Magnetosomes as Potential Nanocarriers for Cancer Treatment.

Magnetotactic bacteria (MTBs) and their organelles, magnetosomes, are intriguing options that might fulfill the criteria of using bacterial magnetosomes (BMs). The ferromagnetic crystals contained in BMs can condition the magnetotaxis of MTBs, which is common in water storage facilities. This review provides an overview of the feasibility of using MTBs and BMs as nanocarriers in cancer treatment. More evidence suggests that MTBs and BMs can be used as natural nanocarriers for conventional anticancer medicines, antibodies, vaccine DNA, and siRNA. In addition to improving the stability of chemot

Bullet-shaped magnetosomes and metagenomic-based magnetosome gene profiles in a deep-sea hydrothermal vent chimney.

Magnetosome-producing microorganisms can sense and move toward the redox gradient and have been extensively studied in terrestrial and shallow marine sediment environments. However, given the difficulty of sampling, magnetotactic bacteria (MTB) are poorly explored in deep-sea hydrothermal fields. In this study, a deep-sea hydrothermal vent chimney from the Southern Mariana Trough was collected using a remotely operated submersible. The mineralogical and geochemical characterization of the vent chimney sample showed an internal iron redox gradient. Additionally, the electron microscopy of parti

The magnetosome model: insights into the mechanisms of bacterial biomineralization.

Though the most ready example of biomineralization is the calcium phosphate of vertebrate bones and teeth, many bacteria are capable of creating biominerals inside their cells. Because of the diversity of these organisms and the minerals they produce, their study may reveal aspects of the fundamental mechanisms of biomineralization in more complex organisms. The best-studied case of intracellular biomineralization in bacteria is the magnetosome, an organelle produced by a diverse group of aquatic bacteria that contains single-domain crystals of the iron oxide magnetite (Fe3O4) or the iron sulf

Biosynthesis of magnetosome-nanobody complex in Magnetospirillum gryphiswaldense MSR-1 and a magnetosome-nanobody-based enzyme-linked immunosorbent assay for the detection of tetrabromobisphenol A in water.

In this study, two mutant strains, TBC and TBC+, able to biosynthesize a novel functional magnetosome-nanobody (Nb), were derived from the magnetotactic bacteria Magnetospirillum gryphiswaldense MSR-1. The magnetosome-Nbs biosynthesized by TBC+ containing multi-copies of the Nb gene had a higher binding ability to an environmental pollutant, tetrabromobisphenol A (TBBPA), than those biosynthesized by TBC containing only one copy of the Nb gene. The magnetosome-Nbs from TBC+ can effectively bind to TBBPA in solutions with high capacity without being affected by a broad range of NaCl and methano

Genetically engineered bacterial magnetosomes as optimized tracers for magnetic particle imaging.

Medical imaging relies on tracer materials to enable accurate visualization and diagnosis of diseases. Magnetic Particle Imaging (MPI) is an innovative tomographic modality that offers exceptional sensitivity and temporal resolution. These characteristics make MPI particularly promising for clinical applications such as real-time vascular and perfusion imaging, tumor detection, and intraoperative guidance. However, MPI performance has so far been limited by the quality of available tracers, as conventional chemical synthesis provides only restricted control over the size, shape, and magnetic p

Non-pyrogenic highly pure magnetosomes for efficient hyperthermia treatment of prostate cancer.

We report the fabrication of highly pure magnetosomes that are synthesized by magnetotactic bacteria (MTB) using pharmaceutically compatible growth media, i.e., without compounds of animal origin (yeast extracts), carcinogenic, mutagenic, or toxic for reproduction (CMR) products, and other heavy metals than iron. To enable magnetosome medical applications, these growth media are reduced and amended compared with media commonly used to grow these bacteria. Furthermore, magnetosomes are made non-pyrogenic by being extracted from these micro-organisms and heated above 400 °C to remove and denatur

Intracellular transformation and disposal mechanisms of magnetosomes in macrophages and cancer cells.

Magnetosomes are magnetite nanoparticles biosynthesized by magnetotactic bacteria. Given their potential clinical applications for the diagnosis and treatment of cancer, it is essential to understand what becomes of them once they are within the body. With this aim, here we have followed the intracellular long-term fate of magnetosomes in two cell types: cancer cells (A549 cell line), because they are the actual target for the therapeutic activity of the magnetosomes, and macrophages (RAW 264.7 cell line), because of their role at capturing foreign agents. It is shown that cells dispose of mag

Development of magnetosomes-based biosensor for the detection of Listeria monocytogenes from food sample.

Listeriosis through contaminated food is one of the leading causes of premature deaths in pregnant women and new born babies. Here, the authors have developed a magnetosomes-based biosensor for the rapid, sensitive, specific and cost-effective detection of Listeria monocytogenes from food sample. Magnetosomes were extracted from Magnetospirillum sp. RJS1 and then directly bound to anti-Listeriolysin antibody (0.25-1 µg/ml), confirmed in spectroscopy. Listeriolysin (LLO) protein (0.01-7 µg/ml) was optimised in enzyme-linked immunosorbent assay. Magnetosomes was conjugated with LLO antibody (0.2

Magnetosome mediated oral Insulin delivery and its possible use in diabetes management.

Our study investigates the effect of magnetosome mediated oral Insulin delivery on diabetic induced rat models. The study involves the development of Magnetosome-Insulin (MI) conjugates by direct and indirect (by means of PEG) coupling method and further characterized by microscopic and spectroscopic analysis. The in vivo oral delivery of magnetosome-Insulin conjugate against streptozotocin-induced rat models and its efficiency was investigated. The impact of MI showed a remarkable change in the reduction of FBG levels up to 65% than the standard (Insulin). Similarly, the serum parameters: tri

Temporal and spatial resolution of magnetosome degradation at the subcellular level in a 3D lung carcinoma model.

Magnetic nanoparticles offer many exciting possibilities in biomedicine, from cell imaging to cancer treatment. One of the currently researched nanoparticles are magnetosomes, magnetite nanoparticles of high chemical purity synthesized by magnetotactic bacteria. Despite their therapeutic potential, very little is known about their degradation in human cells, and even less so of their degradation within tumours. In an effort to explore the potential of magnetosomes for cancer treatment, we have explored their degradation process in a 3D human lung carcinoma model at the subcellular level and wi

Chain organization of dimple-ended magnetite magnetosomes supports field-dependent cell linking in unicellular magnetotactic bacteria.

Magnetotactic bacteria (MTB) are generally thought to navigate as individual cells by using intracellular magnetosome chains that provide a stable magnetic moment for orientation along magnetic field lines. Whether nonclassical magnetosome geometries and particle-scale magnetic states can support robust chain-level magnetic function, and potentially mediate interactions between cells, remains unresolved. Here, to we report a unicellular magnetotactic bacterium, strain WYHV-1, that biomineralizes comparatively large prismatic magnetite magnetosomes with distinctive dimples at both crystal ends

A cumate-inducible promoter system for tuned gene expression and magnetosome overproduction in magnetotactic bacteria.

Magnetosomes are membrane-bound iron-rich organelles synthesized by magnetotactic bacteria as sensors for navigation in the Earth's magnetic field. In the well-studied model Magnetospirillum gryphiswaldense, their multistep biosynthesis is tightly controlled by > 30 specific genes clustered within a genomic magnetosome island. The lack of versatile genetic tools for strong, tunable, and orthogonal transcriptional control has impeded the analysis, engineering, and transplantation of this complex biosynthetic pathway. In addition, recent studies revealed an intricate transcriptional architecture

Magnetosome-incorporated silk fibroin-based magnetic hydrogel for efficient tumor magnetic hyperthermia.

We developed a magnetosome-incorporated silk fibroin-based magnetic hydrogel (MSMH) for efficient tumor magnetic hyperthermia. The MSMH was fabricated by integrating biologically synthesized magnetosomes with glycidyl methacrylate-modified silk fibroin (SFGMA), enabling injectable deployment and rapid photo-crosslinking. The as-fabricated MSMH exhibits excellent magnetothermal conversion performance, achieving prominent temperature rises under alternating magnetic fields (AMF). MSMH can also serve as a T₂-weighted MRI contrast agent (r2 = 173.57 mM-1 s-1 at 0.5 T), enabling imaging-guided ther

Essential magnetosome proteins MamI and MamL from magnetotactic bacteria interact in mammalian cells.

To detect cellular activities deep within the body using magnetic resonance platforms, magnetosomes are the ideal model of genetically-encoded nanoparticles. These membrane-bound iron biominerals produced by magnetotactic bacteria are highly regulated by approximately 30 genes; however, the number of magnetosome genes that are essential and/or constitute the root structure upon which biominerals form is largely undefined. To examine the possibility that key magnetosome genes may interact in a foreign environment, we expressed mamI and mamL as fluorescent fusion proteins in mammalian cells. Loc

Set-up of a pharmaceutical cell bank of Magnetospirillum gryphiswaldense MSR1 magnetotactic bacteria producing highly pure magnetosomes.

We report the successful fabrication of a pharmaceutical cellular bank (PCB) containing magnetotactic bacteria (MTB), which belong to the Magnetospirillum gryphiswaldense MSR1 species. To produce such PCB, we amplified MTB in a minimal growth medium essentially devoid of other heavy metals than iron and of CMR (Carcinogenic, mutagenic and reprotoxic) products. The PCB enabled to acclimate MTB to such minimal growth conditions and then to produce highly pure magnetosomes composed of more than 99.9% of iron. The qualification of the bank as a PCB relies first on a preserved identity of the MTB c

Ultrasound- and magnetic field-triggered destruction of insulin- and lysozyme-derived amyloid fibrils via biogenic magnetosomes.

Amyloid fibrils, highly ordered protein aggregates, are implicated in numerous diseases, including neurodegenerative and systemic amyloidoses. Proteins such as insulin and lysozyme, though not directly associated with neurodegeneration, form amyloid fibrils in metabolic and systemic contexts and serve as well-established models for studying amyloid formation and destruction. Fibrils formed by diverse proteins share common morphological traits and a conserved cross-β sheet structure, making these models broadly relevant. Their high stability presents a major therapeutic challenge, yet the same

Co-delivery of doxorubicin and recombinant plasmid pHSP70-Plk1-shRNA by bacterial magnetosomes for osteosarcoma therapy.

To explore a novel combination of chemotherapy, gene therapy, and thermotherapy for osteosarcoma, a targeted heat-sensitive co-delivery system based on bacterial magnetosomes (BMs) was developed. The optimal culture conditions of magnetotactic bacteria (MTB) AMB-1 and characterization of BMs were achieved. A recombinant eukaryotic plasmid heat shock protein 70-polo-like kinase 1-short hairpin RNA (pHSP70-Plk1-shRNA) under transcriptional control of a thermosensitive promoter (human HSP70 promoter) was constructed for gene therapy. No evidence for the putative nitric oxide sensor NsrR as a key regulator of magnetosome formation in Magnetospirillum gryphiswaldense.

Magnetotactic bacteria produce membrane-bound organelles known as magnetosomes, which consist of chains of magnetite crystals and function as sensors for orientation within the Earth's magnetic field. Magnetosome biosynthesis is a complex, multistep process that depends on iron availability and suboxic conditions. However, the expression of the ca. 30 core magnetosome biosynthetic genes has previously been described as constitutive and largely unaffected by environmental conditions. A recent study by Pang et al., published in this journal, reported the identification of a transcriptional regul

Biodegraded magnetosomes with reduced size and heating power maintain a persistent activity against intracranial U87-Luc mouse GBM tumors.

BACKGROUND: An important but rarely addressed question in nano-therapy is to know whether bio-degraded nanoparticles with reduced sizes and weakened heating power are able to maintain sufficient anti-tumor activity to fully eradicate a tumor, hence preventing tumor re-growth. To answer it, we studied magnetosomes, which are nanoparticles synthesized by magnetotactic bacteria with sufficiently large sizes (~ 30 nm on average) to enable a follow-up of nanoparticle sizes/heating power variations under two different altering conditions that do not prevent anti-tumor activity, i.e. in vitro cellula
