Intermolecular interactions of the malate synthase of Paracoccidioides spp.

BACKGROUND: The fungus Paracoccidioides spp is the agent of paracoccidioidomycosis (PCM), a pulmonary mycosis acquired by the inhalation of fungal propagules. Paracoccidioides malate synthase (PbMLS) is important in the infectious process of Paracoccidioides spp because the transcript is up-regulated during the transition from mycelium to yeast and in yeast cells during phagocytosis by murine macrophages. In addition, PbMLS acts as an adhesin in Paracoccidioides spp. The evidence for the multifunctionality of PbMLS indicates that it could interact with other proteins from the fungus and host.

Discovery of novel inhibitors for malate synthase of Mycobacterium Tuberculosis from natural products.

Tuberculosis (TB) caused by Mycobacterium tuberculosis (Mtb) remains a global public health threat, particularly due to dormant Mtb, which necessitates prolonged drug treatment. Mycobacterium tuberculosis malate synthase (MtbMS) is a key rate-limiting enzyme in the glyoxylate shunt, essential for the survival of dormant Mtb but absent in the host. Using target-based virtual screening and biochemical approaches, we identified novel natural inhibitors of MtbMS. Molecular docking by Schrödinger and subsequent manual selection identified 11 compounds as potential inhibitors. Molecular dynamics (MD

Structural and energetic understanding of novel natural inhibitors of Mycobacterium tuberculosis malate synthase.

Persistent infection by Mycobacterium tuberculosis requires the glyoxylate shunt. This is a bypass to the tricarboxylic acid cycle in which isocitrate lyase (ICL) and malate synthase (MS) catalyze the net incorporation of carbon during mycobacterial growth on acetate or fatty acids as the primary carbon source. To identify a potential antitubercular compound, we performed a structure-based screening of natural compounds from the ZINC database (n = 1 67 740) against the M tuberculosis MS (MtbMS) structure. The ligands were screened against MtbMS, and 354 ligands were found to have better dockin

The malate synthase of Paracoccidioides brasiliensis is a linked surface protein that behaves as an anchorless adhesin.

BACKGROUND: The pathogenic fungus Paracoccidioides brasiliensis is the agent of paracoccidioidomycosis (PCM). This is a pulmonary mycosis acquired by inhalation of fungal airborne propagules that can disseminate to several organs and tissues leading to a severe form of the disease. Adhesion and invasion to host cells are essential steps involved in the internalization and dissemination of pathogens. Inside the host, P. brasiliensis may use the glyoxylate cycle for intracellular survival. RESULTS: Here, we provide evidence that the malate synthase of P. brasiliensis (PbMLS) is located on the f

Glyoxylate detoxification is an essential function of malate synthase required for carbon assimilation in Mycobacterium tuberculosis.

The glyoxylate shunt is a metabolic pathway of bacteria, fungi, and plants used to assimilate even-chain fatty acids (FAs) and has been implicated in persistence of Mycobacterium tuberculosis (Mtb). Recent work, however, showed that the first enzyme of the glyoxylate shunt, isocitrate lyase (ICL), may mediate survival of Mtb during the acute and chronic phases of infection in mice through physiologic functions apart from fatty acid metabolism. Here, we report that malate synthase (MS), the second enzyme of the glyoxylate shunt, is essential for in vitro growth and survival of Mtb on even-chain

Effect of partial obstruction of the rabbit urinary bladder on malate dehydrogenase and citrate synthase activity.

Previous studies have demonstrated that partial outlet obstruction in rabbits induced a significant decrease in oxidative metabolism in urinary bladder smooth muscle. The current experiments were designed to determine whether the decreased oxidative metabolism of obstructed bladder tissue is associated with alterations in the activities of specific mitochondrial enzymes. The activities of two important enzymes in the tricarboxylic acid cycle, malate dehydrogenase and citrate synthase, were measured in samples of bladder body and base from normal bladders and in bladders from rabbits in which p

Primary and post-irradiation inactivation of the sulfhydryl enzyme malate synthase: correlation of protective effects of additives.

The presence of additives during X-irradiation of malate synthase led to radioprotective effects against primary and post-irradiation inactivation. Pronounced effects were provided by typical scavengers, sulfhydryl reagents and specific ligands (substrates, products, analogues). The results show that scavenging and specific protection are responsible for the protective efficiency of additives. Scavengers delete noxious species formed during irradiation or post-radiationem. Sulfhydryl reagents may act as repair substances. Specific ligands protect the active site of the enzyme and the essential

Modeling, molecular docking, probing catalytic binding mode of acetyl-CoA malate synthase G in Brucella melitensis 16M.

There are enormous evidences and previous reports standpoint that the enzyme of glyoxylate pathway malate synthase G (MSG) is a potential virulence factor in several pathogenic organisms, including Brucella melitensis 16M. Where the lack of crystal structures for best candidate proteins like MSG of B. melitensis 16M creates big lacuna to understand the molecular pathogenesis of brucellosis. In the present study, we have constructed a 3-D structure of MSG of Brucella melitensis 16M in MODELLER with the help of crystal structure of Mycobacterium tuberculosis malate synthase (PDB ID: 2GQ3) as tem

Computational insights into Candida albicans Malate synthase: impact of cofactor and substrates on enzyme conformation and tunnel formation.

Candida albicans, an opportunistic pathogen, uptakes host-derived carbon sources for its survival and causes candidiasis infection. However, host defences such as macrophages restrict the C. albicans' sugar uptake via phagocytosis, triggering the glyoxylate pathway for adaptation in nutrient-limited conditions. Within macrophages, the C. albicans malate synthase (CaMLS1) exploits the available acetyl-CoA (ACOA), which condenses in presence of Mg2+ with glyoxylate (GOXL) to form malate, supporting its survival. The absence of structural studies highlights the need for in silico interventions to

The glyoxylate cycle in rat epiphyseal cartilage: the effect of vitamin-D3 on the activity of the enzymes isocitrate lyase and malate synthase.

The effect of vitamin-D deficiency and subsequent vitamin-D replacement on the metabolism of rat epiphyseal growth plate cartilage was studied. Biochemical analyses showed the presence of the two unique glyoxylate cycle enzymes isocitrate lyase and malate synthase in cartilage. The activity of these enzymes was markedly increased after treatment with the vitamin. Additionally, rat cartilage showed the capacity to oxidize fatty acid in the presence of cyanide. This cyanide-insensitive fatty acid oxidation is characteristic of peroxisomal B-oxidation rather than mitochondrial B-oxidation. Vitami

An integrated computational-experimental approach identifies Malate Synthase G as therapeutic target to disrupt carbon metabolism in chronic Pseudomonas aeruginosa infection.

Pseudomonas aeruginosa is a leading cause of nosocomial infections, particularly in individuals with a compromised immune system. Due to its strong adaptive ability, P. aeruginosa tends to develop antibiotic resistance and establish chronic infection, making its eradication through traditional antibiotics challenging. Thus, the development of novel therapeutic targets and corresponding inhibitors is urgently required. We conducted bioinformatics analyses of gene-chip datasets (GSE10362, GSE21966) of P. aeruginosa from patients with cystic fibrosis (CF) in the Gene Expression Omnibus (GEO) data

Dynamics of Citrate Synthase and Malate Dehydrogenase Gene Expression, Enzymatic Activity, and Physicochemical Changes During Postharvest Storage of Soursop (Annona muricata L.) GUANAY-1 Fruit.

Soursop (Annona muricata L.) is a climacteric tropical fruit belonging to the Annonaceae family, widely cultivated in tropical and subtropical regions due to its economic importance, nutritional value, and abundance of bioactive compounds. Its pulp is highly appreciated for fresh consumption and food processing; however, its high respiration rate and ethylene production accelerate ripening, resulting in rapid quality deterioration and a short postharvest shelf life. During ripening, soursop fruit undergoes physiological, biochemical, and molecular changes associated with organic acid metabolis

Alkaloids as inhibitors of malate synthase from Paracoccidioides spp.: receptor-ligand interaction-based virtual screening and molecular docking studies, antifungal activity, and the adhesion process.

Paracoccidioides is the agent of paracoccidioidomycosis. Malate synthase plays a crucial role in the pathogenicity and virulence of various fungi, such as those that are human pathogens. Thus, an inhibitor of this enzyme may be used as a powerful antifungal without side effects in patients once these enzymes are absent in humans. Here, we searched for compounds with inhibitory capacity against the malate synthase of Paracoccidioides species (PbMLS). The three-dimensional (3D) structure of PbMLS was determined using the I-TASSER server. Compounds were selected from the ZINC database. Based on t

Activity of Krebs cycle enzymes in mdx mice.

INTRODUCTION: Duchenne muscular dystrophy (DMD) is a degenerative disease of skeletal, respiratory, and cardiac muscles caused by defects in the dystrophin gene. More recently, brain involvement has been verified. Mitochondrial dysfunction and oxidative stress may underlie the pathophysiology of DMD. In this study we evaluate Krebs cycle enzymes activity in the cerebral cortex, diaphragm, and quadriceps muscles of mdx mice. METHODS: Cortex, diaphragm, and quadriceps tissues from male dystrophic mdx and control mice were used. RESULTS: We observed increased malate dehydrogenase activity in th

Dibutylchloromethyltin chloride, a potent inhibitor of electron transport in plant mitochondria.

Dibutylchloromethyl tin chloride (DBCT) inhibits coupled and uncoupled respiration of mitochondria from potato tubers, cauliflower florets and etiolated mung bean hypocotyls with succinate and L-malate but not with external NADH or TMPD/ascorbate as substrates. Using potato and cauliflower mitochondria. DBCT at 200 pmole/mg of protein gives complete inhibition only in KCl-based media and at pH 6.8. DBCT has no effect on the internal pH of mung bean mitochondria, but does cause a decrease in the membrane potential. Electron transport through the alternative oxidase is not inhibited, neither is

Effect of semi-permeable membrane coverage on carbon dioxide emissions during composting: Insights into carbon metabolism pathways, dominated by the tricarboxylic acid cycle.

The tricarboxylic acid (TCA) cycle is the main carbon metabolism pathway leading to carbon loss in the form of carbon dioxide (CO2). Research on the CO2 emission reduction mechanism based on the TCA cycle under different composting strategies is relatively lacking. This study investigated the effects of semi-permeable membrane coverage on CO2 emissions and pathways of carbon metabolism during three batches of large-scale dairy manure aerobic composting by using gas chromatography and high-throughput sequencing technologies. The results showed that the CO2 emission rate peaked at the initial st

Metabolic crosstalk between the mitochondrion and the nucleus is essential for Toxoplasma gondii infection.

Toxoplasma gondii, an intracellular pathogenic protist with a remarkable ability to infect a wide range of host cells, displays an equally exceptional design of its carbon metabolism. There are, however, critical gaps in our understanding of the metabolic network in T. gondii. We characterized the mito-nuclear metabolism and organelle coupling during its acute infection (lytic cycle). The major enzymes of the TCA cycle, i.e., citrate synthase (CS1), succinyl-CoA synthase alpha subunit (SCSα), succinate dehydrogenase (SDHA) and FAD malate dehydrogenase (MDH-FAD) located in the parasite mitochon
