Protection by zinc-metallothionein (ZnMT) against cadmium-metallothionein-induced nephrotoxicity.

In contrast to inorganic Cd, acute iv administration of Cd bound to metallothionein (CdMT) concentrates in renal tissue. This uptake of CdMT produces functional and morphological changes in kidneys, similar to those observed after chronic exposure to inorganic Cd. In order to examine the importance of the metal component of MT in the renal uptake of MT, the renal concentration of 35S after administration of [35S]ZnMT and [35S]CdMT was compared. Renal uptake of 35S from both CdMT and ZnMT was very rapid, with peak concentrations observed 15-30 min after administration. 35S in kidneys increased

Anti-metallothionein IgG and levels of metallothionein in autistic families.

Metallothioneins (MTs) are a family of small proteins containing 61-68 amino acids with an unusually high concentration of cysteine. MT-1, the most functional and active MT in humans, has the ability to react with and enhance the detoxification of a number of metals including zinc, mercury, copper and cadmium. MT dysfunction may result, then, in many of the aetiological syndromes observed in autistic children, such as the leaky gut. It has been proposed that allergic autoimmune reactions occurring after exposure to heavy metals, may contribute to some symptoms associated with autism. Therefore

Diabetes and metallothionein.

Diabetes is a widespread disease, and its development and toxic effects on various organs have been attributed to increased oxidative stress. Metallothionein (MT) is a group of intracellular metal-binding and cysteine-rich proteins, being highly inducible in many tissues. Although it mainly acts as a regulator of metal homeostasis such as zinc and copper in tissues, MT was found to be a potent antioxidant and adaptive (or stress) protein to protect cells and tissues from oxidative stress. Studies showed that zinc-induced or genetically enhanced MT synthesis in the pancreas prevented the develo

Metallothionein-protein interactions.

Metallothioneins (MTs) are a family of universal, small proteins, sharing a high cysteine content and an optimal capacity for metal ion coordination. They take part in a plethora of metal ion-related events (from detoxification to homeostasis, storage, and delivery), in a wide range of stress responses, and in different pathological processes (tumorigenesis, neurodegeneration, and inflammation). The information on both intracellular and extracellular interactions of MTs with other proteins is here comprehensively reviewed. In mammalian kidney, MT1/MT2 interact with megalin and related receptor

Metallothionein and neurodegenerative diseases.

Neurodegenerative diseases, which mainly include Alzheimer's disease, Parkinson's disease, amyotrophic lateral sclerosis, Wilson's disease, and Huntington's disease, are a group of disorders characterized by loss of neurons in the brain and spinal cord. However, the underlying pathogenetic mechanisms of these disorders remain unclear. The metal ion hypothesis is considered a possible cause of a variety of neurodegenerative diseases. This hypothesis posits that the homeostatic imbalance of metal ions leads to oxidative stress, neuroinflammation, excessive aggregation of pathological proteins, a

Toxicological aspects of metallothionein.

Metallothionein (MT) is expressed to a certain extent in almost all mammalian tissues. The biological significance of MT is related to its various forms MT-1, MT-2, MT-3 and MT4. For MT-1 several isoforms of the protein exist and it is likely that these isoforms are related to various functions involved in developmental processes occurring at various stages of gestation. Toxicokinetics and biochemistry of essential and toxic metals such as cadmium, zinc, mercury and copper in organs e.g. kidney, CNS, are often related to metallothionein. It is debated whether there is a relation or not for oth

Role of metallothioneins in disease.

Metallothioneins are small proteins (6,000 to 10,000 mw) with similar amino acid compositions, high content of sulfhydryl amino acids and no aromatic amino acids. Emax at 250 nm is due to cadmium mercaptide bond. Synthesis is induced by a number of metals including zinc, cadmium, mercury, copper, bismuth, gold and silver. These proteins function as a mechanism of intracellular storage of some essential metals such as zinc and copper. Binding of potentially toxic metals like cadmium may be protective; but with saturation of protein by metal, toxicity may occur. Excessive cadmium-thionein may ha

Metallothionein-I-transgenic mice are not protected from acute cadmium-metallothionein-induced nephrotoxicity.

Mice pretreated with Zn have increased renal metallothionein (MT) levels and are protected from CdMT nephrotoxicity. To determine whether MT is important in this Zn-induced protection against CdMT-induced nephrotoxicity, MT-transgenic mice that have high levels of MT in their kidneys (10-fold over control mice) have been studied to determine whether they are resistant to CdMT-induced nephrotoxicity. Mice were injected with CdMT (0.1-0.6 mg Cd/kg, iv) and kidney injury was evaluated 24 hr later. CdMT produced renal toxicity in a dose-dependent manner. At a nephrotoxic dose of CdMT (0.4 mg Cd/kg

Metallothionein in human seminal plasma.

Metallothionein (MT) concentrations were measured in the seminal plasma of 4 fertile and 35 infertile men and in the hydrocele and spermatocele fluids. The relationship between MT content and sperm density, total number of sperm per ejaculate, sperm motility and abnormal form rates, leukocyte count and zinc levels in seminal plasma, as well as the relationship between MT and serum follicle-stimulating hormone, luteinizing hormone, testosterone, and prolactin were examined. MT was not detected in the hydrocele and spermatocele fluids. MT levels were related to zinc levels and to the leukocyte c

Metallothionein expression in carcinoma of the gallbladder.

AIMS: Metallothioneins (MT) are sulphur-rich, low molecular-weight, intracellular metal-binding proteins with a possible role in carcinogenesis of some human tumours. Metallothionein expression in gallbladder cancer has not been studied previously. METHODS AND RESULTS: Immunohistochemical expression of metallothionein was studied in 42 gallbladders (27 cases of carcinoma of the gallbladder, eight chronic cholecystitis and seven cases of normal gallbladder). Metallothionein expression was significantly higher in cases with carcinoma of the gallbladder (70.37%) as compared to chronic cholecysti

Expression and regulation of brain metallothionein.

Many, but not all, zinc-containing neurons in the brain are a subclass of the glutamatergic neurons, and they are found predominantly in the telencephalon. These neurons store zinc in their presynaptic terminals and release it by a calcium-dependent mechanism. These "vesicular" pools of zinc are viewed as endogenous modulators of ligand- and voltage-gated ion channels. Metallothioneins (MTs) are low molecular weight zinc-binding proteins consisting of 25-30% cysteine, with no aromatic amino acids or disulfide bonds. The areas of the brain containing high contents of zinc such as the retina, th

[Role of metallothionein in cancer chemotherapy].

Metallothionein (MT) shows a protective effect against toxic actions of some antitumor drugs and gamma-irradiation in animals. Preadministration of bismuth subnitrate, an MT inducing drug, significantly reduced side effects of antitumor drugs without affecting antitumor activity of the drugs. This specific effect of bismuth on the toxicity of antitumor drugs appears to be attributable to its specific induction of MT in normal tissues, but not in tumor tissue. Preinduction of MT synthesis in the lung also prevented mice from carcinogenesis caused by cisplatin and melphalan in the lung. On the o

Metallothionein and occupational exposure to cadmium.

The relationship between metallothionein (MT), chronic exposure to cadmium (Cd), and renal function was investigated in 53 men who were occupationally exposed to Cd. The aim was to determine if MT is a potential biological monitor for chronic exposure to Cd which would be useful for preventing Cd nephropathy. In this study MT excretion, serum MT, and serum creatinine concentrations were significantly higher in subjects with abnormal renal function who had been exposed to Cd. MT excretion was also linearly related on an individual basis to protein excretion, beta 2-microglobulin (beta 2-M) excr

Photoprotective role of metallothionein in UV-injury - metallothionein-null mouse exhibits reduced tolerance against ultraviolet-B.

We have shown previously that injection of cadmium chloride (Cd(2+)) depletes the number of ultraviolet B (UVB)-induced sunburn cells (SBC) in the mouse skin in vivo and that Cd(2+) treatment enhances UVB resistance in cultured keratinocytes in vitro, indicating the photoprotective role of Cd(2+)-induced metallothioneins (MTs) with antioxidant property against UVB injury. However, there has been no direct evidence for the role of MTs in UV protection. To improve our understanding of MTs in photoprotection, MT-null mouse deficient in MT-1 and MT-2 genes was studied. Skin explants were prelimina

[Studies of metals and metallothionein in tissue].

This paper is a review of three topics related to bio-trace metals. First, the transfer of metals into tissues of patients with chronic diseases treated with hemodialysis is examined. Such diseases include chronic hepatitis, diabetes, and chronic renal failure. In these diseases, metal contents from fingernails were flexible but non-specific. Toxicity may appear as the amount of heavy metals in tissues of patients with chronic renal failure treated with hemodialysis. For example, cadmium and lead were not excreted from the blood of patients during the hemodialysis treatment, and, therefore, th

Development of a radioimmunoassay for human metallothionein.

We report the development of a radioimmunoassay (RIA) for human metallothionein (MT). The MT was isolated from the liver of a patient with primary biliary cirrhosis and used to raise high titer antibodies in rabbits. The RIA was developed using Bolton-Hunter labeled MT as label and purified MT as standard. The detection limit of the assay was 0.4 ng/ml and concentrations between 0.8 and 100 ng/ml could be routinely measured. Dilutions of plasma samples containing high endogenous MT concentrations and tissue extracts in assay buffer showed parallelism with the standard curve and MT added to pla
