Development of monoclonal antibodies specific to Marek disease virus-EcoRI-Q (Meq) for the immunohistochemical diagnosis of Marek disease using formalin-fixed, paraffin-embedded samples.

Marek disease (MD) is a viral disease characterized by the development of lymphoma in poultry. Although morphologic confirmation of lymphoma is used to diagnose MD, immunohistochemical detection of MD virus-EcoRI-Q (Meq), which is a viral protein that is expressed exclusively in MD tumor cells, would further improve the accuracy of diagnosis. We developed monoclonal antibodies (mAbs) that specifically detect Meq by immunohistochemistry (IHC) using formalin-fixed, paraffin-embedded (FFPE) sections. We evaluated the sensitivity and specificity of 14 mAbs that we produced, using FFPE samples of M

Late Marek's disease in adult chickens inoculated with virulent Marek's disease virus.

Recently, excessive losses from Marek's disease (MD) have been noted in adult laying flocks over the age of 40 weeks. We defined these late outbreaks in adult chickens as "late MD", and experimentally reproduced the disease in adult SPF P2 line (50-week-old) or commercial line (74-week-old) chickens inoculated with a virulent strain of Marek's disease virus (MDV). Commercial line chickens were given MDV vaccines (HVT and CVI 988) at hatch. The occurrence of MD was evaluated periodically by the evidence of neurologic signs such as paralysis, torticollis, ataxia, and/or nervous tics, as well as

Prevention of Marek's disease: a review.

Marek's disease (MD) is a highly infectious neoplastic condition of chickens caused by a herpesvirus. The virus is cell associated in tumors and in all organs except in the feather follicle where enveloped infectious virions egress from the body. From this source, infection is spread horizontally by the airborne route to the environment and to other chickens. Vertical transmission from dam to offspring does not occur or at best is very rare. The nonpathogenic herpesvirus of turkeys (HTV) is ubiquitous in turkeys and is probably spread horizontally by the airborne route. When chickens are inocu

Differential attenuation of Marek's disease virus-induced tumours and late-Marek's disease virus-induced immunosuppression.

Marek's disease virus (MDV) is a herpesvirus that induces lymphoma and a variety of non-neoplastic syndromes in chickens. Furthermore, very virulent plus (vv+) MDVs induce a form of immunosuppression (late-MDV-IS) that might involve both neoplastic and non-neoplastic mechanisms. The objective of this study was to evaluate whether the attenuation of MDV-induced tumours and late-MDV-IS occurs simultaneously or can be dissociated. The immunosuppressive ability of three viruses derived from vv+ MDV strain 686 (wild-type 686, the somewhat attenuated molecular clone 686-BAC, and the nononcogenic mol

Role of the spleen in the pathogenesis of Marek's disease.

The importance of the spleen for the pathogenesis of Marek's disease (MD) was investigated. The spleen was not required for infection of other organs or for the development of latently infected peripheral blood lymphocytes (PBL). Splenectomised (Sx) chickens had infected lymphocytes in the thymus, bursa and PBL by 6 or 7 days after intratracheal exposure to MDV. However, splenectomy did delay the spread of infection since intact birds had positive cells in these tissues 2 to 3 days earlier. Splenectomy reduced the number of visceral tumours in JM-10- but not in GA-5-infected chickens. Splenect

Ultrasonographic features of Marek's disease in a chicken.

A 4-year-old chicken was presented with a history of anorexia, depression, and blindness. An ultrasound examination of the coelomic cavity was performed that revealed splenomegaly, hepatic nodules, and hypoechoic thickening of the intestinal wall. Ultrasonography of the coelomic cavity was done and revealed splenomegaly, nodular hepatic changes, and hypoechoic thickening of the intestinal wall. A diagnosis of Marek's disease was made based on the history and extension of the abdominal organ changes and confirmed by histopathology. This study describes an ultrasonographic appearance of Marek's

Two Cases of Marek's Disease in Backyard Turkeys.

In two independent submissions, a 3-yr-old, dead Bourbon Red turkey tom from a zoo and a Royal Palm turkey hen from a backyard flock were submitted for necropsy. Both birds had been kept together with chickens. Findings of the necropsy of the first turkey were an enlarged and dark liver with many pale white foci and a few small white nodules, pale and enlarged spleen, prominent thymus, mottled and pale kidneys, and pale and enlarged testes. Findings of the necropsy of the second turkey were a dark and mildly enlarged liver and severely enlarged, firm, and pale kidneys. Histopathology revealed

Attenuation of a recombinant Marek's disease virus lacking the meq oncogene and evaluation on its immune efficacy against Marek's disease virus.

SC9-2 is a recombinant Marek's disease virus (MDV) strain lacking the meq oncogene. Previous study demonstrated that SC9-2 virus provides good protection against challenge with a very virulent MDV rMd5, but it induces immunosuppressive effects in specific pathogen-free (SPF) chickens. In the present study, SC9-2 was serially passaged on chicken embryo fibroblast (CEF) cell cultures. The pathogenicity and immune efficacy of SC9-2/10th and SC9-2/40th against rMd5 were evaluated. Animal experimental results showed that SC9-2/10th and SC9-2/40th showed no lethality or tumorigenicity in SPF chicken

Purinergic signaling during Marek's disease in chickens.

Purinergic receptors (PRs) have been reported as potential therapeutic targets for many viral infections including herpesviruses, which urges the investigation into their role in Marek's disease (MD), a herpesvirus induced cancer in chickens that is an important pathogen for the poultry industry. MD is caused by MD virus (MDV) that has a similar viral life cycle as human varicella zoster virus in that it is shed from infected epithelial skin cells and enters the host through the respiratory route. In this report, PR responses during natural MDV infection and disease progression was examined in

In vitro infection of lymphocytes with Marek's disease virus.

Suspension cultures of splenic lymphocytes, incubated at 41 degrees C, became infected with Marek's disease virus (MDV) following exposure to a) other infected lymphocytes, b) infected chicken kidney monolayer cultures, or c) cell-free MDV. Both viral antigen expression and virus isolation could be demonstrated after more than 40 passages made by the addition of fresh spleen cells at 2- to 3-day intervals. Susceptibility of spleen cells from bursectomized chickens was markedly lower than that of cells from intact birds. Furthermore, when spleen cell suspensions were depleted of cells having ch

Some effects of silical treatment on Marek's disease.

Treatment of newly hatched chicks with silica by the intraperitoneal route delayed the onset of mortalities due to the JM strain of Marek's disease (MD' virus inoculated at 6 days of age. During the 88-day observation period fewer silica-treated chicks died of MD, but this difference was not usually statistically significant. Silica treatment had no effect on the susceptibility of 4-week-old birds. Silica treatment reduced the antibody response to MD but, in general, not significantly. The antibody response to bovine serum albumin was significantly enhanced if measured by the indirect hemagglu

Evaluation of the Protection Efficacy of a Serotype 1 Marek's Disease Virus-Vectored Bivalent Vaccine Against Infectious Laryngotracheitis and Marek's Disease.

Laryngotracheitis (LT) is a highly contagious respiratory disease of chickens that produces significant economic losses to the poultry industry. Traditionally, LT has been controlled by administration of modified live vaccines. In recent years, the use of recombinant DNA-derived vaccines using turkey herpesvirus (HVT) and fowlpox virus has expanded, as they protect not only against the vector used but also against LT. However, HVT-based vaccines confer limited protection against challenge, with emergent very virulent plus Marek's disease virus (vv+MDV). Serotype 1 vaccines have been proven to

Application of lentinan in suppression of Marek's disease virus infection.

Marek's disease virus (MDV) is an extremely widespread avian immunosuppressive virus. In recent years, many reports have shown that there are still cases of MDV infection and immunosuppression after immunization with the vaccine. Consequently, there is a need to develop alternative complementary approaches for strengthening the efficacy of MDV prevention and control measures. Lentinan (LNT) is a macromolecular compound with immune-enhancing activity extracted from shiitake mushrooms. To explore the value and effectiveness of administering LNT through drinking water in the prevention and contro

Increased virulence of Marek's disease virus field isolates.

The continuation of an apparent evolutionary trend of Marek's disease virus (MDV) towards greater virulence may explain recent increased losses from Marek's disease (MD) in vaccinated flocks. To address this question, the virulence of 31 isolates of serotype 1 MDV obtained from layer or broiler flocks between 1987 and 1995 were characterized. Each isolate was cultured in duck embryo fibroblasts for four to six passages, and ascertained to be free from contamination with avian retroviruses, chicken anemia virus, and MDVs of other serotypes. The viruses, along with prototype viruses JM/102W and

Occurrence of Marek's Disease in Backyard Chicken Flocks in Vietnam.

Marek's disease (MD) is a common lymphomatous and neuropathic disease of birds, especially chickens, and has caused significant losses to chicken production as a result of high mortality and morbidity. This study aims to determine the status of MD in backyard flocks in the Mekong Delta of Vietnam and to analyze clinical cases occurring during a year. The study was carried out from August 2018 to July 2019, during which time 16 suspected cases of chicken flocks with MD were observed, 40 chickens were subjected to anatomopathological examination, and PCR was performed for diagnosis of MD and det

Recombinant Marek's disease virus type 1 provides full protection against very virulent Marek's and infectious bursal disease viruses in chickens.

Marek's disease virus (MDV) is a preferred vector in the construction of recombinant vaccines. However, bivalent vaccine based on MDV that confers full protection against both very virulent Marek's and infectious bursal disease virus (IBDV) infections in chickens has not been produced. Here we developed a system utilizing overlapping fosmid DNAs transfection that rescues an MDV type 1 (MDV1) vaccine strain. Using this system, we inserted the IBDV VP2 gene at MDV1 genome sites UL41, US10 and US2. The VP2 protein was stably expressed in the recombinant MDV-infected cells and self-assembled into

Marek's Disease in a Flock of Japanese Quails (Coturnix japonica) in Germany.

A case of increased mortality was investigated at a German farm housing Japanese quails (Coturnix japonica) for egg production. Different age groups were kept in aviaries in one barn. The quail chicks had diarrhea and the adults were emaciated, some also with diarrhea and conjunctivitis. Postmortem examination showed gross tumorlike lesions in 7 of 15 adult quails examined, with ocular lesions in 2 of them. On histopathological examination, infiltrates of monomorphic round cells were found in liver, spleen, lung, and proventriculus. In the eyes of two quails, similar cells were infiltrating th

Cellular immune response to Marek's disease: listeriosis as a model of study.

The immune response of chickens to Listeria monocytogenes was studied as a potential model for cell-mediated immunocompetence. Chickens genetically resistant and susceptible to Marek's disease (MD) did not differ in their ability to survive Listeria, although during the early stages of infection the bacteria replicated more readily in MD-susceptible chickens. MD-susceptible chickens responded earlier than MD-resistant chickens, and with equal or increased intensity, in assays of various components of the cell-mediated reaction. These assays included T-cell activation, delayed-type hypersensiti

Differences in CD8αα and cecal microbiome community during proliferation and late cytolytic phases of Marek's disease virus infection are associated with genetic resistance to Marek's disease.

Marek's disease (MD) is an important neoplastic disease of chickens caused by Marek ': s disease virus (MDV), a highly oncogenic alphaherpesvirus. In this study using two chicken lines, one resistant and another susceptible to MD, splenic T cells and cecal microbiome were profiled to gain a better understanding of primary differences in these lines. The percent of splenic CD4+ T cells were similar regardless of MDV challenge status in both bird lines. In contrast, CD8αα profiles were different (P < 0.005) between chicken lines under naïve status and under MDV challenge, suggesting that CD8αα T
