MAFF is a transcription factor that regulates contraction-induced skeletal muscle transcription.

Skeletal muscle adapts to exercise through rapid transcriptional remodeling, but regulators that link contractile activity to these gene programs remain unclear. Here we show that MAFF, a small MAF transcription factor, is consistently induced by acute exercise in both human and mouse skeletal muscle, with induction restricted to muscles recruited by the exercise modality. In C2C12 cells, Maff was induced by in vitro electrical pulse stimulation model. Using CRISPR/Cas9-mediated Maff knockout C2C12 cells, we found that loss of Maff reduced myogenic fusion with accumulation of unfused nuclei. R

Regulation of the MAFF transcription factor by proinflammatory cytokines in myometrial cells.

The MAF (proto-)oncogene family of basic-leucine zipper transcription factors plays crucial roles in the control of mammalian gene expression and development. Here we analyzed the regulation of the human MAFF gene, coding for a small MAF transcription factor, in uterine smooth muscle cells. We found that MAFF transcript levels are induced by proinflammatory cytokines in PHM1-31 myometrial cells. We observed an important induction by interleukin 1 beta (IL1B) and a weaker upregulation by tumor necrosis factor (TNF), whereas interleukin 6 (IL6) treatment had no effect. Time course experiments re

Transcription Factor MAFF (MAF Basic Leucine Zipper Transcription Factor F) Regulates an Atherosclerosis Relevant Network Connecting Inflammation and Cholesterol Metabolism.

BACKGROUND: Coronary artery disease (CAD) is a multifactorial condition with both genetic and exogenous causes. The contribution of tissue-specific functional networks to the development of atherosclerosis remains largely unclear. The aim of this study was to identify and characterize central regulators and networks leading to atherosclerosis. METHODS: Based on several hundred genes known to affect atherosclerosis risk in mouse (as demonstrated in knockout models) and human (as shown by genome-wide association studies), liver gene regulatory networks were modeled. The hierarchical order and r

MafF Is an Antiviral Host Factor That Suppresses Transcription from Hepatitis B Virus Core Promoter.

Hepatitis B virus (HBV) is a stealth virus that exhibits only minimal induction of the interferon system, which is required for both innate and adaptive immune responses. However, 90% of acutely infected adults can clear the virus, suggesting the presence of additional mechanisms that facilitate viral clearance. Here, we report that Maf bZIP transcription factor F (MafF) promotes host defense against infection with HBV. Using a small interfering RNA (siRNA) library and an HBV/NanoLuc (NL) reporter virus, we screened to identify anti-HBV host factors. Our data showed that silencing of MafF led

Regulation of CXCL1 chemokine and CSF3 cytokine levels in myometrial cells by the MAFF transcription factor.

Cytokines play key roles in a variety of reproductive processes including normal parturition as well as preterm birth. Our previous data have shown that MAFF, a member of the MAF family of bZIP transcription factors, is rapidly induced by pro-inflammatory cytokines in PHM1-31 myometrial cells. We performed loss-of-function studies in PHM1-31 cells to identify MAFF dependent genes. We showed that knockdown of MAFF significantly decreased CXCL1 chemokine and CSF3 cytokine transcript and protein levels. Using chromatin immunoprecipitation analyzes, we confirmed CXCL1 and CSF3 genes as direct MAFF

PLK3-Activated Mitochondrial Apoptosis Pathway in the Pathogenesis of Sepsis-Associated Acute Kidney Injury.

Acute kidney injury (AKI), a prevalent complication of sepsis, sorely needs effective interventions. Yet, our grasp of the mechanisms behind sepsis-induced AKI is far from complete, hindering the development of targeted therapies. Cecal ligation and puncture (CLP) was used to induce sepsis in mice, followed by treatment with the PLK3 antagonist R406 to assess the effects of PLK3 inhibition on inflammatory responses and renal damage. A lipopolysaccharide (LPS)-induced injury model in HK-2 cells was developed to explore the impact of LPS on lysosomal membrane permeability (LMP) and PLK3 expressi

MAFF regulates ferroptotic sensitivity through iron homeostasis and fatty acid synthesis.

Ferroptosis is an iron-dependent form of regulated cell death driven by lipid peroxidation. In this study, we identify the transcription factor v-maf musculoaponeurotic fibrosarcoma oncogene homolog F (MAFF) as a key regulator that promotes breast cancer progression while simultaneously sensitizing tumor cells to ferroptosis. Integrated RNA-sequencing and chromatin immunoprecipitation (ChIP)-sequencing revealed that MAFF directly regulates genes involved in iron and fatty acid metabolism. MAFF enhances intracellular iron availability by transactivating SLC11A2 and NCOA4, expanding the labile i

Early growth response 2 activates MAF bZIP transcription factor F to protect β-cells from oxidative stress in type 1 diabetes.

BACKGROUND: Dysfunction of insulin-secreting pancreatic β-cells is a crucial factor in the development and advancement of type 1 diabetes (T1D). This investigation delves into the role of early growth response 2 (Egr2) in the function of pancreatic β-cells and explores its functional mechanism. METHODS: Expression of Egr2 in the pancreatic tissues of streptozotocin (STZ)-treated mice and mouse MIN6 pancreatic β-cells was examined using immunohistochemistry and RT-qPCR. Gain-of-function assays of Egr2 were performed to analyze its role in insulitis severity in mice. In vitro, the viability, ap

Small MAF genes variants and chronic myeloid leukemia.

Chronic myeloid leukemia (CML) is one of the most frequent hematological neoplasia worldwide. The abnormal accumulation of reactive oxygen species may be an important factor in CML development. The transcription factor NRF2 can regulate the transcription of a battery of antioxidant and detoxificant genes after heterodimerizing with small-Maf proteins. Although the participation of NRF2 in the development of chronic degenerative diseases has been thoroughly studied, the role of small-Maf genes has not been documented. We have identified polymorphisms in the three MAF genes (F, G and K) and asse

MAFF mediates transcriptional activation of CFLAR to reduce regulatory T cell apoptosis and inhibit diabetic retinopathy progression.

BACKGROUND: Regulatory T cells (Tregs) regulate inflammation, promote regeneration, and may prevent diabetic retinopathy (DR). This study investigated whether MAFF inhibited DR progression by suppressing Treg apoptosis through CFLAR activation. METHODS: Bioinformatics was utilized to predict differentially expressed genes in peripheral blood mononuclear cells from DR patients and type 2 diabetes mellitus patients. A mouse model of DR was induced through a high-fat diet and streptozotocin. Mice were subjected to genetic intervention of CFLAR overexpression to observe retinal vascular infiltrat

Melatonin Enhances the Myometrial Contraction Through MT2-PKC-NRF2/MAFF Signaling Pathway in Sows.

Spontaneous parturition in both humans and sows occurs predominantly at night. Melatonin is thought to facilitate nocturnal parturition in humans by synergistically enhancing myometrial contractions with oxytocin, but the precise underlying mechanisms remain unclear. In this study, in vivo analyses showed that melatonin levels in both serum and myometrium were significantly increased in laboring sows, accompanied by increased melatonin synthesized in the myometrium. Moreover, exogenous melatonin administration to pregnant sows potentially shifted parturition time. Functional studies demonstrat

Establish a recombinant yeast detection system to study the effect of MIP on transactivation function of hMafF in US2-driven gene transcription.

The human gene MafF (hMafF) is a member of bZip transcription factor Maf family, but it alone cannot activate its target genes. In 2006, a novel hMafF interacting protein (MIP) was identified. Transient transfection assay in Hela cells suggested that co-expression of MIP and hMafF could activate US2-driven transcription. In this work, we constructed a series of plasmids and transformed YM4271 yeast strain to establish a recombinant yeast detection system. In this system, MIP's expression level could be regulated using glucose incubation or galactose-induced incubation. The expression level of

Monocytes differentiate along two alternative pathways during sterile inflammation.

During inflammation, monocytes differentiate within tissues into macrophages (mo-Mac) or dendritic cells (mo-DC). Whether these two populations derive from alternative differentiation pathways or represent different stages along a continuum remains unclear. Here, we address this question using temporal single-cell RNA sequencing in an in vitro model, allowing the simultaneous differentiation of human mo-Mac and mo-DC. We find divergent differentiation paths, with a fate decision occurring within the first 24 h and confirm this result in vivo using a mouse model of sterile peritonitis. Using a

Transcription factors as essential regulators of myometrial smooth muscle cell function during pregnancy and labor.

Parturition is a tightly regulated process coordinated by myometrial smooth muscle cells, which generate the uterine contraction required for birth. Preterm labor, defined as labor occurring before 37 weeks of gestation, remains a significant clinical challenge due to its association with adverse short and long-term risks on neonatal and maternal health. Earlier and recent studies provide strong evidence that transcription factors are performing crucial functions in uterine smooth muscle cells. In this review, we focus on the PR/ER, PPAR, NF-κB, AP-1, NFE2L2, and MAFF transcription factors and

The FACT complex facilitates expression of lysosomal and antioxidant genes through binding to TFEB and TFE3.

TFEB (transcription factor EB) and TFE3 (transcription factor binding to IGHM enhancer 3) orchestrate the cellular response to a variety of stressors, including nutrient deprivation, oxidative stress and pathogens. Here we describe a novel interaction of TFEB and TFE3 with the FAcilitates Chromatin Transcription (FACT) complex, a heterodimeric histone chaperone consisting of SSRP1 and SUPT16H that mediates nucleosome disassembly and assembly, thus facilitating transcription. Extracellular stimuli, such as nutrient deprivation or oxidative stress, induce nuclear translocation and activation of

Induction of endogenous Nrf2/small maf heterodimers by arsenic-mediated stress in placental choriocarcinoma cells.

Exposure to inorganic arsenic has been associated with various forms of cancer, nervous system pathogenesis, and vascular diseases, as well as reproductive and developmental toxicity. Here, the effect of inorganic arsenic on placental JAR choriocarcinoma cells was assessed. The nuclear protein levels of the CNC transcription factor Nrf2 were strongly induced in the presence of arsenic. Dosage response experiments showed that 0.5 microM of arsenic is sufficient to augment Nrf2 levels. The expression of the Nrf2 dimerization partners MafG and MafK appeared not to be modulated by arsenic, whereas
