Solubility enhancement studies on lurasidone hydrochloride using mixed hydrotropy.

Low aqueous solubility is a major problem faced during formulation development of new drug molecules. Lurasidone HCl (LRD) is an antipsychotic agent specially used in the treatments of schizophrenia and is a good example of the problems associated with low aqueous solubility. Lurasidone is practically insoluble in water, has poor bioavailability and slow onset of action and therefore cannot be given in emergency clinical situations like schizophrenia. Hence, purpose of this research was to provide a fast dissolving oral dosage form of Lurasidone. This dosage form can provide quick onset of act

Effects of Temperature and Ionic Strength of Dissolution Medium on the Gelation of Amorphous Lurasidone Hydrochloride.

PURPOSE: Amorphous lurasidone hydrochloride (LH) showed decreased dissolution behavior in comparison to crystalline LH owing to gelation during dissolution as reported in our previous study. The current study aims to investigate external factors including temperature and ionic strength on the gelation and hence the dissolution of amorphous LH. METHODS: Dissolution tests of amorphous LH were performed under different temperatures and buffer ionic strengths. The formed gels were characterized by rheology study, texture analysis, PLM, SEM, DSC, XRPD and FTIR. RESULTS: With the increase of tempe

Charge-assisted bond N+H mediates the gelation of amorphous lurasidone hydrochloride during dissolution.

Lurasidone hydrochloride (LH), the hydrochloride form of lurasidone with a charge-assisted bond N+H, is an atypical antipsychotropic agent for the treatment of schizophrenia. As a BCS class II drug, LH has a low oral bioavailability mainly due to its poor water solubility and low dissolution. In order to improve its solubility, amorphization of LH was performed and characterized. Unexpectedly, the dissolution rate of amorphous LH was much lower than that of crystalline LH. In addition, the amorphous LH powders quickly aggregated when contacting the dissolution media (water, 37°C), and formed a

Lurasidone hydrochloride-loaded microparticle-based long-acting injectable: formulation optimisation, characterisation, and payload release kinetics-a proof of concept study.

Lurasidone hydrochloride is a promising antipsychotic agent. The currently marketed formulation (Latuda®) of lurasidone hydrochloride exhibits extremely poor bioavailability and requires daily dosing. Current research focuses on the development and optimisation of a lurasidone hydrochloride-loaded poly(lactic-co-glycolic acid) microparticle-based long-acting depot formulation. The optimised batch of microparticles showed 32.81 ± 1.46 µm particle size, 95.74 ± 3.19% an entrapment efficiency, and a drug loading of 27.68 ± 0.95%. Assessment of Nitrosamine Impurities in Lurasidone Hydrochloride Using APCI-LC-MS/MS in MRM Mode for Regulatory Compliance and Stability Evaluation.

RATIONALE: Nitrosamine impurities, such as N-nitroso diethylamine (NDEA), N-nitroso dimethylamine (NDMA), and 1-nitroso piperazine (1-NP), have raised serious health concerns due to their carcinogenic and mutagenic properties. The pharmaceutical industry faces increasing regulatory pressure to monitor these compounds at trace levels. This study addresses the need for a robust analytical method to ensure the safety of lurasidone hydrochloride (LUR), an antipsychotic drug. METHODS: A sensitive and specific LC-MS/MS method was developed using atmospheric pressure chemical ionization (APCI) in po

Systematic literature review and network meta-analysis of lurasidone, brexpiprazole and cariprazine for schizophrenia.

A systematic review was undertaken to identify randomized controlled trials (RCTs) comparing the efficacy and safety of lurasidone, brexpiprazole and cariprazine (selected because of a shared safety profile) with each other or placebo in adult patients with schizophrenia. Key outcomes included: Positive and Negative Syndrome Scales (PANSS), Clinical Global Impression-Severity (CGI-S) scores and cardiovascular and metabolic parameters. A feasibility assessment evaluated the trials' suitability for inclusion in a Bayesian network meta-analysis (NMA). Random effects models were used. In total, 11

Factors associated with increased risk of lurasidone-induced somnolence: Two case-control studies based on one bioequivalence trial in healthy volunteers.

Somnolence is a common adverse effect of antipsychotic drugs used to treat psychotic disorders. It causes problems in many areas of life, such as gainful employment, driving, childcare, and social interactions. Somnolence is a major problem for a relatively new antipsychotic drug, lurasidone, whose dose-effect relationship remains unclear. Based on data from a bioequivalence study of two 40 mg lurasidone hydrochloride tablets, we designed two case-control studies to explore the correlation between somnolence and exposure to lurasidone and determine the factors associated with lurasidone-induce

Bilateral pedal edema associated with lurasidone: a case report.

Lurasidone, an atypical antipsychotic, is currently approved for treatment of schizophrenia and bipolar depression. Little is known about whether lurasidone might also cause pedal edema. A 55-year-old female patient had been diagnosed with bipolar I disorder from the age of 26. She had been prescribed escitalopram and quetiapine 300 mg/day for her persistent depressive mood. Later, she took lurasidone plus escitalopram to treat depressive episode. Consequently, she developed mild bilateral swelling over the lower legs and ankles. After lurasidone was discontinued, the bilateral pedal edema was

Lurasidone (latuda®): an atypical antipsychotic.

Lurasidone is a new atypical antipsychotic that has demonstrated positive effects on psychosis, mood, and cognition. This improved efficacy and safety profile for the treatment of schizophrenia. Its overall tolerability profile seems to be comparable to the other atypical antipsychotics. Perhaps its more potent blockade on the 5HT7 receptor will give it more of an advantage in treating the negative symptoms as well as improve cognitive and depressive symptoms associated with schizophrenia. Additionally, it does not appear to have any significant adverse impact on the metabolic profile, such as

[Long-term efficacy and safety of lurasidone in the treatment of schizophrenia].

Lurasidone is a new second-generation antipsychotic approved in March 2014 by the European Medicines Agency for the treatment of schizophrenia. Lurasidone has demonstrated its efficacy in long-term studies. It has been shown to reduce significantly the risk of relapse in comparison with placebo in patients with schizophrenia. In comparator study, lurasidone was noninferior to quetiapine XR in risk for relapse. In open-label studies, lurasidone was associated with sustained improvement in efficacy measures observed and well-tolerated inpatients with schizophrenia who had switched to lurasidone

Lurasidone for schizophrenia.

Lurasidone (Latuda-Sunovion) is a new oral second-generation antipsychotic licensed for treating adults with schizophrenia. Although less likely to cause extrapyramidal adverse effects than first-generation antipsychotics, some second-generation antipsychotics cause weight gain that may increase the risk of diabetes, hypertension and raised lipid levels.1,2 Lurasidone is promoted as offering a balance between efficacy and tolerability, with reported negligible effects on weight and minimal effects on glucose and cholesterol.3 Here we review evidence for lurasidone's efficacy and safety, and co
