Dissecting Maize Aerial Meristems.

The plant kingdom displays a wide array of shapes, and this morphology is tightly linked to function and productivity. For example, leaf size, shape, and arrangement around the stem define canopy architecture, influencing light interception and photosynthetic capacity. Understanding how final morphology is determined is, therefore, not only important for our basic understanding of various plant species, but also an essential tool in the development of new crop varieties. All aerial organs in flowering plants arise on the flanks of meristems throughout development. In Zea mays (maize), these me

Potential bud bank responses to apical meristem damage and environmental variables: matching or complementing axillary meristems?

Soil nutrients, dormant axillary meristem availability, and competition can influence plant tolerance to damage. However, the role of potential bud banks (adventitious meristems initiated only after injury) is not known. Examining Central European field populations of 22 species of short-lived monocarpic herbs exposed to various sources of damage, we hypothesized that: (1) with increasing injury severity, the number of axillary branches would decrease, due to axillary meristem limitation, whereas the number of adventitious shoots (typically induced by severe injury) would increase; (2) favorab

Core pathways controlling shoot meristem maintenance.

Essential to the function of shoot meristems in plants to act as sites of continuous organ and tissue formation is the ability of cells within the meristem to remain undifferentiated and proliferate indefinitely. These are characteristics of the stem cells within meristems that are critical for their growth properties. Stem cells are found in tight association with the stem cell niche-those cells that signal to maintain stem cells. Shoot meristems are unique among stem cell systems in that the stem cell niche is a constantly changing population of recent daughter stem cells. Recent progress fr

Single cell-derived multicellular meristem: insights into male-to-hermaphrodite conversion and de novo meristem formation in Ceratopteris.

Land plants alternate between asexual sporophytes and sexual gametophytes. Unlike seed plants, ferns develop free-living gametophytes. Gametophytes of the model fern Ceratopteris exhibit two sex types: hermaphrodites with pluripotent meristems and males lacking meristems. In the absence of the pheromone antheridiogen, males convert to hermaphrodites by forming de novo meristems, although the mechanisms remain unclear. Using long-term time-lapse imaging and computational analyses, we captured male-to-hermaphrodite conversion at single-cell resolution and reconstructed the lineage and division a

Auxin Involvement in Ceratopteris Gametophyte Meristem Regeneration.

Growth and development of the Ceratopteris hermaphroditic gametophytes are dependent on cell proliferation in the marginal meristem, which when destroyed will regenerate at a new location on the body margin. We established a laser ablation method to destroy a single initial cell in the meristem. Ablation caused the cessation of cell proliferation accompanied by the disappearance of the expression of an auxin synthesis gene (CrTAA2) and a cell proliferation marker gene (CrWOXB). New meristem regeneration occurred within a predictable distance from the original two days post-ablation, signified

Navigating shoot apical meristem plasticity for plant adaptation.

Plants exhibit striking developmental plasticity, driven by stem cell populations within meristematic tissues such as the shoot apical meristem (SAM). While the genetic networks governing stem cell homeostasis are well-characterized, recent studies reveal that the SAM also functions as an active environmental sensor to modulate growth plasticity. This Tansley review synthesizes advances in how the SAM perceives and integrates key environmental signals - including light, temperature, oxygen, and humidity - to direct adaptive growth. We detail the molecular mechanisms, such as photoreceptor-medi

Hormonal architects: Plant steroids shape meristems.

In plants, the shoot and root apical meristems drive post-embryonic development by tightly coordinating stem cell maintenance, cell proliferation, and differentiation. Among the hormonal regulators governing these processes, brassinosteroids (BRs) have emerged as important modulators of meristem function. Although BRs are classically associated with promoting cell expansion, accumulating evidence demonstrates that they also play crucial roles in regulating cell division and meristem activity. In this review, we provide a comparative perspective on BR function in the shoot and root apical meris

Methyl-Salicylate Clearing and Imaging of Maize Meristems.

The final morphology of a plant is determined by much earlier events that occur during development. Meristems are critical for the development of the aerial parts of plants, such as leaves and stems. The morphology of these meristems, which can vary significantly across genetic backgrounds, correlates with phenotypic traits in Zea mays (maize) that influence yield, such as leaf initiation rate and kernel row number. Traditional methods for morphological analysis of meristems, including histological sectioning and fine dissection, often require considerable technical expertise. To address these

Repatterning of the inflorescence meristem in Gerbera hybrida after wounding.

The Asteraceae plant family is characterized by inflorescences, called flower heads or capitula that may combine hundreds of individual florets into a single flower-like structure. The florets are arranged in a regular phyllotactic pattern with Fibonacci numbers of left- and right-winding spirals. Such a pattern may be disrupted due to physical constraints or by wounding occurring during the early meristem development. Recovery from wounding re-establishes patterning although the mechanisms have remained elusive. In this study, we applied Gerbera hybrida as a model system and established metho

The bZIP Transcription Factor PERIANTHIA: A Multifunctional Hub for Meristem Control.

As sessile organisms, plants are exposed to extreme variations in environmental conditions over the course of their lives. Since plants grow and initiate new organs continuously, they have to modulate the underlying developmental program accordingly to cope with this challenge. At the heart of this extraordinary developmental plasticity are pluripotent stem cells, which are maintained during the entire life-cycle of the plant and that are embedded within dynamic stem cell niches. While the complex regulatory principles of plant stem cell control under artificial constant growth conditions begi

Fresh Tissue Scanning Electron Microscopy (SEM) of Maize Meristems.

In plants, final organ shape and position influence function and whole plant productivity. In Zea mays (maize), leaf, ear, and tassel morphology are all key agronomic features influencing yield. These aerial structures originate from stem cell populations within meristems. Understanding how meristems are regulated and how stem cells differentiate over developmental time to form mature leaves, ears, and tassels is important for identifying novel routes for maize improvement. High-resolution imaging of meristem morphology is a key component of developmental studies. Scanning electron microscopy

Cytogenotoxic activity of herbicidal and fungicidal pesticides on Triticum aestivum root meristem.

Phytotoxicity, cytotoxicity and genotoxicity of pesticides with various mechanisms of targeted activity were studied in a hydroponic culture of 2-day-old seedlings of Triticum aestivum. All studied pesticides (with the exception of metribuzin) exhibited dose-dependent phytotoxicity (inhibited the growth of the main root and reduced the yield of root biomass). All studied pesticides did not affect mitotic index in the root apex meristem but did affect the duration of some phases of mitosis. Herbicides increased, while fungicides, on the contrary, decreased the duration of the cytokinesis phase.

Kinetics of DNA Repair in Vicia faba Meristem Regeneration Following Replication Stress.

The astonishing survival abilities of Vicia faba, one the earliest domesticated plants, are associated, among other things, to the highly effective replication stress response system which ensures smooth cell division and proper preservation of genomic information. The most crucial pathway here seems to be the ataxia telangiectasia-mutated kinase (ATM)/ataxia telangiectasia and Rad3-related kinase (ATR)-dependent replication stress response mechanism, also present in humans. In this article, we attempted to take an in-depth look at the dynamics of regeneration from the effects of replication i

A genetic program for rapid initiation of axillary meristems at the onset of flowering.

Shoot branching patterns and their diversity arise from variable activities of axillary meristems (AMs). A common feature of flowering plants is delayed AM initiation and growth during the vegetative phase, followed by a sudden and rapid acceleration of apical AM initiation during the reproductive phase. The mechanisms underlying this pronounced developmental shift are poorly understood. During tomato floral transition, the primary shoot meristem (PSM) terminates in a flower after producing two spatially adjacent AMs: one that will generate a flower and the other a vegetative side shoot. EdU-Based Quantitative Analysis of Cell Proliferation in Arabidopsis Root Meristems.

The division of a mother cell into two daughter cells is a fundamental process in biology, involving the transfer of genomic information to the next generation of cells. In plant roots, cell divisions occur in the root apical meristem, a specialized tissue located at the plant's root tip. Cell proliferation rates in the root apical meristem shape root growth, thereby contributing to general development as well as the acclimatization to stressful environmental conditions through phenotypic plasticity. Here, we present a protocol for the microscopical analysis of cell proliferation in Arabidopsi

Molecular characterization of the duplicated meristem identity genes HvAP1a and HvAP1b in barley.

The vernalization gene VRN-1 has been identified as a MADS-box transcription factor orthologous to the meristem identity gene APETALA1 (AP1). A single copy of this gene was found in diploid wheat, but 2 copies were reported in barley. In this study, we present a detailed characterization of these 2 copies to understand their respective roles in the vernalization response. We identified 2 groups of barley bacterial artificial chromosomes (BACs), each containing 1 AP1 copy designated hereafter as HvAP1a and HvAP1b. A physical map of the VRN-H1 region showed that the HvAP1a BACs were part of the

Localized nitrate uptake by NRT1.8 antagonizes root meristem growth inhibition upon cadmium stress.

Heavy metal pollution and overuse of nitrogen fertilizer have essentially deteriorated soil environments, though how these two factors interact with each other to impact plant growth remains largely unknown. Here, we reported polar localization of NRT1.8 towards the soil side at epidermal cells adjacent to lateral root caps (LRC). The combined treatment of Cd and high nitrate induced NRT1.8 expression at root tips, leading to uptake of more NO3- from LRC into inner cells, meanwhile, unloading of NO3- from the xylem to collaboratively maintain NO3- level in the root tip. The root length and mer

A GRX-SOD module maintains superoxide homeostasis and meristem development in maize.

The continuous initiation of plant aerial organs relies on the maintenance of dynamic stem cell homeostasis within the shoot apical meristem (SAM). Although reactive oxygen species (ROS) have emerged as crucial signals determining cell fate, how plants precisely maintain SAM-specific ROS homeostasis remains to be elucidated. Here, we report that a redox regulatory module comprising the maize CC-type glutaredoxin MSCA1 and its homologs regulates meristem architecture by maintaining superoxide homeostasis in the SAM. We found that the superoxide anion (O2•-) is specifically enriched in the centr

Divide et impera: boundaries shape the plant body and initiate new meristems.

485 I. 485 II. 486 III. 491 IV. 491 V. 495 495 References 495 SUMMARY: Boundaries, established and maintained in different regions of the plant body, have diverse functions in development. One role is to separate different cell groups, for example the differentiating cells of a leaf primordium from the pluripotent cells of the apical meristem. Boundary zones are also established during compound leaf development, to separate young leaflets from each other, and in many other positions of the plant body. Recent studies have demonstrated that different boundary zones share similar properties. They

CENL1 expression in the rib meristem affects stem elongation and the transition to dormancy in Populus.

We investigated the short day (SD)-induced transition to dormancy in wild-type hybrid poplar (Populus tremula x P. tremuloides) and its absence in transgenic poplar overexpressing heterologous PHYTOCHROME A (PHYA). CENTRORADIALIS-LIKE1 (CENL1), a poplar ortholog of Arabidopsis thaliana TERMINAL FLOWER1 (TFL1), was markedly downregulated in the wild-type apex coincident with SD-induced growth cessation. By contrast, poplar overexpressing a heterologous Avena sativa PHYA construct (P35S:AsPHYA), with PHYA accumulating in the rib meristem (RM) and adjacent tissues but not in the shoot apical meri
