Surface dose measurement by optically stimulated luminescent dosimeter: A phantom study.

INTRODUCTION: Radiotherapy is the standard treatment for breast cancer patients after surgery. However, radiotherapy can cause side effects such as dry and moist desquamation of the patient's skin. The dose calculation from a treatment planning system (TPS) might also be inaccurate. The purpose of this study is to measure the surface dose on the CIRS thorax phantom by an optically stimulated luminescent dosimeter (OSLD). METHODS: The characteristics of OSLD were studied in terms of dose linearity, reproducibility, and angulation dependence on the solid water phantom. To determine the surface

Chemosensitivity testing using microplate adenosine triphosphate-based luminescence measurements.

During the last two decades, novel nonclonogenic methods for pretherapeutic chemosensitivity testing have been developed that are likely to overcome major technical limitations of older assays such as low evaluability rates, low degree of standardization and reproducibility, lack of technical robustness, and poor methodological efficacy. Among these, the microplate adenosine triphosphate (ATP)-based tumor chemosensitivity assay (ATP-TCA) has gained particular merits for ex vivo chemosensitivity testing of native nonhematological tumors including cancers of the breast, ovary, gastrointestinal t

Measuring Nicotine in Water Contaminated by Cigarette Butts: Evaluation of a Luminescent Organometallic Network.

Cigarette butts are the world's leading source of litter, disrupting water quality and the normal development of ecosystems. Tobacco use in its various forms is the leading preventable cause of death, illness, and disability worldwide. Nicotine (NCT), a natural alkaloid found in tobacco plants, is a psychoactive substance and the primary component responsible for the addictive properties of such products. Since NCT is an analyte with low native fluorescence, the use of a fluorophore that interacts with it is necessary for its quantification by molecular fluorescence. This work describes, for t

Characterization of optically stimulated luminescence dosemeters to measure organ doses in diagnostic radiology.

OBJECTIVE: The aim of this study was to assess the characteristics of an optically stimulated luminescence dosemeter (OSLD) for use in diagnostic radiology and to apply the OSLD in measuring the organ doses by panoramic radiography. METHODS: The dose linearity, energy dependency and angular dependency of aluminium oxide-based OSLDs were examined using an X-ray generator to simulate various exposure settings in diagnostic radiology. The organ doses were then measured by inserting the dosemeters into an anthropomorphic phantom while using three panoramic machines. RESULTS: The dosemeters demon

In vitro tests of the validity of singlet oxygen luminescence measurements as a dose metric in photodynamic therapy.

Singlet oxygen ((1)O(2)) is widely believed to be the major cytotoxic agent involved in photodynamic therapy (PDT). We showed recently that measurement of the weak near infrared luminescence of (1)O(2) is possible in cells in vitro and tissues in vivo. Here, we investigated the relationship between the integrated luminescence signal and the in vitro PDT response of AML5 leukemia cells sensitized with aminolevulinic acid-induced protoporphyrin IX (PpIX). Sensitized cell suspensions were irradiated with pulsed 523 nm laser light at average fluence rates of 10, 25, or 50 mWcm(-2) and, (1)O(2) lum

Effects of halothane, enflurane, and isoflurane on measurements of Ca(2+) by calcium electrode and aequorin luminescence.

We assessed the possible effects of the volatile halogenated anesthetics halothane, enflurane, and isoflurane on Ca(2+) electrode measurements and on the Ca(2+) sensitivity of the bioluminescent protein aequorin. In Ca(2+)-EGTA buffers of different pCa values (7. 870, 6.726, 6.033, 4.974, 4.038, and 2.995) and in serial Ca(2+) dilutions (10(-4), 10(-3), and 10(-2) M), halothane, enflurane, and isoflurane each caused a concentration-dependent and reversible increase in the absolute value of the negative electrode potential. Isoflurane and enflurane had larger effects than halothane. Neither of

Measuring Live-Cell mRNA Translational Dynamics with Split Luminescent Tagging in HEK293 Cells.

The swift vaccine development to combat COVID-19 illustrated the potential for messenger RNA (mRNA) therapeutics to transform drug development. Like mature mRNA, in vitro transcribed mRNA possesses the same elements including a 5' cap, untranslated regions (UTRs), coding sequence and a poly(A) tail. Previous work studying the effects these components have on mRNA translation has primarily utilized highly engineered reporter proteins which exhibit efficient translation and protein stability. With the structural elements of each mRNA differentially affecting their translation, it is imperative t

Effect of anesthesia on the signal intensity in tumors using BOLD-MRI: comparison with flow measurements by Laser Doppler flowmetry and oxygen measurements by luminescence-based probes.

BOLD-contrast functional magnetic resonance imaging (MRI) was used to assess the evolution of tumor oxygenation and blood flow after administration of four different anesthetics: pentobarbital (60 mg/kg), ketamine/xylazine (80/8 mg/kg), fentanyl/droperidol (0.078/3.9 mg/kg), and isoflurane (1.5%). Gradient echo sequences were carried out at 4.7 Tesla in a TLT tumor model implanted in the muscle of NMRI mice. In parallel experiments, tumor blood flow and tumor pO2 were measured using the OxyLite/OxyFlo probe system. A comparison was made with the changes occurring in the skeletal muscle (host t

An Open-Source, Excitation-Resolved UV-NIR Imaging Platform for Bioluminescence, Luminescence-Decay, and Chemical Discrimination Measurements.

Multispectral imaging is a cornerstone of chemical imaging, with bioluminescence, fluorescence, and phosphorescence imaging underpin much of preclinical and analytical chemical measurements. However, conventional implementations are typically proprietary, costly, and resolve spectral content from the reflectance of a broadband source, without control over the excitation spectrum. This architecture cannot isolate excitation-dependent photophysical processes. Spectral discrimination through sequentially resolved narrowband excitation, detected on a single broadband-sensitive camera, instead enab

Use of Mitochondrial Delayed Luminescence Measurements From Brain Tissue to Optimize Photobiomodulation Prarameters in Alzheimer's Disease Mice.

The therapeutic potential of photobiomodulation (PBM) for Alzheimer's disease (AD) was evaluated by examining β-amyloid accumulation, microglial activation, and memory function. Mitochondrial delayed luminescence (m-DL), an indirect mitochondrial marker, was measured by irradiating 2 J/cm2 of 808 nm near-infrared light to the exposed brain surface of anesthetized 5XFAD mice. Based on m-DL findings, behavioral PBM was applied transcranially to the intact scalp using the same fluence at 30, 40, and 80 Hz with respective duty cycles and durations. The 80 Hz setting produced the longest m-DL decay

Anticancer drug-DNA interactions measured using a photoinduced electron-transfer mechanism based on luminescent quantum dots.

A sensing system based on the photoinduced electron transfer of quantum dots (QDs) was designed to measure the interaction of anticancer drug and DNA, taking mitoxantrone (MTX) as a model drug. MTX adsorbed on the surface of QDs can quench the photoluminescence (PL) of QDs through the photoinduced electron-transfer process; and then the addition of DNA will bring the restoration of QDs PL intensity, as DNA can bind with MTX and remove it from QDs. Sensitive detection of MTX with the detection limit of 10 nmol L(-1) and a linear detection range from 10 nmol L(-1) to 4.5 micromol L(-1) was achie

Bioluminescence Measurement of Time-Dependent Dynamic Changes of CYP-Mediated Cytotoxicity in CYP-Expressing Luminescent HepG2 Cells.

We sought to develop a cell-based cytotoxicity assay using human hepatocytes, which reflect the effects of drug-metabolizing enzymes on cytotoxicity. In this study, we generated luminescent human hepatoblastoma HepG2 cells using the mouse artificial chromosome vector, in which click beetle luciferase alone or luciferase and major drug-metabolizing enzymes (CYP2C9, CYP2C19, CYP2D6, and CYP3A4) are expressed, and monitored the time-dependent changes of CYP-mediated cytotoxicity expression by bioluminescence measurement. Real-time bioluminescence measurement revealed that compared with CYP-non-ex

A new measure of xenobiotic toxicity to the fist-line human defence system from the time-resolved phagocyte luminescence.

A new measure of toxicity based on stochastic modelling of single photon-counting processes, representing time-resolved phagocyte luminescence of xenobiotic-perturbed human neutrophils, has been constructed. The stochastic measure of toxicity has been verified by the QSAR method, and then compared and contrasted with the traditional toxicity measure used in bio- and chemiluminescent research. Phenol and benzene homologues were chosen as perturbers due to their importance from the viewpoint of ecotoxicity and occupational medicine.

The luminescence immunoassay S-100: a sensitive test to measure circulating S-100B: its prognostic value in malignant melanoma.

In this study we measured S-100B using a recently developed luminometric immunoassay with a detection limit of 0.02 microg l(-1). By measuring serum S-100B concentrations in 58 apparently healthy individuals a reference value of 0.16 microg l(-1) was found. To assess the sensitivity of the assay we measured levels of S-100B protein in the serum of 251 patients with cutaneous malignant melanoma before the start of treatment. Only one of 179 patients with limited disease had a serum concentration higher than the reference value, whereas elevated levels were seen in 79% of patients with metastasi

Development and Application in Stroke of an Aldehyde nanoparticle-based Amplified Luminescent Proximity Homogeneous Assay for Rapid Quantitative Measurement of Serum Copeptin.

The timing of onsets is a critical factor in preventing disability in stroking patients. Developing a faster and more accurate diagnostic method is essential. Copeptin, as a stable surrogate for arginine vasopressin, has emerged as a novel and reliable serum biomarker for stroke. Therefore, to facilitate prompt stroke detection, this work attempts to develop a quick and precise technique for identifying copeptin in human serum. For homogenous analysis, a novel amplified luminescence method based on aldehyde was created. This method employs the double antibody sandwich technique to detect copep

Comparison of M22-based ELISA and human-TSH-receptor-based luminescence assay for the measurement of thyrotropin receptor antibodies in patients with thyroid diseases.

Previously, a new procedure for measuring serum TSH receptor autoantibodies (TRAb) was reported in which the autoantibodies inhibit binding of a human monoclonal thyroid stimulating antibody M22 to TSHR-coated ELISA plate wells (TRAb ELISA). The aim of the present study was to evaluate the clinical performance of this assay in comparison to the second generation TRAb assay (TRAb LIA) based on the recombinant human TSH-receptor and chemiluminescence technology (TRAb LIA). Among the 158 patients, 84 patients suffered from Graves' disease (GD), 34 patients had Hashimoto's thyroiditis (HT), and 40

Total oxidant-scavenging capacities of plasma from glycogen storage disease type Ia patients as measured by cyclic voltammetry, FRAP and luminescence techniques.

It has been suggested that the very low incidence of atherosclerosis in glycogen storage disease type Ia (GSD Ia) subjects might be attributed to elevated levels of uric acid, one of the potent low molecular- weight antioxidants found in plasma. The present communication describes a use of two analytical methods-cyclic voltammetry and ferric reducing ability of plasma-and also two chemiluminescence methods to evaluate the total oxidant-scavenging capacities (TOSC) of plasma from GSD Ia patients. Our results verified the elevation of TOSC in GSD Ia patients and we propose the inclusion of lumin

A Novel Luminescence-Based Serum Bactericidal Assay for Vibrio cholerae Reduces Assay Variation, Is Time- and Cost-Effective, and Directly Measures Continuous Titer Values.

Cholera remains a significant public health burden worldwide, and better methods for monitoring cholera incidence would enhance the effectiveness of public health interventions. The serum bactericidal assay (SBA) has been used extensively for Vibrio cholerae vaccine assessments and serosurveillance. Current SBA approaches for V. cholerae rely on colony enumeration or optical density (OD600nm) readings to measure viable bacteria following complement-mediated lysis. These methods provide titer values that are constrained to discrete dilution values and rely on bacterial outgrowth, which is time
