Melatonin, melatonin receptors and melanophores: a moving story.

Melatonin (5-methoxy N-acetyltryptamine) is a hormone synthesized and released from the pineal gland at night, which acts on specific high affinity G-protein coupled receptors to regulate various aspects of physiology and behaviour, including circadian and seasonal responses, and some retinal, cardiovascular and immunological functions. In amphibians, such as Xenopus laevis, another role of melatonin is in the control of skin coloration through an action on melanin-containing pigment granules (melanosomes) in melanophores. In these cells, very low concentrations of melatonin activate the Mel(1

Melanophore: A biosensor for water pollution analysis.

Chemical pollutants bioaccumulate in aquatic animals and can reach humans, causing adverse health effects. Pollution of aquatic ecosystems from industrialization and anthropogenic xenobiotics is a global issue. Poikilothermic animals possess pigment cells called integumentary melanophores, derived from neural crest cells, which contain melanin and can detect environmental pollution. These melanophores serve as effective bioassay tools for pharmacological drugs and chemical contaminants. Studies show pollutants like Aroclor 1242 and naphthalene influence crustacean color changes, indicating pol

Evidence for separate receptors for melanophore stimulating hormone and catecholamine regulation of cyclic AMP in the control of melanophore responses.

1. Skins of the lizard Anolis carolinensis darken in vitro in response to melanophore stimulating hormone (MSH), a peptide hormone, as well as to catecholamines. These hormones darken Anolis skins by dispersing the sub-cellular organelle, the melanosome, out into the dendritic processes of the dermal melanophores.2. Dibutyryl cyclic AMP and methylxanthines also darken skins. In addition, methylxanthines are synergistic with both catecholamines and MSH in causing skin darkening. These data suggest that the dispersion of melanosomes within melanophores in response to both MSH and catecholamines

Melatonin agonists induce phosphoinositide hydrolysis in Xenopus laevis melanophores.

Melatonin, the principal hormone of the vertebrate pineal gland, has been implicated in a variety of neurobiological processes such as circadian rhythmicity and reproductive function. One of the earliest described actions of melatonin was its ability to cause pigment translocation in the dermal melanophores of amphibians. Melatonin binding sites have been identified in the brain of many species and in pigmented tumour cell lines; however, the dermal melanophores of the frog Xenopus Laevis possess the highest known density of melatonin binding sites. These cells are the source from which a mela

A dominant mutation in tyrp1A leads to melanophore death in zebrafish.

Melanin biosynthesis in vertebrates depends on the function of three enzymes of the tyrosinase family, tyrosinase (Tyr), tyrosinase-related protein 1 (Tyrp1), and dopachrome tautomerase (Dct or Tyrp2). Tyrp1 might play an additional role in the survival and proliferation of melanocytes. Here, we describe a mutation in tyrp1A, one of the two tyrp1 paralogs in zebrafish, which causes melanophore death leading to a semi-dominant phenotype. The mutation, an Arg->Cys change in the amino-terminal part of the protein, is similar to mutations in humans and mice where they lead to blond hair (in melane

Metabolic energy requirements during teleost melanophore adaptations.

Inhibition of oxidative phosphorylation by 2,4-dinitrophenol or sodium cyanide promotes complete melanosome aggregation in teleost melanophores. This aggregation is not promoted via the alpha-sympathetic receptor sites because it occurs in the presence of tolazoline hydrochloride, an alpha-receptor blocking agent. Interpretation of these results suggests metabolic energy release is required in the centrifugal direction only.

Further studies on the melanophores of periodic albino mutant of Xenopus laevis.

It is still unknown why dermal melanophores disappear during larval development, and why no or very few epidermal melanophores appear during and after metamorphosis, in Xenopus laevis showing periodic albinism (ap). To elucidate these points, we investigated the occurrence of depigmentation in mutant (ap/ap) melanophores during in vitro proliferation and the incidence of melanophore differentiation from mutant melanoblasts in the skin in vitro. During in vitro proliferation of mutant melanophores, ap-type melanosomes decreased in number gradually and instead the number of premelanosomes increa

Microtubule system of isolated fish melanophores as revealed by immunofluorescence microscopy.

The microtubule system of melanophores of the angelfish, Pterophyllum scalare, has been studied using antibodies prepared against purified porcine brain tubulin in indirect immunofluorescence microscopy. Melanophores were freed from the surrounding tissue components of isolated scales by mild enzymatic digestion and then allowed to settle on a glass cover slip. In both the dispersed and the aggregated states large numbers of fluorescent fibers are seen. The number and the astral arrangement of these fibers, which run from the central region to the periphery of the cell, are striking. The syste

Melanin concentrating hormone. I. Influence of nerves and hormones on the control of trout melanophores.

When melanophores on trout scales are cultured in vitro they show a transitory melanin concentration, which can be prevented by addition to the medium of the alpha adrenergic blocker, phentolamine. This indicates the release of endogenous nor-epinephrine from local nerve terminals. This initial phase of melanin aggregation is followed by redispersion and then by a second, more gradual melanin concentration over several days, which is not antagonized by phentolamine. A final melanophore index of between 2-2.5 is attained which may be the resting state of trout melanophores. Using short-term cul

Transcriptional regulation of mitfa accounts for the sox10 requirement in zebrafish melanophore development.

The transcription factor Sox10 is required for the specification, migration and survival of all nonectomesenchymal neural crest derivatives including melanophores. sox10(-/-) zebrafish lack expression of the transcription factor mitfa, which itself is required for melanophore development. We demonstrate that the zebrafish mitfa promoter has sox10 binding sites necessary for activity in vitro, consistent with studies using mammalian cell cultures that have shown that Sox10 directly regulates Mitf expression. In addition, we demonstrate that these sites are necessary for promoter activity in viv

Understanding the ultrastructural aspects of berberine-induced skin-darkening activity in the toad, Bufo melanostictus, melanophores.

Berberine is an active compound of Berberis vulgaris (Daruhaldi) with known multiple pharmacological activities, including antimicrobial, antiviral, anti-inflammatory, cholesterol-lowering, and anticancer effects. The present work aimed to study the ultrastructural effects of berberine to determine its skin-darkening potential using Bufo melanostictus melanophores, which has not been done to date. Light and electron microscopic analysis of isolated dorsal skin melanophores of B. melanostictus has been done after treatment with various concentrations of berberine, along with specific antagonist

Muscarinic cholinoceptors that mediate pigment aggregation are present in the melanophores of cyprinids (Zacco spp.).

Like melanophores of many teleosts, those of the dark chub, Zacco temmincki, and the common minnow, Z. platypus (Cyprinidae, Cypriniformes) responded to norepinephrine (NE) by the aggregation of pigment. It was further found that some melanophores were responsive to acetylcholine (ACh) in the same way. The response to NE was blocked by an alpha-adrenergic blocker, phentolamine, whereas the response to ACh was not. By contrast, two muscarinic cholinoceptor antagonists, namely, atropine and scopolamine, effectively blocked the action of ACh. The pigment aggregation due to the liberated sympathet

Berberine-induced pigment dispersion in Bufo melanostictus melanophores by stimulation of beta-2 adrenergic receptors.

Reduced production of melanin by decreased or the absence of melanocytes leads to various hypopigmentation disorders, and the development of melanogenetic agents for photoprotection and hypopigmentation disorders is one of the top priority areas of research. Hence, the present study was carried out to elucidate the ability of berberine, a principal active ingredient present in the roots of the herb Berberis vulgaris to stimulate pigment dispersion in the isolated skin melanophores of the toad Bufo melanostictus. In the present study, mean melanophore size index of the isolated skin melanophore

Identification of kit-ligand a as the Gene Responsible for the Medaka Pigment Cell Mutant few melanophore.

The body coloration of animals is due to pigment cells derived from neural crest cells, which are multipotent and differentiate into diverse cell types. Medaka (Oryzias latipes) possesses four distinct types of pigment cells known as melanophores, xanthophores, iridophores, and leucophores. The few melanophore (fm) mutant of medaka is characterized by reduced numbers of melanophores and leucophores. We here identify kit-ligand a (kitlga) as the gene whose mutation gives rise to the fm phenotype. This identification was confirmed by generation of kitlga knockout medaka and the findings that the

A comparative ultrastructural and physiological study on melanophores of wild-type and periodic albino mutants of Xenopus laevis.

Pigment of tail-fin melanophores in periodic albino Xenopus laevis tadpoles is dispersed in response to darkness and to alpha-MSH in a manner similar to wild-type melanophores. However, periodic albino tadpoles lack the response to different background conditions and the melatonin-induced aggregation in darkness. The tyrosinase activity in cells of the latter type tadpoles is weak compared to the wild-type cells. Ultrastructural examination of melanophores from periodic albino mutants and cells from wild-type tadpoles shows similar organelles at corresponding sites. A morphological difference

Adenylate Cyclase 5 (Adcy5) Deficiency Impairs Pigment Granule Dispersion in Melanophores and Erythrophores in Nile Tilapia.

Adenylate cyclase 5 (Adcy5) generates cyclic adenosine monophosphate (cAMP) downstream of G protein-coupled receptor signaling, yet its role in vertebrate pigmentation remains incompletely understood. Here, we generated a CRISPR/Cas9-mediated adcy5 knockout line in Nile tilapia (Oreochromis niloticus) to investigate its function in chromatophore biology. Loss of adcy5 resulted in a pronounced disruption of body coloration, characterized by the absence of vertical black bars and a global reduction in pigmentation. Despite this, chromatophore number was largely unaffected, indicating that Adcy5
