Dwarfism mechanism in Malus clonal rootstocks.

The dwarfing mechanism in apple clonal rootstocks is driven by complex interactions between anatomical, hormonal, genetic, and phenolic factors, offering potential for advanced genetic manipulation to optimize tree size and enhance orchard productivity. The widespread adoption of dwarfing rootstocks is pivotal to modern commercial apple (Malus × domestica Borkh) orchards due to their ability to control tree size, shorten the juvenile period, and enhance reproductive growth and overall productivity. The underlying mechanisms of rootstock-induced dwarfism are multifaceted and involve interaction

The genome of the domesticated apple (Malus × domestica Borkh.).

We report a high-quality draft genome sequence of the domesticated apple (Malus × domestica). We show that a relatively recent (>50 million years ago) genome-wide duplication (GWD) has resulted in the transition from nine ancestral chromosomes to 17 chromosomes in the Pyreae. Traces of older GWDs partly support the monophyly of the ancestral paleohexaploidy of eudicots. Phylogenetic reconstruction of Pyreae and the genus Malus, relative to major Rosaceae taxa, identified the progenitor of the cultivated apple as M. sieversii. Expansion of gene families reported to be involved in fruit developm

Towards the Albino Mutant Gene in Malus × Domestica Borkh.

Albino mutation is among the most common phenomena that often causes a water imbalance and disturbs physiological functions in higher species of trees. Albinism frequently occurs in hybridized apples, but almost all seedlings die shortly after germination. In this study, a spontaneous albino mutant on Fuji apple trees was obtained. After bud grafting, new albino shoots with greenish-white leaves grew, although they were slender, small, and died easily. Resequencing analysis indicated that a total of 49.37 Gbp clean data of the albino mutant samples was obtained; its Q30 reached 91.43%, the ave

Molecular characterization of intergeneric hybrids between Malus and Pyrus.

Apple (Malus) and pear (Pyrus) are economically important fruit crops well known for their unique textures, flavours, and nutritional qualities. Both genera are characterised by a distinct pattern of secondary metabolites, which directly affect not only resistance to certain diseases, but also have significant impacts on the flavour and nutritional value of the fruit. The identical chromosome numbers, similar genome size, and their recent divergence date, together with DNA markers have shown that apple and pear genomes are highly co-linear. This study utilized comparative genomic approaches, i

Universal stress protein in Malus sieversii confers enhanced drought tolerance.

Drought is an important environmental factor that can severely affect plant growth and reproduction. Although many genes related to drought tolerance have been studied in economically important crops, very few genes have been functionally identified in Malus sieversii. In this study, we isolated a new gene based on throughput RNA sequencing analysis and constructed genetic expression vectors and transformed in Arabidopsis thaliana for functional verification. The results showed that MsUspA ectopic expression driven by constitutive (CaMV 35S) promoter gave rise to substantial improvements in ab

Malus xiaojinensis MxbHLH30 Confers Iron Homeostasis Under Iron Deficiency in Arabidopsis.

Iron stress adversely impacts plants' growth and development. Transcription factors (TFs) receive stress signals and modulate plant tolerance by influencing the expression of related functional genes. In the present study, we investigated the role of an apple bHLH transcription factor MxbHLH30 in the tolerance to iron stresses. The expression of MxbHLH30 was induced significantly by low-iron and high-iron treatments and MxbHLH30-overexpressed Arabidopsis plants displayed iron-stress-tolerant phenotypes. A determination of physiological and biochemical indexes associated with abiotic stress res

Genome-Wide Identification and Characterization of the JAZ Gene Family in Malus sieversii.

BACKGROUND: Malus sieversii, the wild ancestor of cultivated apples, possesses high stress tolerance and rich nutritional value but suffers from low fruit firmness. Jasmonate-ZIM (JAZ) domain proteins are key repressors of jasmonic acid (JA) signaling, yet their roles in fruit softening remain largely unexplored, especially in M. sieversii. METHODS AND RESULTS: In this study, we performed a genome-wide identification of the JAZ gene family in M. sieversii and characterized their structural features, phylogenetic relationships, chromosomal distribution, promoter cis-elements, expression patter

A New Function of MbIAA19 Identified to Modulate Malus Plants Dwarfing Growth.

The primary determinants of apple (Malus) tree architecture include plant height and internode length, which are the significant criteria for evaluating apple dwarf rootstocks. Plant height and internode length are predominantly governed by phytohormones. In this study, we aimed to assess the mechanisms underlying dwarfism in a mutant of Malus baccata. M. baccata dwarf mutant (Dwf) was previously obtained through natural mutation. It has considerably reduced plant height and internode length. A comparative transcriptome analysis of wild-type (WT) and Dwf mutant was performed to identify and an

Characterization of a SUPERMAN-like Gene, MdSUP11, in apple (Malus × domestica Borkh.).

Arabidopsis SUPERMAN and its family members of its family play important roles in plant growth and floral organ development; yet much less is known about their functions expanding in apple tree development. Previous work has identified 12 SUP-like genes in the apple (Malus × domestica Borkh.) genome, and the MdSUP11 which is expressed in both vegetative and reproductive organs of apple. However, the function of MdSUP11 remains obscure. In this study, the β-glucuronidase expression driven by the MdSUP11 native promoter was detected in roots, young leaves, and floral organs of transgenic Arabido

Revision of the nomenclature of the differential host-pathogen interactions of Venturia inaequalis and Malus.

The apple scab (Venturia inaequalis-Malus) pathosystem was one of the first systems for which Flor's concept of gene-for-gene (GfG) relationships between the host plant and the pathogen was demonstrated. There is a rich resource of host resistance genes present in Malus germplasm that could potentially be marshalled to confer durable resistance against this most important apple disease. A comprehensive understanding of the host-pathogen interactions occurring in this pathosystem is a prerequisite for effectively manipulating these host resistance factors. An accurate means of identification of

Metabolism of Malus halliana Roots Provides Insights into Iron Deficiency Tolerance Mechanisms.

Iron (Fe) deficiency is one of the most common micronutrient imbalances limiting plant growth globally, especially in arid and saline alkali regions due to the decreased availability of Fe in alkaline soils. Malus halliana grows well in arid regions and is tolerant of Fe deficiency. Here, a physiological and metabolomic approach was used to analyze the short-term molecular response of M. halliana roots to Fe deficiency. On the one hand, physiological data show that the root activity first increased and then decreased with the prolongation of the stress time, but the change trend of root pH was

Enzyme-Assisted Supercritical Fluid Extraction of Flavonoids from Apple Pomace (Malus×domestica).

Herein an enzyme-assisted supercritical fluid extraction (EA-SFE) was developed using the enzyme mix snailase to obtain flavonols and dihydrochalcones, subgroups of flavonoids, from globally abundant waste product apple pomace. Snailase, a commercially available mix of 20-30 enzymes, was successfully used to remove the sugar moieties from quercetin glycosides, kaempferol glycosides, phloridzin and 3-hydroxyphloridzin. The resulting flavonoid aglycones quercetin, kaempferol, phloretin and 3-hydroxyphloretin were extracted using supercritical carbon dioxide (scCO2) and minimum amounts of polar c

Insights into the aroma profiles and characteristic aroma of 'Honeycrisp' apple (Malus × domestica).

'Honeycrisp' is a popular apple cultivar because of its superior appearance and flavor. We investigated its aroma profiles and characteristic aroma. Whereas the aroma profiles of developing fruits were dominated by aldehydes, postharvest fruit accumulated abundant esters, alcohols, and sesquiterpenoids. Most of these components showed maxima at the fruit's climacteric peak. There were more types and higher contents of sesquiterpenoids, aldehydes, and esters in the fruit skin than in the pulp, while alcohol contents and types were comparable between the pulp and skin. Aroma extract dilution and

Malus hupehensis NPR1 induces pathogenesis-related protein gene expression in transgenic tobacco.

Most commercially grown apple cultivars are susceptible to fungal diseases. Malus hupehensis has high resistance to many diseases affecting apple cultivars. Understanding innate defence mechanisms would help to develop disease-resistant apple crops. Non-expressor of pathogenesis-related genes 1 (NPR1) plays a key role in regulating salicylic acid (SA)-mediated systemic acquired resistance (SAR). MhNPR1 cDNA, corresponding to genomic DNA and its 5' flanking sequences, was isolated from M. hupehensis. Sequence analysis showed that the regulatory mechanism for oligomer-monomer transition of the M

Functional Identification of MhPYL4 Involved in Iron-Deficiency Stress in Malus Halliana Koehne.

The PYL protein family are crucial sensors of the core signals of abscisic acid (ABA) and significantly influence the plant's response to ABA-mediated abiotic stresses as well as its growth and development. However, research on the role of the MhPYL4 gene in iron (Fe) deficiency in apple trees is limited. Studies have shown that the MhPYL4 gene, when exposed to Fe-deficiency stress, exhibits more rapid transcriptional upregulation than other genes' quickly elevated transcription. However, the precise mechanism by which it alleviates this stress remains unclear. The MhPYL4 gene (ID:103432868),

A Binary-Based Matrix Model for Malus Corolla Symmetry and Its Variational Significance.

Floral symmetry (corolla symmetry) has important biological significance in plant genetics and evolution. However, it is often multi-dimensional and difficult to quantify. Here, we constructed a multi-dimensional data matrix [X Y Z] by extracting three qualitative variables with binary properties (X: corolla regularity of interval and coplanarity; Y: petal regularity of shape and size; Z: petal local regularity of curling and wrinkle) from different dimensions of petals (overall to individual, and then to the local): all petals (corolla), individual petals, and local areas of petals.
