The Complexity of Bovine Leukemia Virus Oncogenesis.

Bovine leukemia virus (BLV) is a retrovirus infecting several bovid species, notably Bos taurus, where it fulfills Koch's postulates for pathogenicity. The virus primarily targets B-lymphocytes, establishing lifelong infections that remain mostly asymptomatic but can progress to lymphocytosis or lymphoma. Transmission occurs through live infected cells via blood, milk, or transplacental routes. Despite a robust antiviral immunity, BLV replicates by producing virions (i.e., the infectious cycle) or inducing mitosis of infected cells (i.e., clonal expansion). The immune system effectively contro

Seroprevalence of bovine immunodeficiency virus and bovine leukemia virus in draught animals in Cambodia.

Since bovine immunodeficiency virus (BIV), known as bovine lentivirus, has been detected in dairy and beef cattle in various countries around the world, a prevalence study of antibodies to BIV and bovine leukemia virus (BLV) was conducted in draught animals in five provinces in Cambodia, where protozoan parasite infections were suspected in some animals. To clarify the status of draught animals including Haryana, Brahman, mixed-breed, local breed cattle and muscle water buffaloes, a total of 544 cattle and 42 buffaloes were tested, and 26.3 and 16.7%, respectively, were found positive for anti

Bovine leukemia virus pre-miRNA genes' polymorphism.

MicroRNAs (miRNAs) are small non-coding RNAs with a wide distribution in nature among the living things. They play a key role both in normal signaling pathways and in pathological ones. Bovine leukemia virus (BLV) is an oncogenic retrovirus of Deltaretrovirus genus causing persistent infection in its natural hosts - cattle, zebu and water buffalo with diverse clinical manifestations through the defeat of B-lymphocytes (B-cells). Ten BLV encoded miRNAs (further miRs-B) transcribed from five different pre-miRNA (further pre-miR-B) genes are abundantly detected in BLV infected B-cells. Here we re

Fetal infection with bovine leukemia virus in sheep.

Several sheep fetuses were thymectomized, and their tails were removed at 58 to 65 days of gestation for tissue culture. Bovine leukemia virus (BLV) antigens were detected in serial culture of tissues from fetuses whose dams and sires were both BLV positive. However, no BLV antigens were detected in serial cultures of tissues from fetuses whose dams were negative but whose sire was positive. Precolostral serums from 3 of 16 neonatal lambs, whose sire and dams were both BLV positive, were BLV antibody positive. Thus, BLV may be vertically transmitted from a positive dam to her lamb via the plac

Development of multipurpose recombinant reporter bovine leukemia virus.

Bovine leukemia virus (BLV) is a global problem that results in significant economic losses to the livestock industry. We developed three virus strains by inserting the HiBiT reporter tag from NanoLuc luciferase (NLuc) into limited sites within BLV molecular clones. Initial analysis for site selection of the tag insertion revealed a permissible site immediately downstream of the viral envelope gene. Therefore, NLuc activity could be used to measure virus copy numbers in the supernatant and the levels of cell infection. Productivity and growth kinetics of the reporter virus were similar to thos

Co-expression of bovine leukemia virus and bovine foamy virus-derived miRNAs in naturally infected cattle.

UNLABELLED: Among the viruses that encode microRNAs (miRNAs) and infect cattle, such as bovine leukemia virus (BLV), bovine foamy virus (BFV), and Bovine herpesviruses, BLV has gained attention due to the critical role that BLV-miRNAs play in inducing lymphosarcoma in infected animals. BLV is highly prevalent in the Americas and negatively affects dairy herds, primarily due to restrictions on the commercialization of dairy products from infected animals and a decrease in milk production. Co-infections involving BLV and BFV appear to be common in cattle. Considering the ability of foamy viruses

Mapping of CD4+ T-cell epitopes in bovine leukemia virus from five cattle with differential susceptibilities to bovine leukemia virus disease progression.

BACKGROUND: Bovine leukemia virus (BLV), which is closely related to human T-cell leukemia virus, is the etiological agent of enzootic bovine leukosis, a disease characterized by a highly prolonged course involving persistent lymphocytosis and B-cell lymphoma. The bovine major histocompatibility complex class II region plays a key role in the subclinical progression of BLV infection. In this study, we aimed to evaluate the roles of CD4+ T-cell epitopes in disease progression in cattle. METHODS: We examined five Japanese Black cattle, including three disease-susceptible animals, one disease-re

Vaccination against δ-retroviruses: the bovine leukemia virus paradigm.

Bovine leukemia virus (BLV) and human T-lymphotropic virus type 1 (HTLV-1) are closely related d-retroviruses that induce hematological diseases. HTLV-1 infects about 15 million people worldwide, mainly in subtropical areas. HTLV-1 induces a wide spectrum of diseases (e.g., HTLV-associated myelopathy/tropical spastic paraparesis) and leukemia/lymphoma (adult T-cell leukemia). Bovine leukemia virus is a major pathogen of cattle, causing important economic losses due to a reduction in production, export limitations and lymphoma-associated death. In the absence of satisfactory treatment for these

Invited review: Bovine leukemia virus-Transmission, control, and eradication.

Bovine leukemia virus (BLV) infection, endemic in North American dairy herds, has production-limiting effects. A literature review of available papers published since 1995 concerning BLV transmission and its control was conducted. Although confirmed transmission routes were reviewed (blood, natural breeding, in utero, colostrum, and milk), there is still a lack of detailed information on other specific risks for transmission (e.g., contact transmission and hoof-trimming knives). Eradication of BLV has been achieved by combined management, segregation, and culling approaches. In contrast, altho

Seroprevalence and Molecular Analysis of Bovine Leukemia Virus in Kazakhstan.

Bovine leukemia virus (BLV) remains a major concern for cattle industries worldwide due to its persistent nature, economic impact, and challenges in control. In this study, we conducted a comprehensive nationwide survey of BLV in Kazakhstan between 2014 and 2024, utilizing serological diagnostics to assess prevalence and characterize viral genotypes (2024). A total of 433,537 serum samples were screened by agar gel immunodiffusion (AGID), revealing an overall seroprevalence of 5.87%, with the highest rates observed in the North Kazakhstan, Kostanay, and East Kazakhstan regions. In 2024, a targ

The effect of bovine leukemia virus on dairy cow longevity.

Bovine leukemia virus (BLV) is a retrovirus of cattle that infects approximately 45% of all US dairy cattle, with about 90% of US dairy herds having at least one infected animal. Studies have found BLV infection to be associated with multiple measures of decreased immune function, which may explain the observed economic losses from milk production, decreased cow longevity, and predisposition to lymphoma and other diseases. Our objective was to measure the association between BLV infection and cow longevity in dairy cow operations. Ninety-one dairy herds from 9 US states volunteered to particip

Induction of syncytia by the bovine C-type leukemia virus.

Bovine buffy coat cells infected with the bovine leukemia virus (BLV) induce syncytia formation in human diploid embryonic lung cells as well as in monolayer cell cultures of bovine, simian, ovine, bat, and caprine origin, but not in mouse fibroblast cells, normall rat kidney cells, or RSV-transformed rat cells. Syncytia were not observed in diploid embryonic lung cells inoculated with bovine buffy coat cells free of BLV. The syncytia-induction effect is associated with the synthesis of complete BLV by the buffy coat cells and is independent of whether these cells are viable, disrupted, normal

Metabolomic evidence of bovine leukemia virus regulation on ferroptosis in bovine mammary epithelial cells.

Bovine leukemia virus (BLV) is a globally prevalent pathogen that can cause enzootic bovine leukosis (EBL), which reduces dairy cows' immunity, interferes with mammary antimicrobial defense capabilities, and exacerbates clinical mastitis. However, the specific mechanisms by which these effects occur remain incompletely understood. Ferroptosis, an iron-dependent non-apoptotic cell death mechanism, exhibits dual roles in viral infections. In this study, wild-type BLV and a miRNA-deficient mutant strain (BLV-ΔmiRNA) were used to infect bovine mammary epithelial cells in vitro, combined with non-t

Specific antiviral effect of violaceoid E on bovine leukemia virus.

Bovine leukemia virus (BLV) infection has spread worldwide causing significant economic losses in the livestock industry. In countries with a high prevalence of BLV, minimizing economic losses is challenging; thus, research into various countermeasures is important for improving BLV control. Because anti-BLV drugs have not been developed, the present study explored a promising chemical compound with anti-BLV activity. Initially, screening of a chemical compound library revealed that violaceoid E (vioE), which is isolated from fungus, showed antiviral activity. Further analysis demonstrated tha

Bovine leukemia virus relation to human breast cancer: Meta-analysis.

Bovine leukemia virus (BLV) is a virus that infects cattle around the world and is very similar to the human T-cell leukemia virus (HTLV), which causes adult T-cell leukemia/lymphoma (ATL). Recently, presence of BLV DNA and protein was demonstrated in commercial bovine products and in humans. BLV DNA is generally found at higher rates in humans who have or will develop breast cancer, according to research done with subjects from several countries. These findings have led to a hypothesis that BLV transmission plays a role in breast cancer oncogenesis in humans. Here we summarize the current kno

Molecular characterization of bovine leukemia virus from Moldovan dairy cattle.

Bovine leukemia virus (BLV) is the causative agent of enzootic bovine leukosis (EBL), a disease that has worldwide distribution. Whilst it has been eradicated in most of Western Europe and Scandinavia, it remains a problem in other regions, particularly Eastern Europe and South America. For this study, in 2013, 24 cattle from three farms in three regions of Moldova were screened by ELISA and nested PCR. Of these cattle, 14 which were PCR positive, and these were molecularly characterized based on the nucleotide sequence of the env gene and the deduced amino acid sequence of the encoded gp51 pr

Circulating immune complexes in bovine leukemia virus (BLV)-infected cattle.

Circulating immune complexes (ICs) were detected in the sera of bovine leukemia virus (BLV)-seropositive cattle. Immune complexes were precipitated in 2.5% polyethylene glycol (PEG) and further dissociated. Bovine leukemia virus antigens, IgG and IgM molecules were detected after solubilization in the presence of sodium dodecyl sulphate, and quantitated by enzyme-linked immunosorbent assay (ELISA) assays. Mean values of IgG and IgM in BLV-containing ICs did not significantly differ from those obtained from ICs originating from BLV-seronegative animals. However, differences were found in the co

Engineered soluble truncated envelope proteins block bovine leukemia virus infection.

Bovine leukemia virus (BLV), a member of the delta retrovirus family, is transmitted horizontally among cows. BLV causes enzootic bovine leukosis and has great economic impact on the cattle industry. Recently, secretory-defective Env proteins (e.g., Refrex-1 and FeLIX) have been detected in domestic cats and shown to possess antiretroviral activity against gammaretroviruses via viral receptor interference. Therefore, we investigated whether BLV-derived molecules engineered similarly exhibit antiviral activity against BLV infection. We generated several proteins consisting of the BLV envelope s

Circulating Cell-Free DNA of Bovine Leukemia Virus: A Promising Biomarker for Enzootic Bovine Leukosis.

Bovine leukemia virus (BLV) causes Enzootic bovine leukosis (EBL) in approximately 1%-5% of infected cattle after a long latent period. Few biomarkers effectively distinguish non-EBL from EBL cattle. Given the rapid turnover of tumor cells, we hypothesized that cell-free DNA (cfDNA) in plasma could serve as a more effective biomarker for EBL diagnosis. We measured the proviral load (PVL) in whole blood and plasma by quantitative PCR targeting LTR and pol. Consistent with previous reports, PVL levels in whole blood in EBL cattle were generally higher than those in non-EBL with some overlap betw
