Immunocytochemical localization of Lewis blood group antigens in human salivary glands.

We demonstrated the immunohistochemical distribution of Le-a and Le-b blood group antigens in human major and minor salivary glands at the ultrastructural level by applying a post-embedding immunogold staining method. In secretors' glands, a faint Le-a reactivity was found only in mucous droplets, whereas Le-b antigen was intensely stained in secretory granules of most mucous cells, in those of intercalated duct cells, in the pale granular matrix of some serous cells, and, when osmication was omitted, in cytoplasmatic vesicles and cell surfaces of striated ducts. In the submandibular gland of

Lewis Blood-group Antigens Are Associated With Altered Susceptibility to Shigellosis.

In a cohort of infants, we found that lack of the Lewis histo-blood group antigen was associated with increased susceptibility to shigellosis. Broadly inhibiting fucosylation in epithelial cells in vitro decreased invasion by Shigella flexneri. These results support a role for fucosylated glycans in susceptibility to shigellosis.

Correlation of humoral immunity to Lewis blood group antigens with renal transplant rejection.

Recent studies suggest that the Lewis blood group antigens are important for human renal transplantation. A possible mechanism by which Lewis blood group incompatibility could influence allograft rejection was investigated by measuring Lewis antibodies in sera from renal transplant recipients and appropriate controls using conventional hemagglutination and a sensitive and specific kinetic enzyme-linked immunosorbent assay (k-ELISA). The two methods yielded identical results with 14 positive control sera known to contain Lewis antibodies and with 43 sera from negative controls. Antibody was not

Role of the Lewis and ABO Blood Group Antigens in Helicobacter pylori Infection.

BACKGROUND: Helicobacter pylori infection is a major risk factor for chronic gastritis and gastric cancer. Some findings show increased frequencies of these diseases in individuals with type O blood and in secretors (expressing Le(b) antigen), but other studies have not found any relationship between blood groups and this infection. Given that H. pylori infection and gastric cancer are common in Iran, the assessment of the pathogenesis of this infection in relation to these blood groups could be valuable. METHODS: In a cross-sectional study, we determined the ABO and Lewis blood groups of par

Expression of the blood-group-related antigens Sialyl Lewis a, Sialyl Lewis x and Lewis y in term placentas of normal, preeclampsia, IUGR- and HELLP-complicated pregnancies.

Lewis antigens belong to the blood group of antigens and mediate cellular adhesion through interaction with selectins. Invasive trophoblasts use an array of adhesion molecules to facilitate cell-cell and cell-extracellular matrix interactions. Here, we examined immunohistochemically the expression of Sialyl Lewis a (sLe(a)), Sialyl Lewis x (sLe(x)) and Lewis y (Le(y)) in term placentas obtained from cases of normal, intrauterine growth retardation (IUGR), preeclamptic (PE) and hemolysis, elevated liver enzymes and low platelets syndrome (HELLP) pregnancies. We report the expression of sLe(x) i

[Immunohistochemical studies on Lewis blood group antigens in carcinomas and adenomas of the sigmoid colon and rectum].

The presence of Lewis blood group antigens (Lewis(a), Lewis(b)) was determined immunohistochemically in 75 carcinomas and 58 adenomas of the sigmoid colon and rectum. 1. The rate of positive Lewis(b) staining of adult normal mucosa was 100% in the ascending colon, 100% in the transverse colon, 25% in the sigmoid colon and 30% in the rectum, respectively. The incidence of Lewis(b) was high in the proximal colon but low in the sigmoid colon and rectum. 2. The rate of positive Lewis(b) staining was 97% in cancer, 57% in adenoma and 26% in normal mucosa, respectively. The difference between the in

Metabolomic profiling of genotype-derived ABO blood group, secretor status, and Lewis antigens and association with pancreatic ductal adenocarcinoma risk.

The ABO locus is associated with pancreatic ductal adenocarcinoma (PDAC). Potential metabolic mechanisms underlying these associations have not been investigated. We examined associations between genotype-derived ABO blood group (rs505922 and rs8176746) and 1478 prediagnostic serum metabolites in 4042 participants from 8 nested case-control studies within the Prostate, Lung, Colorectal, and Ovarian Cancer Screening Trial and Alpha-Tocopherol, Beta-Carotene Cancer Prevention Study using linear regression and fixed-effect meta-analysis. Interrelation between ABH blood group 0, Lewis(B) blood group antigen, Helicobacter pylori infection, and occurrence of peptic ulcer.

BACKGROUND: Helicobacter pylori is a human pathogen that causes chronic gastritis and peptic ulcers. Epidemiological studies demonstrated that individuals who are blood group 0 positive or represent non-secretors of their blood group antigens are more likely to develop peptic ulcers. The Lewis(b) blood group antigen has been reported to mediate the attachment of H. pylori to human gastric mucosa. The aim of this study was to examine the interrelation between Le(a-b+) phenotype, blood group 0, H. pylori infection, and peptic ulcer occurrence. PATIENTS AND METHODS: The study population consiste

Blood group A and B antigen expression in human kidneys correlated to A1/A2/B, Lewis, and secretor status.

BACKGROUND: In the revived interest in crossing ABO barriers in organ transplantation renal A/B antigen expression has been correlated with donor ABO, Lewis, and secretor subtype to predict antigen expression. METHODS: A/B antigen expression was explored by immunohistochemistry in LD renal biopsies. Donor A1/A2/B, Lewis, and secretor status were determined by serology and polymerase chain reaction. RESULTS: In the renal vascular bed, three distinct A antigen expression patterns with a major, minor, and minimal staining distribution, and intensity (designated as types 3+, 1+ and (+) respectiv

Regulation of the oncodevelopmental expression of type 1 chain ABH and Lewis(b) blood group antigens in human colon by alpha-2-L-fucosylation.

Blood group antigen expression in the distal human colon is related to the development of the organ and is modified by malignant transformation. To elucidate the biochemical basis for these changes, we have (a) analysed the activity of glycosyltransferases coded for by the H, Se, Le, X, and A genes, in tissue biopsy specimens from normal and malignant proximal and distal human colon; (b) characterised the glycosphingolipids expressed in the various regions of normal and malignant colon by immunostaining of high performance thin layer chromatography plates; and (c) located the antigens on tissu

Effect of host Lewis and ABO blood group antigen expression on Helicobacter pylori colonisation density and the consequent inflammatory response.

The Lewis(b) blood group antigen has been implicated as a putative receptor for Helicobacter pylori in the gastric mucosa. Furthermore, an increased prevalence of duodenal ulcer was found in non-secretors and it has been suggested that secretor status may influence bacterial colonisation density. Other investigators have hypothesised that severity of antral gastritis may be related to colonisation density of the bacterium alone, and that a critical bacterial load is necessary for the development of duodenal ulcer. Our objectives were to investigate whether a relationship existed between host L

[Immunohistochemical examination of carcinomas in the pancreatoduodenal region and its non-malignant counterparts using monoclonal antibodies against Lewis blood group-related antigens and sialyl Tn antigen].

Forty cases of carcinomas in the pancreatoduodenal region (carcinoma of the ampulla of Vater: 11 cases, carcinoma of the pancreas head (CPH): 12 cases, carcinoma of the inferior common bile duct (CIBD): 17 cases) and its non-malignant counterparts were examined immunohistochemically, using monoclonal antibodies against four Lewis (Le) blood group-related antigens and sialyl Tn antigen. TKH-2 (anti-sialyl Tn) reactivity, which is almost negative in non-malignant counterparts except for goblet cells in the duodenum, is highly positive in carcinomas, therefore, sialyl Tn antigen can be an excelle

Measurements of the binding force between the Helicobacter pylori adhesin BabA and the Lewis b blood group antigen using optical tweezers.

Helicobacter pylori is a world-wide spread bacterium that causes persistent infections and chronic inflammations that can develop into gastritis and peptic ulcer disease. It expresses several adhesin proteins on its surface that bind to specific receptors in the gastric epithelium. The most well-known adhesin is BabA, which has previously been shown to bind specifically to the fucosylated blood group antigen Lewis b (Leb). The adhesion forces between BabA and the Leb antigen are investigated in this work and assessed by means of optical tweezers. A model system for in situ measurements of the

ABO, secretor, and Lewis carbohydrate histo-blood groups are associated with autoimmune neutropenia of early childhood in Danish patients.

BACKGROUND: Autoimmune neutropenia of early childhood (AIN) is caused by autoantibodies directed against antigens on the neutrophil membrane. The ABO, secretor, and Lewis histo-blood group systems control the expression of carbohydrate antigens and have previously been linked to autoimmune diseases. We aimed to investigate the association between genotypes and the risk of AIN in Danish patients. STUDY DESIGN AND METHODS: One hundred fifty-four antibody-positive AIN patients were included. Controls (n = 400) were healthy unrelated Danish blood donors. Molecular determination of ABO, secretor (
