Structure-activity relationship and hypoglycemic activity of tricyclic matrines with advantage of treating diabetic nephropathy.

Forty-three tricyclic matrinic derivatives with a unique scaffold were prepared and evaluated for their stimulation effects on glucose consumption in HepG2 cells. The structure-activity relationship was systematically elucidated for the first time. Among them, compound 17a exhibited the most promising potency, and dose-dependently increased glucose consumption in L6 myotubes. It significantly lowered blood glucose, glucosylated haemoglobin and AGE level, and improved glucose tolerance and insulin resistance in KK-Ay mice as well. More importantly, 17a effectively ameliorated diabetic nephropat

[The pharmacological studies on matrine and oxymatrine].

Matrine and oxymatrine, major components of Sophora flavescens, were confirmed to have a protective effect on restraint and water immersion stress ulcer in mice. Oxymatrine inhibits the formation of some experimental gastric ulcer, pylorus ligation ulcer and indomethacin ulcer, which are considered to relate to acid secretion. Oxymatrine decreases acid secretion in Shay's rats and inhibits gastric motility induced by restraint and water immersion stress when administered intraduodenally. These results suggest that the protective effect of oxymatrine on stress ulcer is possibly due to a decreas

Ocular anti-inflammatory actions of matrine.

Matrine is an alkaloid isolated from the root of Sophora subprostrata (Leguminosae) which has been used as a Chinese medicine for the treatment of inflammation. Instillation of 50 microliter of 1% matrine showed significant inhibition of ocular inflammation induced by lens proteins. Unlike corticosteroids, matrine did not facilitate the IOP recovery in rabbit eyes nor did it change the electrical potential difference across rabbit iris-ciliary body. These results indicate that matrine could become a safer ocular antiinflammatory agent than corticosteroids. Further, matrine was found to markedl

Matrin 3 in neuromuscular disease: physiology and pathophysiology.

RNA-binding proteins (RBPs) are essential factors required for the physiological function of neurons, muscle, and other tissue types. In keeping with this, a growing body of genetic, clinical, and pathological evidence indicates that RBP dysfunction and/or gene mutation leads to neurodegeneration and myopathy. Here, we summarize the current understanding of matrin 3 (MATR3), a poorly understood RBP implicated not only in ALS and frontotemporal dementia but also in distal myopathy. We begin by reviewing MATR3's functions, its regulation, and how it may be involved in both sporadic and familial

Synthesis of a temperature-sensitive matrine-imprinted polymer and its potential application for the selective extraction of matrine from radix Sophorae tonkinensis.

A temperature-sensitive matrine-imprinted polymer was prepared in chloroform by free-radical cross-linking copolymerization of methacrylic acid at 60 °C in the presence of ethylene glycol dimethacrylate as the cross-linker, N-isopropyl acrylamide as the temperature-responsive monomer and matrine as the template molecule. Binding experiments and Scatchard analyses revealed that two classes of binding sites were formed on molecular imprinted polymer (MIP) at 50 °C. Additionally, the thermoresponsive MIP was tested for its application as a sorbent material for the selective separation of matrine

Matrin 3 variants are frequent in Italian ALS patients.

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disorder characterized by loss of motor neurons in the primary motor cortex, brainstem, and spinal cord. Recently, missense variants in MATR3 were identified in familial and sporadic ALS patients, but very few additional ALS patients have been reported so far. The p.S85C MATR3 variant was previously associated to a different phenotype, namely a distal myopathy associated with dysphagia and dysphonia. Here, we assessed the contribution of MATR3 variants in a cohort of 322 Italian ALS patients. We identified 5 different missense MATR3 va

Matrine and oxymatrine: a comprehensive review of multitarget mechanisms and therapeutic potential.

In recent years, research of matrine (MAT) and oxymatrine (OMT) has shifted from merely discovering pharmacological activities to exploring mechanisms, expanding new indications, and improving formulations. However, comparative analyses of their similarities and differences remain limited. Beyond their well-known functions in cancer treatment-such as regulating non-coding RNAs, preventing metastasis, and triggering apoptosis and ferroptosis-these compounds also show notable immunomodulatory effects, particularly in synergizing with immune checkpoint inhibitors to enhance tumor responsiveness.
