Detection of endocytobionts inhabiting the macronucleus of Frontonia paramagna (Ciliophora, Peniculida).

Bacterial endosymbionts of Frontonia, a widely distributed ciliate genus, remain poorly characterized. Here, we investigated the endosymbiotic microbiota of a Shanghai population of Frontonia paramagna using an integrated morphological and molecular approach. Fluorescence in situ hybridization (FISH) targeting the 16S rRNA gene, coupled with V3-V4 high-throughput sequencing, consistently identified Caedimonas as the bacterial symbiont, specifically localized within the host macronucleus. FISH and transmission electron microscopy confirmed this intramacronuclear colonization with high prevalenc

A 63-kDa Periplasmic Protein of the Endonuclear Symbiotic Bacterium Holospora obtusa Secreted to the Outside of the Bacterium during the Early Infection Process Binds Weakly to the Macronuclear DNA of the Host Paramecium caudatum.

The Gram-negative bacterium Holospora obtusa is a macronucleus-specific symbiont of the ciliate Paramecium caudatum. It is known that an infection of this bacterium induces high level expressions of the host hsp60 and hsp70 genes, and the host cell acquires both heat-shock and high salt resistances. In addition, an infectious form of H. obtusa-specific 63-kDa periplasmic protein with a DNA-binding domain in its amino acid sequence is secreted into the host macronucleus after invasion into the macronucleus and remain within the nucleus. These facts suggest that binding of the 63-kDa protein to

Three new Troglodytella and a new Gorilloflasca ciliates (Entodiniomorphida) from mountain gorillas (Gorilla beringei beringei) in Rwanda.

Four new species, Troglodytella gracilis, T. virunga, T. dolium, and Gorilloflasca longior were described from the mountain gorillas in Rwanda. The three Troglodytella species share a retractable adoral ciliary zone, four non-retractable ciliary arches, two broad skeletal plates, a skeletal rod plate, numerous longitudinal cortical grooves, and four contractile vacuoles. The anterior ciliary arch is the longest of the four arches, extending transversely on the left body surface. T. gracilis and T. virunga are characterized by an elongated body, a tail flap, and a wedge-shaped macronucleus. T.

Tentative identification of the species of Balantidium from ostriches (Struthio camelus) as Balantidium coli-like by analysis of polymorphic DNA.

The characteristics of Balantidium from ostriches (Struthio camelus) are similar to those of Balantidium coli; however, the species Balantidium struthionis was proposed on the basis of the host species (ostriches) and the shape of the macronucleus (with a deep depression in one side). In the present work, we have performed morphological and genetic comparisons between isolates of Balantidium from ostriches and B. coli from pigs to determine the specific status of B. struthionis. The morphological characteristics of the trophozoites of Balantidium from ostriches were reviewed in 100 trophozoite

An endosymbiotic Chlorella-bearing ciliate: Platyophrya chlorelligera Kawakami 1989.

The morphology of an endosymbiotic Chlorella-bearing ciliate, Platyophrya chlorelligera Kawakami, 1989, isolated in a suburb of Hiroshima and cultivated in a culture medium constituted of mineral salts, was investigated by means of protargol staining, scanning and transmission electron microscopy. Trypsin treatment was found to be highly effective for observation of resting cysts by transmission electron microscope as it prevents the occurrence of perforations and increases contrast in cell organelles in sections. The ciliate differs from its congeners, especially with respect to the number of

Phylogenetic position of Nyctotherus teleacus isolated from a tortoise (Astrochelys radiata) and its electron microscopic features.

A commensal ciliate was isolated from the stool of a tortoise (Astrochelys radiata). The ciliate was classified as Nyctotherus teleacus, according to its basic morphological features. Electron microscopic observations using cultured N. teleacus (NictoT1 strain) revealed many spherical hydrogenosomes and methanogen-suspected bacteria, together with a characteristic triangular macronucleus containing many spherical chromosomes in the cytoplasm of NictoT1. The results of phylogenetic analysis showed that NictoT1 was included in the cluster of Nyctotheroides spp. (family Nyctotheridae). Nyctothero

New species of Balantidium and Paracichlidotherus (Ciliophora) inhabiting the intestines of four surgeonfish species from the Tuvalu Islands, Pacific Ocean.

Four species of adult herbivorous surgeonfishes (Family Acanthuridae) were collected from the remote South-Pacific island system of Tuvalu. Their intestinal contents were examined, and of four populations of ciliated protists, two new species were discovered and are described. Ciliates were examined after protargol staining and, in some cases, scanning electron microscopy. Members of each population were examined and 10 characters measured for the balantidia, and 13 for the paracichlidotherids. A new Balantidium is described which has an unusually large dextr-oral field of cilia. A new species

[Morphometric and cytogenetic studies of the follicular epithelium thyroid of small mammals under chronic exposure to small doses].

We have detected the genotoxic effect (increase of micronuclei) after chronic exposure to low-level ionizing radiation of the thyroid follicular epithelium of the voles living for many generations in the radioactive territory. At the same time, we also discovered the stimulating effect of chronic low-level ionizing radiation on the activity of morphogenetic processes in the thyroid parenchyma of the animals from natural populations (increased micro-follicles). These data were confirmed by the experiments; chronic irradiation of the thyroid gland of rats at the doses of 5 and 50 cGy revealed an

The First Case of Diarrhoea in Tibetan Sheep, Ovis aries, Caused by Balantidium coli in the Qinghai Tibetan Plateau Area, China.

This study was carried out to determine the pathogen-causing diarrhoea in sheep Ovis aries in the Qinghai Tibetan Plateau Area, China. A trophozoite was identified as species of ciliate alveolates infecting the sheep based on morphological characteristics examined by microscope. It was mostly spherical, colourless and transparent, with many vesicles. Macronucleus and contractile vacuoles could not be distinguished. Size of the trophozoite was 80-180 × 70-150 μm and its surface was covered with cilia. Molecular analysis based on sequences of 18S rRNA and ITS genes confirmed the ciliate species

Genome content reorganization in the non-model ciliate Chilodonella uncinata: insights into nuclear architecture, DNA content, and chromosome fragmentation during macronuclear development.

UNLABELLED: Ciliates are a model lineage for studies of genome architecture given their unusual genome structures. All ciliates have both somatic macronuclei (MAC) and germline micronuclei (MIC), both of which develop from a zygotic nucleus following sex (i.e., conjugation). Nuclear developmental stages are not well documented among non-model ciliates, including Chilodonella uncinata (class Phyllopharyngea), the focus of our work. Here, we characterize nuclear architecture and genome dynamics in C. uncinata by combining 4',6-diamidino-2-phenylindole (DAPI) staining and fluorescence in situ hyb

Cytogenetic monitoring of domestic mammals exposed to wastewaters from the localities of Dladla and Boukallou near Settat, Morocco.

The aim of this work was to know the biological impact of wastewaters on livestock in the localities of Dladla and Boukallou (Town of Settat, Morocco), where wastewaters are largely used by the local population for their agricultural activities. We used the micronucleus (MN) test in cytochalasin B blocked binucleated cells (BNC) to assess the possible induced genetic effects. The cellular proliferation index (PI) was also calculated. Blood samples were collected from sheep, bovines and mules and peripheral blood cultures were made according to our laboratories' standard methodology. The result

Infection with the Endonuclear Symbiotic Bacterium Holospora obtusa Reversibly Alters Surface Antigen Expression of the Host Paramecium caudatum.

It is known that the ciliate Paramecium cell surface including cilia is completely covered by high-molecular-mass GPI-anchored proteins named surface antigens (SAgs). However, their functions are not well understood. It was found that ciliate Paramecium caudatum reversibly changes its SAgs depending on the absence or presence of the endonuclear symbiotic bacterium Holospora obtusa in the macronucleus. Immunofluorescence microscopy with a monoclonal antibody produced SAg of the H. obtusa-free P. caudatum strain RB-1-labeled cell surface of the H. obtusa-free P. caudatum RB-1 cell but not the H.

Porpostoma guamensis n. sp., a philasterine scuticociliate associated with brown-band disease of corals.

Brown band disease of coral is caused by a ciliate that consumes the tissue of the corals in the genus Acropora. We describe the ciliate associated with this disease on Guam, based on: general morphology, division stages, and ciliature observed on live and protargol-stained specimens; modification of the oral structures between divisional stages, observed on protargol-stained specimens; and some aspects of behavior in field and laboratory studies. Porpostoma guamensis n. sp. is elongate and has ciliature typical for the genus; live cells are 70-500 × 20-75 μm; the macronucleus is sausage-like,

Metacollinia emscheri n. sp., a novel sanguicolous apostome ciliate of freshwater amphipods (Gammarus spp.).

We describe a novel sanguicolous parasitic ciliate, Metacollinia emscheri n. sp., found in the freshwater amphipods Gammarus pulex and G. fossarum. This ciliate infected 8.05 % of the amphipods collected in a German stream catchment, the Boye, a tributary of the river Emscher. The ciliate showed morphological characteristics fitting the genus Metacollinia. Different life stages of variable size occurred simultaneously in the hemocoel throughout the hosts' body. The tomont had 40-47 slightly spiraled kineties, a non-ciliated cortical band, a large macronucleus, and contractile vacuoles arranged

Detection of Ichthyophthirius multifiliis (Ichthyophthiriidae) in two wild amphibian species.

Emerging infectious diseases are one of the main threats to global amphibian populations. Frogs and salamanders are already affected by various pathogens, including ranaviruses, Batrachochytrium dendrobatidis, B. salamandrivorans, and helminths. Here, we report the first confirmed cases of Ichthyophthirius multifiliis, a ciliate parasite traditionally considered fish-specific, infecting wild amphibian larvae in a natural setting. As part of a passive surveillance program in Catalonia, five dead amphibians (four S. salamandra larvae and one Rana temporaria post-metamorph) were collected from a

Morphological characterization and intraspecific variation of Zoothamnium intermedium Precht, 1935 (Ciliophora, Peritrichia) attached to calanoid copepods in the Chesapeake Bay, USA.

A redescription of Zoothamnium intermedium Precht, 1935, a peritrich epibiont on copepods, is provided using specimens colonizing Acartia tonsa and Eurytemora affinis in Chesapeake Bay, USA. Bell-shaped zooids of Z. intermedium ranged in size from 31.2-54.7 microm x 16.7-31.3 microm in vivo. A single contractile vacuole and a "C shaped" macronucleus lie in the upper half of the body. Colonies had up to 30 zooids, but most presented two to four zooids. The single myoneme was continuous from the zooids through the lateral branches and basal stalk, terminating 4-73.2 mm before the attachment poin

Characteristics delineation of piscidin 5 like from Larimichthys crocea with evidence for the potent antiparasitic activity.

Several researches reported that piscidin members of teleosts owned strong antiparasitic activity. Cryptocaryon irritans, a type of ectoparasite, could infect most of the marine teleosts. Larimichthys crocea could severely suffer from marine white spot disease caused by C. irritans, and their mortality rate was significantly high. Concentrating on this problem, we have done many related works. Piscidin 5 like (termed Lc-P5L) was another piscidin member isolated from a comparative transcriptome of C. irritans-immuned L. crocea. In the paper, quantitative Real-time PCR (qRT-PCR) showed Lc-P5L wa
