Luteolin and hormesis.

The present paper provides the first integrated assessment of the capacity of luteolin to induce hormetic dose responses. It was shown that luteolin induced hormetic responses in multiple biological systems, including enhancing neuroprotection in various experimental model disease systems, improving wounding healing, especially in experimental models of high-risk population subgroups, such as diabetics, as well as enhancing osteogenesis in models of osteoporosis. The mechanistic basis for the luteolin-induced hormetic dose responses has been demonstrated to commonly involve the upregulation of

Intestinal absorption of luteolin and luteolin 7-O-beta-glucoside in rats and humans.

In this study, we investigated the intestinal absorption of luteolin and luteolin 7-O-beta-glucoside in rats by HPLC. The absorption analysis using rat everted small intestine demonstrated that luteolin was converted to glucuronides during passing through the intestinal mucosa and that luteolin 7-O-beta-glucoside was absorbed after hydrolysis to luteolin. Free luteolin, its conjugates and methylated conjugates were present in rat plasma after dosing. This suggests that some luteolin can escape the intestinal conjugation and the hepatic sulfation/methylation. LC/MS analysis showed that the main

Luteolin for neurodegenerative diseases: a review.

Neurodegenerative diseases such as Parkinson's disease, Alzheimer's disease, Huntington's disease, and multiple sclerosis affect millions of people around the world. In addition to age, which is a key factor contributing to the development of all neurodegenerative diseases, genetic and environmental components are also important risk factors. Current methods of treating neurodegenerative diseases are mostly symptomatic and do not eliminate the cause of the disease. Many studies focus on searching for natural substances with neuroprotective properties that could be used as an adjuvant therapy i

[Pharmacodynamics of luteolin-histamine and receptors response].

The isolated guinea pig ileum were used to investigate the pharmacodynamics of the relationship between luteolin and histamine dose response. The pharmacodynamics parameters were estimated by various methods. The values of the pA2, the slope and the pD'2 were 5.6, 0.6 and 4.4, respectively. The dissociation constants and the maximal response were estimated by the Lineweaver-Burk, Hanes-Woolf, Scatchard or Woolf methods. The estimated values of the parameters were: EAmax not equal to EABmax, KA not equal to alpha KA, KB not equal to alpha KB. The linear intersecting points of the plotting by th

Luteolin as a modulator of skin aging and inflammation.

Luteolin belongs to the group of flavonoids and can be found in flowers, herbs, vegetables and spices. It plays an important role in defending plants, for example against UV radiation by partially absorbing UVA and UVB radiation. Thus, luteolin can also decrease adverse photobiological effects in the skin by acting as a first line of defense. Furthermore, anti-oxidative and anti-inflammatory activities of luteolin were described on keratinocytes and fibroblasts as well as on several immune cells (e.g., macrophages, mast cell, neutrophils, dendritic cells and T cells). Luteolin can suppress pro

Degradation of quercetin and luteolin by Eubacterium ramulus.

The degradation of the flavonol quercetin and the flavone luteolin by Eubacterium ramulus, a strict anaerobe of the human intestinal tract, was studied. Resting cells converted these flavonoids to 3,4-dihydroxyphenylacetic acid and 3-(3,4-dihydroxyphenyl)propionic acid, respectively. The conversion of quercetin was accompanied by the transient formation of two intermediates, one of which was identified as taxifolin based on its specific retention time and UV and mass spectra. The structure of the second intermediate, alphitonin, was additionally elucidated by (1)H and (13)C nuclear magnetic re

Luteolin inhibits CVB3 replication through inhibiting inflammation.

Coxsackievirus B3 (CVB3) infection causes many inflammation-related diseases, such as viral myocarditis and aseptic meningitis. However, no vaccines or drugs have been approved for prevention or therapy of CVB3-induced diseases. In this study, luteolin (3,4,5,7-tetrahydroxyflavone) had been found that could dose-dependently reduce the production of viral progeny and synthesis of CVB3 RNA and protein. The luteolin-mediated inhibition of CVB3 was found to be mechanistically possible, at least in part, through depressing the phosphorylation of p38 MAPK and JNK MAPK, and inhibiting NF-κB nuclear t

The flavonoid luteolin inhibits niacin-induced flush.

BACKGROUND AND PURPOSE: Sustained release niacin effectively lowers serum cholesterol, LDL and triglycerides, while raising HDL. However, 75% of patients experience cutaneous warmth and itching known as flush, leading to discontinuation. Acetylsalicylic acid (aspirin) reduces this flush only by about 30%, presumably through decreasing prostaglandin D2 (PGD2). We investigated whether niacin-induced flush in a rat model involves PGD2 and 5-HT, and the effect of certain flavonoids. EXPERIMENTAL APPROACH: Three skin temperature measurements from each ear were recorded with an infrared pyrometer f

Effects of luteolin and luteolin-morphine co-administration on acute and chronic pain and sciatic nerve ligated-induced neuropathy in mice.

Background Neuropathic pain (NP) is a common condition accompanied by nerve injury. To date, there is no definite treatment approved for this disorder. In addition, many drugs that are used for NP cause adverse reactions. Luteolin is a naturally occurring flavonoid with diverse pharmacological properties such as anti-inflammatory, antioxidant and anticancer. We sought to investigate luteolin effects on chronic, acute and neuropathic pain as well as its potential to increase morphine anti-nociceptive effects in mice. Methods Albino mice (20-25 g) were randomly divided into 14 groups (n=7) inclu

Luteolin Alleviates Axitinib-Induced Cardiomyocyte Apoptosis in Mice.

Axitinib, a tyrosine kinase inhibitor (TKI), is associated with a risk of cardiotoxicity in clinical practice. However, its underlying toxicological mechanisms remain unclear, and effective cardioprotective strategies are lacking. This study aims to investigate the mechanisms of axitinib-induced cardiac injury and evaluate the protective effects of luteolin. In this study, C57BL/6 mice were randomly divided into four groups: Control, Axitinib, Axitinib + Luteolin, and Luteolin alone. Ventricular systolic function (EF/FS/LVEDD/LVESD) was evaluated by echocardiography. Serum cardiac troponin T (

Luteolin ameliorates loperamide-induced functional constipation in mice.

Functional constipation (FC) is one of the most common gastrointestinal disorders characterized by hard stools and infrequent bowel movements, which is associated with dysfunction of the enteric nervous system and intestinal motility. Luteolin, a naturally occurring flavone, was reported to possess potential pharmacological activities on intestinal inflammation and nerve injury. This study aimed to explore the role of luteolin and its functional mechanism in loperamide-induced FC mice. Our results showed that luteolin treatment reversed the reduction in defecation frequency, fecal water conten

Deglucuronidation of a flavonoid, luteolin monoglucuronide, during inflammation.

In this study, we investigated whether luteolin monoglucuronide was converted to free aglycone during inflammation using human neutrophils stimulated with ionomycin/cytochalasin B and rats treated with lipopolysaccharide (LPS). beta-Glucuronidase activity was assayed using 4-methylumbelliferyl-glucuronide and methanol extracts of rat plasma containing luteolin monoglucuronide. The released 4-methylumbelliferone, a fluorescent molecule, was quantified by fluorometry. Deglucuronidation of luteolin monoglucuronide was examined by high-performance liquid chromatography (HPLC) analysis. HPLC analys

Mechanistic Insights into the Neuroprotective Potential of Luteolin: An Overview.

Neurodegenerative disorders, such as Alzheimer's and Parkinson's diseases, are characterized by pathologies such as oxidative stress, neuroinflammation, mitochondrial dysfunction, and protein aggregation. Luteolin, a natural flavonoid, is known for its neuroprotective properties, targeting key pathological mechanisms. This review highlights the multi-target effects of luteolin, as demonstrated by in vitro and in vivo studies. Luteolin reduces oxidative damage by scavenging ROS, enhancing antioxidant enzymes, such as SOD, CAT, and GPx, and activating the Nrf2/ARE pathway. It reduces neuroinflam

Luteolin is an excellent antioxidant found in a wide variety of natural foods, such as honey and pollen. In this work, the effect of the surrounding environments on the structure and antioxidative activity of luteolin was carried out using density functional theory (DFT) calculation. The studied environments are gas, benzene, chloroform, pyridine, acetonitrile, ethanol, DMSO, and water. The structure of the luteolin monomer in different environments was optimized. The hydrogen-bond was especially focused, and the antioxidative capacity of luteolin was analyzed from the thermodynamic aspect. It

Luteolin: a natural product with multiple mechanisms for atherosclerosis.

Atherosclerosis (AS) is a degenerative and proliferative disease characterised by the deposition of lipid and/or fibrous substances within the intima of arteries. The pathological mechanisms of AS involve endothelial cell (EC) injury and dysfunction, vascular smooth muscle cell (VSMC) migration and proliferation, foam cell formation, inflammatory cell recruitment, and abnormal platelet activation and aggregation. In recent years, the incidence and mortality rates of atherosclerotic cardiovascular disease (ASCVD), which has AS as its pathological basis, have shown an upward trend globally. Curr
