[Developmental anomalies of the metencephalon in the craniofacial and vertebral malformation syndrome].

The authors describe a case of a girl aged 9 years with a syndrome of malformations--cleft of the upper lip, maxilla and palate, lack of C2 arch fusion, polycystic lesions of the vermis and cerebellar hemisphere with obliteration of the aqueduct. The syndrome of intracranial hypertension with prevailing cerebellar symptoms was the cause of diagnostic and surgical management. Suboccipital craniotomy was done with insertion of ventriculoatrial valve which restored the normal intracranial pressure. The girl recovered and resumed education.

Agenesis of the mesencephalon and metencephalon with cerebellar hypoplasia: putative mutation in the EN2 gene--report of 2 cases in early infancy.

Congenital absence of the midbrain and upper pons is a rare human malformation. We describe two unrelated infants with this anomaly and cerebellar hypoplasia who were born at term but died in early infancy from lack of central respiratory drive. MRI in both cases disclosed the lesions during life. Neuropathological examination, performed in one, included immunocytochemical studies of NeuN, synaptophysin, vimentin, and glial fibrillary acidic protein (GFAP). Autopsy revealed a thin midline cord passing through the clivus, in place of the midbrain; it corresponded to hypoplastic and fused cortic

Characterization of proopiomelanocortin in the snakeskin gourami (Trichopodus pectoralis) and its expression in relation to food intake.

Proopiomelanocortin (POMC) is the precursor of several hormones involved in physiological systems including feed intake. The snakeskin gourami (Trichopodus pectoralis) POMC complementary DNA (TpPOMC) was cloned and characterized. Phylogenetic analysis showed that TpPOMC was clustered in a major POMC lineage in fish. Analysis of the Ka to Ks ratios for the entire POMC sequence and for each hormonal segment suggested that different POMC-derived peptide segments were subject to different evolutionary pressures. High expression level of TpPOMC was observed in all brain regions, with the highest le

Directed differentiation of human hindbrain neuroepithelial stem cells recapitulates cerebellar granule neurogenesis.

Cerebellar granule neurons (CGNs) are the most abundant neurons in the human brain. Dysregulation of their development underlies movement disorders and medulloblastomas. It is suspected that these disorders arise in progenitor states of the CGN lineage, for which human models are lacking. Here, we have differentiated human hindbrain neuroepithelial stem (hbNES) cells to CGNs in vitro using soluble growth factors, recapitulating key progenitor states in the lineage. We show that hbNES cells are not lineage committed and retain rhombomere 1 regional identity. Upon differentiation, hbNES cells tr

Human cerebellar organoids with functional Purkinje cells.

Research on human cerebellar development and disease has been hampered by the need for a human cell-based system that recapitulates the human cerebellum's cellular diversity and functional features. Here, we report a human organoid model (human cerebellar organoids [hCerOs]) capable of developing the complex cellular diversity of the fetal cerebellum, including a human-specific rhombic lip progenitor population that have never been generated in vitro prior to this study. 2-month-old hCerOs form distinct cytoarchitectural features, including laminar organized layering, and create functional con

The leukemic oncogene tal-2 is expressed in the developing mouse brain.

tal-1 (T-cell acute leukemia-1; also known as SCL) and tal-2 genes belong to a family of basic helix-loop-helix transcription factors and were originally isolated from the breakpoints of chromosomal translocations in human T-cell leukemia cell lines. tal-1 is expressed not only in hematopoietic cells but also in several endothelial structures and the central nervous system during development. On the other hand, the detailed function and the sites of expression of tal-2 have remained obscure. We cloned the tal-2 cDNA from a mouse embryonic cDNA library and examined its expression pattern in the

Diffuse intracranial oligodendroglioma in a cow.

This report describes the clinical, morphological and immunohistochemical findings in an adult cow with cerebral oligodendroglioma. The tumour extended from the metencephalon to the telencephalon, with infiltration of the meninges. Immunohistochemically, the tumour cells lacked expression of a specific tumour antigen, were partially positive for S-100 and tau protein, and were negative for a range of antigens including glial fibrillary acid protein, neuron-specific enolase, myelin basic protein and synaptophysin. This is the first report describing a diffuse, cerebral oligodendroglioma in a co

Local and transient expression of E-cadherin involved in mouse embryonic brain morphogenesis.

We found that E-cadherin (uvomorulin) is transiently expressed in restricted regions of the metencephalon, mesencephalon and diencephalon of mouse embryonic brain. This expression first occurred in parts of the mesencephalon and diencephalon at around E9.5, and subsequently extended to the primordia of cerebellum, the dorsal midline of mesencephalon and some other regions of the embryonic brain. These E-cadherin expressions ceased by E15 except at the dorsal midline. Immunohistological analyses showed that E-cadherin-positive cells are radially arranged in the neural tube and the E-cadherin-po

Cerebellar granule cell migration and folia development require Mllt11/Af1q/Tcf7c.

The organization of neurons into distinct layers, known as lamination, is a common feature of the nervous system. This process, which arises from the direct coupling of neurogenesis and neuronal migration, plays a crucial role in the development of the cerebellum, a structure exhibiting a distinct folding cytoarchitecture with cells arranged in discrete layers. Disruptions to neuronal migration can lead to various neurodevelopmental disorders, highlighting the significance of understanding the molecular regulation of lamination. We report a role Mllt11/Af1q/Tcf7c (myeloid/lymphoid or mixed-lin

Patterns of age-related shrinkage in cerebellum and brainstem observed in vivo using three-dimensional MRI volumetry.

This study investigates the time course and regional differences in age-related volume loss in cerebellum and brainstem. Three-dimensional (3D) magnetic resonance imaging (MRI) volumetry was used to measure the volumes of 11 regions in the cerebellum and three regions in the brainstem in 48 healthy volunteers (age 19.8-73.1 years). Landmark-adjusted lattices were used to divide the cerebellum into three radial (lobules I-V = lingula/lobulus/culmen, lobules VI-VII = declive/folium/tuber, lobules VIII-X = pyramis/uvula/nodulus) and three transverse subdivisions (vermis, medial, lateral hemispher

The pivotal role of irradiation-induced apoptosis in the pathogenesis and therapy of medulloblastoma.

BACKGROUND: Medulloblastoma (MB) is a rare primitive neuroectodermal tumors originating from the cerebellum. MB is the most common malignant primary brain tumor of childhood. MB originates from neural precursor cells in distinctive regions of the rhombic lip, and their maturation occurs in the cerebellum or the brain stem during embryonal development. Also, apoptosis is a programmed cell death associated with numerous physiological as well as pathological regulations. RECENT FINDINGS: Irradiation (IR)-induce apoptosis triggers cell death, with or without intervening mitosis within a few hours

Fibrocartilaginous Embolic Encephalopathy in a Pig.

A 1-year-old boar was investigated after presenting with acute onset collapse and obtundance. No significant gross lesions were observed at post-mortem examination. Histopathological investigation revealed a severe bilateral and multifocal necrotizing encephalopathy with an amorphous material, which obstructed neuroparenchymal vessels in the metencephalon and mesencephalon. Alcian blue staining identified the material as of cartilaginous origin and a diagnosis of cerebral fibrocartilaginous embolism was established. No gross evidence of vertebral disc disease was detected and the origin of the

Neurochemical changes in newborn rat's brain after gestational cadmium and lead exposure.

Gestational administration of cadmium (10 mg/l) and lead (300 mg/l) produced a strong decrease in proteins at birth (-17%) and on day 5 (-31%) as well as in brain lipid amount on both days (-11 and -23%, respectively). At day 5 postnatal the exposure also produced a marked decrease in DNA and RNA concentrations with respect to the control group. On the other hand, we found a significant increase of indoleamine content in all brain areas studied in the cadmium-lead group and so the dopamine and its metabolite in mesencephalon, whereas dopamine levels in metencephalon decreased significantly. Expression pattern of TG-interacting factor 2 during mouse development.

TGIF2 is a close homologue of TGIF that functions as a transcriptional corepressor by interaction with transforming growth factor-beta (TGF-beta)-activated Smads. Mutations of TGIF have been found in holoprosencephaly, a genetic disease affecting forebrain and craniofacial developmental. Here we analyzed the expression pattern of TGIF2 during mouse embryogenesis. Expression of TGIF2 transcripts was detected at high level at E12.5 and E15.5 in the nervous system including neopallial cortex, mesencephalon, metencephalon, medulla oblongata, spinal cord, trigeminal ganglion, vestibulocochlear gang

Developmental trajectories of the human embryologic brain regions.

Vertebrate brains commonly consist of five basic embryologic anatomical regions: telencephalon; diencephalon; mesencephalon; metencephalon; and myelencephalon. The proportions of these regions vary widely across species and developmental stages. Investigation of their growth trajectories, therefore, has the potential to provide an understanding of the substrates of inter-species variation in neuroanatomy and function. To investigate the volumetric growth trajectories, structural magnetic resonance imaging (MRI) scans obtained from 618 healthy children (334 boys, 284 girls; ages 3-17 years old)

The organization of the reptilian formation: a comparative study using Nissl and Golgi techniques.

The brainstem reticular formation has been studied in 16 genera representing 11 families of reptiles. Measurements of Nissl-stained reticular neurons revealed that they are distributed along a continuum, ranging in length from 10 micrometer to 95 micrometer. Reticular neurons in crocodilians and snakes tend to be larger than those found in lizards and turtles. Golgi studies revealed that reticular neurons possess long, rectilinear, sparsely branching dendrites. Small reticular neurons (less than 31 micrometers length) possess fusiform or triangular somata which bear two or three primary dendri

The effect of upper pontine transections on normal cochlear responses and on the protective effects of contralateral acoustic stimulation in barbiturate-anaesthetized normal-hearing guinea pigs.

In barbiturate-anaesthetized guinea pigs with normal cochlear neural sensitivities, upper pontine transections were made to totally isolate the cell bodies of the olivocochlear neurons in the lower brainstem from all higher centres. The effects of this procedure were examined at the cochlea on normal compound action potential (CAP) thresholds and amplitudes, on the temporary threshold shifts (TTS) in CAP sensitivity caused by monaural loud sound exposures, and on the protective effects of low-level contralateral acoustic stimulation (Cody and Johnstone, 1982; Rajan and Johnstone, 1983a, 1988).

Gene discovery and biological insights into anxiety disorders from a large-scale multi-ancestry genome-wide association study.

We leveraged information from more than 1.2 million participants, including 97,383 cases, to investigate the genetics of anxiety disorders across five continental groups. Through ancestry-specific and cross-ancestry genome-wide association studies, we identified 51 anxiety-associated loci, 39 of which were novel. In addition, polygenic risk scores derived from individuals of European descent were associated with anxiety in African, admixed American and East Asian groups. The heritability of anxiety was enriched for genes expressed in the limbic system, cerebral cortex, cerebellum, metencephalo

Immunohistochemical localization of atrial natriuretic factor and autoradiographic distribution of atrial natriuretic factor-binding sites in the brain of the cave salamander Hydromantes genei (Amphibia, Plethodontidae).

The distribution of atrial natriuretic factor (ANF)-like immunoreactivity in the central nervous system of the cave salamander Hydromantes genei (Amphibia, Plethodontidae) was investigated by using antisera raised against rat and human ANF(1-28). Concurrently, the location of ANF-binding sites was determined by autoradiography, using radioiodinated human ANF(1-28) as a tracer. In several regions of the brain, including the olfactory bulb, the preoptic area, the ventral thalamus, the tectum of the mesencephalon, and the choroid plexuses inside the ventricles, a good correlation was observed bet
