Matrix proteins of the teeth of the sea urchin Lytechinus variegatus.

The teeth of the sea urchin Lytechinus variegatus grow continuously. The mineral phase, a high magnesium calcite, grows into single crystals within numerous compartments bounded by an organic matrix deposited by the odontoblasts. Electron microscopic examination of glutaraldehyde-fixed Ethylene Diamine Tetra acetic acid (EDTA) demineralized teeth shows the compartment walls to be organized from multiple layers of cell membrane which might contain cytoplasmic protein inclusions. Proteins extracted during demineralization of unfixed teeth were examined by gel electrophoresis, high performance li

Mineral-related proteins of sea urchin teeth: Lytechinus variegatus.

Sea urchins have a set of five continuously growing teeth, each of which has a very complex structure. The mineral phase is calcite of varying Mg content, depending on the location within a tooth. The calcium carbonate is present in amorphous, plate-like and rod-like forms. It has been hypothesized that the mineral deposition is a matrix-mediated process, similar to that in vertebrate bone and tooth, wherein certain macromolecules within the organic matrix of the mineralized tissue play an important role in nucleating and controlling the growth habit of the mineral crystals. It has also been h

[Abundance of Lytechinus variegatus (Echinoidea: Toxopneustidae) in the island of Cubagua, Venezuela].

The sea urchin roe is among the highest valued seafood items in the world. Increasing demand has extended this fishery worldwide and cases of overfishing have been detected in some countries. On the eastern coast of Venezuela some species of sea urchin, such as Lytechinus variegatus, are consumed as food but studies to determine the abundance and size do no exist. Between February 1998 and April 1999 six sampling stations (depth under 5 m) in Cubagua island (perimeter 25 km) were visited in 11 occasions to study the population density of L. variegatus, with a 0.25 m2 quadrat thrown eight times

Cytotoxic Activity of Six Samples of Brazilian Propolis on Sea Urchin (Lytechinus variegatus) Eggs.

The cytotoxic activities of extracts of four samples of propolis from the state of Minas Gerais (Southeast Brazil) and two from the state of Paraná (South Brazil) were evaluated using sea urchin (Lytechinus variegatus) eggs. Cytotoxic activity was observed, characterized mainly by the inhibition of the first cleavage of newly fertilized eggs. Methanol extracts at 32 µg mL(-1) of all samples were highly active (97-100%). Extracts were also prepared by successive treatments of the samples with hexane, chloroform, ethyl acetate, and methanol. High activity was observed using the ethyl acetate fra

A transcriptomic atlas of adult tissues of the sea urchin Lytechinus variegatus identifies early transitions in gonadogenesis.

Sea urchins have been a well-used model of fertilization and of the molecular biology of embryonic development. Our understanding of the molecular identity of adult tissues, on the other hand, has lagged significantly, a deficiency all too apparent following recent successes in mutational analyses in transgenerational studies. Here we present molecular analyses of gonadogenesis, with particular reference to developmental features emerging when a juvenile first begins to make eggs and sperm. We observed great variation in developmental time to reach fertility in Lytechinus variegatus, with the

On the formation and functions of high and very high magnesium calcites in the continuously growing teeth of the echinoderm Lytechinus variegatus: development of crystallinity and protein involvement.

Sea urchin teeth grow continuously and develop a complex mineralized structure consisting of spatially separate but crystallographically aligned first stage calcitic elements of high Mg content (5-15 mol% mineral). These become cemented together by epitaxially oriented second stage very high Mg calcite (30-40 mol% mineral). In the tooth plumula, ingressing preodontoblasts create layered cellular syncytia. Mineral deposits develop within membrane-bound compartments between cellular syncytial layers. We seek to understand how this complex tooth architecture is developed, how individual crystalli

Induction of innate immune gene expression following methyl methanesulfonate-induced DNA damage in sea urchins.

Sea urchins are noted for the absence of neoplastic disease and represent a novel model to investigate cellular and systemic cancer protection mechanisms. Following intracoelomic injection of the DNA alkylating agent methyl methanesulfonate, DNA damage was detected in sea urchin cells and tissues (coelomocytes, muscle, oesophagus, ampullae and gonad) by the alkaline unwinding, fast micromethod. Gene expression analyses of the coelomocytes indicated upregulation of innate immune markers, including genes involved in NF-κB signalling. Results suggest that activation of the innate immune system fo

Bidirectional Ca²⁺ signaling occurs between the endoplasmic reticulum and acidic organelles.

The endoplasmic reticulum (ER) and acidic organelles (endo-lysosomes) act as separate Ca(2+) stores that release Ca(2+) in response to the second messengers IP3 and cADPR (ER) or NAADP (acidic organelles). Typically, trigger Ca(2+) released from acidic organelles by NAADP subsequently recruits IP3 or ryanodine receptors on the ER, an anterograde signal important for amplification and Ca(2+) oscillations/waves. We therefore investigated whether the ER can signal back to acidic organelles, using organelle pH as a reporter of NAADP action. We show that Ca(2+) released from the ER can activate the

The dopamine receptor antagonist haloperidol disrupts behavioral responses of sea urchins and sea stars.

Despite lacking a brain and having an apparent symmetrically pentaradial nervous system, echinoderms are capable of complex, coordinated directional behavioral responses to different sensory stimuli. However, very little is known about the molecular and cellular mechanisms underlying these behaviors. In many animals, dopaminergic systems play key roles in motivating and coordinating behavior, and although the dopamine receptor antagonist haloperidol has been shown to inhibit the righting response of the sea urchin Strongylocentrotus purpuratus, it is not known whether this is specific to this

Alizarin Red perturbs skeletal patterning and biomineralization via catalase inhibition.

Alizarin Red S (AZ) is a dye that is commonly used in histological studies and textiles. We find that AZ is teratogenic to Lytechinus variegatus sea urchin larvae. Here, we demonstrate that embryos exposed to AZ have abnormal skeletal patterning and perturbed migration of skeletogenic mesenchyme cells, along with mild ectodermal abnormalities and reduced neural connectivity. Temporal transcriptomics reveal delayed development and implicate wide-reaching changes in gene expression with AZ treatment. Particle image velocimetry experiments show that AZ treatment perturbs the normal pattern of flu

Sex specific gene expression is present prior to metamorphosis in the sea urchin.

A profound collaboration between the germline and somatic cells of an organism is the creation of a functional gonad. Here we establish a foundation for studying molecular gonadogenesis in the sea urchin by use of RNA-seq, quantitative mRNA measurements, and in-situ hybridizations throughout the life cycle of the variegated sea urchin, Lytechinus variegatus (Lv). We found through three distinct analyses that the ovary and testis of this echinoderm expresses unique transcripts involved in gametogenesis, and also discovered uncharacterized gene products unique to each gonad. We further developed

SPR Sensor-Based Analysis of the Inhibition of Marine Sulfated Glycans on Interactions between Monkeypox Virus Proteins and Glycosaminoglycans.

Sulfated glycans from marine organisms are excellent sources of naturally occurring glycosaminoglycan (GAG) mimetics that demonstrate therapeutic activities, such as antiviral/microbial infection, anticoagulant, anticancer, and anti-inflammation activities. Many viruses use the heparan sulfate (HS) GAG on the surface of host cells as co-receptors for attachment and initiating cell entry. Therefore, virion-HS interactions have been targeted to develop broad-spectrum antiviral therapeutics. Here we report the potential anti-monkeypox virus (MPXV) activities of eight defined marine sulfated glyca

Anticoagulant and Antithrombotic Properties of Three Structurally Correlated Sea Urchin Sulfated Glycans and Their Low-Molecular-Weight Derivatives.

The anticoagulant and antithrombotic properties of three structurally correlated sea urchin-derived 3-linked sulfated α-glycans and their low molecular-weight derivatives were screened comparatively through various in vitro and in vivo methods. These methods include activated partial thromboplastin time, the inhibitory activity of antithrombin over thrombin and factor Xa, venous antithrombosis, the inhibition of platelet aggregation, the activation of factor XII, and bleeding. While the 2-sulfated fucan from Strongylocentrotus franciscanus was observed to be poorly active in most assays, the 4

Chemical Defense against Herbivory in the Brown Marine Macroalga Padina gymnospora Could Be Attributed to a New Hydrocarbon Compound.

Brown marine macroalga Padina gymnospora (Phaeophyceae, Ochrophyta) produces both secondary metabolites (phlorotannins) and precipitate calcium carbonate (CaCO3-aragonite) on its surface as potential defensive strategies against herbivory. Here, we have evaluated the effect of natural concentrations of organic extracts (dichloromethane-DI; ethyl acetate-EA and methanol-ME, and three isolated fractions) and mineralized tissues of P. gymnospora as chemical and physical resistance, respectively, against the sea urchin Lytechinus variegatus through experimental laboratory feeding bioassays. Tissue regeneration and biomineralization in sea urchins: role of Notch signaling and presence of stem cell markers.

Echinoderms represent a phylum with exceptional regenerative capabilities that can reconstruct both external appendages and internal organs. Mechanistic understanding of the cellular pathways involved in regeneration in these animals has been hampered by the limited genomic tools and limited ability to manipulate regenerative processes. We present a functional assay to investigate mechanisms of tissue regeneration and biomineralization by measuring the regrowth of amputated tube feet (sensory and motor appendages) and spines in the sea urchin, Lytechinus variegatus. The ability to manipulate r

Experimental manipulation of the amount of tubulin available for assembly into the spindle of dividing sea urchin eggs.

Spindle assembly is studied in the eggs of the sea urchin Lytechinus variegatus by experimentally varying the amount of polymerizable tubulin within the egg. Aliquots of fertilized eggs from the same female are individually pulsed for 1-6 min with 1 X 10(-6) M Colcemid at least 20 min before first nuclear envelope breakdown. This treatment inactivates a portion of the cellular tubulin before the spindle is formed. Upon entering mitosis, treated eggs form functional spindles that are reduced in length and birefringent retardation but not width. With increased exposure to Colcemid, the length an

Antiviral activity of marine sulfated glycans against pathogenic human coronaviruses.

Great interest exists towards the discovery and development of broad-spectrum antivirals. This occurs due to the frequent emergence of new viruses which can also eventually lead to pandemics. A reasonable and efficient strategy to develop new broad-spectrum antivirals relies on targeting a common molecular player of various viruses. Heparan sulfate is a sulfated glycosaminoglycan present on the surface of cells which plays a key role as co-receptor in many virus infections. In previous work, marine sulfated glycans (MSGs) were identified as having antiviral activities. Their mechanism of actio

Antiviral activities of four marine sulfated glycans against adenovirus and human cytomegalovirus.

Broad-spectrum antivirals are more needed than ever to provide treatment options for novel emerging viruses and for viruses that lack therapeutic options or have developed resistance. A large number of viruses rely on charge-dependent non-specific interactions with heparan sulfate (HS), a highly sulfated glycosaminoglycan (GAG), for attachment to cell surfaces to initiate cell entry. As such, inhibitors targeting virion-HS interactions have potential to have broad-spectrum antiviral activity. Previous research has explored organic and inorganic small molecules, peptides, and GAG mimetics to di
