Cofactor F420 tail length diversity in specific representatives of the class Methanosarcinia during acetoclastic and methylotrophic methanogenesis: Deciphering structural variations.

OBJECTIVES: Methanogens possess a variety of coenzymes and cofactors that are involved in redox reactions that occur during methanogenesis. One essential cofactor is F420, a deazaflavin derivate with an oligoglutamate side chain which acts as a low redox potential electron carrier and participates in electron transport coupled with energy conservation. This work aimed at characterizing cofactor F420 analogues in eight distinct acetoclastic and methylotrophic pure cultures of the class Methanosarcinia. METHODS: Pure cultures of Methanothrix soehngenii, Methanothrix thermoacetophila, Methanomet

Analysis of Mechanisms for Electron Uptake by Methanothrix harundinacea 6Ac During Direct Interspecies Electron Transfer.

Direct interspecies electron transfer (DIET) is a syntrophic metabolism wherein free electrons are directly transferred between microorganisms without the mediation of intermediates such as molecular hydrogen or formate. Previous research has demonstrated that Methanothrix harundinacea 6Ac is capable of reducing carbon dioxide through DIET. However, the mechanisms underlying electron uptake in M. harundinacea 6Ac during DIET remain poorly understood. This study aims to elucidate the electron and proton flux in M. harundinacea 6Ac during DIET and to propose a model for electron uptake in this o

Methanolobus use unspecific methyltransferases to produce methane from dimethylsulphide in Baltic Sea sediments.

BACKGROUND: In anoxic coastal and marine sediments, degradation of methylated compounds is the major route to the production of methane, a powerful greenhouse gas. Dimethylsulphide (DMS) is the most abundant biogenic organic sulphur compound in the environment and an abundant methylated compound leading to methane production in anoxic sediments. However, understanding of the microbial diversity driving DMS-dependent methanogenesis is limited, and the metabolic pathways underlying this process in the environment remain unexplored. To address this, we used anoxic incubations, amplicon sequencing

Production of methylmercury by methanogens in mercury contaminated estuarine sediments.

Anaerobic bacteria are known to produce neurotoxic methylmercury [MeHg] when elemental mercury [Hg(0)] is provided as the sole mercury source. In this study, we examined the formation of MeHg in anaerobic incubations of sediment collected from the San Jacinto River estuary (Texas, USA) amended with aqueous Hg(0) to investigate the microbial communities involved in the conversion of Hg(0) to MeHg. The results show that the addition of the methanogen inhibitor 2-bromoethanesulfonate (BES) significantly decreased MeHg production. The mercury methylation gene, hgcA, was detected in these sediments

Biostimulation of indigenous communities for the successful dechlorination of tetrachloroethene (perchloroethylene)-contaminated groundwater.

Chlorinated ethenes are of environmental concern with most reports of successful microbial-mediated remediation being associated with major dechlorinating groups such as Dehalococcoides (Dhc) species. However, limited information is available on the community dynamics and dechlorinating activities of indigenous non-Dhc groups. Here, we present evidence of dechlorination of tetrachloroethene (perchloroethylene, PCE) in groundwater samples by indigenous microbial communities. 100 % PCE conversion to ethene was observed in acetate-stimulated 24 week-microcosms (controls; 15 %). Microbial communit

Effect of soil aggregate size on methanogenesis and archaeal community structure in anoxic rice field soil.

In anoxically incubated slurries of Italian rice field soil, CH(4) production is initiated after a lag phase during which ferric iron and sulfate are reduced. The production of CH(4) was affected by the size of soil aggregates used for the preparation of the soil slurry. Rates of CH(4) production were lowest with small aggregates (<50 and 50-100 µm), were highest with aggregates of 200-2000 µm size and were intermediate with aggregates of 2000-15000 µm size. The different amounts of CH(4) accumulated were positively correlated to the concentrations of acetate, propionate and caproate that tran

Potential of direct interspecies electron transfer in synergetic enhancement of methanogenesis and sulfate removal in an up-flow anaerobic sludge blanket reactor with magnetite.

Anaerobic digestion (AD) has been widely applied in the treatment of industrial wastewater containing oxidized sulfur compounds. However, the production of hydrogen sulfide usually limits the syntrophic metabolism proceeded by interspecies hydrogen transfer (IHT), due to its corrosive and toxic properties. The current study was in an attempt to establish direct interspecies electron transfer (DIET) to resist the toxic inhibition from hydrogen sulfide and keep syntrophic metabolism stable. The results showed that, in the presence of magnetite, the methane production was improved about 3-10 fold

Performance of a haloalkaliphilic bioreactor and bacterial community shifts under different COD/SO₄²⁻ ratios and hydraulic retention times.

Sulfur dioxide from flue gas was converted into sulfate after the absorption of alkaline solutions. Haloalkaliphilic microorganisms have been used in reducing sulfate to decrease expenses and avoid sulfide inhibition. The effects of different COD/SO4(2-) ratios and hydraulic retention times (HRTs) on the sulfate removal efficiency and bacterial community were investigated in model experiments. Ethanol showed better performance as an electron donor than lactate. The optimum COD/SO4(2-) ratio and HRT were 4.0 and 18 h, respectively, with respective sulfate removal efficiency and rate of 97.8 ± 1

Elevated water levels drive greenhouse gas mitigation in the riparian zone profile.

Wetlands are critical for climate regulation, with their hyporheic zone serving as sensitive interfaces for groundwater-soil-atmosphere exchange. These zones are active hotspots for carbon-nitrogen cycling and greenhouse gas (GHG) emissions (CO2, CH4, N2O), yet the impact of water level fluctuations on these emissions and their microbial drivers in freshwater wetlands remains poorly understood. This study investigated the spatiotemporal dynamics of GHG emissions and carbon-nitrogen coupling processes along riparian soil profiles of Baiyangdian Lake during water level fluctuations. Methanolobus mangrovi sp. nov. and Methanolobus sediminis sp. nov, two novel methylotrophic methanogens isolated from mangrove sediments in China.

Two methylotrophic methanogens, designated strains FTZ2T and FTZ6T, were isolated from mangrove sediment sampled in Futian Mangrove Nature Reserve in Shenzhen, PR China. Cells of strains FTZ2T and FTZ6T were cocci, with diameters of 0.6-1.0 µm and 0.6-0.9 µm, respectively. Both strains grew on methanol, methylamine, dimethylamine and trimethylamine, but not on acetate, formate, H2/CO2, choline, betaine or dimethyl sulphide. Strain FTZ2T grew at 10-37 °C (optimally at 33 °C), pH 5.5-8.0 (optimally at pH 7.0) and 0-1.03 M NaCl (optimally at 0.17 M NaCl). In contrast, strain FTZ6T grew at 15-42 °

Insights into the effects of anthropogenic activities on oil reservoir microbiome and metabolic potential.

Microbial communities have long been observed in oil reservoirs, where the subsurface conditions are major drivers shaping their structure and functions. Furthermore, anthropogenic activities such as water flooding during oil production can affect microbial activities and community compositions in oil reservoirs through the injection of recycled produced water, often associated with biocides. However, it is still unclear to what extent the introduced chemicals and microbes influence the metabolic potential of the subsurface microbiome. Here we investigated an onshore oilfield in Germany (Field

Resupply, diffusion, and bioavailability of Hg in paddy soil-water environment with flood-drain-reflood and straw amendment.

Mercury (Hg) poses a significant risk in paddy fields, particularly when it is converted to methylmercury (MeHg) and accumulates in rice. However, the bioavailability and resupply kinetics of Hg in the paddy soil-water environment are not well understood. In this study, the diffusive gradients in thin films (DGT) and the 'DGT-induced fluxes in sediments' model (DIFS) were first adopted to investigate the Hg resupply kinetics, diffusion fluxes and bioavailability in a paddy environment subjected to flood-drain-reflood treatment and straw amendment. Our results show that although the straw amend

Methanogenic and Sulfate-Reducing Activities in a Hypersaline Microbial Mat and Associated Microbial Diversity.

Methanogenesis and sulfate reduction are important microbial processes in hypersaline environments. However, key aspects determining substrate competition between these microbial processes have not been well documented. We evaluated competitive and non-competitive substrates for stimulation of both processes through microcosm experiments of hypersaline microbial mat samples from Guerrero Negro, Baja California Sur, Mexico, and we assessed the effect of these substrates on the microbial community composition. Methylotrophic methanogenesis evidenced by sequences belonging to methanogens of the f

The influence of biochar position in a leach bed system anaerobically digesting chicken litter.

As a consequence of the rapidly growing poultry industry, chicken litter is becoming an abundant and problematic waste. Anaerobic digestion of chicken litter can mitigate environmental issues while producing valuable by-products. Recent studies have shown that leach bed reactor (LBR) systems are suitable for processing chicken litter and that anaerobic digestion can be enhanced using biochar. This study investigates the influence of biochar position within an LBR system on anaerobic digestion of chicken litter. Compared to a system without biochar, application of biochar in both the LBR (mixed

Methanomethylovorans uponensis sp. nov., a methylotrophic methanogen isolated from wetland sediment.

A novel mesophilic, methylotrophic, methanogenic archaeon, designated strain EK1(T), was enriched and isolated from wetland sediment. Phylogenetic analysis showed that strain EK1(T) was affiliated with the genus Methanomethylovorans within the family Methanosarcinaceae, and shared the highest 16S rRNA and methyl-coenzyme M reductase alpha-subunit gene sequence similarity with the type strain of Methanomethylovorans hollandica (98.8 and 92.6 %, respectively). The cells of strain EK1(T) were observed to be Gram-negative, non-motile and irregular cocci that did not lyse in 0.1 % (w/v) sodium dode

Methane emission from feather moss stands.

Data from remote sensing and Eddy towers indicate that forests are not always net sinks for atmospheric CH4 . However, studies describing specific sources within forests and functional analysis of microorganisms on sites with CH4 turnover are scarce. Feather moss stands were considered to be net sinks for carbon dioxide, but received little attention to their role in CH4 cycling. Therefore, we investigated methanogenic rates and pathways together with the methanogenic microbial community composition in feather moss stands from temperate and boreal forests. Potential rates of CH4 emission from

Time-series metatranscriptomics reveals differential salinity effects on the methanogenic food web in paddy soil.

Saltwater intrusion and sea level rise (SWISLR) threaten coastal agroecosystems, yet their impact on the methanogenic food web in rice paddies remains virtually unknown. Using "double-RNA" metatranscriptomics (rRNA, mRNA), we investigated salinity effects on the methanogenic community in straw-amended slurries following different preincubation periods. Our results demonstrate that salt stress significantly inhibits methane (CH4) production, with the degree of inhibition varying by the community's successional stage. Salinity fundamentally shifted the methanogenic food web toward Clostridiaceae

Synergy and competition during the anaerobic degradation of N-acetylglucosamine in a methane-emitting, subarctic, pH-neutral fen.

Peatlands are invaluable but threatened ecosystems that store huge amounts of organic carbon globally and emit the greenhouse gasses carbon dioxide (CO2) and methane (CH4). Trophic interactions of microbial groups essential for methanogenesis are poorly understood in such systems, despite their importance. Thus, the present study aimed at unraveling trophic interactions between fermenters and methanogens in a nitrogen-limited, subarctic, pH-neutral fen. In situ CH4 emission measurements indicated that the fen is a source of CH4, and that CH4 emissions were higher in plots supplemented with amm
