Inactivation of filter bound aerosolized MS2 bacteriophages using a non-conductive ultrasound transducer.

The inactivation of viruses that retain their infectivity when transmitted through the air is challenging. To address this issue, this study used a non-contact ultrasound transducer (NCUT) to generate shock waves in the air at specific distances, input voltages, and exposure durations, targeting bacteriophage virus aerosols captured on to H14 HEPA filters. Initially, a frequency of 27.56 kHz (50V) at 25-mm distance was used, which yielded an inactivation efficiency of up to 32.69 ± 12.10%. Other frequencies at shorter distances were investigated, where 29.10 kHz had the highest inactivation ef

Development, Characterization, and Validation of an MS2 Phage-Based Armored RNA Control for Schmallenberg Virus Detection.

Schmallenberg virus (SBV) is a transboundary animal pathogen that causes reproductive disorders in ruminants, necessitating standardized molecular surveillance. Adhering to the Chinese national standard GB/T 43159-2023, we developed armored RNA quality control materials using MS2 bacteriophage technology targeting the conserved SBV S segment and comprehensively characterized their physicochemical properties. Transmission electron microscopy (TEM) showed icosahedral symmetric virus-like particles (VLPs, approximately 25 nm), with the stock concentration determined to be 3.48 × 1010 copies/mL vi

Virology laboratory biohazards: infectious MS2 aerosols emitted from common procedures and accidental laboratory scenarios in a controlled chamber study.

UNLABELLED: Infectious viral aerosols outside primary containment represent a profound yet undercharacterized occupational biohazard in microbiological environments. While the role of viral aerosols in laboratory-acquired infections is widely acknowledged, quantitative data on the concentration and size distribution of viral aerosols generated during common laboratory workflows remain limited. This study addresses these gaps by using a controlled chamber to characterize the generation of aerosols containing either a physical tracer, sodium fluorescein, or a viral pathogen surrogate, bacterioph

Humic acid and tryptophan prolong virus survival and infectivity: Molecular interactions and mechanisms.

Virus-induced infectious diseases represent one of the most significant threats to global public health. Dissolved organic matter (DOM) constitutes a critical component of aquatic environments and often influences the environmental behavior of microorganisms due to its unique structure and properties. However, research in this area remains limited, particularly concerning the effects of DOM on viral persistence in water. This study systematically investigated the influence of humic acid (HA) and tryptophan (Try) on the aggregation and survival characteristics of viruses, and utilized molecular

Escherichia coli bacteriophage vB_EcoS_MS2 effectively prevents biofilm formation but shows limited activity against mature biofilms.

Pathogenic Escherichia coli biofilms pose a significant challenge in veterinary and clinical settings due to their high antibiotic resistance. This study characterizes the E. coli bacteriophage vB_EcoS_MS2 as a potential targeted biocontrol agent against these pathogens. Host range determination revealed high specificity, with vB_EcoS_MS2 lysing only select E. coli strains and demonstrating superior replication efficiency (Efficiency of Plating: 112.57%) on strain C4 compared to its propagation host, MS2. While nutrient limitation strongly induced biofilm formation in in vitro assays, phage vB

Effects of chemically-reductive trace gas contaminants on non-thermal plasma inactivation of an airborne virus.

Transmission of airborne infectious diseases poses great risk for public health and socio-economic stability, thus, there is a need for an effective control method targeting the spread and transmission of pathogenic aerosols. The existence of chemically-reductive trace air contaminants in animal agriculture may affect the oxidation inactivation process of pathogens. In this study, we report how the presence of such gasses impacts the effectiveness of using non-thermal plasma (NTP) within a packed-bed dielectric barrier discharge reactor to inactivate MS2 bacteriophage. Inactivation of the aero

Infectivity reduction efficacy of UV irradiation and peracetic acid-UV combined treatment on MS2 bacteriophage and murine norovirus in secondary wastewater effluent.

Peracetic acid (PAA) is a strong oxidant/bactericide that has been applied in various industries (e.g., food processing, pharmaceuticals, medical device sterilization, etc.) as a disinfectant. There is increasing interest in using PAA for wastewater disinfection because it does not form halogenated byproducts, and no post-treatment quenching is required. Previous studies have demonstrated good efficiency in controlling bacteria in wastewater, but limited information is available for viruses, especially those hosted by mammals (e.g., norovirus). Therefore, a study on the infectivity reduction o

Efficacy of Laundry Practices in Eliminating Mpox Virus From Fabrics.

BACKGROUND: The declaration of mpox as a public health emergency of international concern highlights the need for interventions to interrupt virus transmission, including transmission via fabrics. Current World Health Organization (WHO) guidance on clothes washing is based on a general consensus of virus inactivation; however, there is uncertainty about the efficacy of laundry detergents and disinfectants or the reduction of risk achieved by washing clothes for mpox virus (MPXV) specifically. METHODS: This study investigates the efficacy of manual washing for inactivating MPXV from clothes. U

Preparing for the next pandemic: Efficacy of 2 commercial glycol vapor and aerosol products against aerosolized surrogate viruses and microbes on surfaces.

BACKGROUND: Aerosolized or vaporized glycols have been proposed as a safe and low-cost measure for air sanitization, but limited information is available on their efficacy in real-world conditions. METHODS: We evaluated 2 commercial glycol products, including a dipropylene glycol aerosol spray air sanitizer containing 30% to 60% ethanol and a triethylene glycol product released as a vapor over up to 48 hours. In a simulated patient room with 8 air changes per hour, we tested the efficacy of both products in reducing aerosolized bacteriophages Phi6 and MS2 in air and settling on disks, and the

Inactivation of MS2 bacteriophage on copper film deployed in high touch areas of a public transport system.

Although SARS-CoV-2 is primarily an airborne risk, the COVID-19 pandemic also highlighted the need for self-disinfection surfaces that could withstand the demand of high occupant densities characteristic of public transportation systems. The aim of this study was to evaluate the durability and antiviral activity of a copper film deployed for 90 days in two high touch locations within an active metropolitan bus and railcar. The antiviral efficacy of this copper film after being deployed in transit vehicles for 90 days (deployed copper film) was then compared to new (unused) copper film to deter

Enhanced effectiveness of medium-pressure ultraviolet lamps on human adenovirus 2 and its possible mechanism.

There has been growing concern over human exposure to adenoviruses through drinking water due to their apparent high resistance to UV irradiation and the anticipated widespread use of ultraviolet (UV) disinfection in drinking water treatment processes. However, most inactivation studies on adenoviruses were performed using only one type of UV technology--low-pressure (LP) UV, and little is known about the effectiveness of different UV technologies such as medium- pressure (MP) UV or other polychromatic UV technologies. In this work, the kinetics and extent of inactivation of a human adenovirus

Electrochemical disinfection of toilet wastewater using wastewater electrolysis cell.

The paucity of proper sanitation facilities has contributed to the spread of waterborne diseases in many developing countries. The primary goal of this study was to demonstrate the feasibility of using a wastewater electrolysis cell (WEC) for toilet wastewater disinfection. The treated wastewater was designed to reuse for toilet flushing and agricultural irrigation. Laboratory-scale electrochemical (EC) disinfection experiments were performed to investigate the disinfection efficiency of the WEC with four seeded microorganisms (Escherichia coli, Enterococcus, recombinant adenovirus serotype 5,

Co-generated particles, but not ambient particles, shield bioaerosols from Far-UVC inactivation and increase exposure to viable bioaerosols.

Far-UVC radiation can be an efficient and secure approach for bioaerosol sterilization in occupied high-risk settings like healthcare facilities. In dental clinics, aerosols produced by dental procedures are characterized by the existence of co-generated particles such as grind tooth fragments, restorative materials, and dental calculus. In this study, the influence of particles on far-UVC disinfection efficiency was investigated. First, the susceptibility of aerosolized bacteria and viruses to far-UVC radiation was experimentally measured under three conditions, i.e., without particles, with

Aerosolization of a Human Norovirus Surrogate, Bacteriophage MS2, during Simulated Vomiting.

Human noroviruses (NoV) are the leading cause of acute gastroenteritis worldwide. Epidemiological studies of outbreaks have suggested that vomiting facilitates transmission of human NoV, but there have been no laboratory-based studies characterizing the degree of NoV release during a vomiting event. The purpose of this work was to demonstrate that virus aerosolization occurs in a simulated vomiting event, and to estimate the amount of virus that is released in those aerosols. A simulated vomiting device was constructed at one-quarter scale of the human body following similitude principles. Sim

Validation of digital PCR as a complementary tool to MS2 plaque assay for virucidal activity assessment.

INTRODUCTION: Bacteriophage MS2 is widely applied as a surrogate for pathogenic non-enveloped RNA viruses that are difficult to culture. However, standardized approaches for evaluating disinfectant efficacy using MS2 remain limited. This study aimed to validate dPCR as a rapid quantitative complement to the previously established MS2 plaque assay, assessing their concordance in disinfectant efficacy testing using a previously established double-layer protocol. METHODS: A previously established double-layer MS2 plaque assay was employed. Disinfectant efficacy was tested against two widely used

Purification strategies and impact of surfactant on bacteriophage stability and aggregation.

Bacteriophages are promising bactericidal agents with potential for infection control, but purification methods and long-term storage can compromise their stability. Viral particle aggregation is a known cause of viability loss and may be reduced with surfactants such as Tween 20. This study evaluated the relationship between aggregation and infectivity loss in MS2 and PhiX174 purified by chloroform clarification and PEG concentration, with or without Tween 20, and by Triton X-100, over 55 days. Infectivity and aggregation varied by purification method, but no direct correlation emerged. MS2 a

Inactivation of indicator organisms on different surfaces after urban floods.

The high frequency and intensity of urban floods caused by climate change, urbanisation and infrastructure failures increase public health risks when the flood water contaminated from combined sewer overflows (CSOs) or other sources of faecal contamination remains on urban surfaces. This study contributes to a better understanding of the effects of urban and recreational surfaces on the occurrence of waterborne pathogens. The inactivation of selected indicator organisms was studied under controlled exposure to artificial sunlight for 6 h followed by 18 h in dark conditions. Concrete, asphalt,
