Bone marrow lipofuscin.

Lipofuscins are commonly present in the macrophages of the marrow. In unstained preparations they may be confused with haemosiderin, but they are readily distinguished by fluorescence microscopy. In contrast to the belief that lipofuscins are a manifestation of senility, no age dependence has been demonstrated. Exceptionally large amounts have been found in illnesses accompanied by fever and leucocytosis, in keeping with the concept of their formation from insoluble remains of ingested cell fragments. It is probable that the ;sea-blue histiocytes', described in the literature, are macrophages

Lipofuscin hypothesis of Alzheimer's disease.

The primary culprit responsible for Alzheimer's disease (AD) remains unknown. Aβ protein has been identified as the main component of amyloid of senile plaques, the hallmark lesion of AD, but it is not definitively established whether the formation of extracellular Aβ deposits is the absolute harbinger of the series of pathological events that hit the brain in the course of sporadic AD. The aim of this paper is to draw attention to a relatively overlooked age-related product, lipofuscin, and advance the hypothesis that its release into the extracellular space following the death of neurons may

Catabolism of the Lipofuscin Cycloretinal by MsP1.

Age-related macular degeneration (AMD) is a debilitating eye disease that tends to affect people over the age of 55. Lipofuscins are autofluorescent, toxic byproducts of the visual cycle thought to contribute toward the progression of the disease. Targeting the accumulation of lipofuscin through catabolism may serve as a method for the early treatment of AMD. Thus, an enzymatic approach capable of degrading lipofuscin, cycloretinal (all-trans retinal dimer), was examined. A peroxidase from the organism Marasmius scorodonius (MsP1) has shown capability of degrading this toxic metabolite into no

Lipofuscin in human glaucomatous optic nerves.

Lipofuscin accumulation has been observed in a number of neurodegenerative diseases. We recently found that autofluorescent particles also occur in the aged human optic nerve. In this study we sought to determine the nature of these particles and their correlation with aging, age-related macular degeneration (AMD) and primary open angle glaucoma (POAG). Groups of eight optic nerves from patients diagnosed with primary open angle glaucoma, age-related macular degeneration, age-matched controls and four optic nerves derived from controls younger than 42 years were used for the study. All samples

Lipofuscin-dependent stimulation of microglial cells.

PURPOSE: To examine the reaction of microglial cells (MG) when incubated with lipofuscin (LP) in vitro with emphasis on the immunological reaction of the MG toward LP and the suppression of this reaction by immunomodulatory agents. MG are involved in the pathogenesis of degenerative eye disorders such as age-related macular degeneration (AMD). LP is a heterogeneous waste material that accumulates in the retinal pigment epithelium (RPE) cells with advancing age. LP is known to have toxic effects on RPE cells and therefore an elevated LP-derived fundus autofluorescence is a risk factor for AMD d

Human and rat brain lipofuscin proteome.

The accumulation of an autofluorescent pigment called lipofuscin in neurons is an invariable hallmark of brain aging. So far, this material has been considered to be waste material without particular relevance for cellular pathology. However, two lines of evidence argue that lipofuscin may play a yet unidentified role for pathological cellular functions: (i) Genetic forms of premature accumulation of similar autofluorescent material in neuronal ceroid lipofuscinosis indicate a direct disease-associated link to lipofuscin; (ii) Retinal pigment epithelium cell lipofuscin is mechanistically linke

Lipofuscin and autofluorescence metrics in progressive STGD.

PURPOSE: To evaluate Stargardt disease (STGD) progression and relative lipofuscin levels via autofluorescence image analysis. METHODS: The relationship between focally increased autofluorescence (FIAF), geographic atrophy (GA) and focally decreased autofluorescence (FDAF) was analyzed in serial, registered autofluorescence (AF) scans of 10 patients with STGD (20 eyes, 40 scans; mean follow-up, 2.0 years) using automated techniques. RESULTS: GA progressed uniformly in a transition zone with minimal FIAF. Only 4.3% of FIAF progressed to GA or FDAF, despite significant progression of GA (median

The lipofuscin-iron association in pigmentosis tubae.

Pigmentosis tubae (PT) is a rare condition characterized by the presence of numerous lipofuscin-laden macrophages in the lamina propria of the fallopian tube. Two women, who also had endometriotic ovarian cysts, showed polypoid pigmented tubal mucosae. In addition to lipofuscin, occasional cells showed spotty positivity for iron. Ultrastructural examination of the tubal mucosa showed the lipofuscin-containing bodies, which were similar to lipofuscin-containing lysosomes found in other pigmented conditions. Cytoplasmatic ferritin and hemosiderin in siderosomes were observed in macrophages and e

Lipofuscin of the retinal pigment epithelium: a review.

Accumulation of lipofuscin is one of the most characteristic features of ageing observed in retinal pigment epithelial (RPE) cells. The lipofuscin found in RPE cells differs from that of other body tissues due to the fact that it is mainly derived from the chemically modified residues of incompletely digested photoreceptor outer segments. It is a heterogeneous material composed of a mixture of lipids, proteins, and different fluorescent compounds, the main fluorophore of which has recently been identified as a derivative of vitamin A. Research interest has variously focussed on the roles of ag

Lipofuscin pigments in the spiral ganglion of the rat.

The purpose of the present study was to clarify morphological differences in lipofuscin or so-called age pigments observed in the spiral ganglion cells of both young and adult rat groups and to characterize the size and structure of ceroid pigment granules generated in vitamin-E-deficient rats. The results showed different patterns of lipofuscin distribution in the two groups. The adult rat group had large aggregated lipoid, dark pigment granules of irregular shape in the cytoplasm. In contrast, the young group had small numbers of small, dense homogeneous granules, suggesting higher Schwann c

Central neurocytoma with lipofuscin granules--case report.

A 30-year-old male presented with a large calcified intraventricular mass. The tumor was completely resected via the transventricular route. Light microscopy showed the tumor was composed of small round cells with perinuclear halo, typical of oligodendroglioma. Electron microscopy revealed marked neuronal differentiation, including neurosecretory granules, well-formed numerous synapses, and lipofuscin granules. The tumor was diagnosed as well-differentiated central neurocytoma.

Hermansky-Pudlak syndrome: albinism with lipofuscin storage.

The Hermansky-Pudlak Syndrome, a "tyrosinase positive' form of oculocutaneous albinism, is a triad comprising albinism, a hemorrhagic diathesis and ceroid-lipofuscin storage. A pedigree is presented showing consanguinity with a pattern of pseudodominance. Electroretinography in two isolated Hermansky-Pudlak subjects was distinctly abnormal, showing decreased rod and cone responses (as well as abnormal flicker fusion responses) in one patient, and reduced photopic and scotopic responses in another. The decreased ERG responses are discussed with reference to the known retinal abnormalities in bo

Bisretinoid lipofuscin, fundus autofluorescence and retinal disease.

Retinal pigment epithelium emits an inherent autofluorescence that originates from naturally occurring fluorophores when excited by short-wavelength light (SW-AF) in the spectral range between 400 and 590 nm. Peak excitation is 490 nm. The autofluorescence emission occurs at wavelengths between 520 and 800 nm with a peak of approximately 600 nm. For clinical purposes this emission is recorded as fundus autofluorescence either using a confocal scanning laser ophthalmoscope (cSLO; 488 nm excitation); a modified fundus camera or by ultra-wide-field ophthalmoscopic technology. The topographic dist

Neuronal ceroid-lipofuscin storage in Siamese cats.

The clinico-pathological findings of a nervous disease in two mature Siamese cats are described. In one animal intermittent convulsions and mania over a period of three weeks terminated in death, while in the other irritability and hind leg weakness were the main clinical features. Neuronal cytoplasmic inclusions composed of a complex lipid material were detected microscopically. The ultrastructure and staining reactions of these inclusions in one cat were similar to the curvilinear bodies described in neurovisceral storage disease in humans and in a neuronal lipodystrophy of cattle. The depos

Hepatic Lipofuscin Deposition in Living Liver Donor Candidate.

Living donor liver transplantation (LDLT) is an important therapeutic option, and ensuring donor safety is paramount. We report a case of a woman in her early 50s evaluated as a living liver donor candidate for her husband. Recurrent mild elevations of alanine aminotransferase (ALT) levels prompted further investigation, and a percutaneous ultrasound-guided liver biopsy revealed unexpected and marked lipofuscin deposition within hepatocytes, without evidence of steatosis, inflammation, fibrosis, or cholestasis. Lipofuscin, an age-related pigment, is generally considered benign, with an unclear

Lipofuscin causes atypical necroptosis through lysosomal membrane permeabilization.

Lipofuscin granules enclose mixtures of cross-linked proteins and lipids in proportions that depend on the tissue analyzed. Retinal lipofuscin is unique in that it contains mostly lipids with very little proteins. However, retinal lipofuscin also presents biological and physicochemical characteristics indistinguishable from conventional granules, including indigestibility, tendency to cause lysosome swelling that results in rupture or defective functions, and ability to trigger NLRP3 inflammation, a symptom of low-level disruption of lysosomes. In addition, like conventional lipofuscins, it ap

Lipofuscin deposition causes the pigmentation of apocrine hidrocystoma.

Apocrine hidrocystoma (AHC) is a benign cystic lesion arising from apocrine glands and frequently presents as a blue hae with a whitish gloss. It has been reported that the amount of lipofuscin and the degree of its oxidation determines the pigmentation of AHC. However, little is known about the correlation between clinical color and lipofuscin granules. In this study, we histopathologically investigated three cases of AHC and statistically analyzed the quantitative correlation between the clinical color tone and the amount of cytoplasmic lipofuscin deposits. The color tone was quantified as t

Mitochondrial autophagy and lipofuscin accumulation in aging odontoblasts.

Aging of long-lived post-mitotic cells is characterized as a progressive and irreversible reduction of functional activity. In such cells, mitochondria are organelles critical for bioenergetic supply, whose turnover is mediated by an autophagic-lysosomal pathway. In human teeth, odontoblasts are post-mitotic cells responsible for sensory function and dentin preservation. Here, human odontoblasts were processed for immunohistochemistry with antibodies against mitochondrial (MTCO2) and lysosomal (LAMP2) markers, and comparatively analyzed in two age groups (young-adult and adult) with light and
