The protistan parasite Ichthyophonus sp. occurs in coastal populations of Pacific Herring Clupea pallasii throughout the northeast Pacific region, but the route(s) by which these planktivorous fish become infected is unknown. Several methods for establishing Ichthyophonus infections in laboratory challenges were examined. Infections were most effectively established after intraperitoneal (IP) injections with suspended parasite isolates from culture or after repeated feedings with infected fish tissues. Among groups that were offered the infected tissues, infection prevalence was greater after

As part of a state-wide multispecies survey of amphibian diseases, sampling was conducted at Archbold Biological Station, Venus, Florida, USA, on 15 April 2011. Gross examination of southern toad (Anaxyrus terrestris) larvae was unremarkable, but infections by a mesomycetozoean-like organism were observed in longitudinally sectioned routine haematoxylin and eosin-stained histologic slides. In 100% of the sectioned specimens examined (n = 5), a high density of the organism, representing several developmental stages, was found in the central nervous system, mainly in the spinal cord, brain, reti

Dermocystidium is a pathogenic fungus-like protist that infects many fish species, causing lesions on the skin, gills and other tissues, which may result in systemic disease. In the present study, a new Dermocystidium anguillae infection in the skin and eyes of the koi carp (Cyprinus carpio) from Iran was described using a comprehensive analysis of morphological, histological and molecular data. Filiform hyphal-like cysts were visible on wet mounts. The cysts were filled with spherical to round spores. Wet mount preparations of fresh cyst contents of Dermocystidium anguillae revealed a massive

OBJECTIVE: We compared differentially expressed genes in Chinook Salmon Oncorhynchus tshawytscha with three divergent Ichthyophonus statuses (undetected, subclinical infections, or clinical disease; n = 100) to investigate associated transcriptomic responses. Disease associated with the fish parasite Ichthyophonus sp. was first diagnosed in adult Chinook Salmon from the Yukon River in the late 1980s and has subsequently been implicated in premature host mortality.
METHODS: Chinook Salmon tissue sample collections and Ichthyophonus infection data were leveraged from a multi-agency collaboratio

Raised pale cysts were observed on Blue Ridge Sculpin Cottus caeruleomentum during stream fish community surveys in Catoctin Mountain Park, Maryland. When examined histologically, preserved sculpin exhibited multiple cysts containing spherical endospores with a refractile central body characteristic of Dermocystidium spp. Cysts were not observed on the gills or internally. The portion of the watershed in which affected sculpin were observed contained lower than expected numbers of sculpin, raising concerns about the population effects of this infection. A nearby stream lacked sculpin even thou

Climate change affects marine ecosystems by promoting pathogens that threaten key fish populations. To protect these, monitoring programs must adapt to manage threats and sustain fisheries. Here, we combined traditional PCR methods and transcriptomic analysis from a single drop of blood stored on FTA cards to determine the prevalence of erythrocytic necrosis virus (ENV) and the Ichthyophonus parasite in the Atlantic herring population. Across 2023–2024, 33% of individual blood samples tested positive for ENV and 10% for Ichthyophonus by PCR, with ENV-positive fish more frequently found in estu

OBJECTIVES: The aim of this study was to clarify the contentious taxonomic classification of Rhinosporidium seeberi, the cause of human rhinosporidiosis, which may have treatment implications.
METHODS: PCR was used to amplify the internal transcribed spacer (ITS)-2 region from the genomic DNA of the aetiological agent obtained from a sample of human rhinosporidiosis lesions. The amplicon was sequenced and the organism identified using the Basic Local Alignment Search Tools (BLAST).
RESULTS: Phylogenetic analysis revealed that the aetiological agent clustered along with the R. seeberi isolate

OBJECTIVE: We sought to determine the suitability of a semi-validated quantitative PCR (qPCR) assay for assessing the prevalence and density of Ichthyophonus sp. infections in Yukon River Chinook Salmon Oncorhynchus tshawytscha, representing a new species and location for the assay's application. Ichthyophonus sp. causes severe disease that has been implicated in the en route prespawning mortality of this Chinook Salmon stock. The reduced survival of these fish has caused many hardships for the people who rely on them.
METHODS: We assessed a modified qPCR assay with stage 1 and partial stage

International biodiversity assessments often overlook the role of emerging infectious pathogens in the decline of freshwater fish populations despite the many examples of emerging diseases in other more visible taxa on a global scale. Whilst the introduction of the rosette agent Sphaerothecum destruens in Europe remained an epidemiological enigma, recent findings have shown that this parasite arrived in Europe with the introduction of the healthy carrier Pseudorasbora parva from China nearly 60 years ago and its emergence went unnoticed for over 45 years despite its severe impact on European f

In 2003, the Alaska walleye pollock industry reported product quality issues attributed to an unspecified parasite in fish muscle. Using molecular and histological methods, we identified the parasite in Bering Sea pollock as Ichthyophonus. Infected pollock were identified throughout the study area, and prevalence was greater in adults than in juveniles. This study not only provides the first documented report of Ichthyophonus in any fish species captured in the Bering Sea, but also reveals that the parasite has been present in this region for nearly 20 years and is not a recent introduction. Since the early 1900s, mesomycetozoan parasites have been reported in both European anuran and caudate species. These reports have primarily been descriptive, which has made assessing the impact of these parasites on host populations difficult. Anecdotal reports of dermocystidium-like parasites are becoming widespread across Europe, possibly indicating that these mesomycetozoan parasites are increasing in distribution and/or abundance. This highlights the need for further investigations into the occurrence, pathogenesis and effects on host health of these parasitic infections for free-living a

Small amoeboid cells, believed to be the infectious stage of Ichthyophonus sp., were observed in the bolus (stomach contents) and tunica propria (stomach wall) of Pacific staghorn sculpins and rainbow trout shortly after they ingested Ichthyophonus sp.-infected tissues. By 24-48 hr post-exposure (PE) the parasite morphed from the classically reported multinucleate thick walled schizonts to 2 distinct cell types, i.e., a larger multinucleate amoeboid cell surrounded by a narrow translucent zone and a smaller spherical cell surrounded by a "halo" and resembling a small schizont. Both cell types

Rhinosporidiosis is caused by Rhinosporidium seeberi, a pathogen currently considered a fungus-like parasite of the eukaryotic group Mesomycetozoea. It is usually a benign condition, with slow growth of polypoid lesions, with involvement of the nose, nasopharynx, or eyes. The clinical characteristics of a painless, friable, polypoid mass, usually unilateral, can guide the diagnosis, but the gold standard for diagnosis is histopathological findings. This article reviews the epidemiology, pathobiology, clinical manifestations, diagnostic strategies, and treatment approach for rhinosporidiosis.

Cutaneous fungal infections used to be rare in most developed countries. However, they have become more common due to immunosuppression and globalization. In this report, we summarize the histopathologic findings of the main cutaneous fungal infections that are commonly seen in daily practice, including eumycetoma, sporotrichosis, histoplasmosis, cryptococcosis, coccidioidomycosis, aspergillosis, zygomycosis, phaeohyphomycosis, alternariosis, blastomycosis, paracoccidioidomycosis, lobomycosis, and chromoblastomycosis. We also include protothecosis (despite the fact that the infectious agent of

Rhinosporidiosis is a chronic localized granulomatous disease caused by Rhinosporidium seeberi, an aquatic protistan parasite belonging to a clade, Mesomycetozoea. Infestation of Rhinosporidiosis to the eye and adnexa is termed oculosporidiosis, in such cases, conjunctival mucosa is mostly involved; however in our case, it involved only the lacrimal sac and deeper periorbital tissue and presented as a case of orbital swelling. Surgical excision of the lesion was done, postoperatively dapsone therapy was given for 6 months, and the patient responded very well with no recurrence till date.

Rhinosporidiosis is a chronic granulomatous disease caused by an aquatic protistan parasite in the class of Mesomycetozoea, that is endemic in India and the subcontinent. This is a case report of a rhinosporidiosis presenting in an individual from Myanmar, whom had been working in Malaysia for the past four years. The disease is characterized by the appearance of polypoidal, friable growths that contain numerous spore filled cysts that stain with PAS staining. This disease is rarely seen in Malaysians due to the extensive urbanization in Kuala Lumpur, however the increasing numbers of migrant

Rhinosporidiosis is a rare infection caused by Rhinosporidium seeberi, an organism classified in its own class, mesomycetozoea. It commonly affects mucus membranes namely the nasal mucosa, pharynx and the conjunctiva. We present the case of an 8-year-old female who presented with a flat, red, vascular, fleshy, pedunculated mass arising from the tarsal conjunctiva of the right upper eyelid. The mass was completely excised. On histopathological examination, multiple sporangia were seen in various stages of degeneration, consistent with rhinosporidiosis. The diagnosis of rhinosporidiosis is based

In the past five years, with the use of molecular strategies the phylogenetic affinities of the two more resilient pathogens studied in medical mycology, Lacazia loboi and Rhinosporidium seeberi were finally deciphered. These studies found that L. loboi was the sister taxon to Paraccidioides brasiliensis, and R. seeberi was closely related to protistan spherical aquatic fish pathogens, located at the point were animals diverged from the fungi, in the class Mesomycetozoea. These initial studies indicated that a molecular strategy was the ideal approach to further understand these anomalous path

Rhinosporidiosis is a chronic granulomatous infectious disease caused by Mesomycetozoea Rhinosporidium seeberi. This highly recurrent polypoid lesion has a predilection for the nose and nasopharynx, although other organ systems may be affected. Involvement of the tracheobronchial tree is very rare, and poses a challenge for diagnosis and management. In this report, we present a 30-year-old man with a history of recurrent nasal polyp who presented with cough, shortness of breath, haemoptysis, and a radiological feature of right lung collapse on imaging. He was diagnosed with rhinosporidiosis ba

Ascetosporea is a group of parasitic protists within Endomyxa that primarily infect marine invertebrates and can cause serious damage to fishery resources such as oysters and prawns. Although approximately 50 species have been formally described, environmental DNA (eDNA) studies suggest that their diversity remains largely underestimated, with many lineages yet to be discovered. Their ecological roles and host ranges are also poorly understood, partly because no complete life cycle has been elucidated, likely due to the difficulty of maintaining live cells in laboratory conditions. In this stu
