Membrane properties modulate methane oxidation by particulate methane monooxygenase.

The copper-dependent membrane monooxygenases particulate methane monooxygenase (pMMO) and ammonia monooxygenase (AMO) oxidize methane to methanol and ammonia to hydroxylamine, respectively. These enzymes, which are important targets for biotechnology, reside in intracytoplasmic membranes (ICMs) where they form densely packed hexagonal arrays. While cryoEM structures of pMMO and AMO in ICMs have revealed closely associated lipids, little is known about how specific lipids and membrane morphologies influence activity. Here we show through cryoelectron tomography (cryoET) that three species of me

Mechanisms for Methane and Ammonia Oxidation by Particulate Methane Monooxygenase.

Particulate MMO (pMMO) catalyzes the oxidation of methane to methanol and also ammonia to hydroxylamine. Experimental characterization of the active site has been very difficult partly because the enzyme is membrane-bound. However, recently, there has been major progress mainly through the use of cryogenic electron microscopy (cryoEM). Electron paramagnetic resonance (EPR) and X-ray spectroscopy have also been employed. Surprisingly, the active site has only one copper. There are two histidine ligands and one asparagine ligand, and the active site is surrounded by phenyl alanines but no charge

Direct Methane Oxidation by Copper- and Iron-Dependent Methane Monooxygenases.

Methane is a potent greenhouse gas that contributes significantly to climate change and is primarily regulated in Nature by methanotrophic bacteria, which consume methane gas as their source of energy and carbon, first by oxidizing it to methanol. The direct oxidation of methane to methanol is a chemically difficult transformation, accomplished in methanotrophs by complex methane monooxygenase (MMO) enzyme systems. These enzymes use iron or copper metallocofactors and have been the subject of detailed investigation. While the structure, function, and active site architecture of the copper-depe

Methane and Trichloroethylene Degradation by Methylosinus trichosporium OB3b Expressing Particulate Methane Monooxygenase.

Whole-cell assays of methane and trichloroethylene (TCE) consumption have been performed on Methylosinus trichosporium OB3b expressing particulate methane monooxygenase (pMMO). From these assays it is apparent that varying the growth concentration of copper causes a change in the kinetics of methane and TCE degradation. For M. trichosporium OB3b, increasing the copper growth concentration from 2.5 to 20 muM caused the maximal degradation rate of methane (V(max)) to decrease from 300 to 82 nmol of methane/min/mg of protein. The methane concentration at half the maximal degradation rate (K(s)) a

Radial oxygen loss, rhizospheric methane oxidation and methane emissions from rice roots.

Methane (CH4) produced in flooded paddy soils is emitted by passive diffusion, by ebullition and by diffusion through the rice (Oryza sativa) plant. Counter-diffusing oxygen (O2) via shoot to root and rhizosphere will inhibit methanogenesis and support aerobic rhizospheric methanotrophs and heterotrophs. In this study, we analyzed the anatomical, morphological and physiological traits of roots of several rice varieties and used modeling approaches to assess the potential effects on methane emissions. Modeling showed that traits that enhance internal O2 diffusion: increased cortical porosity, a

Inhibitory effects of sulfur compounds on methane oxidation by a methane-oxidizing consortium.

Kinetic and enzymatic inhibition experiments were performed to investigate the effects of methanethiol (MT) and hydrogen sulfide (H2S) on methane oxidation by a methane-oxidizing consortium. In the coexistence of MT and H2S, the oxidation of methane was delayed until MT and H2S were completely degraded. MT and H2S could be degraded, both with and without methane. The kinetic analysis revealed that the methane-oxidizing consortium showed a maximum methane oxidation rate (Vmax) of 3.7 mmol g-dry cell weight (DCW)(-1) h(-1) and a saturation constant (Km) of 184.1 μM. MT and H2S show competitive i

Homicide by methane gas.

Methane is a suffocating gas, and "methane deaths" are largely the result of suffocation by gas-air displacement after accidental or deliberate exposure. Neither methane gas nor other suffocating gases are a common means of homicide, with the potential exception of the use of gas in chemical weapons or gas chambers. Here, we report the case of a 53-year-old woman who was killed by her husband with methane gas. The man had given his wife a dose of Lorazepam before setting up a hose that conveyed methane from the kitchen into the apartment's bedroom. The man subsequently faked his own suicide, w

Influence of headspace flushing on methane production in Biochemical Methane Potential (BMP) tests.

The influence of headspace flushing on the specific methane (CH4) production of blank samples with just inoculum in Biochemical Methane Potential (BMP) tests was studied. The three most common ways were applied: flushing with nitrogen (N2) gas, flushing with a mixture of N2 and CO2 (80/20 v/v), and no flushing. The results revealed that removing the oxygen is crucial to avoid aerobic respiration, which caused both hindered activity of methanogens and loss of methane potential. Furthermore it was demonstrated that 20% of CO2 in the flush gas increased significantly the methane production by ove

Bacteriol. 91:1924-1931. 1966.-A new coccus-shaped bacterium capable of aerobic growth at the expense of methane or methanol in a mineral salts medium is described. The organism did not grow at the expense of any of the conventional substrates or homologous hydrocarbons tested. It is gram-negative, nonmotile, and thermotolerant. It grows well at 50 C, optimally at 37 C, but does not grow at 55 C. The cells a

Knowledge Graph for Methane Selective Conversion: Revisiting and Predicting Product Selectivity and Methane Conversion.

Selective conversion of methane to carbon-based compounds is promising but currently limited by issues related to scale. Unraveling the interconnected relationships between products selectivity and methane conversion plays a pivotal role in understanding of this complex process. In this work, a knowledge graph (KG) is constructed for methane conversion with the help of a robust large language model basing on the literature reported methane conversion over different catalysts under various conditions. This KG, structured around 11 entity types and 32 relationship types, allows to effectively an

Water Microdroplets-Initiated Methane Oxidation.

The special redox reactivity of water microdroplets causes "mild ignition" of methane gas to form methane oxygenates. The C(sp3)-H bond of methane can be activated by the hydroxyl radical (OH·) or the hydrogen radical (H·) across the air-water interface (AWI) of microdroplets to generate the methyl radical (CH3·). Once CH3· is formed, it undergoes free-radical reactions with O2 in the air, excessive OH· and H· across the AWI, and H2O2 present at the AWI and generated CH3· itself to produce methanol and other species. Production of the methanol and other oxygenates was confirmed by gas chromato

Methanogens, methane and gastrointestinal motility.

Anaerobic fermentation of the undigested polysaccharide fraction of carbohydrates produces hydrogen in the intestine which is the substrate for methane production by intestinal methanogens. Hydrogen and methane are excreted in the flatus and in breath giving the opportunity to indirectly measure their production using breath testing. Although methane is detected in 30%-50% of the healthy adult population worldwide, its production has been epidemiologically and clinically associated with constipation related diseases, like constipation predominant irritable bowel syndrome and chronic constipati

Epilimnetic oligotrophication increases contribution of oxic methane production to atmospheric methane flux from stratified lakes.

Although considerable attention has been paid to the effects of eutrophication on aquatic methane (CH4) emissions to the atmosphere, the ecosystem-level effects of oligotrophication/re-oligotrophication on aquatic CH4 production and subsequent ecological responses remain to be elucidated. It has been hypothesized that dissolved inorganic phosphorus (DIP)-deficient conditions drive the ecosystem to utilize poorly bioavailable organic phosphorus for biomass formation, thereby generating CH4 as a by-product. To test this hypothesis, a mass balance approach was used to estimate in situ oxic methan

Fe(II)Cl2 amendment suppresses pond methane emissions by stimulating iron-dependent anaerobic oxidation of methane.

Aquatic ecosystems are large contributors to global methane (CH4) emissions. Eutrophication significantly enhances CH4-production as it stimulates methanogenesis. Mitigation measures aimed at reducing eutrophication, such as the addition of metal salts to immobilize phosphate (PO43-), are now common practice. However, the effects of such remedies on methanogenic and methanotrophic communities-and therefore on CH4-cycling-remain largely unexplored. Here, we demonstrate that Fe(II)Cl2 addition, used as PO43- binder, differentially affected microbial CH4 cycling-processes in field experiments and

Development of an imidazole salt catalytic system for the preparation of bis(indolyl)methanes and bis(naphthyl)methane.

Imidazolium salts are shown to catalyze the rapid room temperature reaction of indoles or naphthol with aldehydes to provide bis(indolyl)methanes or bis(naphthol)methane in excellent yields and the reaction proceeds optimally in dichloromethane with no base additives. The reaction exhibits a broad substrate tolerance and occurs through nucleophilic activation of the indoles and naphthols through a cation-π interaction.

Evolution of methane permeability and fracture structure of coal: implications for methane extraction in deep coal seams.

A traditional view is that gas in the intact coal is easier to be extracted compared with tectonic coal after stress relief. With the increase in buried depth, however, the structure of coal and the permeability evolution law may change. Thus, the variation of permeability characteristics and the efficiency of gas extraction method at a deeper coal seam need further study. In this paper, the intact coal and tectonic coal in the Qinan coal mine are taken as the research objects. The permeabilities of both coal samples were tested under hydrostatic pressure, and the effective stress was loaded t
