Calcium buffering in axons and axoplasm of Loligo.

1. Ca-selective micro-electrodes were used to measure free Ca concentration in axons and extruded axoplasm. 2. Free Ca in axons immersed in artificial sea water containing 3 mM-Ca averaged 77 nM in freshly dissected axons and 4.9 microM in cyanide- or azide-poisoned axons. 3. Extruded axoplasm maintained a free Ca only a little higher than that of the axons from which it was obtained. 4. Axoplasmic buffering was investigated by titrating isolated axoplasm with CaCl2 or K-EGTA and monitoring the change in free Ca. Energy-dependent and energy-independent components of Ca binding could be recogni

Occupational allergy to squid (Loligo vulgaris).

Occupational allergy from exposure to squid has been rarely described, mainly as contact dermatitis or urticaria. Our report presents the first case of occupational asthma, rhinitis, conjunctivitis and contact urticaria to squid in a 33-year-old seafood production worker, with documented increased eosinophilia in the nasal and tear fluids after specific inhalation challenge test (SICT) with squid. IgE-mediated sensitization to squid was confirmed by positive skin prick test and opened skin test with squid extract. SICT demonstrated a direct and significant link between the exposure to squid an

Morphology and electrical properties of Schwann cells around the giant axon of the squids Loligo forbesi and Loligo vulgaris.

The first successful dye-fills of Schwann cells around the split giant axon of Loligo show them to be spindle-shaped cells ca. 600 microns long and 20 microns wide lying parallel to the axonal axis. There are some 50,000 Schwann cells per cm2 of axonal membrane. Only a small part (ca. 6% of each Schwann cell membrane) is in contact with the periaxonal space, the remainder is overlain by adjacent Schwann cells, or applied to the basal lamina. The mean membrane potential of the Schwann cells in artificial seawater (ASW) varies from around -40 mV in fresh split-axon preparations to around -60 to

Metabolism and transport of strontium in giant axons of Loligo.

1. The strontium content of squid axons averages 2.9 mumol kg-1 axoplasm and squid blood contains 75 mumol Sr l-1. The bulk of the Sr inside axons is bound, possibly within mitochondria. Sr seems to be handled by squid axons in an essentially similar manner to Ca. 2. Sr uptake is increased by electrical activity, by lowering external Na, by depolarization with K and by poisoning with cyanide or FCCP. Uptake is unaffected by ouabain. 3. Sr efflux has a Q10 of 2.2. Efflux can be divided operationally into three components: one dependent on external Ca (or Sr), one dependent on external Na and a

Sodium transport by perfused giant axons of Loligo.

The sodium efflux from perfused squid giant axons has been studied using radioactive sodium, and the sufficient conditions for the maintenance of a potassium- and ouabain-sensitive sodium efflux have been established. The following were found.1. Axons extruded and then perfused with their own axoplasm had a sodium efflux which was sensitive to cyanide, potassium and ouabain and was thus similar to the efflux from intact axons.2. A method for replacing natural axoplasm into fibres previously perfused with artificial axoplasm was developed and used to establish an artificial perfusate that was n

Acid-base regulation in exercising squid (Illex illecebrosus, Loligo pealei).

Squid (Illex illecebrosus, Loligo pealei) were cannulated in the vena cava and swum in a Beamish-type respirometer. Gas tensions and acid-base variables as well as octopine levels were estimated in samples of the mantle and of venous blood collected from quiescent, exercised, and recovered animals. When exhausted, both species exhibited a decrease in vena cava oxygen tensions and a slight alkalosis. With high swimming speeds prior to exhaustion in Illex a slight acidosis developed in the blood, which was linked to a severe intracellular acidosis. Generally, the drop in intracellular pH was lin

Spatial localization of calcium channels in giant fiber lobe neurons of the squid (Loligo opalescens).

Whole-cell voltage clamp was used to investigate the properties and spatial distribution of fast-deactivating (FD) Ca channels in squid giant fiber lobe (GFL) neurons. Squid FD Ca channels are reversibly blocked by the spider toxin omega-Agatoxin IVA with an IC50 of 240-420 nM with no effect on the kinetics of Ca channel gating. Channels with very similar properties are expressed in both somatic and axonal domains of cultured GFL neurons, but FD Ca channel conductance density is higher in axonal bulbs than in cell bodies at all times in culture. Channels presumably synthesized during culture a

Contact chemosensory cues in egg bundles elicit male-male agonistic conflicts in the squid Loligo pealeii.

Male Loligo pealeii engage in frequent agonistic bouts to gain access to female mates while aggregated at communal egg beds. Male squids are attracted to eggs in the field and in the laboratory. It was recently demonstrated that visual detection followed by physical contact with egg capsules elicited male-male aggression. We tested specific physical and chemical features of the egg capsules that may cause this strong behavioral reaction. Male squids were presented with either natural or artificial egg stimuli and scored for four selected behaviors (egg touch, egg blowing, forward-lunge grab, a

Molecular and morphological characterisation of the metacestode Anthobothrium sp. infecting Loligo vulgaris from Tunisian coasts.

Cestode larvae of Anthobothrium sp. were collected from the caecum and stomach of the common squid Loligo vulgaris Lamarck, 1798 (Cephalopoda: Loliginidae) in Tunisian waters (Bay of Bizerte and Gulf of Tunis), with the highest prevalence recorded in the Gulf of Tunis (52.94%). These specimens were morphologically characterised and compared to previously described Anthobothrium larvae from other hosts. Molecular analysis using 28S ribosomal DNA (rDNA), combined with phylogenetic reconstruction (maximum likelihood and pairwise distances), revealed that the sequences cluster with adult specimens

Cleavage furrow formation in a telolecithal egg (Loligo pealii). I. Filaments in early furrow formation.

Formation of the first cleavage furrow in the telolecithal egg of Loligo was studied with the electron microscope. Before the actual furrow forms, a dense filamentous band develops below the plasma membrane from membrane-bounded dense bodies which appear to be Golgi-derived. The egg surface is thrown into a number of longitudinal folds which parallel the furrow and eventually become incorporated into it. These longitudinal folds contain a network of tubules and vesicles. Frequently, multivesiculate bodies are associated with the furrow and possibly give rise to the network of tubules and vesic

The Effect of Molecular Weight on the Antimicrobial Activity of Chitosan from Loligo opalescens for Food Packaging Applications.

The growing requirement for sustainable processes has boosted the development of biodegradable plastic-based materials incorporating bioactive compounds obtained from waste, adding value to these products. Chitosan (Ch) is a biopolymer that can be obtained by deacetylation of chitin (found abundantly in waste from the fishery industry) and has valuable properties such as biocompatibility, biodegradability, antimicrobial activity, and easy film-forming ability. This study aimed to produce and characterize poly(lactic acid) (PLA) surfaces coated with β-chitosan and β-chitooligosaccharides from a

Cadmium content in fresh and canned squid (Loligo opalescens) from the Pacific coastal waters of California (USA).

Cadmium (Cd) levels were determined in 70 samples of mantle tissue and 70 whole individual squid (Loligo opalescens; commercially known as California squid). Samples were collected from the coastal zones of California (USA) during the period 2007/2008. To further investigate consumer exposure to processed fishery products, cadmium concentration was also determined in 200 canned samples of squid. Cd concentrations in raw mantle were low, between 0.01 and 0.29 mg kg(-1) and below the tolerance limit of current regulations (1 mg kg(-1)). Respective concentrations in whole individuals were signifi

Effect of toxin 1 from Androctonus australis Hector on sodium currents in giant axons of Loligo forbesi.

The effects of external application of micromolar concentrations of toxin 1 of the scorpion, Androctonus australis Hector, on the sodium conductance of squid giant axons have been studied quantitatively using the voltage clamp technique. Toxin concentrations which induce long plateau action potentials under current clamp conditions were found to simultaneously decrease the peak conductance and increase the delayed sodium conductance. Return to holding potential level after step depolarizations was accompanied by large exponential tails of current. The toxin-induced maintained sodium conductanc

Backbone and side chain chemical shift assignment of diisopropyl fluorophosphatase (DFPase) from Loligo vulgaris, an organophosphorus-degrading enzyme.

NMR chemical shift assignments are reported for backbone (15N, 1H) and partial side chain (13Cα and β, side chain 1H) atoms of diisopropyl fluorophosphatase (DFPase), a calcium-dependent phosphotriesterase capable of hydrolyzing phosphorus - fluorine bonds in a variety of toxic organophosphorus compounds. Analysis of residues lining the active site of DFPase highlight a number of residues whose chemical shifts can be used as a diagnostic of binding and detection of organophosphorus compounds.

Peripheral injury induces long-term sensitization of defensive responses to visual and tactile stimuli in the squid Loligo pealeii, Lesueur 1821.

Survivable injuries are a common yet costly experience. The ability to sense and respond to noxious stimuli is an almost universal trait, and prolonged behavioral alterations, including sensitization to touch and other stimuli, may function to ameliorate fitness costs associated with injury. Cephalopods can modify their behavior by learned association with noxious electric shock, but non-associative alterations of behavioral responses after tissue injury have not been studied. The aim of this study was to make the first systematic investigations in any cephalopod of behavioral responses and al
