Memory consolidation.

Conscious memory for a new experience is initially dependent on information stored in both the hippocampus and neocortex. Systems consolidation is the process by which the hippocampus guides the reorganization of the information stored in the neocortex such that it eventually becomes independent of the hippocampus. Early evidence for systems consolidation was provided by studies of retrograde amnesia, which found that damage to the hippocampus-impaired memories formed in the recent past, but typically spared memories formed in the more remote past. Systems consolidation has been found to occur

Consolidation of memory.

Animal studies have proven useful in addressing aspects of memory formation and consolidation that cannot be readily answered in research with humans. In particular, they offer the possibility of controlling both the extent and locus of brain lesions, and the exact nature of the experiences to be remembered. Taking advantage of these possibilities, recent studies indicated that the graded retrograde amnesia often seen after lesions to the hippocampal system is not uniform across lesion site and task, nor is it an indication that all of the remembered information available in intact subjects be

Rac1 Constrains Memory Consolidation.

Ras-related C3 botulinum toxin substrate 1 (Rac1) is a small GTPase that regulates actin cytoskeleton dynamics and synaptic plasticity. Rac1 has been implicated in active forgetting, but whether it also constrains the consolidation of new memories remains unclear. Here we show that systemic administration of the Rac1 inhibitor 1A-116 after training in the novel object recognition task markedly extends memory persistence in rats. A single post-training injection of 1A-116 enhanced recognition memory for at least 28 d without altering locomotor- or anxiety-related behaviors. When given after a b

Memory consolidation in sleep disorders.

In recent years sleep-related memory consolidation has become a central topic in the sleep research field. Several studies have shown that in healthy individuals sleep promotes memory consolidation. Notwithstanding this, the consequences of sleep disorders on offline memory consolidation remain poorly investigated. Research studies indicate that patients with insomnia, obstructive sleep apnea, and narcolepsy often exhibit sleep-related impairment in the consolidation of declarative and procedural information. On the other hand, patients with parasomnias, such as sleep-walking, night terrors an

Interacting brain systems modulate memory consolidation.

Emotional arousal influences the consolidation of long-term memory. This review discusses experimental approaches and relevant findings that provide the foundation for current understanding of coordinated interactions between arousal activated peripheral hormones and the brain processes that modulate memory formation. Rewarding or aversive experiences release the stress hormones epinephrine (adrenalin) and glucocorticoids from the adrenal glands into the bloodstream. The effect of these hormones on memory consolidation depends upon binding of norepinephrine to beta-adrenergic receptors in the

Environmental enrichment accelerates cortical memory consolidation.

Environmental enrichment is an established strategy to enhance learning and to build resilience against neurodegeneration. In humans, this is known as 'cognitive reserve'. Though the beneficial effects of exposure to a complex environment in animal models have been well-documented by behavioural, immunohistological and morphological observations, its impact on the functional properties of neuronal populations remains poorly understood. This study aimed to compare the functional encoding and offline memory retrieval dynamics of cortical neurons in enriched and control mice performing a virtual

Human retrograde amnesia and memory consolidation.

This paper reports a reassessment of published literature on the question of whether retrograde amnesia data from patients with severe trauma supports the idea that there is ongoing consolidation of long-lasting memories. That is, memory consolidation continues for decades with older memories being increasingly consolidated, and, thus, more protected from forgetting. Our analysis was limited to patients with specific traumas rather than neurodegenerative conditions that can be complicated by the additional presence of significant anterograde amnesia. These constraints were used because trauma

The REM sleep-memory consolidation hypothesis.

It has been hypothesized that REM (rapid eye movement) sleep has an important role in memory consolidation. The evidence for this hypothesis is reviewed and found to be weak and contradictory. Animal studies correlating changes in REM sleep parameters with learning have produced inconsistent results and are confounded by stress effects. Humans with pharmacological and brain lesion-induced suppression of REM sleep do not show memory deficits, and other human sleep-learning studies have not produced consistent results. The time spent in REM sleep is not correlated with learning ability across hu

Dysfunctional overnight memory consolidation in ecstasy users.

Sleep plays an important role in the consolidation and integration of memory in a process called overnight memory consolidation. Previous studies indicate that ecstasy users have marked and persistent neurocognitive and sleep-related impairments. We extend past research by examining overnight memory consolidation among regular ecstasy users (n=12) and drug naïve healthy controls (n=26). Memory recall of word pairs was evaluated before and after a period of sleep, with and without interference prior to testing. In addition, we assessed neurocognitive performances across tasks of learning, memor

Respiration Modulates Olfactory Memory Consolidation in Humans.

In mammals respiratory-locked hippocampal rhythms are implicated in the scaffolding and transfer of information between sensory and memory networks. These oscillations are entrained by nasal respiration and driven by the olfactory bulb. They then travel to the piriform cortex where they propagate further downstream to the hippocampus and modulate neural processes critical for memory formation. In humans, bypassing nasal airflow through mouth-breathing abolishes these rhythms and impacts encoding as well as recognition processes thereby reducing memory performance. It has been hypothesized that

Acetazolamide impairs fear memory consolidation in rodents.

Acetazolamide (AZ) is an carbonic anhydrase inhibitor, which has been used in the treatment of seizures, mountain sickness and glaucoma. Memory impairment by AZ has been reported in patient interviews; however, the related mechanism is unclear. We applied two fear conditioning paradigms, shuttle avoidance and passive avoidance, in both rats and mice to investigate this clinical anecdote. Adult Wistar rats receiving AZ 1 h before the shuttle avoidance test showed decreased avoidance rates, especially at high dosage. Adult ICR mice receiving AZ both before and after acquisition trials showed the

A generative model of memory construction and consolidation.

Episodic memories are (re)constructed, share neural substrates with imagination, combine unique features with schema-based predictions and show schema-based distortions that increase with consolidation. Here we present a computational model in which hippocampal replay (from an autoassociative network) trains generative models (variational autoencoders) to (re)create sensory experiences from latent variable representations in entorhinal, medial prefrontal and anterolateral temporal cortices via the hippocampal formation. Simulations show effects of memory age and hippocampal lesions in agreemen

TraceLink: a model of amnesia and consolidation of memory.

A model of amnesia is introduced, called TraceLink, that consists of three systems: 1) a trace system (neocortex), 2) a link system (hippocampus), and 3) a modulatory system (hippocampus/fornix/basal forebrain). It aims to explain salient aspects of the neuropsychology of amnesia, such as Ribot gradients in retrograde amnesia, patterns of dissociation between anterograde and retrograde amnesia, recovery from amnesia, and a newly discovered form of amnesia (semantic dementia) that results from certain temporal lobe lesions that do not affect the hippocampus. The model, furthermore, offers a new

Memory consolidation drives the enhancement of remote cocaine memory via prefrontal circuit.

Remote memory usually decreases over time, whereas remote drug-cue associated memory exhibits enhancement, increasing the risk of relapse during abstinence. Memory system consolidation is a prerequisite for remote memory formation, but neurobiological underpinnings of the role of consolidation in the enhancement of remote drug memory are unclear. Here, we found that remote cocaine-cue associated memory was enhanced in rats that underwent self-administration training, together with a progressive increase in the response of prelimbic cortex (PrL) CaMKII neurons to cues. System consolidation was

Selective attention in visual short-term memory consolidation.

Selection of visual short-term memory content takes place during perceptual and postperceptual processing. We studied postperceptual selection in a change detection task, in which spatially distributed cues preceding the memory array marked potential change targets. In the 550-730 ms interval after the memory array onset but not in the following retention interval, event-related potentials showed larger contralateral delay activity when the spatial cues were selective than when they were unselective. This finding implies that distributed precueing strengthens the visual short-term memory repre

Sleep and memory consolidation: Neural and cognitive perspectives.

This chapter synthesizes current evidence on the role of sleep in memory consolidation, emphasizing the interplay between neural oscillations, synaptic plasticity, and cognitive processes. Sleep is conceptualized as an active state, facilitating the encoding, stabilization, and integration of memories into long-term storage. Distinct contributions of sleep stages are highlighted, with slow-wave sleep (SWS) supporting declarative memory consolidation through hippocampal-neocortical replay mechanisms, and Rapid eye movement (REM) sleep enhancing procedural and emotional memory processing. The sy

Upstream CB1R Regulation of β-Adrenergic Memory Consolidation.

Object recognition memory (ORM) plays a key role in identifying familiar items and encoding episodic information. ORM consolidation depends on β-adrenergic receptor (βAR) signaling and is associated with increased BDNF expression in the dorsal hippocampus. Although hippocampal activation of cannabinoid type-1 receptors (CB1Rs) is known to impair ORM consolidation, the mechanisms underlying this effect remain unclear. In this study, we used the novel object recognition task to examine the interaction between CB1Rs and βARs during ORM consolidation in adult male Wistar rats. Intra-dorsal CA1 inf

Theta Stimulation Enhances Early Consolidation of Semantic Memory.

This study investigates the role of theta frequency transcranial alternating current stimulation (tACS) in enhancing the consolidation of newly acquired semantic knowledge. Although postencoding theta oscillatory power increase has been reported to benefit episodic memory consolidation, its influence on semantic memory retention has not been examined. In this experiment, 175 participants were taught fundamental immunology concepts and were then randomly assigned to receive either theta or beta tACS, applied through midline frontal, left parietal, or bilateral parietal montages, or sham stimula

Hippocampal CaMKII Regulates the Consolidation of Recognition Memory.

Object recognition memory (ORM) is a hippocampus-dependent form of memory essential for distinguishing items and constructing episodic representations of the past. Ca2+/calmodulin-dependent protein kinase II (CaMKII) is a serine/threonine-specific protein kinase highly enriched in the hippocampal formation, where it acts as a memory-relevant calcium effector. We found that, in rats, training in an ORM inducing learning task rapidly increased CaMKII autophosphorylation in the CA1 region of the dorsal hippocampus. Moreover, early post-acquisition intra-dorsal CA1 injection of the substrate-compe
