Ephaptic coupling can explain variability in neural activity.

The waxing and waning cortical oscillatory power correlates with function and disease. This cross-trial variability has been thought to be due to neuromodulation, uncertainty encoding, and/or changes in cortical excitability. Here, we report evidence that it is also due to fluctuations in ephaptic influences of mesoscale electric fields. We analyzed LFP data from the PFC recorded during a spatial delay saccade task. We constructed a model that describes the electric field close to the cortical patch given the neural activity that generates it. This revealed that field-to-neuron interactions (e

Adaptive Deep Brain Stimulation for Parkinson's Disease: Navigating the Roadblocks to Clinical Implementation.

Adaptive deep brain stimulation (aDBS) represents an important evolution in the treatment of Parkinson's disease (PD), building on conventional DBS (cDBS) by adjusting stimulation in response to real-time physiological signals. By enabling dynamic targeting of disease-related neural activity, aDBS offers the potential for more precise modulation of motor symptoms. Additional anticipated advantages include reduced stimulation-related side effects and improved energy efficiency, supporting long-term device performance. Although clinical uptake is still at an early stage, growing experience has h

Microelectrode array scaled for human hippocampal slices.

OBJECTIVE: Temporal lobe epilepsy (TLE) is characterized by recurrent seizures originating usually from the hippocampus, and approximately one-third of TLE patients remain refractory to pharmacological interventions. Surgical resection offers a potential cure for refractory TLE cases, with approximately 70% achieving seizure freedom. Still, the pathogenesis of TLE remains poorly understood. Electrophysiological characterization of the resected tissue with microelectrode arrays (MEAs) can help reveal the pathogenesis of TLE, but the commercially available MEAs cover only a small part of the hip

Local Field Potentials in Movement Disorders: A Narrative Review.

<p>Background: Local field potentials (LFPs) represent the summed electrical activity of neuronal populations and provide a critical window into synaptic and oscillatory dynamics across brain networks. In recent decades, advances in deep brain stimulation (DBS) and chronic sensing technology have established LFPs as essential biomarkers for understanding movement disorders and optimizing neuromodulation therapies. Summary: This review synthesizes evidence on LFPs in movement disorders, focusing on methodological aspects, functional characterization, disease-specific findings, and clinical appl

On variability in local field potentials.

Neuronal variability is a fundamental feature of neuronal coding. In spiking activity, across-trial variance (ATV) normalized by mean spike count (i.e., the Fano factor) shows stimulus-induced quenching. ATV has also been studied in electro/magneto-encephalography (E/MEG) without normalization, revealing effects of stimulation and cognition. Here we show that outside of event-related potentials (ERPs), ATV for both EEG and local field potential (LFP) is nearly identical to intra-trial variance (ITV), which equals signal power. ATV-ITV correlation decreases during ERPs, proportional to how much

Fiber-Less, Large-Scale Opto-Electrophysiology Interface for Micro-Scale Interaction of Multiple Brain Regions.

OBJECTIVE: Recent neuroscientific research craves for understanding sophisticated brain networks formed by neuron ensembles across multiple regions. An ideal way to unveil the complex connectome is bidirectionally interacting (simultaneous recording and stimulation) with neurons at high spatiotemporal resolutions. Existing CMOS recording probes cannot provide micro-scale interactions with limited stimulation capability. Although optogenetics can achieve neuron-specific stimulation, conventional methods using optic fibers illuminate a large volume of tissue, resulting in unspecific perturbation

An artefact-resilient wide bandwidth bidirectional graphene neural interface.

The ability to simultaneously record and modulate neural activity is critical for next-generation bidirectional neural interfaces aiming to enable adaptive neuromodulation therapies for neurological disorders. Active graphene transistor technologies are particularly promising for neural recordings, as they extend the ability to monitor brain activity at very low frequencies and support multiplexed operation for high-density neural interfaces. However, their limited charge injection capacity makes them unsuitable for stimulation. In this work, we present a bidirectional neural interface that co

Spatially Extensive LFP Correlations Identify Slow-Wave Sleep in Marmoset Sensorimotor Cortex.

Identifying neural signatures of slow-wave sleep (SWS) is important for a number of reasons including diagnosing potential sleep disorders and examining its role in memory consolidation ( Diekelmann and Born, 2010; Klinzing et al., 2019; Brodt et al., 2023). Studies of sleep in the common marmoset (Callithrix jacchus) have revealed similarities to humans and other nonhuman primates, including distinct sleep stages ( Crofts et al., 2001) and diurnal sleep patterns ( Hoffmann et al., 2012). Advances in applying wireless technology for recording neural activity during natural, unrestrained behavi

A Low-Power PLL-Less Wideband OOK Wireless Neural-Signal Transmitter for Miniaturized Neural Interfaces with In Vivo Validation in Freely Moving Mice.

High-channel-count neural recording requires wireless links with high throughput, low power, and compact implementation, yet commercial protocols and phase-locked loop (PLL)-based transmitters often trade data rate against power and complexity. We present a low-power, PLL-less wideband on-off keying (OOK) neural-signal transmitter fabricated in a 180 nm CMOS process. The transmitter employs a free-running inductor-capacitor voltage-controlled oscillator (LC-VCO), a Gilbert mixer for OOK modulation and reverse isolation, and a current-reuse stacked power amplifier. It consumes 8 mA from a 3.3 V

Information content and interpretation of CSD non-trivially depends on array density.

Objective. We aim to determine whether commonly-used current source density (CSD) estimation techniques provide valid results when used with recordings from the latest generation of high-density arrays and how the density affects signal interpretation.Approach.We first develop an analytic formulation for the contributions of Gaussian current sources to an estimated CSD as a function of current source position and array density. We then simulate local field potential recordings in a biophysically detailed model of a subvolume of rat somatosensory cortex, and compare estimated current source dens

Frequency-Dependent Inhibition during Deep Brain Stimulation of Thalamic Ventral Intermediate Nuclei.

Deep brain stimulation (DBS) of the thalamic ventral intermediate nucleus (Vim) has been a standard therapy for essential tremor. It has been shown that high-frequency (≥100 Hz) DBS suppresses Vim firing and tremor activity; however, the underlying mechanisms are not fully understood. Here, we investigate whether neuronal suppression during high-frequency DBS occurs at cellular levels or is influenced by network-level effects. Using in vivo recordings of Vim neurons, from people of unknown sex, during different DBS frequencies, we detected a positive-going evoked-field potential, quasi-evoked

Local infrared stimulation modulates spontaneous cortical slow wave dynamics in anesthetized rats.

Cortical slow waves are hallmark oscillations of deep sleep and certain anesthetic conditions, yet the neurobiological mechanisms controlling their dynamics remain incompletely understood. Here, we investigated the effects of local near-infrared (NIR) stimulation on slow-wave activity in ketamine/xylazine-anesthetized rats. Using a silicon-based multimodal optrode, we simultaneously delivered NIR light and recorded local field potentials (LFPs) and multi-unit activity (MUA) across cortical layers in the primary somatosensory (S1Tr) and parietal association (PtA) cortices. NIR stimulation induc

Interactions between Hippocampus and Visceral Organs in Sleep and Wakefulness.

Exteroception is attenuated during sleep, while interoceptive signals may remain active. If so, they can potentially trigger responses in their target areas, including the hippocampus, which is known to receive information from the internal organs. We investigated whether the hippocampus is synchronized to various visceral events in sleep and wakefulness. Activity of hippocampal neurons and local field potentials (LFPs) was coregistered with respiration, heart rate, and myoelectric signals of the stomach and duodenum in two adult female cats over multiple sleep-wake cycles. Visceral event-trig

Similar Response Dynamics Represent Opposite Behaviors and Rewards in the Frontal Cortex.

The frontal cortex (FC) plays a pivotal role in adaptively controlling actions and their dynamics in response to incoming sensory signals. We explored FC encoding of identical stimuli and their behavioral consequences when they signified diametrically opposite responses depending on task context. Two groups of female ferrets performed Go-NoGo auditory categorization tasks with opposite contingencies and rewards and diverse stimuli. Remarkably, despite opposite stimulus-action associations, single-unit responses were similar across all tasks, being more sustained and stronger to Target sounds (

Neural Network Connection Characteristics Between Motor Cortex and Spinal Cord During Rat Bipedal Walking.

OBJECT: Bipedal locomotion requires coordinated corticospinal interactions, yet how these networks dynamically reorganize across gait phases remains poorly understood. This study aimed to characterize the phase-dependent restructuring of corticospinal functional networks during rest, stance, and swing in healthy rats. METHODS: Single-unit spiking activity was simultaneously recorded from the hindlimb motor cortex and lumbar spinal cord of healthy rats across three behavioral states: Rest, stance, and swing. Granger causality analysis was applied to local field potentials (LFPs) to assess dire

Local Field Aperiodic Spectral Power Modulated by Deep Brain Stimulation in Parkinson's Disease.

BACKGROUND: Aperiodic spectral broadband power has been described recently as reflecting Parkinson's disease (PD) severity. It has therefore become an increasing focus of research interest in the context of the new adaptive deep brain stimulation (DBS) approach. OBJECTIVE: We aimed to study the influence of DBS on the main parameters of the aperiodic spectral component of local field potentials (LFPs). METHODS: LFPs were recorded from the subthalamic nucleus (STN) in patients with PD (n = 22) during a simple experimental paradigm that included 5 minutes of resting state and a short gait task

The potential of utilizing Local Field Potential (LFP) in diagnosis and management of Parkinsonism's hallmark symptoms: a comprehensive review.

Parkinson’s disease (PD) is a prevalent neurodegenerative disorder that significantly impacts motor functions. Despite advances in diagnostic imaging, accurately diagnosing and understanding PD remains challenging. Local field potentials (LFPs), which represent the collective electrical activity of neuron populations, can provide insights into the pathophysiology of PD, particularly in symptoms such as tremors, dystonia, gait disturbances, sleep disorders, and cognitive dysfunction. Tremors, a hallmark symptom of PD, have been closely studied through LFPs recorded from the subthalamic nucleus

Localizing beta synchronous neurons in the STN using directional DBS recordings and patient-specific biophysical models.

OBJECTIVE: Local field potentials (LFPs) exhibit abnormally elevated beta-band power in the subthalamic nucleus (STN) of patients with Parkinson's disease (PD). To better understand these signals, we coupled advanced biophysical models with experimental LFP recordings to characterize the neural sources underlying beta oscillations in PD patients. METHODS: Patient-specific biophysical models simulated LFPs with detailed representation of the directional DBS electrodes in the local neuroanatomy, as well as the ionic currents generated by STN neural activity. We then used the models to perform s

Spatial memory performance is associated with region-specific coordination of hippocampo-cortical sleep oscillations.

Sleep plays a critical role in memory consolidation, yet how coordination among sleep oscillations across hippocampo-cortical circuits relates to behavioral performance remains incompletely understood. Here, we examined how cross-regional coordination of cardinal sleep oscillations during non-rapid eye movement (nREM) sleep relates to spatial memory performance. Adult rats were trained in an object-place recognition task and allowed to sleep while local field potentials were recorded simultaneously from dorsal and ventral hippocampus (CA1d and CA1v) and retrosplenial, prefrontal, and lateral e

Assessment of Anxiety- and Depression-like Behaviors and Local Field Potential Changes in a Cryogenic Lesion Model of Traumatic Brain Injury.

Patients with traumatic brain injury (TBI) have an elevated risk of developing chronic psychiatric and behavioral disorders, including impairments in motor function, memory deficits, anxiety, and depression. Although a substantial body of work has addressed the treatment and rehabilitation of sensory, motor, and cognitive symptoms after TBI, there is a relative scarcity of comprehensive behavioral assessments targeting neuropsychiatric manifestations in preclinical models. This study aims to investigate the connections between emotional sequelae after TBI and modifications in local field poten
