Host immune response to lumpy skin disease virus.

Lumpy skin disease virus (LSDV) is a transboundary pathogen of cattle characterised by necrotic skin nodules, lymphadenopathy, and fever. First detected in southern Africa a century ago, it has continued to spread and cause significant welfare concerns and economic losses in Asia and, more recently, in Europe. LSDV replicates locally in the skin before dissemination via lymph and blood, causing vasculitis and characteristic cutaneous lesions. Localised innate immune responses lead to infiltration of immune cells in the skin; however, virally encoded proteins interfere with the innate response'

Isolation and Identification of Lumpy Skin Disease Virus.

Lumpy skin disease virus (LSDV), a member of the capripoxvirus genus, induces lumpy skin disease (LSD) in cattle and buffalo populations. The initial documentation of LSD dates back to 1929 in Zambia, with subsequent rapid dissemination within the cattle community in Africa and Asia. Described as the "smallpox" equivalent in cattle, LSD manifests through clinical features such as fever, extensive cutaneous nodules, body wasting, lymph node enlargement, and skin edema. Isolation of LSDV constitutes an important step in evaluating virulence and implementing disease control measures. Skin samples

Seminal transmission of lumpy skin disease virus in heifers.

It is known that lumpy skin disease virus (LSDV) can be shed in bull semen following infection and also that artificial insemination (AI) poses a biosecurity risk. However, it is not known whether the use of LSDV infected semen in AI poses a biosecurity risk. The aim of this study was to investigate whether LSDV, transmitted through semen, can infect cows and their embryos. Two controlled trials were performed simultaneously. Eleven young beef heifers, naïve to LSDV, were synchronized using an OvSynch protocol and inseminated on Day 0 with fresh semen spiked with a field strain of LSDV on day

Non-vector-borne transmission of lumpy skin disease virus.

The transmission of "lumpy skin disease virus" (LSDV) has prompted intensive research efforts due to the rapid spread and high impact of the disease in recent years, especially in Eastern Europe and Balkan countries. In this study, we experimentally evaluate the vaccine-derived virulent recombinant LSDV strain (Saratov/2017) and provide solid evidence on the capacity of the virus for transmission in a vector-proof environment. In the 60-day long experiment, we used inoculated bulls (IN group) and two groups of in-contact animals (C1 and C2), with the former (C1) being in contact with the inocu

Alternative cell lines for the propagation of lumpy skin disease virus.

In our Institute lumpy skin disease virus is grown on primary lamb testis cells for isolation, identification and vaccine production. However, the availability of lambs in Kenya has been seriously reduced over the past few years. This has led to an increase in the cost of using primary lamb testis cells. This study was undertaken to investigate other primary cell lines, which are easily available and provide an equivalent or better yield of lumpy skin disease virus. Foetal bovine muscle (FBM) cells were found to be an adequate alternative for lamb testis cells.

Pathogenicity and virulence of lumpy skin disease virus: A comprehensive update.

Lumpy skin disease (LSD), which was confined to the Africa for many decades, has expanded its geographical distribution to numerous countries across Asia and Europe in recent years. The LSD virus (LSDV) is a relatively poorly studied virus. Its 151 Kb genome encodes 156 open reading frames (ORF); however, the exact number of the proteins encoded by the viral genome and their specific functions remain largely unknown. Arthropod vectors primarily transmit the LSDV mechanically, but the precise nature of these vectors in different regions and their role in transmission is not fully understood. Ho

Characterization of bovine kidney cells infected with lumpy skin disease virus.

Lumpy skin disease virus (LSDV) infection can cause systemic infection in cattle, accompanied by widespread viral dissemination in the skin, respiratory tract, lymphatic tissues, and visceral organs. However, the replication characteristics of the virus and the mechanisms by which it causes damage to host cells remain unclear. In this study, the ultrastructure of LSDV and the expression profile of host immune genes were investigated. The results showed that LSDV replicated efficiently in MDBK cells and induced cellular lesions, with the viral titer reaching approximately 1 × 10⁷ copies/mL. The

Molecular characterization of lumpy skin disease virus in Iran (2014-2018).

Lumpy skin disease was first reported in the western provinces of Iran in 2014, and this was followed by several outbreaks throughout the country. In this study, 10 Iranian lumpy skin disease virus (LSDV) samples collected during the period of 2014-2018 were characterized by sequence analysis of the GPCR, LSDV142, and IL10LP genes. Sequence comparison of the respective genes revealed a close relationship between Iranian LSDV isolates and viruses from Asia and Europe. Interestingly, some nucleotide sequence diversity was also observed in the IL10LP genes of the Iranian field isolates.

Cell death modulation dictates tissue-specific tropism of lumpy skin disease virus.

Lumpy skin disease virus (LSDV) is a critical transboundary pathogen that causes devastating infections in cattle, buffalo, and other ruminants. The virus induces characteristic clinical manifestations, including cutaneous nodules, marked reduction in milk yield, and impaired production performance, leading to severe economic losses in the global livestock sector. Although LSDV exhibits remarkable multi-tissue tropism and persistent viral shedding in various organs, posing significant challenges for disease control, the molecular mechanisms underlying its tissue-specific adaptation remain poor

A high-quality telomere-to-telomere lumpy skin disease virus genome assembly.

Lumpy skin disease virus (LSDV) is an emerging livestock capripoxvirus (CaPV) that continues to cause substantial economic losses across Africa, Asia and Europe. However, important uncertainties remain regarding LSDV genome structure, particularly at the telomeric inverted terminal repeats (ITRs) that are central to host interaction, replication and adaptive evolution. Structural variation at the ITRs is driven by recombination during virus replication. Previous assemblies have relied primarily on short-read sequencing, which provides limited resolution of repeat-rich telomeric regions and pro

Preparation and immune efficacy evaluation of inactivated lumpy skin disease virus vaccine.

Lumpy Skin Disease (LSD), historically restricted to Africa, has in recent years exhibited a marked expansion into several Asian and European countries, resulting in substantial economic impacts on the global cattle industry. In August 2019, the first confirmed outbreak of LSD was documented in the Xinjiang region of China. In response to this emerging threat, our study established primary, basic, and working seed banks utilizing the LSDV/MZGD/2020 strain, which was isolated from Meizhou, Guangdong. These seed banks underwent rigorous validation via sterility assays and evaluation of prolifera

LSDVvac: An immunoinformatics database for vaccine design against lumpy skin disease virus.

Development of an effective vaccine against Lumpy Skin Disease Virus (LSDV) is crucial for protecting livestock. The current study outlines a web-based platform developed to aid the scientific community in designing effective peptide-based vaccines against LSDV. First, we generated all possible overlapping (K-mer value 9 and 15) peptides from the proteins of 73 LSDV strains. Second, after removing redundancy, the obtained peptides were utilized for predicting B-cell and T-cell epitopes. Third, the predicted B-cell and T-cell epitopes were screened for immunogenicity, allergenicity, and toxicit

miRNA profiling of primary lamb testicle cells infected with lumpy skin disease virus.

In this study, miRNA profiling of cells infected with lumpy skin disease virus (LSDV) was conducted for the first time. When compared to mock-infected cells, LSDV-infected primary lamb testicle (LT) cells showed dysregulation of 64, 85, and 85 miRNAs at 12 hours postinfection (hpi), 48 hpi, and 72 hpi, respectively. While some of these miRNAs were found to be dysregulated at a particular time point following LSDV infection, others were dysregulated at all three time points. Analysis of the differentially expressed miRNA-mRNA interaction networks, Gene Ontology analysis of the predicted targets

Construction and Biological Characterization of ORF133-Deletion Mutant of Lumpy Skin Disease Virus.

Lumpy skin disease virus (LSDV), a Capripoxvirus genus member, causes severe cattle disease. Though Capripoxviruses share high nucleotide sequence homology indicating common ancestry, they have evolved distinct host adaptations. The LSDV genome encodes numerous proteins, with ORF133 being LSDV-specific and lacking clear homologs in other Capripoxviruses, implying potential roles in host range and virulence. To explore ORF133's function, this study generated mouse polyclonal antibodies against ORF133 and constructed the ORF133-deleted recombinant virus (LSDVΔORF133-EGFP) via homologous recombin

Construction and characterization of the ORF131 gene deletion strain of lumpy skin disease virus.

Lumpy Skin Disease (LSD), caused by the Lumpy Skin Disease Virus (LSDV), is a legally reportable disease recognized by the World Organization for Animal Health (WOAH) and has resulted in significant economic losses for the global cattle industry. Although several commercial LSDV vaccines are currently available, safer and more effective gene-deleted versions remain lacking. Therefore, screening key functional genes and developing gene-deleted live-attenuated vaccine strains hold substantial research value. In this study, we focused on ORF131, a gene whose function remains unclear. We successfu

Estimating evolutionary changes between highly passaged and original parental lumpy skin disease virus strains.

Research into the phylogenetic relationships of lumpy skin disease virus (LSDV) strains was long overlooked, partially due to its original restricted distribution to sub-Saharan Africa. However, recent incursions into northern latitudes, and a rapid spread causing major economic losses worldwide, have intensified additional research on the disease and the causative virus. This study delineates the phylogeny of LSDV in the context of full genome sequences of strains recovered in the field, as well as strains highly passaged in cell culture. We sequenced the oldest known field strain to date (is
