The 3-methylhistidine content of human tissues.

1. The amount of 3-methylhistidine (3-MeH) has been measured in eighty-eight samples of tissue taken post-mortem from five adults. 2. The highest concentration (mumol/g fat-free dry weight) of 3-MeH was in skeletal muscle (3.31 +/- 0.05); intermediate values (2-3) were found in cardiac muscle and those tissues containing smooth muscle; and low values (less than 1) occurred in parenchymal tissues such as liver and kidney. 3. There was little variation between the mean 3-MeH content of striated muscles in different individuals, and no significant difference between the 3-MeH concentrations of st

Urinary 3-methylhistidine and urinary 3-methylhistidine/creatinine ratio in Duchenne-muscular dystrophy in hemizygotes and in gene-carriers.

The informative value of urinary 3-methylhistidine excretion and urinary 3-methylhistidine/creatinine ratio was investigated in DMD hemizygote male children (n = 13) and in gene-carrier mothers. A significant increase of the urinary 3-methylhistidine/creatinine ratio was found in DMD hemizygotes. There was no significant correlation between serum CK and the clinical stages of DMD and the above mentioned laboratory parameters. These parameters were not found suitable in genetic counselling concerning the DMD gene-carrier status.

[The effect of the small intestine on 3-methylhistidine metabolism in the human].

20 male adult Sprague-Dawley rats were fed exclusively parenterally. After achieving metabolic equilibrium they received a duodenoileostomy and subtotal resection leaving only 8-10% of the small gut. On the 1st postoperative day the urinary 3-MH excretion rose to 1.5-1.7 times the preoperative level, but on the 12th-14th postoperative day it fell again and was equal to the preoperative basal level. A control group of 10 rats undergoing a small gut anastomosis without resection yielded similar results. We conclude that the small gut source does not make a significant contribution to 24h-urinary

Muscle protein breakdown in thyrotoxicosis assessed by urinary 3-methylhistidine excretion.

Urinary 3-methylhistidine (3MH) excretion has been studied in 20 thyrotoxic patients before, during and after treatment. 3MH excretion was high on diagnosis and fell significantly, when the patients had been rendered euthyroid, to levels measured in a control group. There was a significant linear correlation between 3MH excretion and free thyroxine index. The calculated muscle protein breakdown and 3MH/creatinine molar ratio also fell significantly after treatment of thyrotoxicosis.

3-Methylhistidine excretion as an index of myofibrillar protein catabolism in neuromuscular disease.

Myofibrillar protein catabolism has been calculated in a variety of neuromuscular diseases from the amount of 3-methylhistidine excreted in the urine. It was found to be significantly raised in Duchenne type muscular dystrophy, motor neurone disease, polymyositis, and thyrotoxic myopathy. In Becker type muscular dystrophy the level was slightly raised. It was normal in scapuloperoneal and limb girdle dystrophy, dystrophia myotonica, extrapyramidal disease, and multiple sclerosis. It was significantly decreased in hypothyroid myopathy.

Eimeria acervulina infection elevates plasma and muscle 3-methylhistidine levels in chickens.

To assess muscle breakdown during avian coccidiosis, the level of the nonmetabolizable amino acid 3-methylhistidine (3MH) was determined in muscle, plasma and excreta from chickens infected with Eimeria acervulina. The changes in 3MH levels during infection were assessed at 1-29 days postinoculation (DPI) in animals given 5 x 10(5) oocysts per bird. The effect of levels of parasitism were evaluated at 8 DPI in birds receiving 5 x 10(3), 5 x 10(4), 5 x 10(5) or 1 x 10(6) oocysts each. The 3MH levels of plasma, muscle, and excreta samples were determined by high-pressure liquid chromatography af

Muscle protein breakdown in uncontrolled diabetes as assessed by urinary 3-methylhistidine excretion.

In an attempt to evaluate muscle protein catabolism in patients with uncontrolled diabetes, urinary excretion of 3-methylhistidine was measured in eight diabetic subjects, during poor control and after achievement of satisfactory control. The results were compared with the excretion values of ten healthy subjects fed a similar amount of meat. In the diabetic patients in poor metabolic control, 3-methylhistidine excretion was significantly increased compared with the healthy subjects, and returned to normal when a satisfactory glycaemic control was achieved. No significant differences were obse

Clinical usefulness of urinary 3-methylhistidine excretion in indicating muscle protein breakdown.

Urinary excretion of the post-translationally modified amino-acid 3-methylhistidine, derived from the contractile proteins actin and myosin, was measured in patients with conditions associated with nitrogen loss. The ratio of 3-methylhistidine:creatinine excretion, a measure of the fractional catabolic rate of myofibrillar protein was increased in severe injury, thyrotoxicosis, neoplastic disease, prednisolone administration, and sometimes Duchenne muscular dystrophy. In myxoedema, osteomalacia, and hypothermia the ratio was decreased; and starvation, elective operations, and rheumatoid arthri

Leucine metabolism in thyrotoxicosis: plasma aminogram and 3-methylhistidine excretion before and after treatment.

Previous studies have suggested increased protein catabolism and altered muscle metabolism in hyperthyroid patients. In this experiment we investigated parameters of protein and leucine metabolism before and after treatment of hyperthyroidism. While confined in a metabolic ward, patients' daily caloric intake was based on the resting energy expenditure and an allowance for 16 hours of light physical activity. We found no significant difference in plasma aminogram and urinary 3-methylhistidine excretion (an index of protein catabolism) before and after treatment. On the other hand, hyperthyroid

[3-Methylhistidine as a parameter for the determination of muscle proteolysis in the post-stress syndrome and in diabetes mellitus].

The renal excretion of 3-methylhistidine was measured in healthy human volunteers under different diets. The excretion of 3-methylhistidine and total nitrogen was increased in 8 surgical patients. These findings are referred to an increased proteolysis mainly in the skeletal muscle. In four insulin-dependent diabetic patients (IDDM) the excretion of 3-methylhistidine into urine was increased only when referred to body weight. The interpretation of the 3-methylhistidine/creatinin ratio is discussed. In diabetic patients the total nitrogen excretion seems to be a better parameter for protein tur

Muscle protein catabolism in diabetes: 3-methylhistidine excretion in the spontaneously diabetic "BB" rat.

The muscle protein lost in uncontrolled diabetes may be due to decreased synthesis, increased catabolism, or to any combination of alteration in these rates that results in net loss. Differing methods of examining these rates in vivo and in vitro have given conflicting results. We assessed the rate of catabolism of proteins containing 3-methylhistidine (3-MH) by measurement of its urinary excretion in spontaneously diabetic "BB" Wistar rats. Prior to overt diabetes, rates of excretion were appropriate to the age of the rats (1.46 +/- 0.15 mumole/day), with 34%-47% as the nonacetylated form. Ac

Isotopic decay of urinary or plasma 3-methylhistidine as a potential biomarker of pathologic skeletal muscle loss.

BACKGROUND: Skeletal muscle loss accompanying aging or cancer is associated with reduced physical function and predicts morbidity and mortality. 3-Methylhistidine (3MH) has been proposed as a biomarker of myofibrillar proteolysis, which may contribute to skeletal muscle loss. METHODS: We hypothesized that the terminal portion of the isotope decay curve following an oral dose of isotopically labeled 3MH can be measured non-invasively from timed spot urine samples. We investigated the feasibility of this approach by determining isotope enrichment in spot urine samples and corresponding plasma s

Urinary 3-methylhistidine excretion in juvenile-onset diabetics: evidence of increased protein catabolism in the absence of ketoacidosis.

Urinary 3-methylhistidine excretion (an indicator of protein catabolism) was measured in ten diabetic patients and in age and weight matched control subjects. The diabetic group, while receiving their usual insulin dose, excreted 42% more 3-methylhistidine than the control group (2.7 versus 1.9 mumole/kg body weight/24 hr). When the insulin dose of the diabetic subject was reduced by 15% or 25%, the concentrations of blood and urinary glucose were significantly increased by the rate of urinary 3-methylhistidine excretion was not increased further. Effect of glucocorticoid administration on the rate of muscle protein breakdown in vivo in rats, as measured by urinary excretion of N tau-methylhistidine.

The role of glucocorticoids in regulating the rate of muscle protein breakdown was evaluated by measuring excretion of N(tau)-methylhistidine during administration of various doses of corticosterone to adrenalectomized rats. Groups of rats received daily subcutaneous injections of 0, 0.2, 0.5, 1.0, 5.0 or 10.0mg of corticosterone/day per 100g body wt. for 7 days, followed by 3 days without hormone treatment, after which they were killed. A group with intact adrenal glands served as an additional control. All animals were pair-fed with the untreated adrenalectomized group. No significant differ

Thyroid status and muscle protein breakdown as assessed by urinary 3-methylhistidine excretion: study in thyrotoxic patients before and after treatment.

Urinary 3-methylhistidine (3MH) excretion was studied in nine thyrotoxic patients before and after treatment. Urinary creatinine (Cr) output was also measured and was low in the thyrotoxic subjects before treatment. Thus, although urinary output of 3MH was not greater than among the control population when expressed per subject, it was significantly elevated when expressed as the ratio of 3MH to Cr; this ratio fell significantly, reaching normal control values after a euthyroid state was obtained. In one patient who became hypothyroid, the 3MH/Cr ratio fell under the control value. There was a

Modification of glucocorticoid-induced changes in myofibrillar protein turnover in rats by protein and energy deficiency as assessed by urinary excretion of Ntau-methylhistidine.

The effects of differing degrees of experimental protein-energy malnutrition on the response of myofibrillar protein turnover rates to administration of corticosteroid has been studied in two experiments on rats. The basal control diet, offered ad lib. in each case, contained 40 g protein/kg, and other groups received diets containing 62 X 5, 95 or 220 g protein/kg at 0 X 67, 1 or 1 X 5 times the level of the control energy intake. Daily administration of 25 or 30 mg corticosterone/kg body-weight after 18 d pre-feeding caused an increase in plasma protein, glucose and insulin concentrations, b

Beta adrenoceptor agonists, clenbuterol, and isoproterenol retard denervation atrophy in rat gastrocnemius muscle: use of 3-methylhistidine as a marker of myofibrillar degeneration.

The effects of beta adrenergic agonists, clenbuterol (2 mg/kg body weight/d) and isoproterenol (12 mg/kg body weight/d), in normal innervated and denervated rat gastrocnemius muscle were investigated. The daily administration of beta adrenergic agonists to normal innervated rats for a short period (7 d) resulted in the hypertrophy of gastrocnemius as confirmed from the measurement of total tissue protein contents. The development of denervation atrophy witnessed a stimulation in the expression of acid and alkaline phosphatases, pointing to an enhanced myofibrillar degeneration. An administrati

Total and net muscle protein breakdown in infection determined by amino acid effluxes.

The present investigation was undertaken to study whether, in human infection of varying severity, peripheral 3-methylhistidine efflux and urinary excretion are associated with net protein degradation and to estimate the protein synthesis rate from the combined effluxes of 3-methylhistidine, tyrosine, and phenylalanine. Quadruplicate femoral arteriovenous differences of 3-methylhistidine, tyrosine, and phenylalanine were multiplied by leg plasma flow in 15 infected patients.
