Lysinoalanine crosslinking in the extracellular flagellar hook of Synergistota.

UNLABELLED: Bacterial motility is driven by the flagellar motor, a conserved macromolecular complex that enables cells to navigate chemical gradients. In spirochetes, motility depends on periplasmic flagella whose function is supported by a unique post-translational modification (PTM) in the hook protein FlgE: a covalent lysinoalanine (Lal) crosslink between adjacent subunits. This modification has been shown to be conserved across the phylum and is required for motility. In spirochetes, Lal is thought to provide additional hook stability needed to meet the demands of flagellar rotation in the

Structure and chemistry of lysinoalanine crosslinking in the spirochaete flagella hook.

The flagellar hook protein FlgE from spirochaete bacteria self-catalyzes the formation of an unusual inter-subunit lysinoalanine (Lal) crosslink that is critical for cell motility. Unlike other known examples of Lal biosynthesis, conserved cysteine and lysine residues in FlgE spontaneously react to form Lal without the involvement of additional enzymes. Oligomerization of FlgE via its D0 and Dc domains drives assembly of the crosslinking site at the D1-D2 domain interface. Structures of the FlgED2 domain, dehydroalanine (DHA) intermediate and Lal crosslinked FlgE subunits reveal successive sna

Inhibitors of lysinoalanine crosslinking in the flagella hook as antimicrobials against spirochetes.

Spirochetes are especially invasive bacteria that are responsible for several human diseases, including Lyme disease, periodontal disease, syphilis and leptospirosis. The flexible hook of these PFs contains a post-translational modification in the form of a lysinoalanine (Lal) crosslink between adjacent subunits of FlgE, which primarily comprise the hook. Spirochaete flagella hook proteins self-catalyse a lysinoalanine covalent crosslink for motility.

Spirochaetes are bacteria responsible for several serious diseases, including Lyme disease (Borrelia burgdorferi), syphilis (Treponema pallidum) and leptospirosis (Leptospira interrogans), and contribute to periodontal diseases (Treponema denticola)(1). These spirochaetes employ an unusual form of flagella-based motility necessary for pathogenicity; indeed, spirochaete flagella (periplasmic flagella) reside and rotate within the periplasmic space(2-11). The universal joint or hook that links the rotary motor to the filament is composed of ∼120-130 FlgE proteins, which in spirochaetes form an u

Dual-Frequency Ultrasound-Assisted Glycation Modulates Beef Myofibrillar Protein Gels and Inhibits Lysinoalanine Formation.

Ultrasound-assisted xylose glycation is found to be an effective method to improve the gel properties, functional properties and structural characteristics of meat proteins, and to some extent minimize the generation of protein cross-linking. The effects of dual-frequency ultrasound-assisted (20/25, 20/28, 25/28, 20/40, 25/40, 28/40, 28/68, and 40/68 kHz) xylose glycation on beef myofibrillar proteins (MPs) were evaluated in the study. Compared with the control, dual-frequency ultrasound promoted glycation in a frequency-dependent manner. The highest grafting degree was observed at 20/25 and 2

Phenolic Acids Inhibiting Lysinoalanine Cross-Linking in Soybean Protein Isolate via Covalent Site Blocking and Conformational Stabilization.

Alkali-heat treatment is widely used to functionalize protein-based food materials but inevitably induces lysinoalanine (LAL), a harmful nonnatural cross-linked amino acid. Herein, p-hydroxybenzoic acid, 3,5-dihydroxybenzoic acid, and gallic acid were covalently grafted onto alkali-heat-treated soy protein isolate (SPI) models to elucidate the topology-dependent inhibition of LAL formation. The inhibitory effect increased with the hydroxyl number, with gallic acid showing the highest inhibition rate (68.25%). Covalent conjugation stabilized SPI conformation by reducing reactive-site exposure,

An ELISA for discovering protein-protein interaction inhibitors: blocking lysinoalanine crosslinking between subunits of the spirochete flagellar hook as a test case.

The inhibition of a specific protein-protein interaction is often difficult to achieve in targeted drug design. We report the development and optimization of a general-purpose, readily implemented enzyme-linked immunosorbent assay (ELISA) for high-throughput screening to identify small- molecule inhibitors of protein interactions. This ELISA does not involve the use of any capture antibodies, probes, or compounds coated on the plate and represents a general strategy to identify inhibitors of a given protein-protein interaction. We demonstrate its utility in blocking lysinoalanine crosslinking

Mechanistic basis for inhibition of the extended-spectrum β-lactamase GES-1 by enmetazobactam and tazobactam.

β-Lactamase-catalysed hydrolysis is the primary form of β-lactam antibiotic resistance in Gram-negative bacteria. The penicillanic acid sulfone (PAS) enmetazobactam is thought to inhibit extended-spectrum β-lactamases (ESBLs) by fragmentation of an initial acyl-enzyme to form an active-site lysinoalanine cross link. We investigate interactions of enmetazobactam and its congener tazobactam with GES-1, an ESBL with structural features of carbapenem-hydrolysing β-lactamases. Crystal structures show different breakdown products of the two inhibitors covalently bound to the catalytic Ser70, assigne

Protein reactions during food processing and storage--their relevance to human nutrition.

The main reactions of protein components during food processing and storage were presented and discussed. Since most data are available from the reactions of lysine with other food components these results are mainly demonstrated. Using furosine as indicator the formation and presence of the early Maillard products fructoselysine, lactuloselysine or maltuloselysine in several food systems was measured. Also lysinoalanine is mentioned. A further, recently detected compound, N-epsilon-carboxymethyllysine, appears to be also of biological and technological interest. Several foods contained up to

Nonenzyme browning and its effect on protein nutrition.

The nonenzyme browning involves the thermal decomposition of sugars, the caramelization, the decomposition of oxi-acids, the so called "Maillard reaction" between amino acids and carbohydrates, the reaction between oxidized fats and proteins, and those alterations which take place by the alkaline treatment of proteins. The Maillard reaction is of secondary importance in the case of foodstuffs and fodders with low carbohydrate contents (meats, meat meal, fish meal). By the heat treatment, the sulphur-containing amino acids of proteins (cystine, methionine) are damaged primarily because of oxida

Nutritional consequences of food processing.

A variety of methods are used to process foods: if they are not edible, to render them so; to permit storage; to alter texture and flavor; to destroy microorganisms and other toxins. These methods include heating (baking, cooking, frying, microwaving), freezing, and high pH. It is a paradox of nature that the processing of foods can improve, nutrition, quality, and safety; yet, occasionally these processing alternatives can lead to the formation of anti-nutritional and toxic compounds. These multi-faceted consequences of food processing result from molecular interactions among nutrients and wi

Food processing and storage as a determinant of protein and amino acid availability.

Protein is perhaps the most reactive of the major food components. During food processing, the essential amino acids, lysine, tryptophan, methionine and cyst(e)ine, may react with other food components causing a loss in amino acid bioavailability and sometimes a reduction in the digestibility of the whole protein molecule. This review first discusses some recent developments concerning protein-polyphenol reactions, racemization and lysinoalanine formation, and then describes the reactions of proteins and reducing sugars (the Maillard reaction) in greater detail. We report on the chemistry of t

Design, Validation, and Application of an Enzyme-Coupled Hydrogen Sulfide Detection Assay.

Hydrogen sulfide (H2S) is a key metabolite in biosynthesis and is increasingly being recognized as an essential gasotransmitter. Owing to its diffusible and reactive nature, H2S can be difficult to quantify, particularly in situ. Although several detection schemes are available, they have drawbacks. In efforts to quantify sulfide release in the cross-linking reaction of the flagellar protein FlgE, we developed an enzyme-coupled sulfide detection assay using the Escherichia coli O-acetylserine sulfhydrylase enzyme CysM. Conversion of HS- to l-cysteine via CysM followed by derivatization with th

Studies on enmetazobactam clarify mechanisms of widely used β-lactamase inhibitors.

β-Lactams are the most important class of antibacterials, but their use is increasingly compromised by resistance, most importantly via serine β-lactamase (SBL)-catalyzed hydrolysis. The scope of β-lactam antibacterial activity can be substantially extended by coadministration with a penicillin-derived SBL inhibitor (SBLi), i.e., the penam sulfones tazobactam and sulbactam, which are mechanism-based inhibitors working by acylation of the nucleophilic serine. The new SBLi enmetazobactam, an N-methylated tazobactam derivative, has recently completed clinical trials. Biophysical studies on the me

Defatted hempseed meal altered the metabolic profile of fermented yogurt and enhanced the ability to alleviate constipation in rats.

BACKGROUND: Hempseeds (Cannabis sativa L.) are rich in easily digestible proteins, fats, polyunsaturated fatty acids, and insoluble fiber and are of high nutritional value. Probiotics have been found to relieve constipation, which solves a health problem that constantly troubles a lot of people. Therefore, the changes in the metabolites of fermented yogurt with or without 10% defatted hempseed meal (10% SHY or 0% SHY respectively) were studied and their laxative effects were examined through animal experiments. RESULTS: Amino acids and peptides, terpene glycosides, carbohydrates, lineolic aci
