Lycoris species have been known since long ago as a multi-utility ethnomedicinal herbal in China. It has been reported to exhibit a number of properties such as anticancer, neuroprotective, and antibacterial activities. In the present study, the anticancer efficacy of dichloromethane extracts of Lycoris aurea (DELA), was evaluated both in vivo and in vitro using murine sarcoma 180 cells. To evaluate the effects of DELA on apoptotic cell death, flow cytometry and Western blotting were performed. DELA demonstrated promising inhibition effects on sarcoma 180 cells in vitro and a 53.49% inhibitory

Lycoris radiata is the main source of galanthamine, a clinical drug used in Alzheimer's disease; however, the galanthamine content in L. radiata is low. Lycoris aurea is another Lycoris species with high galanthamine content. Fungal endophytes can enhance plant secondary metabolite accumulation; thus, we compared the fungal communities in these two Lycoris species to identify certain fungal taxa in L. aurea capable of enhancing galanthamine accumulation. Several fungal endophytes, which were enriched in, exclusively isolated from L. aurea, or showed significant correlations with galanthamine,

Lycoris radiata, commonly known to contain alkaloids such as lycorine and galanthamine, is toxic and is easily accessible in East Asian countries, including Japan. Lycorine is a central nervous system and gastrointestinal irritant, whereas galanthamine, a carbamate, exhibits reversible acetylcholinesterase inhibition. Symptoms of Lycoris radiata poisoning are generally acute and resolve quickly. We present a case of an 83-year-old woman with impaired consciousness that presented to our hospital. The patient's presentation initially was not significant; however, a rapid progression of symptoms

LrPAL is a novel full-length cDNA isolated from Lycoris radiata by degenerate oligonucleotide primer PCR (DOP-PCR), 3'- and 5'-RACE approaches, harbours an open reading frame (ORF) encoding a 708 amino acid product. Sequence alignment showed that the deduced amino acid sequence of LrPAL shared more than 80% identity with other PAL sequences reported in Arabidopsis thaliana and other plants. RT-PCR revealed that LrPAL transcripts were higher in bud flowers and wilting flowers (5 days after blooming) than in blooming flowers. The transcript levels of LrPAL in leaves were significantly induced by

Heat stress (HS) has become an increasingly severe constraint on plant growth, quality and yield under global warming. However, the thermotolerance mechanisms of Lycoris radiata (L. radiata) remain underexplored. Here, we found that high temperature (42°C) induced rapid leaf wilting and rolling L. radiata leaves, accompanied by decreased chlorophyll content, reduced Fv/Fm and relative water content, and increased membrane damage and ROS accumulation. Further transcriptome analysis revealed significant enrichment the term of "response to heat", and we mainly focused on DREB-A2 transcription fac

Vasodepressor mechanism of lycorenine (an alkaloid of Lycoris radiata Herb.) was investigated in anesthetized rats. Lycorenine (1--10 mg/kg i.v.) produced dose-related decreases in blood pressure and heart rate and tachyphylaxis developed with repeated injections. In the blood-perfused rat hindquarters, lycorenine (62.5--500 micrograms i.a.) produced dose-related decreases both in mean blood pressure and in perfusion pressure, and the lycorenine-induced decrease in perfusion pressure was abolished by phenoxybenzamine or hexamethonium. Lycorenine (more than 1 mg/kg i.v.) blocked the pressor res

Fungal infections not only cause extensive agricultural damage but also result in serious diseases in the immunodeficient populations of human beings. Moreover, the increasing emergence of drug resistance has led to a decrease in the efficacy of current antifungals. Thus, screening of new antifungal agents is imperative in the fight against antifungal drug resistance. In this study, we show that an endophytic bacterium, Burkholderia gladioli HDXY-02, isolated from the medicinal plant Lycoris aurea, showed broad-spectrum antifungal activity against plant and human fungal pathogens. An antifunga

ETHNOPHARMACOLOGICAL RELEVANCE: The bulb of Lycoris radiata (L'Hér.) Herb. (Amaryllidaceae) has been widely utilized in Traditional Chinese Medicine (TCM) for its properties of "clearing heat and relieving toxicity" (Qing Re Jie Du) and "eliminating phlegm" (Qu Tan), and is frequently prescribed for throat inflammation, abscesses, and ulcers. Although lycorine is known as the main active alkaloid, its specific activity against the avian respiratory coronavirus, infectious bronchitis virus (IBV), remains unexplored, despite the plant's long-standing use for respiratory ailments.

Depressive amnesia, involving memory impairment and mood dysregulation, frequently co-occurs with depression and neurodegenerative diseases. Methylglyoxal (MGO), a reactive glycolytic byproduct, contributes to depressive-like behaviors and cognitive deficits. This study evaluated the therapeutic potential of 2',4',6'-trimethoxyacetophenone (TMA), a bioactive compound from Lycoris sanguinea var. koreana, in a mouse model of MGO-induced depressive amnesia. Mice received MGO (60 mg/kg) followed by TMA (5 or 20 mg/kg), and behavioral tests were conducted to assess mood, cognition, and locomotor ac

The flowers of Lycoris longituba are distinguished by their fragrant petals and dynamic colour variations. Floral volatile compounds play vital roles in plant biology; however, the biosynthetic pathways and regulatory mechanisms underlying fragrance production in L. longituba remain inadequately understood. In this study, metabolomic and transcriptomic analyses were integrated to characterize the variation of 526 volatile organic compounds (VOCs) in L. longituba florets. Metabolite profiling revealed that terpenoids constituted the predominant class of VOCs, while sweet and green aromas were t

Plants of the Amaryllidaceae family have been used as therapeutic agents against CNS related maladies such as Alzheimer's disease. The known primary alkaloid constituents have significant biological activity. We identified the Lycoris alkaloids lycorine and lycoricidinol from Amaryllidaceae using a real-time reporter gene assay system based on NIH3T3 cells. These alkaloids have a wide spectrum of pharmacological actions and dose-dependently lengthen the circadian period. When cells that had been incubated with lycorine or lycoricidinol were washed and then incubated without these alkaloids, pe

Galanthamine, an acetylcholinesterase inhibitor marketed as a hydrobromide salt (Razadyne®, Reminyl®) for the treatment of Alzheimer's disease (AD), is obtained from Amaryllidaceae plants, especially those belonging to the genera Leucojum, Narcissus, Lycoris and Ungernia. The growing demand for galanthamine has prompted searches for new sources of this compound, as well as other bioactive alkaloids for the treatment of AD. In this paper we report the isolation of the new alkaloid 11β-hydroxygalanthamine, an epimer of the previously isolated alkaloid habranthine, which was identified using NMR

Cholestatic liver disease (CLD) is defined by impaired bile synthesis and excretion, which in turn results in progressive liver injury or fibrosis, however, there are currently no effective therapeutic drugs. Lycorine is an alkaloid isolated from Lycoris bulbs, exhibiting antiviral, anti-inflammatory, analgesic, and cardioprotective properties. However, there has been no reports concerning its effects on CLD so far. We assessed the hepatoprotective potential of lycorine on intrahepatic cholestasis mouse models induced by α-naphthyl isothiocyanate (ANIT) and multidrug resistance protein 2 knock

Glioblastoma multiforme (GBM) is the highest grade of glioma. Tumours, including GBM, possess reprogrammed metabolism, such as altered aerobic glycolysis and aberrant energy production. Lycorine hydrochloride (LH) was extracted from the bulb of Lycoris radiata. The previous study indicated that LH exerts antiviral, anti-inflammatory and antitumour effects. However, the effect of LH on GBM and the underlying molecular mechanism remain unclear. Our study revealed that LH restrained chemoresistant GBM cells growth by inhibiting PDK3 expression in vitro and in vivo. Functionally, LH inhibited the

BACKGROUND: Lycorine hydrochloride (LH), an alkaloid extracted from the bulb of the Lycoris radiata, is considered to have anti-viral, anti-malarial, and anti-tumorous effects. At present, the underlying mechanisms of LH in gastric cancer remain unclear. MCL1, an anti-apoptotic protein of BCL2 family, is closely related to drug resistance of tumor. Therefore, MCL1 is considered as a potential target for cancer treatment.
METHODS: The effect of LH on gastric cancer was assessed in vitro (by MTT, BrdU, western blotting…) and in vivo (by immunohistochemistry).
RESULTS: In this study, we showed

Ovarian cancer remains the most lethal gynecological malignancy worldwide, largely owing to poor prognosis associated with chemotherapy resistance and cancer stem cell-driven recurrence. This study comprehensively investigates the antitumor effects of lycorine, a natural alkaloid derived from Lycoris radiata, in human ovarian cancer models. Our results demonstrate that lycorine significantly inhibits ovarian cancer cell proliferation, induces apoptosis, and suppresses cancer stemness through modulation of the PI3K/AKT signaling pathway. In vitro, lycorine treatment reduced the expression of st
