Biochemical, Bioluminescence and Bioreporter Properties of Functionally Improved Firefly Luciferases.

Firefly luciferase (Fluc) is one the most widely used bioluminescent reporters for biological imaging and gene expression analysis, yet its limited stability and high assay cost remain major constraints limiting broader applications. Here, we engineered Fluc variants (N116H, M118L and N116H/M118L) with improved catalytic performance, thermostability, and soluble expression. The N116H variant exhibited the most pronounced enhancement, including a 4.6-fold increase in kcat, a 2.7-fold higher soluble expression yield, a 3.5-fold longer half-life at 37 °C, and 1.25-fold enhanced quantum yield rela

Secreted luciferases as a minimally invasive 3R-compliant tool for accurate monitoring of tumor burden.

Preclinical mouse models are indispensable in cancer research, providing insights into tumor biology and therapeutic responses. This protocol describes a minimally invasive blood-based tumor monitoring approach using secreted luciferases for longitudinal tracking of tumor burden in transplantable xenografts and genetically engineered mouse models. Unlike intracellular luciferases used in bioluminescence imaging, secreted luciferases are actively released into circulation, enabling precise quantification from microliter-scale blood samples. We describe a transplantable model, where tumor cells

A recombinant rabies virus expressing luciferase.

A recombinant Rabies virus (RV) expressing firefly luciferase (rRV-luc) was generated by an improved reverse genetics system. Its biological properties were compared with those of the parental RV. The rRV-luc grew in BHK-21 cells similarly to RV, but its virulence for mice was weaker as shown by the lower infectious titers in brain. Rising infectious titers of rRV-luc during its passaging in BHK-21 cells indicated a virus adaptation, while the luciferase (luc) expression was stable. These results suggest that the recombinant RV carrying luc gene might prove a useful tool for further analysis o

Expression and characterization of the Renilla luciferase with the cumulative mutation.

Luciferase from Renilla reniformis (RLuc) is a good research tool as a reporter protein and bioimaging probes, yielding blue light using the substrate coelenterazine. However, the applications are limited since RLuc is unstable under various conditions. Therefore, an attempt was made to increase RLuc thermostability. In this study, 5 mutations reported previously [1] and one mutation obtained using site-directed mutagenesis were combined. As a result of this combination, the thermostability effect increased, with the mutant showing approximately 10 °C higher stability. Furthermore, the mutant

Factors affecting the cellular expression of bacterial luciferase.

The in vivo expression of cellular bacterial luciferase has been defined as the luciferase expression quotient, measured as the ratio of the bioluminescence intensity in vivo to the in vitro activity of luciferase in crude cell extracts. The expression is greater in the presence of inhibitors of the electron transport system such as cyanide and N-heptyl-4-hydroxy-quinoline and also at lower oxygen tensions. The higher expression of the cellular luciferase under these conditions is postulated to be due to an increase in the intracellular levels of reduced coenzymes which enhance both the reduct

Luciferase Reporter Assay for MicroRNA Target Validation.

MicroRNAs (miRNAs) are small, non-coding RNA molecules, typically 22-25 nucleotides in length, that modulate gene expression after transcription. They exert their function by binding to complementary sequences within the 3' untranslated regions (3' UTRs) of target messenger RNAs (mRNAs), leading to mRNA degradation or translational repression. Over the past decade, miRNAs have emerged as critical key mediators of essential cellular functions, such as cell growth, lineage commitment, programmed cell death, and adaptation to stress. Dysregulated miRNA expression has been linked to the developmen

A reproducible and affordable method of conducting luciferase assay.

Commercially available glow luciferase assay kits are widely popular and convenient to use. However, concerning high-throughput screening, commercial kits are limited by huge running costs. As an alternative to commercial luciferase assay kits, this study presents a cost-effective and efficient methodology of performing a simple and rapid laboratory flash luciferase assay. The proposed luciferase assay method has a versatile use ranging from screening lysates in a microplate reader for quantitative assay as well as screening live cells qualitatively or quantitatively under an imaging system.

Novel luciferase-opsin combinations for improved luminopsins.

Previous work has demonstrated that fusion of a luciferase to an opsin, to create a luminescent opsin or luminopsin, provides a genetically encoded means of manipulating neuronal activity via both chemogenetic and optogenetic approaches. Here we have expanded and refined the versatility of luminopsin tools by fusing an alternative luciferase variant with high light emission, Gaussia luciferase mutant GLucM23, to depolarizing and hyperpolarizing channelrhodopsins with increased light sensitivity. The combination of GLucM23 with Volvox channelrhodopsin-1 produced LMO4, while combining GLucM23 wi

Luciferase as a model for the site of inhaled anesthetic action.

UNLABELLED: The in vivo potencies of anesthetics correlate with their capacity to suppress the reaction of luciferin with luciferase. In addition, luciferin has structural resemblances to etomidate. These observations raise the issues of whether luciferin, itself, might affect anesthetic requirement, and whether luciferase resembles the site of anesthetic action. Because the polar luciferin is unlikely to cross the blood-brain barrier (we found that the olive oil/water partition coefficient was 100 +/- 36 x 10(-7)), we studied these issues in rats by measuring the effect of infusion of lucifer

Luciferase-dependent oxygen consumption by bioluminescent vibrios.

Oxygen uptake due to luciferase in two luminous Vibrio species was estimated in vivo by utilizing inhibitors having specificities for luciferase (decanol) and cytochromes (cyanide). Cyanide titration of respiration revealed a component of oxygen uptake less sensitive to cyanide which was completely inhibitable by low concentrations of decanol. From this it was estimated that in vivo luciferase is responsible for less than 12% (Vibrio harveyi) or 20% (Vibrio fischeri) of the total respiration. From these data in vivo bioluminescent quantum yields are estimated to be not lower than 1.7 and 2.6%,

A panel of Trypanosoma brucei strains tagged with blue and red-shifted luciferases for bioluminescent imaging in murine infection models.

BACKGROUND: Genetic engineering with luciferase reporter genes allows monitoring Trypanosoma brucei (T.b.) infections in mice by in vivo bioluminescence imaging (BLI). Until recently, luminescent T.b. models were based on Renilla luciferase (RLuc) activity. Our study aimed at evaluating red-shifted luciferases for in vivo BLI in a set of diverse T.b. strains of all three subspecies, including some recently isolated from human patients. METHODOLOGY/PRINCIPAL FINDINGS: We transfected T.b. brucei, T.b. rhodesiense and T.b. gambiense strains with either RLuc, click beetle red (CBR) or Photinus py

Mutagenesis and Structural Studies Reveal the Basis for the Activity and Stability Properties That Distinguish the Photinus Luciferases scintillans and pyralis.

The dazzling yellow-green light emission of the common North American firefly Photinus pyralis and other bioluminescent organisms has provided a wide variety of prominent research applications like reporter gene assays and in vivo imaging methods. While the P. pyralis enzyme has been extensively studied, only recently has a second Photinus luciferase been cloned from the species scintillans. Even though the enzymes share very high sequence identity (89.8%), the color of the light they emit, their specific activity and their stability to heat, pH, and chemical denaturation are quite different w

Evaluation of LIPS (luciferase immunoprecipitation system) for serodiagnosis of Toxoplasmosis.

Development of reliable, quantitative technologies for serodiagnosis of Toxoplasma gondii infection remains desirable. The luciferase immunoprecipitation system (LIPS) is a relatively simple, highly sensitive, and rapid quantitative immunoassay. The major advantages of this assay over ELISA are a wider dynamic range, shorter overall assay time, and less sample volume. In this study, we aimed to use this method for the serodiagnosis of toxoplasmosis. Recombinant Toxoplasma antigens (dense granule antigens GRA6, GRA7, and GRA8 and bradyzoite antigen BAG1) fused with nanoluciferase (Nluc, a small

Assessment of immunity to mycobacterial infection with luciferase reporter constructs.

Protective immunity to mycobacterial infection is incompletely understood but probably involves the coordinated interaction of multiple cell types and cytokines. With the aim of developing assays that might provide a surrogate measure of protective immunity, we have investigated the use of recombinant mycobacteria carrying luciferase reporter enzymes to assess the effectiveness of antimycobacterial immunity in model systems. Measurement of luminescence was shown to provide a rapid and simple alternative to the counting of CFU as a means of monitoring mycobacterial viability. We describe optimi

Construction of luciferase-expressing Neospora caninum and drug screening.

BACKGROUND: Neospora caninum is an apicomplexan parasite that is particularly responsible for abortions in cattle and neuromuscular disease in dogs. Due to the limited effectiveness of currently available drugs, there is an urgent need for new therapeutic approaches to control neosporosis. Luciferase-based assays are potentially powerful tools in the search for antiprotozoal compounds, permitting the development of faster and more automated assays. The aim of this study was to construct a luciferase-expressing N. caninum and evaluate anti-N. caninum drugs. METHODS: Luciferase-expressing N. ca

Opiate analgesics' dual role in firefly luciferase activity.

The effects of three opiate analgesics, isolated from opium, on the firefly luciferase enzyme have been studied. Morphine (MN), 6-acetylmorphine (MAM), and diacetylmorphine (DAM) inhibited the enzyme activity at different levels. At lower concentrations, MN and MAM enhanced enzyme activity, effecting inhibition at higher concentrations. However, DAM inhibited the enzyme activity at all concentrations investigated. The stimulating activity of MN and MAM is attributed to the hydrophilic interaction of the proton donor-acceptor type with the polar regions of the luciferase located outside the bin

Effects of surfactants on luminescence intensity and duration of the minimal luciferase picALuc.

Bioluminescence is an oxidation-mediated chemical reaction involving the luciferase enzyme and the luciferin substrate. To date, various artificial luciferases have been developed, and their luminescence properties have been investigated. However, very few studies have focused on the solvent environment in which these luminescent reactions occur, and the details of their influence on luminescent phenomena remain unclear. In this study, we investigated the effects of various surfactants on the activity of the luciferase picALuc. To this end, we analyzed the effects of adding diverse surfactants

A novel luciferase-based assay for quantifying coronavirus-induced syncytia.

Coronaviruses can induce cell‒cell fusion that results in the formation of multinucleated syncytia through the interaction of viral spike proteins with host cell receptors. Quantifying syncytial formation is crucial for screening potential efficacious antiviral compounds. However, some traditional methods for syncytial quantification are often labor-intensive and limited by a low-throughput capacity. Therefore, we developed a novel high-throughput assay for the efficient quantification of syncytial formation induced by feline coronavirus (FCoV) and SARS-CoV-2. This assay, which is based on the
