Subcellular Potassium Ion-Mediated Structural Coordination Between Mesophyll Cells and Chloroplasts Determines CO2 Mesophyll Conductance.

Potassium (K) homeostasis critically orchestrates photosynthetic performance through mesophyll conductance (gm) regulation-a key bottleneck limiting CO2 assimilation under K deficiency. While K deprivation reduces gm via anatomical alterations, the mechanistic interplay between subcellular K+ distribution and chloroplast-level structural determinants remains elusive. Field experiments using two Brassica napus L. cultivars with contrasting K utilization efficiencies (KUtE) were employed to resolve how subcellular K+ dynamics regulate gm through hierarchical structural modifications. Under mild

Regulation of cytoplasmic streaming in Vallisneria mesophyll cells.

Induction and cessation of the rotational cytoplasmic streaming in Vallisneria mesophyll cells could be controlled by external stimuli. In cells that had been kept in darkness the cytoplasm remained quiescent. However, when the cells were treated in the dark with EGTA solution (10 mM or 20 mM buffered with 10 mM-Tris-maleate at pH 7.0), rotational cytoplasmic streaming was induced. When the cells were transferred again to artificial pond water in the dark, the induced streaming was inhibited; that is, only 50% of the observed cells exhibited active streaming after 2 h. When the cells were irra

Endopolyploidy in the mesophyll and bundle sheath cells of C4 Poaceae.

BACKGROUND AND AIMS: Most C4 plants share photosynthetic reactions between mesophyll and bundle sheath cells. Bundle sheath cells in these C4 plants are enlarged and have higher organelle content, and these traits are considered crucial enablers of the evolution of C4 photosynthesis. Endopolyploidy, the occurrence of varying ploidy levels in the somatic tissues of an organism, has been implicated in the enlargement of cells. Endopolyploidy may therefore contribute to the enlargement of bundle sheath cells, but it remains elusive as to whether the nuclei of bundle sheath cells are endopolyploid

Maintenance of High Photosynthetic Rates in Mesophyll Cells Isolated from Papaver somniferum.

The establishment and maintenance of high rates of photosynthetic CO(2) incorporation in mesophyll cells of Papaver somniferum (opium poppy) depend on a regime of dark and light periods immediately following isolation, as well as carefully adjusted conditions of isolation. Analysis of the incorporation pattern of (14)CO(2) by the isolated cells indicates an initial "stress-response" period of approximately 20 hours characterized by increased respiratory-type metabolism and diminished photosynthesis. Under the favorable regime, this period is followed by rapid recovery and the reinstatement of

Extracellular components implicated in the stationary organization of the actin cytoskeleton in mesophyll cells of Vallisneria.

In mesophyll cells of Vallisneria gigantea Graebner, an aquatic angiosperm, the association of the plasma membrane with the cell wall at the end wall has been reported to be indispensable for the mechanism that maintains the stationary organization of the bundles of microfilaments (MFs) [Masuda et al. (1991) Protoplasma 162: 151]. To identify putative extracellular components that might play a crucial role in this mechanism, we examined the effects of two exogenously applied synthetic hexapeptides, GRGDSP and ARYDEI, which include an RGD and an RYD motif, respectively. The RGD motif is known a

Nitric oxide-induced Na+/H+ exchange activity confers salt tolerance in pea (Pisum sativum L.) mesophyll cells.

Salinity stress severely hinders global agricultural productivity, and the issue will only increase under current climate scenarios. Due to complexity of salinity tolerance traits, crop breeding for salt tolerance remains a highly challenging task. The exogenous application of growth regulators, such as nitric oxide (NO), is considered a viable practical alternative to boost crop yield and quality under conditions of soil salinity. Numerous papers reported beneficial role of exogenous NO application on plant growth under salt stress, but very few explored the mechanistic basis of this process.

Reductions in mesophyll conductance under drought stress are influenced by increases in cell wall chelator-soluble pectin content and denser microfibril alignment in cotton.

Plants commonly undergo leaf morphoanatomy and composition modifications to cope with drought stress, and these tend to reduce mesophyll conductance to CO2 diffusion (gm), a key limitation to photosynthesis. The cell wall appears to play a crucial role in this reduction, yet the specific effect of cell wall component on gm and the underlying regulatory mechanisms of cell wall thickness (Tcw) variation are not well understood. In this study, we subjected cotton plants to varying levels of water deficit to investigate the impact of leaf cell wall component and the arrangement patterns of microfi

Spatial multi-omics reveals the putative biosynthetic sites and distribution patterns of key flavor-contributing metabolites in tea leaves.

Tea (Camellia sinensis) quality is determined by key metabolites including catechins, caffeine, theanine, and volatiles, yet their biosynthesis and spatial distribution at the cellular level remain poorly understood. Here, we constructed a cell-type-resolved spatial multi-omics atlas of tea leaves from C. sinensis var. sinensis 'Jinxuan' and C. sinensis var. assamica 'Yinghong No. 9' by integrating single-nucleus RNA sequencing (snRNA-seq), physically isolated cell-type multi-omics, and matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI). Cytosolic acidification and oxidation are the toxic mechanisms of SO2 in Arabidopsis guard cells.

SO2/H2SO3 can damage plants. However, its toxic mechanism has still been controversial. Two models have been proposed, cytosolic acidification model and cellular oxidation model. Here, we assessed the toxic mechanism of H2SO3 in three cell types of Arabidopsis thaliana, mesophyll cells, guard cells (GCs), and petal cells. The sensitivity of GCs of Chloride channel a (CLCa)-knockout mutants to H2SO3 was significantly lower than those of wildtype plants. Expression of other CLC genes in mesophyll cells and petal cells were different from GCs. Treatment with antioxidant, disodium 4,5-dihydroxy-1,

Plant Nuclei Move to Escape Ultraviolet-Induced DNA Damage and Cell Death.

A striking feature of plant nuclei is their light-dependent movement. In Arabidopsis (Arabidopsis thaliana) leaf mesophyll cells, the nuclei move to the side walls of cells within 1 to 3 h after blue-light reception, although the reason is unknown. Here, we show that the nuclear movement is a rapid and effective strategy to avoid ultraviolet B (UVB)-induced damages. Mesophyll nuclei were positioned on the cell bottom in the dark, but sudden exposure of these cells to UVB caused severe DNA damage and cell death. The damage was remarkably reduced in both blue-light-treated leaves and mutant leav

Effect of heat shock on ultrastructure and calcium distribution in Lavandula pinnata L. glandular trichomes.

The effects of heat shock (HS) on the ultrastructure and calcium distribution of Lavandula pinnata secretory trichomes are examined using transmission electron microscopy and potassium antimonate precipitation. After 48-h HS at 40°C, plastids become distorted and lack stroma and osmiophilic deposits, the cristae of the mitochondria become indistinct, the endoplasmic reticulum acquires a chain-like appearance with ribosomes prominently attached to the lamellae, and the plasma and organelle membranes become distorted. Heat shock is associated with a decrease in calcium precipitates in the tricho

C4-like metabolism and HCO3- use in submerged leaves of Ottelia cordata lacking Kranz anatomy at both low and high CO2 concentrations.

To date, only a few submerged plants have been reported to perform C4 and CAM. Ottelia cordata is a heteroblastic aquatic plant developing both submerged and floating leaves throughout its life cycle. Previous research found that, besides HCO3- use, the submerged leaves of O. cordata can also perform C4 metabolism. However, it remains unclear how the HCO3- use or the C4 pathway, and the anatomical structure, respond to varying CO2 environments. Therefore, we examined the anatomical structures and carbon-concentrating mechanisms in O. cordata submerged leaves under high (HC) and low CO2 (LC) co

Multiple metal accumulation within a manganese-specific genus.

PREMISE OF THE STUDY: Plants that strongly accumulate metals may be practically beneficial, and also serve as novel resources for increasing fundamental understanding of plant biology. Australian Gossia (Myrtaceae) species are delineated by a conspicuous affinity for the heavy metal manganese (Mn), which is a micronutrient crucial to photosynthesis. This genus includes several Mn hyperaccumulators such as G. bidwillii. Unusually, in G. bidwillii foliar Mn is most highly concentrated in photosynthetic cells, an observation thus far restricted to foliar-Mn accumulation in Mn hyperaccumulators. R

The petal identity gene PhDEF has a major binding and regulatory action in the epidermis.

In flowering plants, floral organ identity is specified by the combinatorial action of homeotic genes. While the role of these genes in the early specification of organ identity is well established, their late function throughout floral organ development and in specific cell types is much less characterized. In particular, since plant organs are structured in clonally independent cell layers, whether and how homeotic identity interacts with cell layer identity is unknown. We have previously identified cell layer-specific mutants for the petal identity gene PhDEF in petunia flowers, resulting i

Mass Spectrometry Imaging Combined With Single-Cell Transcriptional Profiling Reveals the Multidimensional Spatial Distributions and Biosynthetic Pathways of Medicinal Components in Andrographis paniculata.

The synthesis and accumulation of active ingredients in medicinal plants are distributed in specific organs, tissues, and cell types, which are important for the exploitation of medicinal plants. However, the fine distribution of active ingredients is difficult to know. Here, the system of mass spectrometry imaging (MSI) integrated with single-cell RNA sequencing was established for the first time in Andrographis paniculata (A. paniculata), a medicinal plant widely utilised in China and Southeast Asia. MSI shows specific distribution of andrographolides in A. paniculata, with higher accumulati

Rho GTPase overexpression impacts leaf internal architecture and mesophyll conductance in Arabidopsis.

Leaves are built from multiple cell types and are structured to enable the conversion of carbon dioxide and water into sugars in the process of photosynthesis. Understanding how cell architecture impacts the movement of CO2 within leaves may provide means to improve photosynthesis. Here, we examined the impact of mesophyll cell architecture on air networks and air permeability by employing high-resolution tomography data in leaves of Arabidopsis thaliana wild-type plants and in plants with altered Rho of Plant (ROP)-GTPase-related activities. We employed high-resolution tomography to character

Single-cell RNA-sequencing profiles reveal the developmental landscape of hawthorn leaves.

Hawthorn (Crataegus spp.) is an economically and medicinally important genus in Rosaceae. Its leaves accumulate valuable secondary metabolites, including flavonoids, and have long been used in traditional Chinese medicine. Despite available chromosome-level genome assemblies and bulk transcriptome data, the cellular composition of hawthorn leaves, their developmental trajectories, and the cell-type-specific distribution of biosynthetic gene expression remain uncharacterized. Here we report the first single-cell transcriptomic atlas of hawthorn leaves, generated by integrating protoplast-based

Palisade cell geometry in relation to leaf optical and photosynthetic properties in Viburnum.

The optical properties of lobed palisade mesophyll cells remain poorly understood despite their presence in diverse taxa and the critical role of the palisade layer in leaf-light interactions and carbon assimilation. Using microcomputed tomography, 3D ray tracing simulations, and physiological experiments, we tested the interactions among palisade cell geometry, chloroplast localization, light directional quality, and leaf optical and photosynthetic performance in the model taxon Viburnum. Simulations showed that lobed cells shifted between absorptance- and transmittance-dominated states depen

Deep single-cell proteomics reveals cellular heterogeneity in the cold response of Arabidopsis leaves.

Mass spectrometry (MS)-based proteomic analysis at single-cell resolution in complex plant tissues is challenging. Here, we present a highly sensitive single-cell proteomics (SCP) workflow that integrates key steps including single-cell sorting, sample pretreatment, MS detection, and data processing, with specific optimizations tailored for plant cells. Using this customized workflow, we achieved great identification depth, quantifying approximately 4,200 protein groups from a single mesophyll cell. In the proof-of-principle application in the cold response research of Arabidopsis leaves, we a
