The Leydig cell hypoplasia is a rare and well defined form of male pseudohermaphroditism with autosomal recessive inheritance pattern. An inadequate fetal testicular Leydig cell differentiation and, consequently a low androgenic production during intra uterine and post natal periods, result in absence or incomplete virilization in patients with 46,XY karyotype. These patients exhibit a wide clinical spectrum, ranging from complete female external genitalia to male external genital with micropenis, low serum testosterone levels associated with high LH levels. Inactivating mutations of the LH/hC

With the global population aging at an accelerated pace, testicular aging and its associated male health issues have garnered increasing attention. Testicular aging encompasses both age-related physiological decline and premature or pathological aging caused by various pathogenic factors. It is primarily characterized by reduced androgen secretion (mainly testosterone) and impaired spermatogenesis. Leydig cells, the key cells in the testes responsible for synthesizing and secreting testosterone, play a critical role in maintaining testicular function, and their dysfunction is one of the major

Leydig cells are the primary source of androgens in the mammalian testis. It is established that the luteinizing hormone (LH) produced by the anterior pituitary is required to maintain the structure and function of the Leydig cells in the postnatal testis. Until recent years, a role by the thyroid hormones on Leydig cells was not documented. It is evident now that thyroid hormones perform many functions in Leydig cells. For the process of postnatal Leydig cell differentiation, thyroid hormones are crucial. Thyroid hormones acutely stimulate Leydig cell steroidogenesis. Thyroid hormones cause p

From 64 consecutive autopsies of patients with neither testicular nor hormonal pathology, 26 showed extraparenchymal Leydig cells, located mainly in the epididymis and in the spermatic cord. The ultrastructural study of these specimens plus those obtained from 2 patients affected with functional testicular tumors leads to the following conclusions: (1) The origin of ectopic Leydig cells is not interstitial Leydig cells having infiltrated the testicular nerves and migrated along them towards ectopic locations. (2) The ectopic Leydig cells are considered to develop from undifferentiated precurso

This review is about the study of the testis Leydig cells formation and development in prenatal and postnatal periods. Leydig cells of testis are the main place of synthesis and secretion of androgens including testosterone--the main male sexual hormone. Testosterone plays an important role in male reproduction regulation. There are two types (two populations) of Leydig cells during ontogenesis. The first type is fetal Leydig cells, which appear and function in the prenatal masculinization period of the male urogenital system. Another type is adult Leydig cells, which originate during sexual m

We report on a 54-year-old male patient with Leydig cell tumor of testis, an incidental finding following orchiectomy for a testicular tumor. The pathological findings met the established histologic criteria for a benign tumor. The patient required no other treatment. The results of hormonal analyses were hormonal. We discuss recently proposed histologic and hormonal criteria to distinguish the benign from the malignant form of this tumor type. The foregoing are useful in determining the therapeutic approach.

In our search for markers to identify and study ovarian Leydig cells, we utilized immunohistochemical techniques and visualized the results using conventional and confocal microscopy. We successfully employed steroidogenic factor- 1 (SF1), androgen receptor (AR), and class III β-tubulin as markers. SF1 and AR specifically highlighted the intraneural cell precursors of Leydig cells, which were previously identified in a published case of mature cystic teratoma of the ovary, and the adult ovarian Leydig cells. Furthermore, the transient expression of class III β-tubulin could be associated with

Leydig cells, located in the loose interstitial tissue of seminiferous tubules, are the major site for androgen synthesis and secretion, and play an important role in the reproductively and fertility of males. The dysfunction of Leydig cells may lead to various male diseases, such as primary hypogonadism, cryptorchidism, and hypospadias. This review outlines the recent findings concerning the generation, development and regulation of Leydig cells.

It was demonstrated by light and electron microscopical methods that Leydig cells also exist outside of the testis. In the spermatic cord and near the intraabdominal testicular vessels cells were observed with the characteristics of intratubular Leydig cells. Such characteristics were intranuclear precrystalline inclusions, mitochondria with tubular interior structures and lipid droplets, and a well developed smooth and rough endoplasmatic reticulum. The last two items can be regarded as signs of hormonal activity. The extratesticular Leydig cells in the spermatic cord are located within nerve

Adrenal-like lipoid-rich Leydig cells, which could be found in a cryptorchid testis, were investigated by light and electronmicroscopy. There were nodular and diffuse proliferation of these adrenal-like cells in the interstitium of the testis. Electronmicroscopically these cells are fasciculated and characterized by large liposomes, many tubulovesicular mitochondria, and a large smooth endoplasmatic reticulum. But the presence of crystals of Reinke in these cells underlined their relationship to Leydig cells. The clinical history of this case is characterised by an extreme adipositas (167 kg)

Male aging is accompanied by reduced testosterone production by the Leydig cells, the testosterone-producing cells of the testis. The mechanism by which this occurs is unknown. Based on the observations that reactive oxygen is capable of damaging components of the steroidogenic pathway and that reactive oxygen is produced during steroidogenesis itself, we hypothesized that long-term suppression of steroidogenesis might inhibit or prevent age-related deficits in Leydig cell testosterone production. To test this, we administered contraceptive doses of testosterone to groups of young (3 months ol

The presence of ovarian hilar cells in anencephaly has been investigated and compared with the presence of testicular Leydig cells in the same condition. The examination of 29 anencephalic ovaries and 49 controls showed that up to 36 weeks of gestation there was a preponderance of ovarian hilar cells in anencephalic monsters as compared with controls. Similar observations were made with regard to ovarian follicles which showed more pronounced maturational changes than controls. However, during the last month of gestation both these processes were more marked in the controls. These findings are

Leydig cell hyperplasia and Leydig cell tumours of the ovary are rare. We present two cases in which patients had increased blood levels of testosterone and frank hirsutism. Imaging showed minimal abnormalities. After adrenal disease had been ruled out, they underwent a bilateral oophorectomy. One case showed a Leydig cell hyperplasia, the other a Leydig cell tumour. An androgen producing tumour should be excluded in every woman with evidence of hirsutism or frank virilization and markedly elevated testosterone levels. Adrenal disease with androgen hypersecretion can be suspected by detailed c

Leydig cells contain significant amounts of constitutively produced steroidogenic acute regulatory protein (STAR; STARD1). Hormone-induced STAR plays an essential role in inducing the transfer of cholesterol into the mitochondria for hormone-dependent steroidogenesis. STAR acts at the outer mitochondrial membrane, where it interacts with a protein complex, which includes the translocator protein (TSPO). Mutations in STAR cause lipoid congenital adrenal hyperplasia (lipoid CAH), a disorder characterized by severe defects in adrenal and gonadal steroid production; in Leydig cells, the defects ar

Drugs have been shown to adversely affect male fertility and recently anti-hypertensive drugs were added to the list. The anti-fertility effects of amlodipine, a calcium channel blocker, are well-illustrated in in vivo experiments but lack an in vitro proof. The present study was designed to experimentally elucidate the effects of amlodipine on Leydig cell steroidogenesis and intracellular calcium in vitro. Leydig cells of Sprague-Dawley rats were isolated and purified by Percoll. Cells were incubated for 3h with/without amlodipine in the presence/absence of LH, dbcAMP, Pregnenolone and 25-Hyd

Cells identical to Leydig cells were found within a peritubular boundary layer and even inside a basal lamina of seminiferous tubules in three male patients (two with inguinal cryptorchism and one with infertility). The seminiferous tubules of all patients showed a moderate to marked thickening of the boundary layer and a complete loss of spermatogenic cells. The "ectopic Leydig cells" were characterized by the presence of Reinke crystals or an extensively developed smooth endoplasmic reticulum. These cells were believed to have differentiated in situ from myoid cells within the boundary layer

Leydig cell hypoplasia(LCH) or agenesis, which has an autosomal recessive inheritance pattern, is a well defined form of male pseudohermaphroditism resulting from inadequate fetal testicular Leydig cell differentiation. These patients exhibit a wide clinical spectrum that ranges from phenotypic females to males with micropenis, and have low levels of serum testosterone and elevated levels of LH. Recently, it has become clear that defective male sex differentiation in patients with LCH is caused by deficient LH receptor signal transduction. The fetal Leydig cells of LCH patients fail to respond
