Wave damping by giant kelp, Macrocystis pyrifera.

BACKGROUND AND AIMS: The increased likelihood and severity of storm events has brought into focus the role of coastal ecosystems in provision of shoreline protection by attenuating wave energy. Canopy-forming kelps, including giant kelp (Macrocystis pyrifera), are thought to provide this ecosystem service, but supporting data are extremely limited. Previous in situ examinations relied mostly on comparisons between nominally similar sites with and without kelp. Given that other factors (especially seafloor bathymetry and topographic features) often differ across sites, efforts to isolate the ef

Heat stress induces organelle alterations in Macrocystis pyrifera gametophytes.

Kelp, brown macroalgae in the order Laminariales, provide ecosystem services vital to ocean biodiversity. However, kelp forests worldwide are declining due to abiotic stressors such as ocean warming. In this study, we present results from high-resolution confocal microscopy and in vivo imaging system imaging using protocols developed to visualize kelp gametophyte cells exposed to heat-stress treatments. Imaging revealed chloroplast mislocalization, fragmentation, and subsequent loss of chloroplasts in heat-stressed gametophyte cells. Additionally, nuclei exhibited fragmentation and a progressi

Experimental evidence for early bacterial inheritance in the giant kelp, Macrocystis pyrifera.

Bacteria associated with macroalgae influence the development, health, and ecological interactions of their hosts. Although vertical transmission of bacterial symbionts has been proposed in some algal species, evidence remains limited. In this study, we combined 16S rRNA gene amplicon sequencing, fluorescence in situ hybridization (FISH), and transmission electron microscopy (TEM) to assess bacterial presence and localization throughout early developmental stages of the brown macroalga Macrocystis pyrifera. Amplicon sequencing detected bacterial taxa in reproductive sori, released spores, and

Fucoidan from Macrocystis pyrifera has powerful immune-modulatory effects compared to three other fucoidans.

Fucoidan, a sulfated polysaccharide purified from brown algae, has a variety of immune-modulation effects, such as promoting activation of dendritic cells (DCs), natural killer (NK) cells and T cells, and enhancing anti-viral and anti-tumor responses. However, the immune-modulatory effect of fucoidan from different seaweed extracts has not been thoroughly analyzed and compared. We analyzed fucoidans obtained from Ascophyllum nodosum (A. nodosum), Macrocystis pyrifera (M. pyrifera), Undaria pinnatifida (U. pinnatifida) and Fucus vesiculosus (F. vesiculosus) for their effect on the apoptosis of

Effects of Heat Waves and Light Deprivation on Giant Kelp Juveniles (Macrocystis pyrifera, Laminariales, Phaeophyceae).

Due to climate change, the incidence of marine heat waves (MHWs) has increased, yet their effects on seaweeds are still not well understood. Adult sporophytes of Macrocystis pyrifera, the species forming the iconic giant kelp forests, can be negatively affected by thermal stress and associated environmental factors (e.g., nutrient depletion, light deprivation); however, little is known about the tolerance/vulnerability of juvenile sporophytes. Simultaneously to MHWs, juveniles can be subjected to light limitation for extended periods of time (days-weeks) due to factors causing turbidity, or ev

Applications of environmental DNA monitoring for seaweed reproductive phenology: A case study with giant kelp (Macrocystis pyrifera).

Monitoring the seasonal reproductive cycles of seaweeds is crucial for effective population and ecosystem management, as well as mariculture seedstock collection. Traditional methods, such as visual monitoring by SCUBA diving or snorkeling, are costly, labor-intensive, and limited in temporal and spatial coverage. This study explores substituting these methods with environmental DNA (eDNA) techniques for giant kelp (Macrocystis pyrifera, order Laminariales). This laboratory study aimed to determine the minimum detectable concentration of zoospores and sporophyte tissue needed for detecting the

Latitudinal variation in chemical composition and biological activities of fucoidans from Macrocystis pyrifera (brown algae): first insights along the Chilean coast.

In this study the chemical composition of fucoidans from Macrocystis pyrifera seaweed collected along a latitudinal gradient on the Chilean coast was investigated. Three water-soluble fucoidans were extracted and characterized using different physico-chemical techniques. Their antioxidant, anticoagulant, and cytotoxic activities were also assessed. The results showed that three extracted fucoidans contained a high degree of sulfation and had a high molecular weight. The sugar analysis revealed the presence of fucose, mannose, galactose, glucuronic acid, and glucose monosaccharides at different

Micronutrients and kelp cultures: evidence for cobalt and manganese deficiency in southern california deep seawater.

It has been suggested that naturally occurring copper and zinc concentrations in deep seawater are toxic to marine organisms when the free ion forms are overabundant. The effects of micronutrients on the growth of gametophytes of the ecologically and commercially significant giant kelp (Macrocystis pyrifera) were studied in defined media. The results indicate that toxic copper and zinc ion concentrations as well as cobalt and manganese deficiencies may be among the factors controlling the growth of marine organisms in nature.

Light limitation and water velocity modify the impacts of simulated marine heatwaves on juvenile giant kelp.

Coastal regions are complex habitats, where multiple natural and anthropogenic drivers can interact to affect the survival and growth of marine organisms. The giant kelp Macrocystis pyrifera is sensitive to increasing seawater temperatures and susceptible to marine heatwaves. Light availability and hydrodynamics can also affect the growth, morphology, and resilience of this species. In this experiment, juvenile sporophytes of M. pyrifera from Scorching Bay, Wellington, Aotearoa, New Zealand, a were exposed to a combination of simulated marine heatwaves at one of four different temperatures (20

Genomic analyses of giant kelp reveal restricted connectivity across the Southern Hemisphere and striking diversity in Aotearoa (New Zealand).

The macroalga Macrocystis pyrifera ("giant" or "bladder" kelp) plays a vital role in cold-temperate marine ecosystems but faces increasing threats from ongoing environmental change. Understanding its population diversity and connectivity is essential for predicting future range shifts and assessing the ecological consequences of such shifts in both present and novel ranges. Here, we used high-resolution genotyping-by-sequencing (GBS) to investigate the population structure of M. pyrifera within and among populations in Aotearoa (New Zealand), Lutruwita (Tasmania), and Falkland Islands/Malvinas

Synergistic Effects of Sulfated Polysaccharides from Mexican Seaweeds against Measles Virus.

Sulfated polysaccharides (SPs) extracted from five seaweed samples collected or cultivated in Mexico (Macrocystis pyrifera, Eisenia arborea, Pelvetia compressa, Ulva intestinalis, and Solieria filiformis) were tested in this study in order to evaluate their effect on measles virus in vitro. All polysaccharides showed antiviral activity (as measured by the reduction of syncytia formation) and low cytotoxicity (MTT assay) at inhibitory concentrations. SPs from Eisenia arborea and Solieria filiformis showed the highest antiviral activities (confirmed by qPCR) and were selected to determine their

Comparison of in vitro and ex vivo osteogenic efficacy of fucoidans extracted from three brown macroalgae of the Pacific Ocean.

Brown macroalgae possesses bioactive compounds including fucoidan, a sulfated polysaccharide that can affect bone cells based on its origin and composition. This study examined the potential of fucoidan isolated from three macroalgal species - Macrocystis pyrifera, Sargassum muticum, and Undaria pinnatifida - to modulate the bone remodeling process and evaluated their comparative effectiveness. Fucoidans were extracted under acidic media, and chemically and structurally characterized before comparing their bioactivity on bone cells at different stages. Fucoidans moderately decreased the surviv

Population genomics reveals strong impacts of genetic drift without purging and guides conservation of bull and giant kelp.

Kelp forests are declining in many parts of the northeast Pacific.1,2,3,4 In small populations, genetic drift can reduce adaptive variation and increase fixation of recessive deleterious alleles,5,6,7 but natural selection may purge harmful variants.8,9,10 To understand evolutionary dynamics and inform restoration strategies, we investigated genetic structure and the outcomes of genetic drift and purging by sequencing the genomes of 429 bull kelp (Nereocystis luetkeana) and 211 giant kelp (Macrocystis sp.) from the coastlines of British Columbia and Washington. We identified 6 to 7 geographica

Pheromone-induced spawning of Pacific herring. I. Behavioral characterization.

A spawning pheromone in the milt (semen) and testes of the Pacific herring, Clupea harengus pallasi, triggers spawning in sexually mature fish of both sexes and is thought to facilitate school spawning of this species. We found the response to the pheromone to be a stereotyped behavioral sequence consisting of a graded extension of the gonadal papilla, release of gametes, and spawn deposition behavior. The response is triggered by an olfactory stimulus, as demonstrated by the elimination of the response by occlusion of the nares. Stimulus concentrations of an approximate 1:500 dilution of fres
