Cellular responses to beating hydrogels to investigate mechanotransduction.

Cells feel the forces exerted on them by the surrounding extracellular matrix (ECM) environment and respond to them. While many cell fate processes are dictated by these forces, which are highly synchronized in space and time, abnormal force transduction is implicated in the progression of many diseases (muscular dystrophy, cancer). However, material platforms that enable transient, cyclic forces in vitro to recreate an in vivo-like scenario remain a challenge. Here, we report a hydrogel system that rapidly beats (actuates) with spatio-temporal control using a near infra-red light trigger. Sma

Model of cellular mechanotransduction via actin stress fibers.

Mechanical stresses due to blood flow regulate vascular endothelial cell structure and function and play a key role in arterial physiology and pathology. In particular, the development of atherosclerosis has been shown to correlate with regions of disturbed blood flow where endothelial cells are round and have a randomly organized cytoskeleton. Thus, deciphering the relation between the mechanical environment, cell structure, and cell function is a key step toward understanding the early development of atherosclerosis. Recent experiments have demonstrated very rapid (∼100 ms) and long-distance

SERPINE1 (PAI-1) Regulation in Mechanotransduction-Associated Cellular Senescence.

Cellular senescence is a complex biological process characterized by irreversible cell-cycle arrest and acquisition of the senescence-associated secretory phenotype (SASP). Beyond biochemical stress, increasing evidence indicates that mechanical cues from the extracellular microenvironment contribute to the regulation of senescence; however, the molecular mechanisms linking mechanotransduction to senescent phenotypes remain incompletely understood. SERPINE1 (plasminogen activator inhibitor-1, PAI-1) has emerged as a key mechanosensitive effector induced downstream of mechanotransduction signal

Static and Dynamic: Evolving Biomaterial Mechanical Properties to Control Cellular Mechanotransduction.

The extracellular matrix (ECM) is a highly dynamic system that constantly offers physical, biological, and chemical signals to embraced cells. Increasing evidence suggests that mechanical signals derived from the dynamic cellular microenvironment are essential controllers of cell behaviors. Conventional cell culture biomaterials, with static mechanical properties such as chemistry, topography, and stiffness, have offered a fundamental understanding of various vital biochemical and biophysical processes, such as cell adhesion, spreading, migration, growth, and differentiation. At present, novel

Biochemical Pathways of Cellular Mechanosensing/Mechanotransduction and Their Role in Neurodegenerative Diseases Pathogenesis.

In this review, we shed light on recent advances regarding the characterization of biochemical pathways of cellular mechanosensing and mechanotransduction with particular attention to their role in neurodegenerative disease pathogenesis. While the mechanistic components of these pathways are mostly uncovered today, the crosstalk between mechanical forces and soluble intracellular signaling is still not fully elucidated. Here, we recapitulate the general concepts of mechanobiology and the mechanisms that govern the mechanosensing and mechanotransduction processes, and we examine the crosstalk b

A SUN1-ApoD feedback loop promotes cellular aging via microtubule-nuclear mechanotransduction.

Alterations in nuclear envelope proteins such as SUN1 and prelamin A are known hallmarks of cellular aging, but how they cause aging phenotypes beyond the expressing cells is unknown. With a tissue culture version of parabiosis, we found that aged fibroblasts secrete an activity that induces aging-related polarity defects in young fibroblasts. Secretomics identified the factor as apolipoprotein D (ApoD), a circulating protein whose levels are known to rise with age. ApoD increased SUN1 levels and disrupted polarity in fibroblasts via SUN1-promoted coupling of microtubules to the nucleus. In tu

Vibroacoustic disease: biological effects of infrasound and low-frequency noise explained by mechanotransduction cellular signalling.

At present, infrasound (0-20 Hz) and low-frequency noise (20-500 Hz) (ILFN, 0-500 Hz) are agents of disease that go unchecked. Vibroacoustic disease (VAD) is a whole-body pathology that develops in individuals excessively exposed to ILFN. VAD has been diagnosed within several professional groups employed within the aeronautical industry, and in other heavy industries. However, given the ubiquitous nature of ILFN and the absence of legislation concerning ILFN, VAD is increasingly being diagnosed among members of the general population, including children. VAD is associated with the abnormal gro

Controllable ligand spacing stimulates cellular mechanotransduction and promotes stem cell osteogenic differentiation on soft hydrogels.

Hydrogels with tunable mechanical properties have provided a tremendous opportunity to regulate stem cell differentiation. Hydrogels with osteoid (about 30-40 kPa) or higher stiffness are usually required to induce the osteogenic differentiation of mesenchymal stem cells (MSCs). It is yet difficult to achieve the same differentiation on very soft hydrogels, because of low environmental mechanical stimuli and restricted cellular mechanotransduction. Here, we modulate cellular spatial sensing of integrin-adhesive ligands via quasi-hexagonally arranged nanopatterns to promote cell mechanosensing

Packing-driven mechanotransduction: local crowding overrides adhesion and stiffness cues for YAP activation in cellular collectives.

The regulation of mechanotransduction is crucial for various cellular processes, including stem cell differentiation, wound healing and cancer progression. While the activation of mechanotransduction has been extensively studied in single cells, it remains unclear whether similar mechanisms extend to mechanotransduction in multicellular collectives. Here, by focusing on Yes-associated protein (YAP), known as the master regulator of mechanotransduction, we reveal that the local packing fraction of cells acts as the primary determinant of YAP activation in cell collectives. We further show that

Substrate stiffness orchestrates epithelial cellular heterogeneity with controlled proliferative pattern via E-cadherin/β-catenin mechanotransduction.

UNLABELLED: Epithelial cellular heterogeneity has been observed in pathological tissues with abnormal matrix stiffness and cells cultured on rigid substrates. However, it remains unclear how matrix stiffness influences cellular heterogeneity formation in multi-cellular population. Here, we demonstrated that cellular heterogeneity regulated by substrate stiffness is evident starting from the initial single-cell stage (indicated by cellular Young's modulus and morphology) until the resulting multi-cellular stage (indicated by cellular functions) through distinguished proliferative patterns. Analysis of bone architecture sensitivity for changes in mechanical loading, cellular activity, mechanotransduction, and tissue properties.

Bone has an architecture which is optimized for its mechanical environment. In various conditions, this architecture is altered, and the underlying cause for this change is not always known. In the present paper, we investigated the sensitivity of the bone microarchitecture for four factors: changes in bone cellular activity, changes in mechanical loading, changes in mechanotransduction, and changes in mechanical tissue properties. The goal was to evaluate whether these factors can be the cause of typical bone structural changes seen in various pathologies. For this purpose, we used an establi

Biomimetic photonic crystal cellular force microscopy resolves spatiotemporal force dynamics and mechanotransduction during epithelial-mesenchymal transition.

Epithelial-mesenchymal transition (EMT) is a fundamental biological process driving kidney fibrosis. Although biochemical networks are well defined, clinical interventions targeting these pathways often show limited efficacy. Emerging evidence confirms the vital role of cellular forces and the physical microenvironment (e.g., matrix stiffness) in EMT progression. Yet, traditional probing methods such as traction force microscopy (TFM) and atomic force microscopy (AFM) frequently neglect this aspect. Such techniques are also hindered by their requirement for exogenous labels or invasive probes.

Loss of novel mechanosensitive ion channel TMEM63A impairs osteogenic differentiation and proliferation via disruption of cellular mechanotransduction.

Skeletal homeostasis depends critically on the local mechanobiological microenvironment. While well-known mechanosensitive ion channels like Piezo1 and Piezo2 translate physical forces into immediate membrane ion fluxes, the proximal sensors regulating medium- and long-term osteogenic pathways remain poorly understood. Low-intensity pulsed ultrasound (LIPUS) is widely used clinically to accelerate fresh fracture repair, yet its underlying upstream mechanotransduction machinery is not fully characterized. Here, we identify TMEM63A - a stretch-activated cation channel - as a crucial mechanosenso

Peyronie's disease and mechanotransduction: an in vitro analysis of the cellular changes to Peyronie's disease in a cell-culture strain system.

INTRODUCTION: Penile traction therapy (PTT) has gained considerable popularity as a noninvasive treatment option in Peyronie's disease (PD) to improve penile curvature and length. However the exact mechanism of action of PTT is unknown and as yet unproven. AIM: Using a cyclic strain culture system, we investigate the cellular changes to the tunical tissues following equibiaxial tractional forces. The observed protein alteration following mechanical traction should replicate the changes observed in tunical tissue following PTT. METHODS: Flexcell FX-5000(TM) Tension Plus system (Flexcell Inter

Molecular Mechanisms of Endothelial Shear Stress Mechanotransduction in Health and Disease.

Endothelial cells translate hemodynamic forces into biochemical signals that regulate vascular homeostasis through a process called mechanotransduction. Over the past decades, the study of this physiological process has received significant attention, leading to the identification of molecular structures that detect changes in shear stress. However, the functional capacity of these mechanosensors is compromised under conditions such as obesity, diabetes, and aging, resulting in impaired mechanotransduction. Such impaired mechanotransduction leads to endothelial dysfunction, a key early compone

Identification of Elongated Primary Cilia with Impaired Mechanotransduction in Idiopathic Scoliosis Patients.

The primary cilium is an outward projecting antenna-like organelle with an important role in bone mechanotransduction. The capacity to sense mechanical stimuli can affect important cellular and molecular aspects of bone tissue. Idiopathic scoliosis (IS) is a complex pediatric disease of unknown cause, defined by abnormal spinal curvatures. We demonstrate significant elongation of primary cilia in IS patient bone cells. In response to mechanical stimulation, these IS cells differentially express osteogenic factors, mechanosensitive genes, and signaling genes. Considering that numerous ciliary g

Mechanotransduction gone awry.

Cells sense their physical surroundings through mechanotransduction - that is, by translating mechanical forces and deformations into biochemical signals such as changes in intracellular calcium concentration or by activating diverse signalling pathways. In turn, these signals can adjust cellular and extracellular structure. This mechanosensitive feedback modulates cellular functions as diverse as migration, proliferation, differentiation and apoptosis, and is crucial for organ development and homeostasis. Consequently, defects in mechanotransduction - often caused by mutations or misregulatio

Barrier organ mechanomedicine and therapeutic insights.

Mechanical forces regulate development, homeostasis, and repair in the skin, lung, and cornea-external barrier organs that are exposed to stretch, shear, and stiffness. Dysregulated mechanotransduction drives fibrosis, inflammation, and impaired repair via conserved pathways [Piezo1 (Piezo-type mechanosensitive ion channel 1), TRPV4 (transient receptor potential vanilloid 4), and integrin-YAP (Yes-associated protein)]. Targeting these circuits with small molecules, biologics, or stiffness-tuned biomaterials offers a novel category of cross-organ therapies. As mechanosensitive pathways and mech

Building a tissue: Mesenchymal and epithelial cell spheroids mechanical properties at micro- and nanoscale.

Cell transitions between the epithelial and mesenchymal phenotypes provide the regulated morphogenesis and regeneration throughout the ontogenesis. The tissue mechanics and mechanotransduction play an essential role in these processes. Cell spheroids reproduce the cell density of native tissues and represent simple building blocks for the tissue engineering purposes. The mechanical properties of mesenchymal and epithelial cells have been extensively studied in 2D monolayer cultures, but have not been sufficiently compared in spheroids. Here, we have simultaneously applied several techniques to
