Metabolic reprogramming in cholangiocarcinoma.

Metabolic reprogramming is a hallmark of cancer and allows tumour cells to meet the increased energy demands required for rapid proliferation, invasion, and metastasis. Indeed, many tumour cells acquire distinctive metabolic and bioenergetic features that enable them to survive in resource-limited conditions, mainly by harnessing alternative nutrients. Several recent studies have explored the metabolic plasticity of cancer cells with the aim of identifying new druggable targets, while therapeutic strategies to limit the access to nutrients have been successfully applied to the treatment of som

Immune metabolic reprogramming in MASLD.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a major global health challenge driven by the interplay between metabolic dysregulation and chronic low-grade inflammation. Beyond simple hepatic lipid accumulation, MASLD is characterized by extensive remodeling of the liver immune microenvironment in response to metabolic stress and persistent immunological activation. Recent advances in the field of immunometabolism have revealed that metabolic pathways not only shape immune cell identity and functions, but are also themselves influenced by immune responses, creating a reci

Metabolic reprogramming in liver fibrosis.

Chronic liver diseases, primarily metabolic dysfunction-associated steatotic liver disease (MASLD), harmful use of alcohol, or viral hepatitis, may result in liver fibrosis, cirrhosis, and cancer. Hepatic fibrogenesis is a complex process with interactions between different resident and non-resident heterogeneous liver cell populations, ultimately leading to deposition of extracellular matrix and organ failure. Shifts in cell phenotypes and functions involve pronounced transcriptional and protein synthesis changes that require metabolic adaptations in cellular substrate metabolism, including g

Histone deacetylases in cancer metabolic reprogramming.

Cancer cells undergo extensive metabolic reprogramming to sustain rapid proliferation and adapt to heterogeneous tumour microenvironments. These metabolic alterations are tightly linked to epigenetic regulation, which reshapes gene expression and cellular signalling mechanisms. Among epigenetic regulators, histone deacetylases (HDACs) have emerged as key modulators of cancer metabolic reprogramming. Along with their canonical roles in histone deacetylation, HDACs regulate non-histone substrates, including metabolic enzymes and transcription factors, thereby coordinating transcriptional and met

Mitochondrial metabolic reprogramming in diabetic cardiomyopathy.

Diabetes mellitus represents a major global health challenge and is strongly associated with cardiovascular complications, among which diabetic cardiomyopathy (DCM) is a major contributor to heart failure. Increasing evidence indicates that mitochondrial dysfunction plays a central role in DCM pathogenesis. However, mitochondrial abnormalities in the diabetic heart reflect not merely cellular injury but a coordinated process of mitochondrial metabolic reprogramming, characterized by altered substrate utilization, impaired oxidative phosphorylation, and disruption of mitochondrial quality contr

PRMT1-mediated metabolic reprogramming promotes leukemogenesis.

Copious expression of protein arginine methyltransferase 1 (PRMT1) is associated with poor survival in many types of cancers, including acute myeloid leukemia. We observed that a specific acute megakaryocytic leukemia (AMKL) cell line (6133) derived from RBM15-MKL1 knock-in mice exhibited heterogeneity in Prmt1 expression levels. Interestingly, only a subpopulation of 6133 cells expressing high levels of Prmt1 caused leukemia when transplanted into congenic mice. The PRMT1 inhibitor, MS023, effectively cured this PRMT1-driven leukemia. Seahorse analysis revealed that PRMT1 increased the extrac

Metabolic Reprogramming in Tumor Endothelial Cells.

The dynamic crosstalk between the different components of the tumor microenvironment is critical to determine cancer progression, metastatic dissemination, tumor immunity, and therapeutic responses. Angiogenesis is critical for tumor growth, and abnormal blood vessels contribute to hypoxia and acidosis in the tumor microenvironment. In this hostile environment, cancer and stromal cells have the ability to alter their metabolism in order to support the high energetic demands and favor rapid tumor proliferation. Recent advances have shown that tumor endothelial cell metabolism is reprogrammed, a

Metabolic reprogramming in cancer and senescence.

The rising trend in global cancer incidence has caused widespread concern, one of the main reasons being the aging of the global population. Statistical data show that cancer incidence and mortality rates show a clear upward trend with age. Although there is a commonality between dysregulated nutrient sensing, which is one of the main features of aging, and metabolic reprogramming of tumor cells, the specific regulatory relationship is not clear. This manuscript intends to comprehensively analyze the relationship between senescence and tumor metabolic reprogramming; as well as reveal the impac

Podocyte Metabolic Reprogramming and Targeted Therapy.

Podocytes, highly specialized glomerular epithelial cells, are essential for maintaining the filtration barrier integrity, yet they are particularly susceptible to metabolic stress. Recent advances have identified metabolic reprogramming as a central driver of podocyte injury in diverse glomerular diseases, including diabetic kidney disease and FSGS. Pathologic stimuli, such as hyperglycemia, lipotoxicity, oxidative stress, and inflammatory cytokines, lead to profound alterations in podocyte metabolism, encompassing dysregulation of lipid, glucose, amino acid, and ion handling and activation o

The metabolic escape: how tumor metabolic reprogramming drives drug resistance.

In the last few years, metabolic reprogramming has been recognized as a fundamental characteristic of cancer, and is also acknowledged as a crucial cause to drug resistance, which consistently acts as a significant barrier in cancer treatment by allowing tumor cells to adapt and escape various therapies. This review gives a systematically investigation of how metabolic reprogramming contributes to drug resistance in cancer, including aerobic glycolysis (also known as the Warburg effect), lactate metabolism, glutamine addiction, lipid synthesis reprogramming, mitochondrial and ion metabolic cha

Diagnosis-dependent metabolic reprogramming of follicular fluid.

BACKGROUND: The follicular fluid (FF) microenvironment plays a critical role in oocyte maturation and embryo development, reflecting local ovarian activity and systemic metabolic status. While metabolic alterations in FF have been described in different diseases, comparative analyses across different infertility-related disorders remain limited. OBJECTIVE: This study aimed to characterize and compare amino acid profiles in FF from patients undergoing in vitro fertilization (IVF) with insulin resistance (IR), endometriosis (EM), thyroid dysfunction (TD) and to identify disease-specific metabol

Biomaterials for metabolic reprogramming in skeletal disorders.

Metabolic reprogramming is a central regulator of cell fate and tissue homeostasis. By systematically remodeling metabolic pathways, cells adapt to specific physiological or pathological conditions to support survival and proliferation, often through enhanced glycolysis and lactate accumulation. Emerging evidence suggests that lactate accumulation can drive histone and nonhistone lactylation, thereby reshaping gene regulation and promoting inflammatory activation, matrix catabolism, and cellular senescence, which contribute to the initiation and progression of skeletal disorders such as osteoa

Metabolic reprogramming promotes Staphylococcus aureus serum resistance.

Staphylococcus aureus is a leading cause of bloodstream infections causing an estimated 300,000 deaths worldwide. Using a functional genomics approach, our group previously identified that adaptation of S. aureus to human serum is polygenic, with a clinically occurring non-synonymous mutation (V76I) in dihydrolipoamide dehydrogenase (lpdA1/pdhD) improving bacterial survival. In this work, we establish that improved serum survival of strains expressing PdhD V76I is underpinned by enhanced resistance to host-derived antimicrobials, including antimicrobial peptides and host-defence fatty acids wh

Metabolic Reprogramming in HIV-Associated Neurocognitive Disorders.

A significant number of patients infected with HIV-1 suffer from HIV-associated neurocognitive disorders (HAND) such as spatial memory impairments and learning disabilities (SMI-LD). SMI-LD is also observed in patients using combination antiretroviral therapy (cART). Our lab has demonstrated that the HIV-1 protein, gp120, promotes SMI-LD by altering mitochondrial functions and energy production. We have investigated cellular processes upstream of the mitochondrial functions and discovered that gp120 causes metabolic reprogramming. Effectively, the addition of gp120 protein to neuronal cells di

Metabolic reprogramming in the OPA1-deficient cells.

OPA1, a dynamin-related GTPase mutated in autosomal dominant optic atrophy, is essential for the fusion of the inner mitochondrial membrane. Although OPA1 deficiency leads to impaired mitochondrial morphology, the role of OPA1 in central carbon metabolism remains unclear. Here, we aim to explore the functional role and metabolic mechanism of OPA1 in cell fitness beyond the control of mitochondrial fusion. We applied [U-13C]glucose and [U-13C]glutamine isotope tracing techniques to OPA1-knockout (OPA1-KO) mouse embryonic fibroblasts (MEFs) compared to OPA1 wild-type (OPA1-WT) controls. Furtherm

The cancer metabolic reprogramming and immune response.

The overlapping metabolic reprogramming of cancer and immune cells is a putative determinant of the antitumor immune response in cancer. Increased evidence suggests that cancer metabolism not only plays a crucial role in cancer signaling for sustaining tumorigenesis and survival, but also has wider implications in the regulation of antitumor immune response through both the release of metabolites and affecting the expression of immune molecules, such as lactate, PGE2, arginine, etc. Actually, this energetic interplay between tumor and immune cells leads to metabolic competition in the tumor ec
