OsWHY1 Negatively Modulates Rice Immunity Against Magnaporthe oryzae.

WHIRLY proteins, known for their DNA/RNA-binding capabilities conferred by a highly conserved WHIRLY domain, are integral to plant defense mechanisms. However, the role of OsWHY1 in rice immunity remains to be clarified. We examined OsWHY1 expression after treatment with exogenous hormones, PAMPs, and Magnaporthe oryzae. We generated OsWHY1-overexpressing rice plants and performed yeast two-hybrid (Y2H), bimolecular fluorescence complementation (BiFC), transcriptome, and RT-qPCR analyses. OsWHY1 expression was upregulated by exogenous hormones and PAMPs, with a significant increase at 24 h pos

MoGT2 Is Essential for Morphogenesis and Pathogenicity of Magnaporthe oryzae.

Magnaporthe oryzae causes the rice blast disease, which is one of the most serious diseases of cultivated rice worldwide. Glycosylation is an important posttranslational modification of secretory and membrane proteins in all eukaryotes, catalyzed by glycosyltransferases (GTs). In this study, we identified and characterized a type 2 glycosyltransferase, MoGt2, in M. oryzae Targeted gene deletion mutants of MoGT2 (mogt2Δ strains) were nonpathogenic and were impaired in vegetative growth, conidiation, and appressorium formation at hyphal tips. Moreover, MoGT2 plays an important role in stress tol

Molecular and biological characterisation of Magnaporthe oryzae botourmiavirus 15.

In this study, we have identified and characterized a novel fungal virus, designated Magnaporthe oryzae botourmiavirus 15 (MoBV15), which was isolated from the rice blast fungus. The genome of MoBV15 is a positive-sense, single-stranded RNA (+)ssRNA 3,228 nucleotides in length with a GC content of 51.4%. It features a single open reading frame (ORF) predicted to encode a putative RNA-dependent RNA polymerase (RdRp). The 5' and 3' ends of the MoBV15 genomic RNA exhibit reverse complementarity and may form a panhandle structure-a feature rarely observed. On tomato-oatmeal medium, the virus‑carry

Oligomycins inhibit Magnaporthe oryzae Triticum and suppress wheat blast disease.

Oligomycins are macrolide antibiotics, produced by Streptomyces spp. that show antagonistic effects against several microorganisms such as bacteria, fungi, nematodes and the oomycete Plasmopara viticola. Conidiogenesis, germination of conidia and formation of appressoria are determining factors pertaining to pathogenicity and successful diseases cycles of filamentous fungal phytopathogens. The goal of this research was to evaluate the in vitro suppressive effects of two oligomycins, oligomycin B and F along with a commercial fungicide Nativo® 75WG on hyphal growth, conidiogenesis, conidial ger

Insights into antifungal mechanism of cumin aldehyde against Magnaporthe oryzae.

BACKGROUND: Cumin essential oil from Cuminum cyminum is a promising alternative to synthetic fungicides because of its broad-spectrum antifungal activity and environmental compatibility. However, its major active constituent and antifungal mechanism remain unclear. Using Magnaporthe oryzae Guy11, the causal agent of rice blast as model, this study identified the major antifungal constituent of cumin essential oil and investigated its mode of action. RESULTS: Twelve major constituents were identified by gas chromatography-mass spectrometry, with cumin aldehyde as the predominant component (42.

Genetics of polymorphism in nitrogen-induced-susceptibility of rice to Magnaporthe oryzae.

Nitrogen fertilization is essential for crop yield but can increase susceptibility to rice blast, a phenomenon known as Nitrogen-Induced Susceptibility (NIS). The genetic architecture governing natural variation in NIS remains poorly characterized. To address this, we performed a genome-wide association study (GWAS) using a diverse panel of 268 rice accessions from the rice diversity panel 2, under controlled nitrogen regimes 0N (control), 1N (100 mg/L NH4NO3), 2N (200 mg/L NH4NO3). Our analyses defined a NIS Index (NISI), which revealed extensive phenotypic polymorphism in nitrogen responsive

Biological and biorational management of blast diseases in cereals caused by Magnaporthe oryzae.

Blast diseases, caused by the fungal pathogen Magnaporthe oryzae, are among the most destructive diseases that occur on at least 50 species of grasses, including cultivated cereals wheat, and rice. Although fungicidal control of blast diseases has widely been researched, development of resistance of the pathogen against commercially available products makes this approach unreliable. Novel approaches such as the application of biopesticides against the blast fungus are needed for sustainable management of this economically important disease. Antagonistic microorganisms, such as fungi and probio

Integrated omics analyses reveal natamycin disrupts oxidative phosphorylation pathways in Magnaporthe oryzae.

Magnaporthe oryzae, the causative agent of rice blast, is a devastating fungal pathogen that threatens global rice production. While natamycin is a known antimicrobial compound of microbial origin, its efficacy and precise mode of action against M. oryzae have remained largely unexplored. In this study, treatment with various concentrations of natamycin potently inhibited mycelial growth by 16.43% to 100%, with an EC₅₀ value of 11.77 mg/L. Pathogenicity of M. oryzae was also markedly reduced, declining to 4.1% of the control level under both in vitro and greenhouse conditions. To elucidate the

Abscisic acid negatively interferes with basal defence of barley against Magnaporthe oryzae.

BACKGROUND: Plant hormones are well known regulators which balance plant responses to abiotic and biotic stresses. We investigated the role of abscisic acid (ABA) in resistance of barley (Hordeum vulgare L.) against the plant pathogenic fungus Magnaporthe oryzae. RESULTS: Exogenous application of ABA prior to inoculation with M. oryzae led to more disease symptoms on barley leaves. This result contrasted the finding that ABA application enhances resistance of barley against the powdery mildew fungus. Microscopic analysis identified diminished penetration resistance as cause for enhanced susce

Septin-mediated plant cell invasion by the rice blast fungus, Magnaporthe oryzae.

To cause rice blast disease, the fungus Magnaporthe oryzae develops a pressurized dome-shaped cell called an appressorium, which physically ruptures the leaf cuticle to gain entry to plant tissue. Here, we report that a toroidal F-actin network assembles in the appressorium by means of four septin guanosine triphosphatases, which polymerize into a dynamic, hetero-oligomeric ring. Septins scaffold F-actin, via the ezrin-radixin-moesin protein Tea1, and phosphatidylinositide interactions at the appressorium plasma membrane. The septin ring assembles in a Cdc42- and Chm1-dependent manner and form

cAMP Regulates Infection Structure Formation in the Plant Pathogenic Fungus Magnaporthe grisea.

Magnaporthe grisea, the causal agent of rice blast, is one of the most destructive fungal pathogens of rice throughout the world. Infection of rice by M. grisea requires the formation of an appressorium, a darkly pigmented, dome-shaped structure. The germ tube tip differentiates into an appressorium following germination of conidia on a leaf surface. When conidia germinate on growth medium or other noninductive surfaces, the emerging germ tube does not differentiate and continues to grow vegetatively. Little is known about the endogenous or exogenous signals controlling the developmental proce

Carbonic anhydrases contribute to mitochondrial function, conidial development, and pathogenicity of Magnaporthe oryzae.

Carbonic anhydrases (CAs) are metalloenzymes widely present in biological systems, capable of catalyzing the reversible hydration of carbon dioxide (CO₂) to form bicarbonate (HCO3⁻) and protons (H+). This biochemical reaction plays a crucial role in various physiological processes. Previous studies have shown that MoCA1, a carbonic anhydrase in Magnaporthe oryzae, is localized in the mitochondria and is involved in conidial development and pathogenicity. However, the functions of other carbonic anhydrases in M. oryzae, such as MoCA2, MoCA4, and MoCA6, remain largely unexplored. Through subcell

Transmission of the G143A QoI-resistance point mutation through anastomosis in Magnaporthe grisea.

BACKGROUND: Soon after the introduction of Qo inhibitor fungicides in 1996, the point mutation leading to the amino acid exchange glycine to alanine at the 143 position of the mitochondrial cytochrome b gene was identified as the main cause of resistance. The present study describes the role of anastomosis in the transmission of the G143A mutation in Magnaporthe grisea. RESULTS: Two M. grisea mutants were co-cultivated on oatmeal agar and also co-inoculated on barley leaves. The mutants differed by the presence of the G143A mutation in one isolate and a disrupted AOX gene by insertion of a hy

Signaling defense responses of upland rice to avirulent and virulent strains of Magnaporthe oryzae.

Rice blast (Magnaporthe oryzae) can cause large losses in crop yields, especially in upland rice systems. Avirulent strains of M. oryzae can induce resistance to subsequent attacks by virulent strains in plants. This study aimed to investigate the defense responses in upland rice challenged with a virulent strain of M. oryzae after acclimation with an avirulent strain. The avirulent strain decreased rice blast severity in the challenged plants. Induced resistance was characterized by a hypersensitive response and early accumulation of phenolic compounds. Scanning electron microscopy showed tha

Exogenous anti-MoPMK1 microRNA and precursor formats reduce Magnaporthe oryzae infection in rice.

Exogenous RNA application is emerging as a useful strategy to investigate plant-pathogen interactions. Most studies against fungal pathogens have focused on long double-stranded RNAs, whereas the potential of artificial miRNA and miRNA-precursor formats remains less explored. Here, we targeted the Magnaporthe oryzae MAP kinase gene MoPMK1, a central regulator of appressorium-mediated penetration, invasive growth and host tissue colonization, using three exogenous RNA formats: chemically synthesized mature miRNA duplexes, in planta-produced anti-MoPMK1-containing small RNA fractions and in vitr

A putative rRNA methyltransferase Mrm1 regulates mitochondrial dynamics and pathogenicity in Magnaporthe oryzae.

Rice blast disease, a major global threat to staple crops, is caused by the ascomycete fungus Magnaporthe oryzae. This pathogen has complex mechanisms to invade rice, with mitochondrial function crucial for infection energy. Our study looks at the impact of Mrm1, a putative rRNA methyltransferase, on mitochondrial dynamics and pathogenicity of M. oryzae. Mrm1 deficiency delays appressorium formation and reduces turgor pressure for host penetration and infection hypha expansion. The N-terminal sequence of Mrm1, with a mitochondrial targeting sequence (MTS), is vital for its localization and fun

MoMih1 is indispensable for asexual development, cell wall integrity, and pathogenicity of Magnaporthe oryzae.

Asexual spore serves as essential inoculum of rice blast during the disease cycle, and differentiation of young conidium from conidiophore is intimately regulated by cell cycle. Mih1 encodes a dual-specificity phosphatase that involved in the G2/M transition of the mitotic cell cycle by regulating the Cdk1 activity in eukaryotes. Till now, the roles of Mih1 homologue, however, remain unclear in Magnaporthe oryzae. We here functionally characterized the Mih1 homologue MoMih1 in M. oryzae. MoMih1 is localized to both the cytoplasm and nucleus and can physically interact with the CDK protein MoCd

Activation of the signalling mucin MoMsb2 and its functional relationship with Cbp1 in Magnaporthe oryzae.

Various surface signals are recognized by Magnaporthe oryzae to activate the Pmk1 MAP kinase that is essential for appressorium formation and invasive growth. One of upstream sensors of the Pmk1 pathway is the MoMsb2 signalling mucin. However, the activation of MoMsb2 and its relationship with other sensors is not clear. In this study, we showed that the cleavage and transmembrane domains are essential for MoMsb2 functions. Cleavage of MoMsb2 was further confirmed by western blot analysis, and five putative cleavage sites were functionally characterized. Expression of the extracellular region

Evolutionary constraints and regulatory plasticity shape host specialization in the Magnaporthe oryzae species complex.

Rice blast caused by Magnaporthe oryzae threatens global rice production, and wheat blast emergence highlights the pathogen's capacity for host shifts. Although numerous studies have described M. oryzae genome organization and infection mechanisms, critical questions remain regarding the evolutionary drivers of stable host specialization. Importantly, the blast pathogen comprises a species complex of genetically differentiated, host-adapted lineages rather than single homogeneous species. To address this knowledge gap, we integrate evidence from evolutionary genomics, transcriptomics, and meta
