Effects of CO2 and H2 limitations on Methanococcus maripaludis.

Hydrogenotrophic methanogens play a key role in anaerobic ecosystems by catalyzing the bioconversion of hydrogen (H2) and carbon dioxide (CO2) into methane (CH4) and water. This metabolic process is the basis of biological power-to-methane (PtM) technology, a promising solution for the long-term storage of surplus renewable energy as CH4. Its successful application can be improved through a deeper understanding of methanogen physiology, particularly the metabolic response to intermittent substrate supply of H2 or CO2. In batch cultures of PtM-relevant strains, mesophilic Methanococcus maripalu

Toward complete carbon utilization: Improved methane yield from formate and hydrogen co-feeding through constitutive formate dehydrogenase-gene expression in Methanothermobacter thermautotrophicus ΔH.

Formate is emerging as a relevant intermediate in carbon capture and utilization technologies. However, its low energy density limits its value as an energy carrier. Some hydrogenotrophic methanogens can reduce formate to the established energy carrier methane. The stoichiometric limitation of formate disproportionation is that 75 % of the carbon is released as carbon dioxide, and achieving a complete carbon utilization requires co-feeding hydrogen. However, hydrogen-dependent genetic regulation of formate metabolism inhibits simultaneous formate and hydrogen utilization in hydrogenotrophic me

Steady state characteristics of acclimated hydrogenotrophic methanogens on inorganic substrate in continuous chemostat reactors.

A Monod model has been used to describe the steady state characteristics of the acclimated mesophilic hydrogenotrophic methanogens in experimental chemostat reactors. The bacteria were fed with mineral salts and specific trace metals and a H(2)/CO(2) supply was used as a single limited substrate. Under steady state conditions, the growth yield (Y(CH4)) reached 11.66 g cells per mmol of H(2)/CO(2) consumed. The daily cells generation average was 5.67 x 10(11), 5.25 x 10(11), 4.2 x 10(11) and 2.1 x 10(11) cells/l-culture for the dilutions 0.071/d, 0.083/d, 0.1/d and 0.125/d, respectively. The ma

Developing a robust biomethanation process by improving oxygen tolerance of Methanothermobacter wolfeii strain BSEL.

Application of biomethanation for conversion of CO2 and H2 to renewable natural gas (RNG) is constrained by the strong O2 sensitivity of methanogenic archaea. In biogas facilities, O2 enters through leaks, sampling ports, and intentional micro-aeration for hydrogen sulfide control, creating intermittent oxidative conditions that can destabilize a downstream biomethanation process for biogas upgrading to RNG. An O2-tolerant biocatalyst reduces reliance on strict anaerobic techniques for handling the biocatalyst as well as purification of the gases used for operating biomethanation units. For re

Investigating the role of gut microbiota in hemorrhagic stroke: Evidence from causal analysis.

BACKGROUND: Hemorrhagic stroke is potentially fatal and debilitating. Previous studies have indicated a potential correlation between gut microbiota and hemorrhagic stroke. METHODS: We conducted a two-sample Mendelian randomization (MR) study to assess the potential causal effects of gut microbiota on hemorrhagic stroke, including nontraumatic intracranial hemorrhage (ntICH), intracerebral hemorrhage (ICH), and subarachnoid hemorrhage (SAH). The inverse variance weighted (IVW) method was employed as the primary MR evaluation approach. Complementary methods of MR‒Egger, simple mode, weighted m

Isocyanides as a Chemical Tool for Identifying [Fe]-Hydrogenase-Dependent Methanogenesis.

In the hydrogenotrophic methanogens cultivated under nickel-sufficient standard medium (~5 µM Ni2+), two cytosolic [NiFe]-hydrogenases, F420-reducing hydrogenase (Frh) and heterodisulfide reductase (Hdr)-associating hydrogenase (Mvh), mainly provide electrons to the methanogenic pathway. In Methanothermobacter marburgensis under strictly nickel-limited conditions (< 50 nM Ni2+), which are encountered in certain natural environments, Frh and Mvh are strongly downregulated. Under these conditions, a coupled reaction with [Fe]-hydrogenase (Hmd) and F420-dependent methylene-tetrahydromethanopterin

Core cooperative metabolism in low-complexity CO2-fixing anaerobic microbiota.

Biological conversion of carbon dioxide into methane has a crucial role in global carbon cycling and is operated by a specialised set of anaerobic archaea. Although it is known that this conversion is strictly linked with cooperative bacterial activity, such as through syntrophic acetate oxidation, there is also a limited understanding on how this cooperation is regulated and metabolically realised. In this work, we investigate the activity in a microbial community evolved to efficiently convert carbon dioxide into methane and predominantly populated by Methanothermobacter wolfeii. Through mul

Analyzing the causal relationship between gut microbiota, metabolites, and constipation: A Mendelian randomization study.

A growing body of evidence suggests that the gut microbiota (GM) and its metabolites may play a role in the development of constipation; however, further investigation is necessary to ascertain the precise relationship. Genetic instruments were derived from large-scale genome-wide association studies (GWAS) of European populations. Pooled summary statistics for GM (n = 18,340) and metabolites (n = 7824) were derived from public GWAS, while constipation data (n = 47,696) were obtained from the FinnGen database R10. Mendelian randomization (MR) analyses were performed using data from GWAS to det

Nutritional and biochemical characterization of Methanospirillum hungatii.

To ascertain its physiological similarity to other methanogenic bacteria, Methanospirillum hungatii, the type species of the genus, was characterized nutritionally and biochemically. Good growth occurred in a medium consisting of mineral salts, cysteine sulfide reducing buffer, and an H2-CO2 (80:20) atmosphere. Addition of amino acids and B vitamins stimulated growth. Cell-free extracts contained methylcobalamin-coenzyme M methyltransferase, methylreductase, and formate hydrogenlyase. Cells contained coenzyme M and coenzyme F420. Coenzyme F420 was required for formate hydrogenlyase activity. C

Impact of trace metal supplementation on anaerobic biological methanation under hydrogen and carbon dioxide starvation.

Biomethanation is a crucial process occurring in natural and engineered systems which can reduce carbon dioxide to methane impacting the global carbon cycle. However, little is known about the effect of on-and-off gaseous provision and micronutrients on bioconversion. Here, anaerobic microbiomes underwent intermittent feeding with incremental starvations and selective metal supplementation to assess the impact of hydrogen and carbon dioxide availability on microbial physiology. Resilience was tested under differential cultivations in basal medium supplemented with either nickel or cobalt. Nick

Quantitative analysis of amino acid excretion by Methanothermobacter marburgensis under N2-fixing conditions.

Methanogenic archaea (methanogens) possess fascinating metabolic characteristics, such as the ability to fix molecular nitrogen (N2). Methanogens are of biotechnological importance due to the ability to produce methane (CH4) from molecular hydrogen (H2) and carbon dioxide (CO2) and to excrete proteinogenic amino acids. This study focuses on analyzing the link between biological methanogenesis and amino acid excretion under N2-fixing conditions. Among five hydrogenotrophic, autotrophic methanogens, Methanothermobacter marburgensis was prioritized and further cultivated in closed batch cultivati

Exploring genetic adaptation and microbial dynamics in engineered anaerobic ecosystems via strain-level metagenomics.

Genetic heterogeneity exists within all microbial populations, with sympatric cells of the same species often exhibiting single-nucleotide variations that influence phenotypic traits, including metabolic efficiency. However, the evolutionary dynamics of these strain-level differences in response to environmental stress remain poorly understood. Here, we present a first-of-its-kind study tracking the adaptive evolution of an anaerobic, carbon-fixing microbiota under a controlled engineered ecosystem focused on carbon dioxide bioconversion into methane. Leveraging strain-resolved metagenomics wi

Markerless mutagenesis enables isoleucine biosynthesis solely from threonine in Methanothermobacter marburgensis.

UNLABELLED: The archaeal model microorganism Methanothermobacter marburgensis has been studied for methane production for decades. However, genetic modifications are required to harness M. marburgensis for the generation of novel archaeal cell factories for industrial-scale production of commodity and high-value chemicals. Only the development of tools for genetic engineering opens up this possibility. Here, we present the establishment of the first markerless mutagenesis system for genetic modification of M. marburgensis. This system allows the recycling of positive selection markers and enab

Phenyl Acid Induced Inhibition of Methanogenesis in CO2 Reducing Organisms.

During anaerobic digestion (AD) of lignocellulose- and protein-rich substrates known to contain a high load of aromatic compounds, various undesired intermediates can arise, which can accumulate and cause serious disturbances during the cascade-like AD process. The phenyl acids phenyl-acetic-(PAA), phenyl-propionic-(PPA), and phenyl-butyric acid (PBA) are such intermediates suspected to negatively affect the microbial community, resulting in a decreased biogas yield. In the present study, the impact of PAA, PPA, and PBA on the metabolism of CO2 reducing methanogens was investigated. The mesoph

Crystallization and preliminary X-ray characterization of two thermostable DNA nucleases.

Temperature-tolerant organisms are an important source to enhance the stability of enzymes used in biotechnological processes. The DNA-cleaving enzyme exonuclease III from Escherichia coli is used in several applications in gene technology. A thermostable variant could expand the applicability of the enzyme in these methods. Two homologous nucleases from Archaeoglobus fulgidus (ExoAf) and Methanothermobacter thermoautrophicus (ExoMt) were studied for this purpose. Both enzymes were crystallized in different space groups using (poly)ethylene glycols, 2,4-methyl pentandiol, dioxane, ethanol or 2

Assessing the methanogenic activity of microbial communities enriched from a depleted reservoir.

Using a depleted gas reservoir as a natural reactor is a novel approach for microbial methanation of hydrogen (H2) and carbon dioxide (CO2) into methane (CH4). This approach, known as underground biomethanation reactor (UMR), could enable the simultaneous valorization of geologically sequestered CO2 and the excess renewable energy, stored in the form of H2 in the same formation as the CO2. In this study, we explore the possibility to trigger biomethanation from formation water sample by testing various carbon sources (CO2, trypticase peptone, glucose, and acetate) in batch test with a defined

Microbiome and resistome of the European bison (Bison bonasus).

After facing extinction in the early 20th century, populations of the two remnant genetic lines of European bison are now under continuous health monitoring. Faecal samples were taken from five Polish and one German herd of European bison over the course of several years. Through metagenomic sequencing, the bacterial and archaeal microbiome as well as the resistome of these samples could be characterized. Significant differences were mainly found between the bacterial microbiome of samples taken from droppings as opposed to rectal samples. Apart from this, the microbiome and resistome had low

Physiological and genomic characterization of Methanothermobacter thermautotrophicus strain Z-73A, an isolate from deep underground.

Underground biomethanation represents a promising solution for green energy storage. An isolate of Methanothermobacter thermautotrophicus was obtained in previous study from an underground gas reservoir where it played a key role in a successful in-situ biomethanation experiment. In this study, the physiological and environmental tolerances of the strain are characterised. Optimal methane evolution rates were observed at 55-65 °C and pH 6.5-7.0, methane production was significantly increased by agitation (up to 2.29 mmol CH4·h-1·L-1). The strain showed higher tolerance to NH4Cl (up to 0.6 M) t

Electron flow in hydrogenotrophic methanogens under nickel limitation.

Methanogenic archaea are the main producers of the potent greenhouse gas methane1,2. In the methanogenic pathway from CO2 and H2 studied under laboratory conditions, low-potential electrons for CO2 reduction are generated by a flavin-based electron-bifurcation reaction catalysed by heterodisulfide reductase (Hdr) complexed with the associated [NiFe]-hydrogenase (Mvh)3-5. F420-reducing [NiFe]-hydrogenase (Frh) provides electrons to the methanogenic pathway through the electron carrier F420 (ref. 6). Here we report that under strictly nickel-limited conditions, in which the nickel concentration

Diversity of electron-bifurcating CO2-fixing supercomplexes in methanogens.

In the hydrogenotrophic methanogenic pathway, formylmethanofuran dehydrogenase (Fmd) reduces and fixes CO2, driven by low-potential electrons provided by electron-bifurcating heterodisulfide reductase (Hdr) complexed with electron-donating proteins such as Mvh hydrogenase. Here, we report the structure of a C2-symmetric (Mvh-Hdr)2-Fmd4 supercomplex from a Class I methanogen, Methanothermobacter marburgensis, which is architecturally different from the previously reported ring-shaped D3-symmetric supercomplex of a methanogen belonging to phylogenetically distinct Class II methanogens.
