Unveiling the Potential of Magainin Derivatives against Candida Species.

The increasing antifungal resistance, including Candida albicans resistance, highlights the urgent need for novel antifungal agents. Antimicrobial peptides are promising alternative antimicrobial agents, but most work focuses on antibacterial studies, including Magainin II and its derivative Pexiganan (MSI-78). Here, we evaluated a Pexiganan peptide library and identified MSI-78 (4-20) as a lead-hit antifungal agent against multiple Candida species, including fluconazole-resistant C. auris isolates. Importantly, it improved survival and reduced fungal burden in C. albicans-infected Galleria me

Lack of magainin-like activity in human cervical tissue.

PROBLEM: The cervix plays an integral role in innate immunity of the human reproductive tract. Magainins are antimicrobial and spermicidal peptides recently described in human submandibular glands. We investigated the human cervix for magainin-like peptides. METHOD: Histologic sections of frozen and paraffin embedded cervical tissue from nine subjects were separately incubated with two rabbit, polyclonal, anti-magainin antibodies (CB-2 and CB-7) to investigate for magainin-like activity in the human cervix. RESULTS: There was no specific staining of cervical columnar cells within the endocer

Magainin-like immunoreactivity in human submandibular and labial salivary glands.

Magainins, antimicrobial peptides secreted by granular glands of frog skin, may be related to the high resistance to infections of this epithelial surface. The oral mucosa of healthy individuals is another tissue in which infection is not frequent, probably owing to the activity of potent salivary and mucosal defense mechanisms. To investigate if magainin-like factors are a component of these oral defense mechanisms, human and animal minor (mucosal) and major salivary glands were examined by immunohistochemistry, using a polyclonal rabbit anti-magainin antibody. Cryostat sections of (para) for

The effects of magainin 2, cecropin, mastoparan and melittin on Brucella abortus.

The effect of the alpha-helical polycationic peptides magainin 2, melittin, mastoparan and cecropin on the viability of Brucella abortus 544 (type species), B. abortus S19 (vaccine strain) and B. abortus S2308 (vaccine challenge strain) was determined. Rough mutants of these strains and the rough candidate vaccine strain B. abortus RB51 were also tested. S. typhimurium was used as a control. The peptides did not affect the viability of B. abortus smooth strains but some of the peptides affected viability of the rough strains. Magainin 2 at a concentration of 100 micrograms ml-1 did not reduce

Magainin 2 induces bacterial cell death showing apoptotic properties.

Magainin 2 is pore-forming antimicrobial peptide on lipid matrix of bacterial membrane, secreted from the skin of the African clawed frog Xenopus laevis. The aim of this study was to investigate a new concept for antibacterial mechanisms called bacterial apoptosis-like cell death. We examined the morphological changes induced by magainin 2 in Escherichia coli, regarding apoptosis. Specifically, phosphatidylserine externalization from the inner to outer membrane surface was detected by Annexin V staining, and DNA fragmentation and chromatin condensation was detected by TUNEL and DAPI assay. We

The cytotoxic effect of magainin II on the MDA-MB-231 and M14K tumour cell lines.

Many studies have highlighted the tumoricidal properties of some natural peptides known to have antimicrobial virtues. Also, the increasingly higher resistance to conventional antibiotics has become a global public health issue, and the need for new antibiotics has stimulated interest in finding and synthesizing new antimicrobial peptides, which may also be used as chemotherapeutic agents. Relying on the literature, the purpose of our in vitro research was to assess the tumoricidal potential of magainin II on a series of tumour cell lines, namely, MDA-MB-231 (breast adenocarcinoma) and M14K (h

Charge-dependent insertion of the antimicrobial peptide magainin 2 into lipid monolayers probed by neutron reflectivity.

Many antimicrobial peptides act by disrupting cellular membranes, with therapeutic selectivity arising from their ability to discriminate between bacterial and mammalian cells, a distinction largely governed by membrane surface charge. Here, we investigate the interfacial organisation and membrane insertion of the cationic antimicrobial peptide magainin 2 using specular neutron reflectometry on lipid monolayers that mimic bacterial and mammalian membranes. Negatively charged dipalmitoylphosphatidylglycerol (DPPG) and zwitterionic dipalmitoylphosphatidylcholine (DPPC) were used as respective re

Investigating specific bacterial resistance to AMPs by using a magainin I-resistant Escherichia coli model.

Antimicrobial peptides (AMPs) are multifunctional compounds that may show antimicrobial and immunomodulatory activities. With the rapid increase in the incidence of multidrug-resistant bacteria, there is an enormous interest in AMPs as templates for the production of new antibiotics. However, there are concerns that the therapeutic administration of AMPs can select resistant strains. In order to distinguish between resistant and non-resistant strains and verify resistance specificity to AMPs, in this study a magainin I-resistant Escherichia coli model was used. First, the identity of all strai

Therapeutic effect of pH responsive Magainin II modified azithromycin plus curcumin micelles in different depth models of MRSA infection.

Methicillin-resistant Staphylococcus aureus (MRSA) is a major pathogen responsible for serious infections in humans. The overuse of antibiotics has led to the evolution of resistance genes in bacteria. This study aimed to develop a pH-responsive micelle, loaded with therapy drugs and modified with antimicrobial peptides, to treat drug-resistant bacterial infections at varying depths. pH-responsive micelles containing azithromycin and curcumin, modified with Magainin II, were prepared using the thin-film dispersion method. The physicochemical properties of the micelles were characterized, and t

Antibacterial properties of Magainin II peptide onto 304 stainless steel surfaces: A comparison study of two dopamine modification methods.

Marine biofouling is perplexing the development of marine industry, and the traditional antifouling methods are restricted by the requirements of marine environmental friendliness. Marine bacteria attachment is the initial stage of marine fouling and it can be effectively reduced by reducing bacterial attachment. In this study, two modification methods were reported to synthesize antibacterial surfaces based on the different order of Magainin Ⅱ (MAG Ⅱ) modification. The preparation of SS-DA-M was generated by modifying the 304 stainless steel (304 SS) surface with dopamine firstly and then gra

Antibacterial activities of rhodamine B-conjugated gelsolin-derived peptides compared to those of the antimicrobial peptides cathelicidin LL37, magainin II, and melittin.

The growing number of antibiotic-resistant bacteria necessitates the search for new antimicrobial agents and the principles by which they work. We report that cell membrane-permeant rhodamine B (RhB)-conjugated peptides based on the phosphatidylinositol-4,5-bisphosphate binding site of gelsolin can kill the gram-negative organisms Escherichia coli and Pseudomonas aeruginosa and the gram-positive organism Streptococcus pneumoniae. RhB linkage to the QRLFQVKGRR sequence in gelsolin was essential for the antibacterial function, since the unconjugated peptide had no effect on the bacteria tested.

Purification and properties of antimicrobial peptides from skin secretions of the Eritrea clawed frog Xenopus clivii (Pipidae).

Five peptides with antimicrobial activity were isolated from norepinephrine-stimulated skin secretions of the tetraploid frog Xenopus clivii Peracca, 1898 (Pipidae). Characterization of the peptides demonstrated that they are structurally similar to magainins (2 peptides), caerulein-precursor fragments, CPF (2 peptides), and xenopsin-precursor fragments, XPF (1 peptide) that have been previously isolated from other species of the genus Xenopus. The magainins and the XPF peptide were active only against the Gram-negative microorganism Escherichia coli whereas the CPF peptides were also active a

Common mechanism unites membrane poration by amyloid and antimicrobial peptides.

Poration of bacterial membranes by antimicrobial peptides such as magainin 2 is a significant activity performed by innate immune systems. Pore formation by soluble forms of amyloid proteins such as islet amyloid polypeptide (IAPP) is implicated in cell death in amyloidoses. Similarities in structure and poration activity of these two systems suggest a commonality of mechanism. Here, we investigate and compare the mechanisms by which these peptides induce membrane leakage and bacterial cell death through the measurement of liposome leakage kinetics and bacterial growth inhibition. For both sys
