Karrikin (KAR) signaling plays crucial roles in plant development, regulating key traits such as photomorphogenesis, root hair development, and arbuscular mycorrhizal symbiosis. SUPPRESSOR OF MAX2 1 (SMAX1), a key negative regulator of the KAR pathway, has been functionally studied in several plant species. However, the SMAX1 protein remains poorly understood in Medicago truncatula, an important model legume, which limits the comprehensive understanding and application of the KAR pathway in legumes. In this study, we identified the single SMAX1 ortholog MtSMAX1 in M. truncatula. Sequence align

Symbiotic nitrogen (N) fixation entails successful interaction between legume hosts and rhizobia that occur in specialized organs called nodules. N-fixing legumes have a higher demand for phosphorus (P) than legumes grown on mineral N. Medicago truncatula is an important model plant for characterization of effects of P deficiency at the molecular level. Hence, a study was carried out to address the alteration in metabolite levels of M. truncatula grown aeroponically and subjected to 4 weeks of P stress. First, GC-MS-based untargeted metabolomics initially revealed changes in the metabolic prof

INTRODUCTION: Antimicrobial peptides (AMPs), key components of innate immunity, offer broad-spectrum activity against diverse pathogens. In Medicago truncatula, over 700 nodule-specific cysteine-rich (NCR) peptides with highly diverse sequences and physicochemical properties are produced in the symbiotic cells of root nodules, where cationic members block bacterial cell division and display potent antimicrobial activity in vitro. In contrast, anionic NCRs typically lack antimicrobial effects, and NCR147-a neutral peptide-is the only known non-cationic NCR that shows weak bactericidal activity.

This section details the optimized methods for single-cell RNA sequencing (scRNA-seq) in Medicago truncatula roots and nodules. We describe a detailed experimental workflow, comprising plant growth and bacteria application, protoplast isolation, quality control, and library processing according to 10× Genomics®, suitable for both long- and short-read sequencing. This protocol can serve as a starting point for establishing robust, high-quality scRNA-seq workflows to study nodulation and root development in legumes.

Mercury (Hg) contamination is increasing worldwide in both wild ecosystems and agricultural soils due to natural processes, but mostly to anthropic activities. The molecular mechanisms involved in Hg toxicity and tolerance in plants have been extensively studied; however, the role of flavonoids in response to Hg stress remains to be investigated. We conducted a metabolomic study to analyze the changes induced at the secondary metabolite level in three Hg-tolerant and one Hg-sensitive Medicago truncatula cultivars. A total of 46 flavonoid compounds, classified into five different flavonoid fami

Compound leaves show more complex patterns than simple leaves, and this is mainly because of a specific morphogenetic process (leaflet initiation and arrangement) that occurs during their development. How the relevant morphogenetic activity is established and modulated to form a proper pattern of leaflets is a central question. Here we show that the trifoliate leaf pattern of the model leguminous plant Medicago truncatula is controlled by the BEL1-like homeodomain protein PINNATE-LIKE PENTAFOLIATA1 (PINNA1). We identify PINNA1 as a determinacy factor during leaf morphogenesis that directly rep

Nitrate reductase (NR) is a key enzyme in nitrate assimilation, yet its function within nodules remains poorly understood. In Medicago truncatula, 3 NR genes, MtNR1, MtNR2, and MtNR3, exhibit distinct evolutionary origins and regulatory features. Phylogenetic analyses indicate that NR3-type genes, originated from a duplication of NR1 within the inverted repeat-lacking clade (IRLC) legumes, have lost the conserved phosphorylation sites critical for post-translational regulation. To assess the functional significance of these isoforms, we characterized single and double nr mutants obtained throu

The symbiosome, a temporary plant organelle enabling nitrogen fixation in legume-rhizobia symbiosis, consists of a plant-derived symbiosome membrane (SM), symbiosome space (SS), and enclosed bacteroid. Here, we isolate and purify symbiosomes from Medicago truncatula-Sinorhizobium meliloti root nodules and perform label-free quantitative mass spectrometry to profile protein abundances in the symbiosomes. We identify 1,018 M. truncatula proteins, including 829 in the SM and 457 in the SS. Combined with transport assays, our data reveal multiple dicarboxylate transporters in the SM that potential

The milestone of compound leaf development is the generation of separate leaflet primordia during the early stages, which involves two linked but distinct morphogenetic events: leaflet initiation and boundary establishment for leaflet separation. Although some progress in understanding the regulatory pathways for each event have been made, it is unclear how they are intrinsically coordinated. Here, we identify the PINNATE-LIKE PENTAFOLIATA2 (PINNA2) gene encoding a newly identified GRAS transcription factor in Medicago truncatula. PINNA2 transcripts are preferentially detected at organ boundar

One or more effectors in the labial saliva (LS) of generalist Noctuid caterpillars activate plant signaling pathways to modulate jasmonate (JA)-dependent defense responses; however, the exact mechanisms involved have yet to be elucidated. A potential candidate in this phytohormone interplay is the ethylene (ET) signaling pathway. We compared the biochemical and molecular responses of the model legume Medicago truncatula and the ET-insensitive skl mutant to herbivory by fourth instar Spodoptera exigua (Hübner) caterpillars with intact or impaired LS secretions. Cellular oxidative stress increas

The formation of nitrogen-fixing symbiotic nodules is a complex process that involves extensive transcriptional reprogramming of legume roots to enable intracellular accommodation of host compatible rhizobia. This process requires coordinated regulation of hundreds to thousands of genes, including the induction of highly expressed genes, traditionally referred to as nodulin genes. While the functions of many nodulins have been elucidated, a substantial number remained poorly characterized, or their functions are still unclear. One such gene, NOD25, was originally identified based on differenti

Ethylene is a well-established negative regulator of nodulation, yet how ethylene biosynthesis and perception are spatially coordinated during early symbiotic signaling remains unresolved. Here, we investigate the dynamics of ethylene responses in Medicago (Medicago truncatula) using transcriptomics, promoter-reporter analyses, loss-of-function approaches and a synthetic reporter. We show that the activity of the ethylene-responsive EBSn reporter shifts from inner root tissues under nonsymbiotic conditions to the outer cortex and epidermis following rhizobial inoculation, revealing a spatial r

Plant growth and development are intricately regulated by molecular mechanisms, with the mitogen-activated protein kinase (MAPK) signaling cascade and its associated modules being pivotal. In this study, we identified and characterized a member of the MAPKKK family, MtMAP3Kδ6, from Medicago truncatula. This gene, classified within the B3 subgroup of the MAPKKK family, was expressed across various tissues during plant growth. The knockout mutant of MtMAP3Kδ6 displayed dwarfism, characterized by reduced branching and smaller leaf size, whereas overexpression of MtMAP3Kδ6 in Medicago truncatula l

Legumes have unique features, such as compound inflorescences and a complex floral ontogeny. Thus, the study of regulatory genes in these species during inflorescence and floral development is essential to understand their role in the evolutionary origin of developmental novelties. The SUPERMAN (SUP) gene encodes a C2H2 zinc-finger transcriptional repressor that regulates the floral organ number in the third and fourth floral whorls of Arabidopsis thaliana. In this work, we present the functional characterization of the Medicago truncatula SUPERMAN (MtSUP) gene based on gene expression analysi

Diverse forms of leaves are present in nature. However, the regulatory mechanisms that underpin the development of diverse leaf forms remain enigmatic. The initiation of leaf primordia from the periphery of shoot apical meristem (SAM) requires downregulation of the class 1 knotted-like homeobox KNOXI proteins. In plants with simple leaves, this downregulation is permanent, consistent with leaves being determinant organs. In most of plants with compound leaves, the KNOXI proteins are reactivated in developing leaf primordia, and this reactivation is required for the development of compound leav

Direct and indirect defences against feeding induced by chewing (Spodoptera littoralis) and piercing-sucking (Tetranychus urticae) herbivores, as well as components of signal transduction, were investigated in the model legume Medicago truncatula. Emitted volatiles, representing a mechanism of indirect defence, were measured and identified by gas chromatography/mass spectrometry (GC-MS). As elements of direct defence, the accumulation of phenolic compounds and of reactive oxygen species (ROS) was assessed using microscopic techniques. Gibberellin (GA) plays a controversial role in the legume-rhizobium symbiosis. Recent studies have shown that the GA level in legumes must be precisely controlled for successful rhizobial infection and nodule organogenesis. However, regulation of the GA level via catabolism in legume roots has not been reported to date. Here, we investigate a novel GA inactivating C20-GA2-oxidase gene MtGA2ox10 in Medicago truncatula. RNA sequencing analysis and quantitative polymerase chain reaction revealed that MtGA2ox10 was induced as early as 6 h post-inoculation (hpi) of rhizobia and reached peak transcr

Plant height is a vital agronomic trait that greatly determines crop yields because of the close relationship between plant height and lodging resistance. Legumes play a unique role in the worldwide agriculture; however, little attention has been given to the molecular basis of their height. Here, we characterized the first dwarf mutant mini plant 1 (mnp1) of the model legume plant Medicago truncatula. Our study found that both cell length and the cell number of internodes were reduced in a mnp1 mutant. Using the forward genetic screening and subsequent whole-genome resequencing approach, we c

Defense pathways and stress responses induced under Cd stress were illustrated in roots of hydroponically grown Medicago truncatula seedlings. Actually, the ascorbate-glutathione and antioxidative system, secondary metabolism events including peroxidases, phenolic compounds, and lignification launching, and developmental modifications were described. Cd (100 μM) initially increased reactive oxygen species, enhanced antioxidative (total SOD, CAT, and PRX) and ascorbate-glutathione-related metabolism enzymes (APX and MDAR), except in A17 and TN1.11. Aphids cause significant yield losses in agricultural crops worldwide. Medicago truncatula, a model legume, cultivated pasture species in Australia and close relative of alfalfa (Medicago sativa), was used to study the defence response against Therioaphis trifolii f. maculate [spotted alfalfa aphid (SAA)]. Aphid performance and plant damage were compared among three accessions. A20 is highly susceptible, A17 has moderate resistance, and Jester is strongly resistant. Subsequent analyses using A17 and A20, reciprocal F1s and an A17×A20 recombinant inbred line (RIL) population revealed that this
