A scanning flow cell approach for generation of electrocatalyst material libraries via electrodeposition.

Multi-element material systems offer great property-tailoring possibilities for electrocatalysis. Experimental exploration of multi-dimensional composition spaces requires high-throughput methods for targeted synthesis and testing. We present a scanning flow cell setup that can synthesize material libraries of multi-component alloys via electrodeposition, test the electrocatalytic activity of the samples immediately after synthesis, and measure the stability under electrocatalytic conditions with online dissolution monitoring. Using this setup, we explored the Au-Pd-Pt ternary space with 45 sa

Application of High-Throughput Seebeck Microprobe Measurements on Thermoelectric Half-Heusler Thin Film Combinatorial Material Libraries.

In view of the variety and complexity of thermoelectric (TE) material systems, combinatorial approaches to materials development come to the fore for identifying new promising compounds. The success of this approach is related to the availability and reliability of high-throughput characterization methods for identifying interrelations between materials structures and properties within the composition spread libraries. A meaningful characterization starts with determination of the Seebeck coefficient as a major feature of TE materials. Its measurement, and hence the accuracy and detectability

Isolating Viral and Host RNA Sequences from Archival Material and Production of cDNA Libraries for High-Throughput DNA Sequencing.

The vast majority of surgical biopsy and post-mortem tissue samples are formalin-fixed and paraffin-embedded (FFPE), but this process leads to RNA degradation that limits gene expression analysis. As an example, the viral RNA genome of the 1918 pandemic influenza A virus was previously determined in a 9-year effort by overlapping RT-PCR from post-mortem samples. Using the protocols described here, the full genome of the 1918 virus was determined at high coverage in one high-throughput sequencing run of a cDNA library derived from total RNA of a 1918 FFPE sample after duplex-specific nuclease t

Understanding Self-Assembly of Silica-Precipitating Peptides to Control Silica Particle Morphology.

The most advanced materials are those found in nature. These evolutionary optimized substances provide highest efficiencies, e.g., in harvesting solar energy or providing extreme stability, and are intrinsically biocompatible. However, the mimicry of biological materials is limited to a few successful applications since there is still a lack of the tools to recreate natural materials. Herein, such means are provided based on a peptide library derived from the silaffin protein R5 that enables rational biomimetic materials design. It is now evident that biomaterials do not form via mechanisms ob

A novel biofunctionalizing peptide for metallic alloy.

OBJECTIVES: Improving biocompatibility of metallic alloy biomaterials has been of great interest to prevent implant associated-diseases, such as stent thrombosis. Herein a simple and efficient procedure was designed to biofunctionalize a biomaterial surface by isolating a SUS316L stainless steel binding peptide. RESULTS: After three rounds of phage panning procedure, 12 mer peptide (SBP-A; VQHNTKYSVVIR) was identified as SUS316L-binding peptide. The SBP-A peptide formed a stable bond to a SUS316L modified surface and was not toxic to HUVECs. The SBP-A was then used for anti-ICAM antibody modi

Multisize CdSe nanocrystal/polymer nanocomposites for selective vapor detection identified from high-throughput screening experimentation.

We have implemented high-throughput spectroscopic screening tools for the investigation of vapor-selectivity of CdSe semiconductor nanocrystals of different size (2.8- and 5.6-nm diameter) upon their incorporation in a library of rationally selected polymeric matrices. This library of resulting sensing materials was exposed to polar and nonpolar vapors in air. Each of the sensing materials demonstrated its own photoluminescence vapor-response patterns. Two criteria for the evaluation of vapor responses of the library of sensing materials included the diversity and the magnitude of sensing resp

Unraveling the Potential of Small Molecule Heparin Glycomimetics in Neuroregenerative Therapeutics.

Heparin and heparan sulfate (HS) glycosaminoglycans (GAGs) are essential regulators of neurotrophic signaling. However, their therapeutic applications are hindered by structural heterogeneity, batch variability, and anticoagulant activity. Thus, there is a need for well-defined glycomimetics that replicate the function of native HS in regenerative medicine. Here, we synthesized HS glycomimetics through a modular strategy that enables the installation of sulfate groups at the designated positions along the sugar backbone. These glycomimetics selectively bind and stabilize neurotrophins, such as

Identification of Biofilm Inhibitors by Screening Combinatorial Libraries of Metal Oxide Thin Films.

With the rise in nosocomial infections worldwide, research on materials with an intrinsic ability to inhibit biofilm formation has been generating a great deal of interest. In the present work, we describe how thin film material libraries generated by pulsed laser deposition can be used for simultaneously screening several novel metal oxide mixtures that inhibit biofilm formation in a common human pathogen, Salmonella enterica serovar Typhimurium. We discovered that in a material library constructed using two metal oxides, the net effect on biofilm formation can be modeled as an addition of th

Research in the Time of COVID: Virtual Fellowships at the Linda Hall Library.

When COVID-19 prevented scholars from traveling to libraries, archives, and museums, staff at those institutions devised creative strategies for providing remote access to their holdings. This article examines one such initiative launched by the Linda Hall Library of Science, Engineering, and Technology in Kansas City, Missouri. The Library's prioritization of print over archival materials, experience fulfilling document delivery requests, and investments in digitization infrastructure facilitated the transition to a completely virtual fellowship program during the 2021-22 academic year. Design of highly specific ligands of fibrin for therapeutic applications.

The work presented here is aimed at designing high-affinity ligands for the fibrin gamma (312-324) epitope. This epitope is specific for fibrin recognition, as it is exposed only on the fibrin surface, while in fibrinogen it is buried in the protein bulk. This property makes it very useful for therapeutic applications. In fact, it may be exploited in driving systems for targeted delivery of thrombolytic drugs toward the specific compartment where they are needed. It will then allow avoidance of serious unwanted side effects produced by a conventional systemic administration. The approach chose

Host-Directed Discovery of Dynamic-Covalent Guests From Dynamic Combinatorial Libraries.

Dynamic covalent chemistry and host-guest recognition are complementary tools for constructing adaptive molecular systems and functional materials, yet their integration has been hindered by the lack of general strategies for efficiently discovering host-compatible dynamic-covalent guests. Here we report a host-directed screening strategy that directly identifies optimal imine guests from dynamic combinatorial libraries. Upon addition of the macrocyclic host, selective host-guest binding drives thermodynamically controlled library redistribution, leading to significant constitutional amplifica

Orthopedic tissue regeneration: cells, scaffolds, and small molecules.

Orthopedic tissue regeneration would benefit the aging population or patients with degenerative bone and cartilage diseases, especially osteoporosis and osteoarthritis. Despite progress in surgical and pharmacological interventions, new regenerative approaches are needed to meet the challenge of creating bone and articular cartilage tissues that are not only structurally sound but also functional, primarily to maintain mechanical integrity in their high load-bearing environments. In this review, we discuss new advances made in exploiting the three classes of materials in bone and cartilage reg

Facile Preparation of a Glycopolymer Library by PET-RAFT Polymerization for Screening the Polymer Structures of GM1 Mimics.

Commercialized oligosaccharides such as GM1 are useful for biological applications but generally expensive. Thus, facile access to an effective alternative is desired. Glycopolymers displaying both carbohydrate and hydrophobic units are promising materials as alternatives to oligosaccharides. Prediction of the appropriate polymer structure as an oligosaccharide mimic is difficult, and screening of the many candidates (glycopolymer library) is required. However, repeating polymerization manipulation for each polymer sample to prepare the glycopolymer library is time-consuming. Herein, we report

Performance of non-formalin fixed paraffin embedded samples in hybrid capture and amplicon next-generation sequencing panels.

BACKGROUND: Genomic profiling using next-generation sequencing (NGS) is fundamental for driving prognostic and therapy in cancer. Formalin-fixed paraffin embedded (FFPE) tissue is the widely used material, whereas non-FFPE may represent an alternative. However, studies comparing the NGS performance of non-FFPE materials to FFPE are still lacking in the literature. The objective of this study was to characterize in non-FFPE preparations the nucleic acid yield and NGS performance on both a capture-based and an amplicon-based NGS platform. NGS quality metrics obtained from non-FFPE preparations w

Genetically Encoded Fragment-Based Discovery from Phage-Displayed Macrocyclic Libraries with Genetically Encoded Unnatural Pharmacophores.

Genetically encoded macrocyclic peptide libraries with unnatural pharmacophores are valuable sources for the discovery of ligands for many targets of interest. Traditionally, generation of such libraries employs "early stage" incorporation of unnatural building blocks into the chemically or translationally produced macrocycles. Here, we describe a divergent late-stage approach to such libraries starting from readily available starting material: genetically encoded libraries of peptides. A diketone linchpin 1,5-dichloropentane-2,4-dione converts peptide libraries displayed on phage to 1,3-diket

Hydroxyapatite Scaffold and Bioactive Factor Combination as a Tool to Improve Osteogenesis, In Vitro and In Vivo Experiments Using Phage Display Technology.

Mesenchymal stem cells have been widely investigated in the field of regenerative medicine and also used as a model to study the differentiation-induction properties of a variety of biomaterials. This study evaluates the osteoinductive potential of novel hydroxyapatite scaffolds functionalized with a phage-displayed peptide (SC1) selected via biopanning for its similarity to bone matrix proteins. The peptide, identified through sequence alignment as a mimotope of osteonectin (SPARC), was used to functionalize scaffolds. Results from SC1 were gathered at different time points (14, 28 and 46 day

Synthetic materials trained tactile sensing enables quantitative perception of Young's modulus and Poisson's ratio.

Accurately perceiving object softness remains challenging for tactile sensors, as current approaches estimate Young's modulus from object deformation in response to an applied pressure while overlooking the pivotal role of Poisson's ratio in shaping that deformation. Here we present a tactile sensor that quantitatively and accurately detects both Young's modulus and Poisson's ratio, achieved by constructing a broad materials library spanning diverse Young's modulus-Poisson's ratio and capturing materials' surface bulging height-pressure trajectories. Using these trajectories as machine-learnin

Redesigning library orientation for first-year medical students during the pandemic.

BACKGROUND: Prior to 2020, library orientation for first-year medical students at Weill Cornell Medicine took the form of an on-site treasure hunt competition. Due to the COVID-19 pandemic, the orientation for the MD class of 2024 was shifted to an all-virtual format. This shift mandated a full redesign of the library orientation. CASE PRESENTATION: The Samuel J. Wood Library sought to preserve the excitement and fun of the treasure hunt in the new virtual format. The competition was redesigned as a Zoom meeting using breakout rooms, with library faculty and staff serving as team facilitators

Fabrication and Characterization of Drug-Loaded Conductive Poly(glycerol sebacate)/Nanoparticle-Based Composite Patch for Myocardial Infarction Applications.

Heart tissue engineering is critical in the treatment of myocardial infarction, which may benefit from drug-releasing smart materials. In this study, we load a small molecule (3i-1000) in new biodegradable and conductive patches for application in infarcted myocardium. The composite patches consist of a biocompatible elastomer, poly(glycerol sebacate) (PGS), coupled with collagen type I, used to promote cell attachment. In addition, polypyrrole is incorporated because of its electrical conductivity and to induce cell signaling. Results from the in vitro experiments indicate a high density of c
