In order to investigate the role of bacterioferritin (Bfr) in the biomineralization of magnetite by microorganisms, we have cloned and sequenced the bfr genes from M. magnetotacticum. The organism has two bfr genes that overlap by one nucleotide. Both encode putative protein products of 18 kDa, the expected size for Bfr subunits, and show a strong similarity to other Bfr subunit proteins. By scanning the DNA sequence databases, we found that a limited number of other organisms, including N. gonorrhea, P. aeruginosa, and Synechocystis PCC6803, also have two bfr genes. When the sequences of a nu

Magnetotactic bacteria produce membrane-bound organelles known as magnetosomes, which consist of chains of magnetite crystals and function as sensors for orientation within the Earth's magnetic field. Magnetosome biosynthesis is a complex, multistep process that depends on iron availability and suboxic conditions. However, the expression of the ca. 30 core magnetosome biosynthetic genes has previously been described as constitutive and largely unaffected by environmental conditions. A recent study by Pang et al., published in this journal, reported the identification of a transcriptional regul

We report the successful fabrication of a pharmaceutical cellular bank (PCB) containing magnetotactic bacteria (MTB), which belong to the Magnetospirillum gryphiswaldense MSR1 species. To produce such PCB, we amplified MTB in a minimal growth medium essentially devoid of other heavy metals than iron and of CMR (Carcinogenic, mutagenic and reprotoxic) products. The PCB enabled to acclimate MTB to such minimal growth conditions and then to produce highly pure magnetosomes composed of more than 99.9% of iron. The qualification of the bank as a PCB relies first on a preserved identity of the MTB c

Magnetotactic bacteria (MTB) are of special scientific interest due to the formation of magnetosomes, intracellular membrane-enveloped magnetite crystals arranged into a linear chain by a dedicated cytoskeleton. Magnetotaxis relies on the formation and proper inheritance of these unique magnetic organelles, both of which need to be coordinated with the segregation of other cellular content such as chromosomes or motility and chemotaxis related structures. Thus, elaborated mechanisms are required in MTB to coordinate and maintain a high level of spatial and temporal subcellular organization dur

Magnetotactic bacteria (MTB) are worth studying because of magnetosome biomineralization. Magnetosome biogenesis in MTB is controlled by multiple genes known as magnetosome-associated genes. Recent advances in bioinformatics provide a unique opportunity for studying functions of magnetosome-associated genes and networks that they are involved in. Furthermore, various types of bioinformatics analyses can also help identify genes associated with magnetosome biogenesis. To predict novel magnetosome-associated genes in the extended CtrA regulon, we analyzed expression data of Magnetospirillum magn

In this study, two mutant strains, TBC and TBC+, able to biosynthesize a novel functional magnetosome-nanobody (Nb), were derived from the magnetotactic bacteria Magnetospirillum gryphiswaldense MSR-1. The magnetosome-Nbs biosynthesized by TBC+ containing multi-copies of the Nb gene had a higher binding ability to an environmental pollutant, tetrabromobisphenol A (TBBPA), than those biosynthesized by TBC containing only one copy of the Nb gene. The magnetosome-Nbs from TBC+ can effectively bind to TBBPA in solutions with high capacity without being affected by a broad range of NaCl and methano

Nutrient imbalances, soil salinity, and shrinking arable land threaten global food security, driving demand for sustainable biofertilizer alternatives to chemical inputs. Aquatic ecosystem-derived biofertilizers such as Magnetospirillum gryphiswaldense (MSR-1) are promising sustainable substitutes and show strong agricultural potential due to their stress tolerance, adaptability, and plant growth-promoting traits. This study investigated the ability of MSR-1 to enhance the growth and productivity of tomato and paddy under normal, iron-deficient, and saline conditions. MSR-1 was cultured in mod

The soil-water interface is replete with photic biofilm and iron minerals; however, the potential of how iron minerals promote biotic nitrate removal is still unknown. This study investigates the physiological and ecological responses of photic biofilm to hematite (Fe2O3), in order to explore a practically feasible approach for in-situ nitrate removal. The nitrate removal by photic biofilm was significantly higher in the presence of Fe2O3 (92.5%) compared to the control (82.8%). Results show that the presence of Fe2O3 changed the microbial community composition of the photic biofilm, facilitat

Magnetotactic bacteria (MTB) are generally thought to navigate as individual cells by using intracellular magnetosome chains that provide a stable magnetic moment for orientation along magnetic field lines. Whether nonclassical magnetosome geometries and particle-scale magnetic states can support robust chain-level magnetic function, and potentially mediate interactions between cells, remains unresolved. Here, to we report a unicellular magnetotactic bacterium, strain WYHV-1, that biomineralizes comparatively large prismatic magnetite magnetosomes with distinctive dimples at both crystal ends

Magnetosome chains biosynthesized by magnetotactic bacteria combine high magnetic responsivity with efficient magnetothermal conversion but face limitations due to fragile assembly and potential biosafety risks. Here, we report a biomimetic Stöber-based encapsulation strategy that preserves the native chain architecture while imparting structural robustness and enhanced safety. The resulting B-MNC exhibit a coercivity (33.6 mT), approximately 5× higher than magnetosomes, 1.4× higher than intact magnetotactic bacteria, and 2.2× higher than chemically synthesized analogs (C-MNC22). Enzymatic nanomotors self-propel by converting free molecular energy from chemical reactions. While initially used mainly as individual particles, these systems are increasingly implemented as swarms, where collective effects enable adaptable navigation in complex environments, broader coverage and enhanced local agent concentration. However, most studies have focused on planar swarming, and collective systems that combine magnetic navigation with enzyme-powered motion remain largely underexplored. Here, as a mechanistic proof-of-principle, we introduce urease-powered nanomotors based on biopr

Medical imaging relies on tracer materials to enable accurate visualization and diagnosis of diseases. Magnetic Particle Imaging (MPI) is an innovative tomographic modality that offers exceptional sensitivity and temporal resolution. These characteristics make MPI particularly promising for clinical applications such as real-time vascular and perfusion imaging, tumor detection, and intraoperative guidance. However, MPI performance has so far been limited by the quality of available tracers, as conventional chemical synthesis provides only restricted control over the size, shape, and magnetic p

Bacterial magnetic particles (BMPs) are of interest as potential carriers of bioactive macromolecules, drugs, or liposomes. In this study, a high-pressure homogenizer was used to disrupt Magnetospirillum gryphiswaldense strain MSR-1 cells, and BMPs were purified. BMPs were labeled with fluorescence reagent 1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocianin perchlorate (DiI) and injected into the tail vein of BALB/c nude mice. Distribution of fluorescence signals of DiI-BMPs in vivo was examined using a whole-body fluorescence imaging system. The result showed that fluorescence signals were

We report the fabrication of highly pure magnetosomes that are synthesized by magnetotactic bacteria (MTB) using pharmaceutically compatible growth media, i.e., without compounds of animal origin (yeast extracts), carcinogenic, mutagenic, or toxic for reproduction (CMR) products, and other heavy metals than iron. To enable magnetosome medical applications, these growth media are reduced and amended compared with media commonly used to grow these bacteria. Furthermore, magnetosomes are made non-pyrogenic by being extracted from these micro-organisms and heated above 400 °C to remove and denatur

Magnetotactic bacteria have evolved the remarkable capacity to biomineralize chains of magnetite [Fe(II)Fe(III)2O4] nanoparticles that align along the geomagnetic field and optimize their navigation in the environment. Mechanisms enabling magnetite formation require the complex action of numerous proteins for iron acquisition, sequestration in dedicated magnetosome organelles, and precipitation into magnetite. The MamP protein contains c-type cytochromes called magnetochrome domains that are found exclusively in magnetotactic bacteria. Ablation of magnetochromes in MamP prevents bacteria from

Magnetosomes are membrane-bound iron-rich organelles synthesized by magnetotactic bacteria as sensors for navigation in the Earth's magnetic field. In the well-studied model Magnetospirillum gryphiswaldense, their multistep biosynthesis is tightly controlled by > 30 specific genes clustered within a genomic magnetosome island. The lack of versatile genetic tools for strong, tunable, and orthogonal transcriptional control has impeded the analysis, engineering, and transplantation of this complex biosynthetic pathway. In addition, recent studies revealed an intricate transcriptional architecture

To detect cellular activities deep within the body using magnetic resonance platforms, magnetosomes are the ideal model of genetically-encoded nanoparticles. These membrane-bound iron biominerals produced by magnetotactic bacteria are highly regulated by approximately 30 genes; however, the number of magnetosome genes that are essential and/or constitute the root structure upon which biominerals form is largely undefined. To examine the possibility that key magnetosome genes may interact in a foreign environment, we expressed mamI and mamL as fluorescent fusion proteins in mammalian cells. Loc

This work evaluated the fates of linear alkylbenzene sulfonate (LAS), chemical oxygen demand (COD), ammonia nitrogen (NH4+-N), and total nitrogen (TN) when treating greywater (GW) in an oxygen-based membrane biofilm reactor (O2-MBfR). An influent ratio of chemical oxygen demand to total nitrogen (COD/TN) of 20 g COD/g N gave the best removals of LAS, COD, NH4+-N and TN, and it also had the greatest EPS accumulation in the biofilm. Higher EPS and improved performance were linked to increases in the relative abundances of bacteria able to biodegrade LAS (Zoogloea, Pseudomonas, Parvibaculum, Magn

Biofilm colonies are typically resistant to general antibiotic treatment and require targeted methods for their removal. One of these methods includes the use of nanoparticles as carriers for antibiotic delivery, where they randomly circulate in fluid until they make contact with the infected areas. However, the required proximity of the particles to the biofilm results in only moderate efficacy. We demonstrate here that the nonpathogenic magnetotactic bacteria Magnetosopirrillum gryphiswalense (MSR-1) can be integrated with drug-loaded mesoporous silica microtubes to build controllable micros
