic stem and progenitor cells (HSPCs) is a major challenge. Current therapeutic strategies in genetic disorders or hematological malignancies are hindered by high costs, low accessibility, and high off-target toxicities. Layer-by-layer nanoparticles (LbL NPs) are modular systems with tunable surface properties to enable highly specific targeting. In this work, we developed LbL NPs that target HSPCs via antibody functionalization with reduced off-target uptake by circulating myeloid cells. NPs layered with poly(acrylic acid), a bioinert po

Layer-by-layer assembly of Au and CdS nanoparticles on the surface of bacterial cells for photo-assisted bioanodes in microbial fuel cells.

Surface modification of exoelectrogens with photoelectric materials is a promising way for achieving photo-assisted microbial fuel cells (MFCs). However, the poor conductivity of most photoelectric materials inevitably hampers the electron transfer inside bacterial biofilms. Herein, by utilizing the electrostatic layer-by-layer assembly strategy, the conductive Au nanoparticles (NPs) and photo-responsive CdS NPs were alternatively modified onto the surface of Escherichia coli for photo-assisted bioanodes in MFCs. The CdS layer was found to protect the bacterial cells from light illumination-in

pH-Responsive, Charge-Reversing Layer-by-Layer Nanoparticle Surfaces Enhance Biofilm Penetration and Eradication.

Microbes entrenched within biofilms can withstand 1000-fold higher concentrations of antibiotics, in part due to the viscous extracellular matrix that sequesters and attenuates antimicrobial activity. Nanoparticle (NP)-based therapeutics can aid in delivering higher local concentrations throughout biofilms as compared to free drugs alone, thereby enhancing the efficacy. Canonical design criteria dictate that positively charged nanoparticles can multivalently bind to anionic biofilm components and increase biofilm penetration. However, cationic particles are toxic and are rapidly cleared from c

Layer‑by‑layer biointerface engineering of biopolymer-nanoparticle hybrid coatings for durable antifouling and antibiofilm performance.

The layer-by-layer (LbL) assembly method using nanomaterials and naturally derived cationic and anionic polysaccharides is an actively researched field, driven to meet the demand for eco-friendly antifouling coatings and to improve current available alternatives. In this work, we present a strategy involving the activation of a polyethylene substrate to construct a four-layered coating consisting of an alginate-iron oxide nanoparticle hybrid (layer 1), bacterial cellulose (layer 2), an alginate-zinc sulfide nanoparticle hybrid (layer 3), and a protease extracted from Penaeus vannamei shrimp wa

Stacked-structure-dependent photoelectrochemical properties of CdS nanoparticle/layered double hydroxide (LDH) nanosheet multilayer films prepared by layer-by-layer accumulation.

Inorganic multilayer films were prepared by layer-by-layer accumulation of positively charged layered double hydroxide (LDH) nanosheets and negatively charged CdS nanoparticles of different sizes. Nanoparticles were densely immobilized on LDH sheets to form a monolayer without coalescence into larger particles. The absorbance and photoluminescence intensity of immobilized CdS particles were enlarged with an increase in the accumulation number of the film. Hybrid films produced by accumulation of both monolayers of CdS particles (diameter: 5 nm) and those of smaller CdS particles (2.1 nm) exhib

Layer-by-layer coated chitosan-CRISPR/Cas9 mTOR nanoparticles: A novel approach to inhibit lens epithelial cell proliferation and migration for preventing posterior capsule opacification.

Posterior capsular opacification (PCO) is the most common complication following cataract surgery and a significant cause of vision impairment. PCO arises from the proliferation, migration, and epithelial-mesenchymal transition (EMT) of residual lens epithelial cells (LECs), driven by an activated mTOR signalling pathway. Previous research has demonstrated that inhibiting mTOR activity effectively reduces LEC proliferation and EMT in rabbit models. However, achieving sustained mTOR inhibition remains a challenge. In this study, we encapsulated the CRISPR/Cas9 system targeting mTOR into chitosa
