Mercury methylation by the methanogen Methanospirillum hungatei.

Methylmercury (MeHg), a neurotoxic substance that accumulates in aquatic food chains and poses a risk to human health, is synthesized by anaerobic microorganisms in the environment. To date, mercury (Hg) methylation has been attributed to sulfate- and iron-reducing bacteria (SRB and IRB, respectively). Here we report that a methanogen, Methanospirillum hungatei JF-1, methylated Hg in a sulfide-free medium at comparable rates, but with higher yields, than those observed for some SRB and IRB. Phylogenetic analyses showed that the concatenated orthologs of the Hg methylation proteins HgcA and Hgc

Nutritional and biochemical characterization of Methanospirillum hungatii.

To ascertain its physiological similarity to other methanogenic bacteria, Methanospirillum hungatii, the type species of the genus, was characterized nutritionally and biochemically. Good growth occurred in a medium consisting of mineral salts, cysteine sulfide reducing buffer, and an H2-CO2 (80:20) atmosphere. Addition of amino acids and B vitamins stimulated growth. Cell-free extracts contained methylcobalamin-coenzyme M methyltransferase, methylreductase, and formate hydrogenlyase. Cells contained coenzyme M and coenzyme F420. Coenzyme F420 was required for formate hydrogenlyase activity. C

Description and genome analysis of a novel archaeon isolated from a syntrophic pyrite-forming enrichment culture and reclassification of Methanospirillum hungatei strains GP1 and SK as Methanospirillum purgamenti sp. nov.

The archaeal isolate J.3.6.1-F.2.7.3T was obtained from an anaerobic enrichment culture, where it may play an important role in methane production during pyrite formation. The new isolate formed a species-level clade with Methanospirillum hungatei strains GP1 and SK, which is separate from the type strain JF-1T. Cultivation-independent surveys indicate the occurrence of this phylogenetic group in sediments and anaerobic digesters. The abundance of this clade appears to be negatively affected by high nitrogen loads, indicating a sensitivity to certain nitrogen compounds that is not known in M.

Isolation and characterization of a motile hydrogenotrophic methanogen from rice paddy field soil in Japan.

A hydrogenotrophic motile methanogen was isolated from flooded Japanese paddy field soil. Anaerobic incubation of the paddy soil on H(2)-CO(2) at 20 degrees C led to the enrichment of symmetrically curved motile autofluorescent rods. The methanogenic strain TM20-1 isolated from the culture was halotolerant and utilized H(2)-CO(2), 2-propanol-CO(2), or formate as a sole methanogenic substrate. Based on the 16S rRNA gene sequence similarity (94.8%) with Methanospirillum hungateii, and on the physiological and phenotypic characteristics, TM20-1 was suggested to be a newly identified species belon

The response of selected members of the archaea to the gram stain.

Archaea possess a broader range of cell envelope structural formats than eubacteria and their cell walls do not contain peptidoglycan. Some archaea have only a single S-layer as their cell wall (e.g. Methanococcus jannaschii and Sulfolobus acidocaldarius), whereas others have multiple layers (e.g. Methanospirillum hungatei). Sometimes there can also be a high proportion of tetraether lipids in membranes to make the envelope more resilient to environmental stress (e.g. Methanococcus jannaschii and Sulfolobus acidocaldarius grown at 70 degrees C). Since the Gram reaction depends on both the stru

The origin of methyl group in methanogen-mediated mercury methylation: From the Wolfe cycle.

Methylmercury (MeHg) is a bioaccumulating neurotoxin mainly produced by anaerobic microorganisms, with methanogen being one of the important methylators. A critical aspect for understanding the mechanism for microbial mercury (Hg) methylation is the origin of the methyl group. However, the origin of methyl group in methanogen-mediated Hg methylation remains unclear. This study aims to identify the source of methyl group for MeHg synthesis in methanogens. Our study revealed that Hg methylation in Methanospirillum hungatei JF-1 is closely related to methanogenesis process, according to the resul

Phenyl Acid Induced Inhibition of Methanogenesis in CO2 Reducing Organisms.

During anaerobic digestion (AD) of lignocellulose- and protein-rich substrates known to contain a high load of aromatic compounds, various undesired intermediates can arise, which can accumulate and cause serious disturbances during the cascade-like AD process. The phenyl acids phenyl-acetic-(PAA), phenyl-propionic-(PPA), and phenyl-butyric acid (PBA) are such intermediates suspected to negatively affect the microbial community, resulting in a decreased biogas yield. In the present study, the impact of PAA, PPA, and PBA on the metabolism of CO2 reducing methanogens was investigated. The mesoph

Pyrite formation from FeS and H2S is mediated through microbial redox activity.

The exergonic reaction of FeS with H2S to form FeS2 (pyrite) and H2 was postulated to have operated as an early form of energy metabolism on primordial Earth. Since the Archean, sedimentary pyrite formation has played a major role in the global iron and sulfur cycles, with direct impact on the redox chemistry of the atmosphere. However, the mechanism of sedimentary pyrite formation is still being debated. We present microbial enrichment cultures which grew with FeS, H2S, and CO2 as their sole substrates to produce FeS2 and CH4 Cultures grew over periods of 3 to 8 mo to cell densities of up to

Evaluation of process performance and retained sludge properties of a psychrophilic UASB reactor for treatment of iso-plophyl alcohol (2-propanol)-containing wastewater.

In this study, a continuous feeding experiment was conducted with synthetic iso-plophyl alcohol (2-propanol)-containing wastewater using a lab-scale psychrophilic UASB reactor to evaluate process performance and retained sludge properties. For smooth acclimation, methanogenic granular sludge was seeded and a proportion of 2-propanol in the synthetic wastewater containing sucrose and volatile fatty acids was increased stepwise from 0% to 30%, 60% and then 90% of COD (chemical oxygen demand). As a result, after a 4-week period for acclimation to 2-propanol degradation, a COD removal rate of 95%

Growth of methanogenic bacteria in pure culture with 2-propanol and other alcohols as hydrogen donors.

Two types of mesophilic, methanogenic bacteria were isolated in pure culture from anaerobic freshwater and marine mud with 2-propanol as the hydrogen donor. The freshwater strain (SK) was a Methanospirillum species, the marine, salt-requiring strain (CV), which had irregular coccoid cells, resembled Methanogenium sp. Stoichiometric measurements revealed formation of 1 mol of CH(4) by CO(2) reduction, with 4 mol of 2-propanol being converted to acetone. In addition to 2-propanol, the isolates used 2-butanol, H(2), or formate but not methanol or polyols. Acetate did not serve as an energy substr

Mercury methylation in methanogenic archaea: A protocol for stabilized cultivation and accurate assessment.

Although methanogenic archaea are among the oldest microorganisms capable of mercury methylation, their contribution to methylmercury (MeHg) production has only recently gained attention. Studies with laboratory-cultivated methanogens elucidate the transformation of inorganic mercury (Hg) into MeHg, thereby uncovering underlying microbial methylation mechanisms. However, this field faces challenges such as significant Hg loss and unstable culture systems, which impede accurate assessment of these processes. This study aims to develop a reliable low-Hg-loss cultivation protocol for Hg methylati

Phenotypic properties and microbial diversity of methanogenic granules from a full-scale upflow anaerobic sludge bed reactor treating brewery wastewater.

Methanogenic granules from an anaerobic bioreactor that treated wastewater of a beer brewery consisted of different morphological types of granules. In this study, the microbial compositions of the different granules were analyzed by molecular microbiological techniques: cloning, denaturing gradient gel electrophoresis and fluorescent in situ hybridization (FISH), and scanning and transmission electron microscopy. We propose here that the different types of granules reflect the different stages in the life cycle of granules. Young granules were small, black, and compact and harbored active cel

Changes in dysbiosis and gene expression in the gut of wharf roach (Ligia spp.) fed with expanded polystyrene.

The East Asian region, with its intensive human and fishery activity, suffers from serious marine plastic pollution along its coastlines. Wharf roaches (Ligia spp.) on the coast of western Japan frequently ingest expanded polystyrene (EPS) and excrete it as microplastic in their feces, yet the effects on their gut remain unclear. Under controlled laboratory exposure, we examined both the host gut transcriptome, and the gut microbiome of wharf roaches fed EPS versus a fasted control group. DESeq2 and PyDESeq2 yielded a high-confidence set of 25 differentially expressed genes, among which xenobi

Discovery of a Microbial Carrier with High Adsorption Affinity for Syntrophic Long-Chain Fatty Acid-Degrading Microorganisms.

Long-chain fatty acid (LCFA) degradation primarily involves several species of Syntrophomonas and hydrogenotrophic methanogens, constituting the rate-limiting step in anaerobic digestion. It is crucial to augment their abundance to enhance LCFA degradation. Utilizing microbial carriers presents an effective strategy to maintain the microorganisms on the surface and prevent their washout from the digester. In this study, we aimed to identify a suitable microbial carrier with a superior adsorption capacity for LCFA-degrading microorganisms. We tested various polymers, poly(vinyl alcohol) (PVA),

Combining high pressure homogenization with free nitrous acid pretreatment to improve anaerobic digestion of sewage sludge.

Single pretreatment of sewage sludge, either physical, chemical or biological, has its own drawbacks in term of poor sanitization, energy intensity and high operational and capital cost. To tackle these drawbacks, combined high pressure homogenization (HPH) and free nitrous acid (FNA) pretreatment for sludge solubilization and further biodegradation in anaerobic digestion was investigated. Synergistic effect of combined HPH (40 MPa) and FNA (2.49 mg/L) pretreatment (HPH-FNA) for improving anaerobic digestion was evaluated, and its effect on archaeal and bacterial community structure was analyz

Microbial-mediated CH4 and CO2 emissions and environmental controls in the fluvial sediment across hydrological seasons.

Fluvial sediments are recognized as critical sources of methane (CH4) and carbon dioxide (CO2) emissions. However, the interactions between microbial communities and carbon emissions in these sediments remain poorly understood. This study investigated carbon fluxes at the water-air interface in the mainstream of the Liao River, integrating environmental parameters and microbial communities within sediments to elucidate seasonal differences in carbon cycling mechanisms. The results highlighted the wet season as a hot period for CH4 and CO2 emissions, with fluxes in the lower reach (CH4: 78.6 ±

Metagenomic analysis of a stable trichloroethene-degrading microbial community.

Dehalococcoides bacteria are the only organisms known to completely reduce chlorinated ethenes to the harmless product ethene. However, Dehalococcoides dechlorinate these chemicals more effectively and grow more robustly in mixed microbial communities than in isolation. In this study, the phylogenetic composition and gene content of a functionally stable trichloroethene-degrading microbial community was examined using metagenomic sequencing and analysis. For phylogenetic classification, contiguous sequences (contigs) longer than 2500 bp were grouped into classes according to tetranucleotide fr

Unravelling methylmercury formation in high-altitude Tibetan thermokarst lakes.

Climate warming accelerates permafrost degradation, which not only increases greenhouse gas emission but also the conversion of permafrost-stored mercury (Hg) to the neurotoxic methylmercury (MeHg) - a global pollutant to humans and ecosystems. While several studies have examined Hg transformation processes in high-latitude Arctic permafrost, little is known regarding the formation and distribution of MeHg in high-altitude permafrost, particularly the thermokarst lakes on the Tibetan Plateau (so-called Earth's third pole). We conducted field investigations and show an elevated MeHg concentrati

Involvement of NADH:acceptor oxidoreductase and butyryl coenzyme A dehydrogenase in reversed electron transport during syntrophic butyrate oxidation by Syntrophomonas wolfei.

Methanogenic oxidation of butyrate to acetate requires a tight cooperation between the syntrophically fermenting Syntrophomonas wolfei and the methanogen Methanospirillum hungatei, and a reversed electron transport system in S. wolfei was postulated to shift electrons from butyryl coenzyme A (butyryl-CoA) oxidation to the redox potential of NADH for H(2) generation. The metabolic activity of butyrate-oxidizing S. wolfei cells was measured via production of formazan and acetate from butyrate, with 2,3,5-triphenyltetrazolium chloride as electron acceptor.
