3D MicroCT Imaging of Medicago sativa Root Nodules.

The symbiotic relationship between the legume Medicago sativa and the soil bacteria Sinorhizobium meliloti results in the formation of nitrogen-fixing root nodules. Traditional destructive methods, including paraffin sectioning, vibratome sectioning, and cryosectioning, have been applied to visualize how bacteria occupy the nodule, making it extremely difficult to obtain reliable three-dimensional information. These approaches are often combined with fluorescent labeling or staining, which can introduce additional stress affecting plant growth and nodule formation. MicroCT has emerged as a rel

Cellular damage induced by cadmium and mercury in Medicago sativa.

Alfalfa (Medicago sativa) plantlets were exposed to Cd or Hg to study the kinetics of diverse stress indexes. In the so-called beaker-size hydroponic system, plantlets were grown in 30 microM of Cd or Hg for 7 d. Oxidative stress took place and increased over time, a linear response being observed with Cd but not with Hg. To improve the sensitivity of the stress assays used, a micro-assay system, in which seedlings were exposed for 24 h, was developed. Phytotoxicity of metals, quantified as growth inhibition, was observed well before there was any change in the non-protein thiol tissue concent

Anti-melanogenic effects of Medicago sativa seed extracts on melanocytes.

OBJECTIVE: Alfalfa (Medicago sativa) is a prominent herbal treatment among Asian countries and its antioxidant and anti-inflammatory effects have already been generally elucidated. Excessive melanin synthesis is one of the major troubles in the cosmetics industry, thus such research has been extensively described. Here, we investigated the anti-melanogenic effects and molecular mechanisms of two types of alfalfa extracts: alfalfa ethanol precipitate (AEP) and alfalfa ethanol supernatant (AES). METHODS: The chemical composition of AEP and AES was analysed using HPAEC-PAD and LC-MS/MS. B16F10 c

Adenylate cyclase activity in a higher plant, alfalfa (Medicago sativa).

An adenylate cyclase activity in Medicago sativa L. (alfalfa) roots was partially characterized. The enzyme activity remains in the supernatant fluid after centrifugation at 105,000 g and shows in crude extracts an apparent Mr of about 84,000. The enzyme is active with Mg2+ and Ca2+ as bivalent cations, and is inhibited by EGTA and by chlorpromazine. Calmodulin from bovine brain or spinach leaves activates this adenylate cyclase.

The utilization of chopped and pelleted lucerne (Medicago sativa) by growing lambs.

1. The efficiency of utilization of the dietary energy and nitrogen contained in a dried lucerne (Medicago sativa cv. Chartainvilliers) given either chopped (CL) or ground (1.96 mm screen) and pelleted (PL), was measured in a comparative slaughter experiment. Growing lambs were given equal amounts of digestible energy in the chopped or pelleted form at each of the three planes of nutrition for a period of 100 d. 2. The initial energy, fat and protein content of both the carcass and the total body of the test lambs was estimated from regression equations between fasted (18 h) live weight and th

Genome-wide identification of rhabdoviral sequences in alfalfa (Medicago sativa L.).

We recently reported the identification of endogenous viral elements (EVEs) originating from the Caulimoviridae family within the alfalfa (Medicago sativa L.) genome. Our subsequent identification of ubiquitous rhabdoviral elements in infected and healthy alfalfa tissues by high-throughput sequencing suggested that the alfalfa genome might be populated with integrated rhabdoviruses as well. Bioinformatics analysis using 26 publicly available alfalfa genomes proved the suggestion accurate. We found multiple nonretroviral segments of the Rhabdoviridae family belonging to the genera Betanucleorha

Diversity of Sinorhizobium strains nodulating Medicago sativa from different Iranian regions.

Alfalfa is believed to have originated in north-western Iran and has a long history of coexistence with its bacterial symbiont Sinorhizobium in soils of Iran. However, little is known about the diversity of Sinorhizobium strains nodulating Iranian alfalfa genotypes. In this study, Sinorhizobium populations were sampled from eight different Iranian sites using three cultivars of Medicago sativa as trap host plants. A total of 982 rhizobial strains were isolated and species were identified showing a large prevalence of Sinorhizobium meliloti over Sinorhizobium medicae. Analysis of salt tolerance

Improved chromium tolerance of Medicago sativa by plant growth-promoting rhizobacteria (PGPR).

BACKGROUND: Soil pollution by heavy metals increases the bioavailability of metals like hexavalent chromium (Cr (VI)), subsequently limiting plant growth and reducing the efficiency of phytoremediation. Plant growth-promoting rhizobacteria (PGPR) have substantial potential to enhance plant growth as well as plant tolerance to metal stress. The aim of this research was to investigate Cr (VI) phytoremediation enhancement by PGPR. RESULTS: The results showed that the 27 rhizobacterial isolates studied were confirmed as Cr (VI)-resistant PGPR, by using classical biochemical tests (phosphate solub

Gene expression and characterization of an antimicrobial peptide from Medicago sativa "Sazova" cultivar.

In recent years, the discovery of new antimicrobial agents has become necessary because of the increase in antibiotic resistance, the development of herbicides and fungicides resistance. Among the antimicrobial agents, antimicrobial peptides (AMPs) stand out due to their stable structure. In this study, the aim was to identify a thermostable AMP from the seeds of M. sativa "Sazova" cultivar and to analyze gene expression during germination. Antimicrobial tests were performed for the seed peptides after heat treatment (85 °C for 10 min), revealing antimicrobial effects against S. aureus, E. col

Arbuscular mycorrhizal fungus regulates cadmium accumulation, migration, transport, and tolerance in Medicago sativa.

Cadmium (Cd) pollution in croplands is a global environmental problem. Measures to improve the tolerance of sensitive crops and reduce pollutant absorption and accumulation are needed in contaminated agricultural areas, and inoculation with rhizosphere microorganisms to regulate plant resistance and heavy metal transport can provide an effective solution. A pot experiment was conducted to analyse the impact of arbuscular mycorrhizal fungi (AMF) on alfalfa oxidase activity, heavy metal resistance genes and transport proteins, metabolism, and other biochemical regulation mechanisms that lead to

Perfluorooctanoic Acid Uptake by Alfalfa (Medicago sativa) and Bioavailability in Sprague-Dawley Rats.

ABSTRACT: Perfluorooctanoic acid (PFOA) is a perfluorinated alkyl substance used as a surfactant in a wide variety of industrial and consumer products. Over the past decade, concern has increased over the presence of PFOA in biosolids from wastewater treatment plants used as fertilizer on agricultural lands because of the potential for PFOA to enter the food chain. In this study, the uptake of 14C-PFOA from soil by alfalfa and 14C-PFOA bioavailability from consumption of this alfalfa was evaluated in Sprague-Dawley rats. Alfalfa leaves accumulated 14C-PFOA up to 4 to 5 μg/g of dry leaf, approx

Characterization of a novel green-revertible albino mutant in alfalfa (Medicago sativa L.).

High-temperature-sensitive leaf color mutants are ideal materials for studying photosynthetic pigment biosynthesis and corresponding response mechanisms under heat stress. Here, we provide the first report of albinism occurrence in alfalfa and characterize the high-temperature albino regreen (har) mutant of alfalfa, which presents albino leaves when exposed to temperatures ≥35 °C and is not specific to developmental stage. Genetic analysis demonstrated that the albino trait exhibits dominant inheritance. Agronomic trait evaluations revealed that the har mutants were slightly but negatively aff

Pancreatic and extra-pancreatic effects of the traditional anti-diabetic plant, Medicago sativa (lucerne).

Medicago sativa (lucerne) is used as a traditional plant treatment of diabetes. In the present study, administration of lucerne in the diet (62.5 g/kg) and drinking water (2.5 g/l) reduced the hyperglycaemia of streptozotocin-diabetic mice. An aqueous extract of lucerne (1 mg/ml) stimulated 2-deoxy-glucose transport (1.8-fold), glucose oxidation (1.7-fold) and incorporation of glucose into glycogen (1.6-fold) in mouse abdominal muscle. In acute 20 min tests, 0.25-1 mg/ml aqueous extract of lucerne evoked a stepwise 2.5-6.3-fold stimulation of insulin secretion from the BRIN-BD11 pancreatic B-c

Protective role of lycopene against salinity-induced oxidative stress in Medicago sativa L. seedlings.

Soil salinization is one of the major challenges to agriculture, reducing productivity and directly affecting plant growth, germination, and physiology, especially in economically important crops such as Medicago sativa. Conventional soil management methods are not always sufficient to mitigate the adverse effects of salinity, highlighting the need for alternative and sustainable strategies. This study evaluated the phytoprotective potential of lycopene (LCP) against NaCl-induced stress in M. sativa. Seeds were treated with 256 µg/mL and 512 µg/mL of LCP combined with 50 mM NaCl and compared t

Using Medicago sativa L. Callus Cell Extract for the Synthesis of Gold and Silver Nanoparticles.

Gold (Au) and silver (Ag) nanoparticles (NPs) are used for drug transport and plant protection due to their insoluble nature and unique properties. To produce health-friendly NPs, toxic solvents should be replaced with plant-based synthesis. Plants, such as alfalfa (Medicago sativa L.), release biomolecules that reduce metal ions and form nanoclusters without free radicals, showing anti-inflammatory and antioxidant properties. In this study, callus cultures of two M. sativa genotypes, 'Kometa' and 'La Bella Campagnola', were exposed to two precursors (AgNO3 and HAuCl4) for 24 and 48 h to asses
