Description of two novel Marinobacter species isolated from saline-alkali soil: Marinobacter alkalisoli sp. nov. and Marinobacter shunpengi sp. nov.

Four Gram-staining negative, non-motile, rod-shape bacteria, named strains GN3S48T, HN1S83, LN3S78T, and M1N3S26, were isolated from the bulk saline soils, in Baotou, China. Among them, strains GN3S48T and HN1S83 could degrade 100 mg l-1n-hexadecane as sole carbon and energy source for their growth. Phylogenetic analyses showed that the four strains always formed two distinct clades: Strain LN3S78T clustered with strain M1N3S26, and strain GN3S48T clustered with strain HN1S83. Nonetheless, all four strains tightly clustered and shared the highest 16S rRNA gene similarities with Marinobacter sp

Marinobacter liaoningensis sp. nov., isolated from the marine sediment.

A Gram-stain-negative, facultatively anaerobic, rod-shaped bacterial strain, namely F4206T, was isolated from the sediments of Beihai Red Beach, China. The optimal temperature, pH, and NaCl concentration of F4206T were 20-40 °C, pH 6.5, and 10%. The digital DNA-DNA hybridization (dDDH), average nucleotide identity (ANI) and average amino acid identity (AAI) values between strain F4206T and its closest relatives ranged from 20.5 to 44.3%, 78.2 to 80.5% and 65.0 to 94.8% respectively, which are well below the established species delineation thresholds of 70%, 95% and 95%. Genomic analysis of Marinobacter flavimaris ZYH30 reveals its role in marine dimethylsulfide cycling.

Dimethylsulfide (DMS) is a volatile organic compound that influences climate change and plays a key role in the global sulfur cycle and chemotaxis. Marine bacteria can produce DMS from dimethylsulfoniopropionate (DMSP) and other sulfur precursors. A highly efficient DMS-producing strain, Marinobacter flavimaris ZYH30, was isolated from the surface sediment of the South China Sea, but the key genes involved in DMS production in its genome remain unknown. Here, we reported the complete genome sequence of M. flavimaris ZYH30 and its genomic potential in DMS cycling. The genome of M. flavimaris ZY

Halophilic lipase does forms catalytically active aggregates: Evidence from Marinobacter sp. EMB5 lipase (LipEMB5).

Lipases are prone to aggregation due to the presence of hydrophobic surface and corresponding hydrophobic interactions. However, like most lipases, halophilic lipases do not tend to aggregate owing to the presence of charged surface. Conversely, Marinobacter sp. EMB5 produces a unique lipase (LipEMB5) which tends to form functionally active aggregate, despite a presence of 2% (w/v) NaCl in the environment. Gel filtration using Sephacryl S-200 column resulted in elution of lipase in the void volume despite a high salt level, suggesting aggregation behavior. The aggregated form of LipEMB5 (172.3

A proteinaceous antimicrobial compound isolated from Marinobacter pelagius, exhibits antagonistic activity against multidrug-resistant Salmonella paratyphi.

Antimicrobial resistance (AMR) poses a serious threat currently to human health and necessitates urgent discovery of novel antimicrobial agents. We hypothesized that the vast biodiversity of the marine environment represents a promising resource for isolating bacteria with potent antagonistic activity against multi-drug resistant pathogens. The primary objective was to purify and characterize the antibacterial protein fractions exhibiting antagonistic activity from marine bacteria. To accomplish this, the present study isolated, identified, and characterized antibacterial compounds produced by

Oligo-heterotrophic Activity of Marinobacter subterrani Creates an Indirect Fe(II) Oxidation Phenotype in Gradient Tubes.

Autotrophic bacteria utilizing Fe(II) as their energy and electron sources for growth affect multiple biogeochemical cycles. Some chemoheterotrophic bacteria have also been considered to exhibit an Fe(II) oxidation phenotype. For example, several Marinobacter strains have been reported to oxidize Fe(II) based on formation of oxidized iron bands in semi-solid gradient tubes that produce opposing concentration gradients of Fe(II) and oxygen. While gradient tubes are a simple and visually compelling method to test for Fe(II) oxidation, this method alone cannot confirm if, and to what extent, Fe(I

Biochemical characterization and mutational analysis of the tetrameric DABA transaminase EctB from the Arctic bacterium Marinobacter sp. CK1.

Ectoine is a chemical chaperone and osmoprotectant discovered in halophilic bacteria where it is used to protect from osmotic stress in high-salt environments. Ectoine has become a valuable product in pharmaceutical, biotechnological, cosmetical, and other applications due to its protective and stabilizing properties. The rate-limiting step of ectoine biosynthesis is catalyzed by the l-2,4-diaminobutyric acid (DABA) transaminase enzyme EctB, which converts aspartate-β-semialdehyde (ASA) to DABA. We studied the structural and functional properties of EctB from a novel halophilic bacterium Marin

Structure-guided evolution of Marinobacter subterrani deacetylase for enhanced herbicide precursor synthesis by substrate-binding pocket and channels engineering.

Deacetylases can directly hydrolyze amide bonds for the environmentally friendly synthesis of the herbicide intermediate O-(3-chloro-2-propenyl)hydroxylamine. However, the rigid halogenated chain of N-[(E)-3-chloroprop-2-enoxy]acetamide (NECA) may hinder productive substrate positioning and limit catalytic efficiency. Accordingly, 20 deacetylases were initially screened, and a deacetylase from Marinobacter subterrani (MsDeac) was identified as the optimal starting scaffold. To improve its catalytic performance toward NECA, a synergistic pocket and tunnel reshaping strategy was subsequently imp

Bacterial volatile organic compound specialists in the phycosphere.

Labile dissolved organic carbon in the surface oceans accounts for about one-fourth of carbon produced through photosynthesis and turns over on average every 3 days, fueling one of the largest engines of microbial heterotrophic production on the planet. Volatile organic compounds are poorly constrained components of dissolved organic carbon. Here, we detected 72 m/z signals, corresponding to unique volatile organic compounds, including petroleum hydrocarbons, totaling ~18.5 nM in the culture medium of a model diatom. In five cocultures with bacteria adapted to grow with this diatom, 1-59 m/z s

The potential for bacteria from carbon-limited deep terrestrial environments to participate in chlorine cycling.

Bacteria capable of dehalogenation via reductive or hydrolytic pathways are ubiquitous. Little is known, however, about the prevalence of bacterial dechlorination in deep terrestrial environments with a limited carbon supply. In this study we analyzed published genomes from three deep terrestrial subsurface sites: a deep aquifer in Western Siberia, the Sanford Underground Research Facility in South Dakota, USA, and the Soudan Underground Iron Mine (SUIM) in Minnesota, USA to determine if there was evidence to suggest that microbial dehalogenation was possible in these environments. Diverse deh

Quorum-sensing inhibitory compounds from extremophilic microorganisms isolated from a hypersaline cyanobacterial mat.

In this study, extremely halophilic and moderately thermophilic microorganisms from a hypersaline microbial mat were screened for their ability to produce antibacterial, antidiatom, antialgal, and quorum-sensing (QS) inhibitory compounds. Five bacterial strains belonging to the genera Marinobacter and Halomonas and one archaeal strain belonging to the genus Haloterrigena were isolated from a microbial mat. The strains were able to grow at a maximum salinity of 22-25 % and a maximum temperature of 45-60 °C. Hexanes, dichloromethane, and butanol extracts from the strains inhibited the growth of

Metal resistance genes enrichment in marine biofilm communities selected by biocide-containing surfaces in temperate and tropical coastal environments.

Microorganisms able to form biofilms in marine ecosystems are selected depending on immersed surfaces and environmental conditions. Cell attachment directly on toxic surfaces like antifouling coatings suggests a selection of tolerant (or resistant) organisms with characteristics conferring adaptive advantages. We investigated if environment would drive metal resistance gene abundance in biofilms on artificial surfaces. Biofilms were sampled from three surfaces (a PVC reference and two antifouling coatings) deployed in three coastal waters with dissimilar characteristics: The Mediterranean Sea

Microbial degradation of short-chain chlorinated paraffins in marine environment: bacterial community responses, functional genes, transformation products, and metabolic mechanisms.

Short-chain chlorinated paraffins (SCCPs) are persistent organic pollutants that pose ecological risks to marine ecosystems. Their impacts on bacterial communities and environmental fate remain poorly understood. In this study, SCCP exposure significantly altered sediment bacterial diversity and composition, enriching key genera such as Halomonas, Pseudomonas, and Marinobacter. Six SCCP-degrading strains from these genera were isolated. Whole-genome and transcriptomic analyses identified key functional genes involved in 1,2,5,6,9,10-hexachlorodecane degradation, including oxidative stress resp

Impact of mixed carbon sources on microbial function and network structure in hypersaline denitrification.

This study evaluated denitrification systems using two carbon sources: glucose alone (G), and a mixture of glucose, methanol, and sodium acetate (GMS), treating synthetic saline wastewater to prove the superior operational efficiency of the mixed carbon sources. Both systems showed excellent nitrogen removal under 80 g-NaCl L-1, but the glucose-based system exhibited unstable nitrate removal at a low C/N ratio of 3. Microbial community composition was distinct between the systems. The GMS system showed the highest organic removal due to the abundance of Marinobacter spp., and the prediction of

Poly(ethylene glycol)-containing hydrogel surfaces for antifouling applications in marine and freshwater environments.

This work describes the fabrication, characterization, and biological evaluation of a thin protein-resistant poly(ethylene glycol) (PEG)-based hydrogel coating for antifouling applications. The coating was fabricated by free-radical polymerization on silanized glass and silicon and on polystyrene-covered silicon and gold. The physicochemical properties of the coating were characterized by infrared spectroscopy, ellipsometry, and contact angle measurements. In particular, the chemical stability of the coating in artificial seawater was evaluated over a six-month period. These measurements indic

Chemical defence in mussels: antifouling effect of crude extracts of the periostracum of the blue mussel Mytilus edulis.

Shells of the blue mussel Mytilus edulis remain free of fouling organisms as long as they possess an intact periostracum, and a multiple antifouling defence that comprises a ripple-like microtopography and the production of chemical antifouling compounds has been suggested previously. This study investigates the chemical defence strategy of blue mussels for the first time. Six crude extracts of the periostracum of intact shells were made using solvents of increasing polarity. These extracts were tested against common fouling organisms in laboratory based bioassays. Non-polar and moderately pol

Biodegradation of benzene homologues in contaminated sediment of the East China Sea.

This study focused on acclimating a microbial enrichment to biodegrade benzene, toluene, ethylbenzene and xylenes (BTEX) in a wide range of salinity. The enrichment degraded 120 mg/L toluene within 5d in the presence of 2M NaCl or 150 mg/L toluene within 7d in the presence of 1-1.5M NaCl. PCR-DGGE (polymerase chain reaction-denatured gradient gel electrophoresis) profiles demonstrated the dominant species in the enrichments distributed between five main phyla: Gammaproteobacteria, Sphingobacteriia, Prolixibacter, Flavobacteriia and Firmicutes. Antarctic Salt-Cones: An Oasis of Microbial Life? The Example of Boulder Clay Glacier (Northern Victoria Land).

The evaporation of a localized, highly saline water body of the Boulder Clay debris-covered glacier, in the Northern Victoria Land, probably generated the accumulation of mirabilite (Na2SO4 × 10H2O) and thenardite (Na2SO4) in a glacier salt-cone. Such an extremely cold and salty environment resembles the conditions on Mars, so it can be considered a terrestrial analog. The study was aimed at gaining a first glimpse at the prokaryotic community associated with Antarctic mirabilite and thenardite minerals and also to find clues about the origin of the salts. For this purpose, samples were analyz

Growth and Toxin Production of Gambierdiscus spp. Can Be Regulated by Quorum-Sensing Bacteria.

Gambierdiscus spp. are the major culprit responsible for global ciguatera fish poisoning (CFP). At present, the effects of microbiological factors on algal proliferation and toxin production are poorly understood. To evaluate the regulatory roles of quorum-sensing (QS) bacteria in the physiology of Gambierdiscus, co-culture experiments with screened QS strains were conducted in this study. Except for the growth-inhibiting effect from the strain Marinobacter hydrocarbonoclasticus, the algal host generally displayed much higher growth potential and toxin production ability with the existence of
